Biological Activity and Chemical Characteristics of Cordyceps militaris Powder Fermented by Several Microscopic Organisms (발효 동충하초의 유용성분 및 생리 활성 작용)

  • The comparative effects of the fibrinolytic action, antioxidative activity, and tyrosinase inhibition of Cordyceps militaris powder and fermented Cordyceps militaris powders were investigated using several microscopic organisms. The nutritional components such as phenolic compounds, flavonoids, and minerals were also measured. The total phenolic compounds and flavonoid concentrations were highest in the Cordyceps militaris powder fermented by Aspergillus oryzae. Major minerals were K, Ca, Mg, and Zn. Native polyacrylamide gel electrophoresis (native-PAGE) analysis of the total protein patterns of Cordyceps militaris powder and fermented Cordyceps militaris powders revealed slight varietal differences. Fibrinolytic activity was highest in the Cordyceps militaris powder fermented by Bacillus subtilis and Aspergillus kawachii. The DPPH radical scavenging activity was slightly stronger in the powder fermented by Monascus purpureus; however, these samples all exhibited a relatively low activity when compared with butylated hydroxytoluene (BHT). Tyrosinase inhibition activity was stronger in the powder fermented by Aspergillus oryzae than in unfermented powder. These results may provide basic data for understanding the biological activities and chemical characteristics of Cordyceps militaris powder fermented by several microscopic organisms for the development of functional foods.

Purification of Two Novel Antimicrobial Peptides from Pyloric Caeca of the Starfish Asterina pectinifera (별불가사리 Asterina pectinifera의 유문맹낭 추출물로부터 새로운 2종류의 항균활성 펩타이드의 정제)

  • PAP-1, a novel antimicrobial peptide isolated from pyloric caeca extract of the starfish Asterina pectinifera was purified and characterized. First, the acidified pyloric caeca extract was put through Sep-Pak C18 solid phase extraction cartridge using a stepwise gradient. Among the eluents, RM 60 (retained materials at 60% methanol) showed good antimicrobial activity against Bacillus subtilis and Escherichia coli D31 and was purified in C18 reversed-phase and ion-exchange high-performance liquid chromatography columns. The purification steps yielded two novel peptides showing strong antimicrobial activities. These peptides were named pyloric caeca A. pectinifera peptide 1 and 2 (PAP-1 and PAP-2). For the characterization of the purified peptides, the molecular weights and amino acid sequences were determined by MALDI-TOF MS and Edman degradation. The molecular weights of PAP-1 and PAP-2 were about 2951.54 Da and 2980.15 Da respectively. The amino acid sequences of PAP-1 and PAP-2 were partially determined: AIQNAGES and AIQNAAES, respectively. PAP-2 is an isoform of PAP-1, differing merely by a single residue at position 6 (glycine or alanine). The comparison of the N-terminal amino acid sequences and molecular weights of the peptides with those of other known antimicrobial peptides revealed that PAP-1 and PAP-2 have no homology with any known peptides. These findings suggest that PAP-1 and PAP-2 play a significant role in the innate defense system of starfish pyloric caeca.

Studies on the Antibacterial Activity and Active Constituents of Sonbanghwalmyoungum (선방활명음(仙方活命飮)의 항균효능(抗菌效能) 및 성분(成分)에 관(關)한 연구(硏究))

