ALTERATION OF HEPATIC 3′-PHOSPHOADENOSINE 5′_ PHOSPHOSULFATE(PAPS) AND SULFATE IN ICR MICE BY XENOBIOTICS THAT ARE SULFATED

  • Phenol, acetaminophen(AA) and salicylamide are all known to be sulfated in rats and mioe. We have previously demonstrated that capacity-limited sulfation of xenobiotics in rats is due to the reduced availability of hepatic PAPS, the co-substrate for sulfation, which in turn is limited by the availability of its precursor, inorganic sulfate. Because species differences have been reported in the extent of sulfation, this study was conducted to determine whether these xenobiotics lower hepatic PAPS and sulfate in ICR mice. All three substrates decreased serum sulfate concentrations in a dose-and time-dependent manner. However. contrary to the observations in rats, phenol markedly increased hepatic PAPS concentraions in a dose-dependent manner, 1 hr after ip injection of 0-4 mmol/kg. Following ip injection of 2 or4 mmol/kg phenol, hepatic PAPS concentraions were enhanced 2-3 fold, 0.52 hr arter dosing and returned to control values 3 hr after dosing, whereas AA and salicylamide had little effect on hepatic PAPS concentrations. In summary. these studies demonstrate that phenol markedly enhances hepatic PAPS concentrations in mice, whereas hepatic PAPS levels are not affected by AA and salicylamide. Our data suggest that 1) hepatic sulfation for high dosages of xenobiotics in ICR mice is not limiod by the availability of co-substrate, and 2) there are significant species differences in the regulation of PAPS between rats and mice.

  • This research has been studied in terms of investigating the reaction behavior of pyrite with a cyclone reactor. The Mathematical model has developed pyrite oxidation and lime sulfation in this reactor. The model assumes a chemical control shrinking core behavior for the pyrite and a fluid film control shrinking core behavior for the lime. The model was solved and characterized numerically. Experiments have been performed to study the influence of reaction parameters such as reactor temperatures, pyrite particle sizes, air flow rates, feeding rates, and mixing ratio of pyrite and lime. The oxidation and sulfation products were characterized chemically and physically.

The development of a fundamental model for pyrite oxidation and lime sulfation in a vertical cyclone reactor. The model assumes a chemical control shrinking core behavior for the pyrite and a fluid film control shrinking core behavior for the lime. The oxygen and sulphur dioxide concentrations and the energy balance for the gas, pyrite and lime particles are solved. The model was solved and characterized numerically. An Empirical Study on the Application of Lead-Acid Batteries to ESSs and Performance Improvement Methods

  • In this paper, an approach to extend the life of lead-acid batteries through the elimination of sulfation with "rest time" using the pulse current of a conventional DPV (Differential Pulse Voltammetry) method is discussed. A new rest time without "0" current in "saw-tooth" pattern pulses is proposed to overcome the "0" current (blackout period), which is a shortcoming of DPV. This will enable the proposed method to be used in the loaded state. In the proposed approach, ESSs (Energy Storage Systems) were discharged for 5 h twice per day for 6 months on weekdays. To observe the changes in the lifespan of the systems, for the same period, the changes in the impedances of lead-acid batteries which were being charged e without charging and discharging were measure and compared. This study is focused on determining the effectiveness of lead-acid batteries as ESSs.

  • Polysaccharide from Sanguisorba officinalis was separated and fractionated using DEAE-Sephadex ion-exchange chromatography and Sephacry HR-200 gel filtration chromatography. One of the fraction(Fr. II) was sulfated and its anticoagulant activity was tested in vitro. Sulfation could increase the clotting time 50 times compared to unsulfated one. Fr. II was hydrolyzed and its composition was analyzed by conjugation with 7-amino-1, 3-naphthalene disulfonic acid using HPLC and electrophoresis. Arabinose and galactose were mainly composed at the ratio of 4 : 1. In addition, xylose and rhamnose were also found.

