Continuous Production Process of Methyl Fructoside Using Alginate-enclosed Microspheres (Alginate-enclosed Microspheres를 이용한 메틸 프룩토시드의 연속생산공정)

  • Methyl fructoside was continuously produced in suspended bed enzyme reactor using alginate-enclosed microspheres biocatalyst which was developed for enzymatic synthesis of methyl fructoside. And the continuous operating conditions were optimized with reactor simulation in order to demonstrate a feasibility of commercialization of the continuous enzymatic production process development. The yield and productivity of methyl fructoside were as high as 47.1%o and $2g/\ell$-hr, respectively. The optimum operating conditions were pH 4.8, 30%(v/v) of methanol content and $2U/m\ell$ of enzyme activity when the initial concentration of sucrose is $0.291mo1/\ell$ at the reaction temperature of $25^{\circ}C$.

Effect of Diluent Salt Concentration and pH on the Enumeration of Vibrio parahaemolyticus by Direct Plating on Selective Agar

  • The maintenance of physiological activity during dilution is very critical for the accurate enumeration of Vibrio spp. in marine samples. We investigated the effect of various diluents on the recovery of Vibrio parahaemolyticus using the direct plate counting and most probable number (MPN) methods. The effects of NaCl (0.85 and 3%) and pH (from 6.6 to 7.4) in diluents based on distilled water or phosphate buffered saline (PBS) were evaluated with three V. parahaemolyticus strains. PBS-3% NaCl (pH 6.6), as opposed to PBS, was the most effective diluent at maintaining viable cell numbers up to 2 log CFU/g during dilution for direct plate counting using on thiosulfate citrate bile salts sucrose (TCBS) selective agar, as well as minimizing the difference in cell numbers between TCBS and non-selective nutrient agar. It also increased counts of V parahaemolyticus inoculated into oysters relative to PBS (p<0.01), suggesting that PBS-3% NaCl (PH 6.6) can reduce the problem of underestimating V. parhaemolyticus counts using PBS alone.

Strain Improvement by Interspecific Fusion of Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 producing $\alpha$-Amylase Inhibitor ($\alpha$-Amylase 저해제 생성균 Streptomyces actuosus DMCJ-49와 Streptomyces minoensis DMCJ-144의 종간 융합에 의한 균주 개량)

  • Streptomyces actuosus DMCJ-49 and Streptomyces minoensis DMCJ-144 produce the .alpha.-amylase inhibitor. Inerspecific protoplast fusion technique was used to increase the productivity of .alpha.-amylase inhibitor. Four auxotrophic mutants were obtained respectively from two strains by N-methyl-N'-nitor-N-nitrosoguanidine(3mg/ml) treatment. The optimum conditions for the protoplast formation of Streptomyces actuosus DMCJ-49 ade was as follows; 1.2% w/v of glycine, 3mg/ml of lysozyme, and 30 min of lysozyme treatment followed by 36 hr. incubation in the protop-last formation medium. In case of DMCJ-144-his those were 1.2%w/v, 3 mg/ml, 30 minutes and 60 hours, respectively. Regeneration was accomplished with hypertonic soft agar medium that contained 0.4M sucrose, 20mM CaCl$_2$, 50 mM MgCl$_2$ and low levels of phosphate. Fusion of protoplasts carrying different auxotrophic markers was achieved by treatment with polyethylene glycol. The optimum concentration of polyethylene glycol 1450 for the production of recombinants was 40%w/v. When the protoplasts was treated with 40% polyethylene glycol for 30 minutes, the frequency of recombinants was 6.5$\times$$10^{-3}$ and the $\alpha$-amylase inhibition activity of $ade^-his^-$ No. 4, which is the fusant with the most improved activity increased from 33 to 125 I.U./ml.

  • Cultures of hairy root induced from ginseng(Panax C.A. Meyer) transformed with Agrobacterium rhizogenes (strain A4, ATCC 15834) were established and morphologically two different hairy root strains (HB1, HB2) were obtained. To determine the optimum growth rate, the hairy root (HB2) was cultured in various liquid medium supplemented with or without plant growth hormone. The growth rate of hairy root cultured on MS medium was 1.3-3.1 times higher than those cultured on other media, and the optimum sucrose concentration and pH were 3-6%, 5.5-6.5, respectively. Also, the growth rate of hairy root was increased when 0.02 M ammonium nitrate, 1.2 mM potassium phosphate (monobasic) and 0.5 mg/l IBA were supplied to liquid medium. The saponin patterns and contents of hairy root (HB2) were determined by TLC and HPLC. The crude saponin contents were 4.67% and the total saponin contents were 1.0%, on dry weight basis.

  • Perfusion culture strategies for high density culture of plant cell suspensions to enhance the productivity of extracellular polysaccharides were investigated. Angelica gigas Nakai cell suspensions were used to produce the extracellular polysaccharide and perfusion parameters were optimized to maximize the production. When the medium exchange was started at the fifth day after inoculation, the maximum cell concentration (23.8 g dry cell weight per liter) was achieved.