  • 약효(藥效) 및 안정성(安定性)이 입증(立證)된 한약(韓藥)으로부터 새로운 항균제(抗菌劑)를 도출(導出)하고자, 임상(臨床)에서 청열작용(淸熱作用)이 있거나 염증성(炎症性) 질환(疾患)과 박테리아의 감염(感染)으로 비롯된 질환(疾患)에 자주 쓰이는 22개(個)의 처방(處方)에 대(對)해 paper disk 법(法)을 사용(使用)하여 항균(抗菌) 효능(效能)을 검색(檢索)하였다. 시료(試料)는 한약(韓藥)의 고용(股用) 방법(方法)을 고려(考慮)하여 물로 추출(抽出)하였으며, 사용(使用) 균주(菌柱)로는 그람 양성균(陽性菌)인 포도상구균(葡萄狀球菌)과 고초균(枯草菌) 그리고 그람 음성균(陰性菌)인 대장균(大腸菌)과 녹농균(綠膿菌)을 사용(使用)하였고, 대조물질(對照物質)로는 tetracycline을 사용(使用)하여 상법(常法)에 의(依)해 실험(實驗)하였다. 22개(個)의 처방(處方) 가운데 백두옹탕(白頭翁湯), 황연해독탕(黃連解毒湯), 증황연황연해독탕(增黃連黃連解毒湯), 사심탕(瀉心湯), 용담사간탕(龍膽瀉肝湯), 선방활명음(仙方活命飮) 등(等)의 처방(處方)이 탁월(卓越)한 항균(抗菌) 활성(活性)을 보였다. 이 가운데 본(本) 연구(硏究)에서는 탕제(湯劑)의 구성(構成) 약재(藥材)가 다른 처방(處方)들에 비(比)해 비교적(比較的) 다양(多樣)한 선방활명음(仙方活命飮)에 대(對)해 활성(活性) 성분(成分)의 규명(糾明) 및 구성(構成) 약재간(藥材間)의 상화작용(相互作用)을 밝히고자 하였다. 선방활명음(仙方活命飮) 5 kg을 메탄올 (MeOH)로 추출(抽出)한 뒤 우수(優秀)한 항균(抗菌) 활성(活性)을 보인 에칠아세테이트 (EtOAc) 분획(分劃)에 대(對)하여 silica gel 및 Sephadex LH-20 column chromatography를 반복(反復)하여 stilbene 계열(系列)의 화합물(化合物)인 Rhapontigenin (Compound 1)과 Rhaponticin (Compound 2)을 얻었다. S. aureus SG 511에 대(對)해 Rhapontigenin은 50 mg/ml 이상(以上) 농도(濃度)에서 항균(抗菌) 효과(效果)를 나타내었고, Rhaponticin은 $200\;mg/m{\ell}$ 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었으며, B. subtilis ATCC 6633에 대(對)해 Rhapontigenin은 50 mg/ml 이상(以上) 농도(濃度)에서 항균(抗菌) 효과(效果)를 나타내었고, Rhaponticin은 200 mg/ml 농도(濃度)에서 우수(優秀)한 항균(抗菌) 효과(效果)를 나타내었으며, 100 mg/ml 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었다. 또한 E. coli 055에 대(對)해서는 Rhapontigenin과 Rhaponticin 모두 200 mg/ml 농도(濃度)에서 미약(微弱)하게 유효(有效)한 항균(抗菌) 효과(效果)를 나타내었다. 이상(以上)의 결과(結果)로 보아 선방활명음(仙方活命飮)의 항균(抗菌) 효능(效能)은 군약(君藥)인 대황(大黃)의 성분(成分) 중(中)의 하나인 stilbene 계열(系列)의 화합물(化合物)인 Rhapontigenin과 Rhaponticin의 작용(作用)에 의(依)한 것이며, 이는 한의학(韓醫學) 방제(方劑) 원리(原理)인 군신좌사(君臣佐使) 이론(理論)에서 군약(君藥)이 주증(主症)에 주(主)로 작용(作用)하는 약물(藥物)이라는 것을 밝혀주는 것이라고 사료(思料)된다.

Carbon Metabolism and Its Global Regulation in Corynebacterium glutamicum (Corynebacterium glutamicum의 탄소대사 및 총체적 탄소대사 조절)

  • In this review, the current knowledge of the carbon metabolism and global carbon regulation in Corynebacterium glutamicum are summarized. C. gluamicum has phosphotransferase system (PTS) for the utilization of sucrose, glucose, and fructose. C. glutamicum does not show any preference for glucose when various sugars or organic acids are present with glucose, and thus cometabolizes glucose with other sugars or organic acids. The molecular mechanism of global carbon regulation such as carbon catabolite repression (CCR) in C. glutamicum is quite different to that in Gram-negative or low-GC Gram-positive bacteria. GlxR (glyoxylate bypass regulator) in C. glutamicum is the cyclic AMP receptor protein (CRP) homologue of E. coli. GlxR has been reported to regulate genes involved in not only glyoxylate bypass, but also central carbon metabolism and CCR including glycolysis, gluconeogenesis, and tricarboxylic acid (TCA) cycle. Therefore, GlxR has been suggested as a global transcriptional regulator for the regulation of diverse physiological processes as well as carbon metabolism. Adenylate cyclase of C. glutamicum is a membrane protein belonging to class III adenylate cyclases, thus it could possibly be a sensor for some external signal, thereby modulating cAMP level in response to environmental stimuli. In addition to GlxR, three additional transcriptional regulators like RamB, RamA, and SugR are also involved in regulating the expression of many genes of carbon metabolism. Finally, recent approaches for constructing new pathways for the utilization of new carbon sources, and strategies for enhancing amino acid production through genetic modification of carbon metabolism or regulatory network are described.