  • EC-2B와 EC-2C 획분은 정향(Eugenia caryophyllata)의 알칼리 추출물로부터 에탄올 침전, cetavlon 처리 및 한외여과를 거쳐 분획하였다. EC-2B 획분은 APTT에서 항응고 활성을 가지는 반면, EC-2C 획분은 APTT와 TT 모두에서 항응고 활성을 가지고 있으며 EC-2B와 EC-2C에서 모두 혈소판 응집억제능을 관찰할 수 있었다. EC-2B와 EC-2C 획분의 경구투여에서 두 획분 모두 독성이 없었으며 EC-2B 획문은 1,000 mg/kg (mouse, intravenours)에서도 독성이 없었으나, EC-2C 획분은 LD50 322 mg/kg 정도의 독성을 가지고 있었다. 두 획분의 in vivo 성에서의 항응고 활성을 60% 생존율을 갖는 dose로 표시한 결과 EC-2B는 131 mg/kgdlsep 비해 EC-2C는 58 mg/kg로 활성의 차이가 in vitro에서 보다 크게 나타났다. 이 두획분을 sulfation시킨 후 활성의 변화를 ex vivo를 통해 확인한 결과 두 획분 모두 활성이 증가하였으며 특히 EC-2B 획분의 활성이 급격히 증가하였다.

A study of the Property Changes of Oil Used in Diesel Engine (디젤 엔진오일의 사용에 따른 물성 변화에 관한 연구)

  • This study is concerned with the property changes(chemical and mechanical) of the used oil in diesel engine, which were sampled from a test engine with dynamometer and various cars. The properties of oil are TAN, viscosity, oxidation, ZDTP depletion factor and etc. Also the relation between the change of chemical and antiwear property of used oil was strongly related to ZDTP depletion factor as wall as the change of TAN(total acid number), sulfation and pentan unsoluble contents which were reated to the formation of protect film in sliding area. The oil used in pick-up(small) truck engine was deteriorated within the shortest distance than that of other cars. The antiwear property of used oil was decreased sharply as the running distance were over 5,000km. The oil used to mini bus was the least to deteriorate of properities for the funning distance.

  • The sulfation of the domestic titaniferous magnetite ore with ammonium sulfate was investigated to find a cyclic process for the production of synthetic rutile and high purity iron oxide and to test the feasibility of ammonium sulfate being an alternative sulfation agent. The proper sulfation conditions were determined to be a temperature of $425^{\circ}C$, 2.5 hours of reaction time, the weight ratio of ammonium sulfate to titaniferous magnetite : 11, and particle size or titaniferous magnetite : -250 mesh. 90.4 % of $TiO_2$ and 85.3 % of iron were extracted from the titaniferous magnetite sulfated under these conditions by the water leaching. From the leachate $TiO_2$ of 93.8 % purity as a mixture of rutile and anatase and ${\alpha}-Fe_2O_3$ of 97.6 % purity were obtained.

  • An In vitro protein sulfation in the soluble fraction of rat brain was charaderized further by an improved method of alkaline hydrolysis and thin layer ceflulose electrophoresis TLE) The protein sulfation was carried out in a reaction system containing [35 S] 3'-phosphoadenosine-5'-phosphosulfate (PAPS), Tris-maleate buffer (pH 8), MgCI$_2$, and soluble proteins from rat brain. The sulfated proteins were precipitated by acetone and alkaline hydrolysis was performed to obtain sulfated amino acids. The hydrolysate was separated further by TLE and the separated residues were identified by fluorography. The Iluorography of one-dimensional The showed at least nine sulfated residues including tryosine-O-sulfate. The other spots were not identified yet positively. General properties of protein sulfotransferases (PST) using this method were re-examined such as effects of concentrations of PAPS, pH, incubation temperature and $Mg^2$+. These results suggest a possible occurrence of several PST corresponding to each sulfated residue in rat brain and that the sulfation can occur not only in tyrosine but also in other residues as well.