  • Bioconversion of aglycone-formed isoflavones from glycoside-formed isoflavones by commercial lactic acid bacteria in fermented soybean paste was evaluated. Enterococcus faecium KCTC 13410 showed the most resistant capacity and Lactobacillus acidophilus KCTC 3925 had a sensitive susceptibility at a high NaCl concentration (13.2%) in fermented soybean paste. Among the 5 strains tested, Lac. acidophilus KCTC 3925 showed the highest relative ratio of aglycone-formed isoflavones to total isoflavones in fermented soybean paste. Production of exopolysaccarides (EPS) by lactic acid bacteria was compared using de Man, Rogosa, and Sharpe medium containing 1% sucrose at $37^{\circ}C$ for 48 h. Among the 5 lactic acid bacteria, Lac. acidophilus KCTC 3925 and Lactobacillus rhamnosus KCTC 3929 were investigated to produce EPS. Based on the results concerning growing susceptibility and conversion of aglycone-formed isoflavones/EPS production, it is anticipated that Lac. acidophilus KCTC 3925 may be used for preparation of Cheonggukjang, which contains relative low NaCl content.

Studies on Persimmon Wine (감술제조에 관한연구)

  • The possibility of wine making from soft and dried persimmon and the effects of treatment for persimmon on the quality of wine have been investigated. Soft persimmon was pretreated by two method of heat and hydrochloric acid treatment. Fermentation mash was composed of 12% persimmon and 15% of sucrose and was fermented at 17$^{\circ}C$ for 36 days. The fermentation using persimmons which were untreated and treated by acid was not successful because of contamination caused by lactic acid and acetic acid bacteria. Acidity of the fermented broth produced from boiled dried acid treated and untreated persimmon was 4,4, 5.0, 5,8 and 13.5ml(0.1N NaOH/10ml broth) respectively. Ethanol concentration of the fermented broth produced from boiled dried acid treated and untreated persimmon was 11.2, 10.0, 9.4 and 6.1% v/v respectively. The wine fermented for 28day's fermentation using boiled persimmon as substate had the best quality and stability. After 36day's fermentation using boild persimon following composition was obtained: 4.65% of total sugar 3.65% of reducing sugar 0.03mg/ml of protin 0.17$\mu$/ml of amino acid and 13.02 $\mu$g/ml of pectin. The pH and yeast cell of the boiled persimmon broth were 3.4 and 5.75 logCFU/ml. Fermented wine using dried persimmon had aslo good quality but the fermentation rate was slow.

  • This study was attempted to investigate the production of L-tyrosine by Brevibacterium flavum ATCC 14067. To select the strain which produce more L-tyrosine, mutants were induced by N-methyl-N'-nitro-nitrosoguanidine (NTG) treatment and phenylalanine auxotrophic mutants were induced by NTG and penicillin treatments. PFP resistant mutant was isolated from a phenylalanine auxotroph by retreatment with NTG and screened for increase of L-tyrosine production. PFP-326 mutant resistant to PPP (100ug/ml) was derived from phenylalanine auxotroph by mutagenesis with NTG and PFP-106 mutant resistant to PFP (1201g/ml) was derived from PFP-326 by mutagenesis with NTG. The composition of media for L-tyrosine production in strain PFP-106 was studied. PFP-106 mutant strain produced 50mg 11 of L-tyrosine while the parent strain produced 0.56mg 11 of L-tyrosine. The optimum composition of medium for L-tyrosine by strain PFP-106 was 10cA sucrose as carbon source, 3% ammonium sulfate as nitrogen source. The optimum cultural condition for producing L-tyrosine by strain PFP-106 was L-phenylalanine at a concentration of 1000g/mg.

  • The metabolic behavior of Debaryomyces hansenii was investigated in terms of substrate utilization and by product formation under different cultural conditions. Debaryomyces hansenii exhibited best growth and most tolerant of increased NaCl, sucrose and potassium sorbate at their optimum pH (5.0). A combination of two or more environmental stresses had stronger inhibitory effects on their growth kinetics, utilization of carbohydrate substrates and the production of organic acids, volatile compounds and other metabolites. Significant amounts of glycerol (0.35-4.4 g/L) and arabitol (0.08-9.8 g/L) were produced by D. hansenii. The main organic acids produced were citric (0.6-1.4 g/L), acetic (0.3-2.8 g/L), fumaric (0.2-1.0 g/L) and malic acids (1.1-1.7 g/L). A range of other compounds such as ethyl acetate, n-propanol, isoamyl alcohol, 2-phenylethanol and acetoin were also produced. The concentration of these compounds varied with the cultural conditions. Such compounds would have specific impacts on food quality in which D. hansenii is found.

A Protocol for High Frequency Plant Conversion from Somatic Embryos of Peanut (Arachis hypogaea L. cv. DRG-12)

  • A protocol was developed for somatic embryogenesis with 100% induction rate from immature zygotic embryo axes of peanut (Arachis hypogaea L. cv. DRG-12) cultured on MS medium containing $18.09\;{\mu}M$ 2,4-D. The frequency of somatic embryogenesis (31.7%) as well as the number of somatic embryos induced per explant (6.6) decreased when the concentration of 2,4-D was increased to $72.4\;{\mu}M$. Morphologically abnormal somatic embryos were observed at a frequency of 43.3% on MS medium containing $72.4\;{\mu}M$ 2,4-D. Somatic embryos isolated from 30-day-old cultures of immature zygotic embryo axes exhibited precocious germination with varied responses when placed on MS basal medium with 3% sucrose. Maximum shoot induction (80.0%) was observed from somatic embryos isolated from 60-day-old cultures of immature zygotic embryo axes when placed as a clump rather than individually on MS medium supplemented with $26.63\;{\mu}M$ BA and $0.54\;{\mu}M$ NAA. Shoots developed from somatic embryos rooted with higher frequency (93.3%) on Blaydes' medium containing $5.4\;{\mu}M$ NAA.