Antioxidation, Antimicrobial and Antithrombosis Activities of Aged Black Garlic (Allium sativum L.) (흑마늘의 항산화, 항균 및 항혈전 활성)

  • In the course of study for development of functional food ingredients from aged black garlic (ABG), heat-treated ripe bulbs of Allium sativum L., the water extracts from raw-garlic (RG) and ABG, and the subsequent organic solvent fractions of ABG were prepared, and their antioxidant, antimicrobial, and antithrombosis activities were compared. The extraction yield of ABG was 4-folds higher than that of RG, and the contents of total polyphenol, total flavonoid, total sugar and reducing sugar in the ABG extract were 4-folds, 1.56-folds, 3.36-folds and 6.75-folds higher than those of the RG extract, respectively. In antioxidation activity assay, the extract of ABG showed minor scavenging activity against DPPH anion, but revealed strong scavenging activity against ABTS cation and nitrite. Especially, the ethylacetate fraction from the ABG extract demonstrated stronger antioxidation activity than the RG extract and other fractions. Although the antimicrobial and antithrombosis activities of the RG extract did not appear in the ABG extract, the ethylacetate fraction from the ABG extract had antibacterial activity against Staphylococcus aureus and Bacillus subtilis, and strong antithrombosis activity via the inhibition of prothrombin, blood coagulation factors and platelet aggregation. All extracts and fractions did not show any hemolytic activity against human red blood cells up to 5 mg/ml. Our results suggest that the ethylacetate fraction of ABG could be applicable to the development of functional food ingredients for antithrombosis agents.

Bacterial Distribution of Kochujang (개량식 및 재래식 고추장의 세균 분포)

  • This study was carried out to investigate the bacterial microflora of commercial and traditional Kochujang. Computer aided idetification systems were used to identify the bacteria in Kochujang. Seven genera such as Bacillus, Corynebacterium, Enterococcus, pasteurella, Pseudomonas, Staphylococcus and Streptococcus in commercial process and 10 genera such as Bacillus, Corynebacterium, Enterococcus, Flavimonas, Flavobacterium, Gemella, Pasteurella, Pseudomonas, Staphylococcus and Streptococcus in traditional process were identified. Distribution of genus Bacillus was $56{\sim}70%$ during fermentation in the commercial process and $38{\sim}50%$ in the traditional process. B. lichenoformis strains identified in this study were classified into three types by their physiological characteristics.

Chemical Composition and Antimicrobial Activities of Houttuynia cordata Thunb. (어성초의 화학성분 및 항미생물 활성)

  • Chemical composition and antimicrobial activity of Houttuynia cordata Thunb. were investigated to develop a natural food preservative from it. Aspartic acid, glutamic acid, glycine and arginine were major amino acids of Houttuynia cordata Thunb., but were present in a trace amount. Free sugars were composed of glucose, fructose, sucrose and maltose and major fatty acids were linolenic acid, linoleic acid, oleic acid, and palmitic acid. Mineral elements were potassium, calcium, magnesium, iron, zinc, and copper. Antimicrobial activities were shown in acidic, neutral and phenolic fraction of Houttuynia cordata Thunb., but not in basic fraction. Among the four fractions, neutral fraction showed the strongest antimicrobial activities against microorganisms tested, such as Pseudomnas aeruginosa, Micrococcus luteus, Bacillus subtilis, Aspergillus parasiticus and Aspergillus flavus. Minimum inhibitory concentration (MIC) and minimum lethal concentration (MLC) of the neutral fraction varied according to microoganisms tested. The lowest values of MIC (0.0075 g eq./mL) and MLC (0.10 g eq./mL) were obtained from Pseudomonas aeruginosa.

Solubility, Antioxidative and Antimicrobial Activity of Chitosan-Ascorbate (키토산-아스코베이트의 용해성, 항산화성 및 항균성)

  • This study was conducted to investigate the solubility, antioxidative and antimicrobial activity of the freeze dried chitosan-ascorbate (CAs) and chitosan-acetate (CAc). In the results of solubility, CAs was soluble over 0.5% in distilled water, vinegar, green tea, soju (distilled liquor), beer and red wine, while it was not soluble in soy sauce, soy milk, milk, orange juice, coffee, sesame oil, soy milk and soybean oil. The solubility of CAc in the liquid foods was similar to those of CAs, but it was soluble less than 0.1% in beer, and formed curd in red wine. Electron donating activity, antioxidative activity and SOD activity of CAs were 48.2, 90.6 and 67.5%, respectively, while the activities of the CAc were 0, 40.0 and 10.0%, respectively. The minimal inhibitory concentrations of CAs and CAc were $200\;{\mu}g/disc$ against Bacillus circulans, Bacillus brevis, Bacillus licheniformis, Bacillus arabitane and Bacillus sterothermophillus, $400\;{\mu}g/disc$ against Escherichia coli O157, Listeria monocytogenous, Bacillus cereus and Bacillus subtilis. There was no significant difference in Hunter's L* value between CAs and CAc $(81.95{\sim}82.97)$, but Hunter's a* and b* values of the CAs was higher than those of CAc. While sour and bitter tastes of CAs were lower than those of CAc, there was no significant difference in astringent taste. From these results, it suggested that CAs has more extensive utility in liquid foods with antimicrobial and antioxidant activity as well as sensory quality compared to CAc.

Antioxidant and Antimicrobial Activities of Extracts from Sarcodon aspratus (능이버섯(Sarcodon aspratus) 추출물의 항산화성과 항균성)

  • The antioxidative and antimicrobial activities were determined on the mushroom (Sarcodon aspratus) extracts in order to find out new food functional components. The antioxidative activities of water and ethanol extracts from the Sarcodon aspratus were measured by peroxide values (POV), electron-donating ability (EDA) using 1,1-diphenyl-2-picryl hydroxyl (DPPH), nitrite-scavenging ability and superoxide dismutase-like activity (SODA) by pyrogallol. The antioxidative activity of the ethanol extract measured by POV was higher than those of the water extract, BHT, and ${\alpha}-tocopherol$. The EDA of the water extract and ethanol extract using DPPH showed the highest values of 76.94% and 73.06%, respectively. The nitrite-scavenging abilities (pH 1.2, 1,000 ppm) of the water and ethanol extracts were 72.61% and 62.69%, respectively, and the nitrite-scavenging ability of the water extract was higher than that of the ethanol extract in all pH values. The SODA of the ethanol extract was higher than that of the water extract. The Sarcodon aspratus extracts had antimicrobial effects on Listeria monocytogenes and Staphylococcus aureus.

Effects of Fermented Soybean upon Anti-inflammation and Intestinal Mucous Membrane Permeability (청국장의 항염증 및 장점막 투과성 개선 효과)

  • Objectives This study was designed to investigate the effects of fermented soybean upon anti-inflammation, cytotoxicity, antioxidant and intestinal mucous membrane permeability by measuring the cell viability, NO (nitric oxide) production, DPPH, Polyphenol, HRP and TEER in cells like Raw 264.7 and HCT 116 using fermented soybean. Methods Raw 264.7 cell and HCT 166 cell were used in this study. And fermented soybean powders were used for the experimental group and soybean powders for the control group. There was inflammation response upon using lipopolysaccharide(LPS). Fermented soybean powders and soybean powders were in a respectively different dose added to the cells with LPS. MTT assay, NO, DPPH and Polyphenol measurement, TEER, HRP were conducted for each cell. The results of this study were presented in mean and standard deviation. Results 1. In Raw 254.7 cells added with $100{\mu}l/ml$ unfermented soybean powders, 104.95% higher than 62.59% was measured. In Raw 254.7 cells added with $100{\mu}l/ml$ fermented soybean powders, there was 74.90% measured higher than 62.59%, which was a significant result. 2. By a gradual increase of unfermented soybean powders like $0.1{\mu}l/ml$, $1.0{\mu}l/ml$, $10{\mu}l/ml$, $100{\mu}l/ml$, the measured NO were also gradually decreased $53.12{\mu}M$, $47.57{\mu}M$, $37.02{\mu}M$, $28.16{\mu}M$. In case of cells added with fermented soybean powders, $43.95{\mu}M$ NO was measured in $0.1{\mu}l/ml$ which is significant, and in other cases, mostly measured over$ 56.72{\mu}M$. 3. It was inferred that fermented soybean powders have anti-inflammatory effects of maintaining intestinal mucous membrane permeability because the measured values of cells in both groups were all higher than $133.62{\Omega}$ measured of cells added with only LPS. And measured values of cells in both groups were all lower than 2.26 measured of cells added with only LPS. 4. In case of experiment DPPH and polyphenol measurement, fermented group was all higher than unfermented group. Conclusion From the results of conducting MTT assay, NO measurement, and TEER, HRP by using cells Raw 264.7 and HCT-116, even though there was no significance in the correlation between cytotoxicity, anti-inflammatory effects, both unfermented soybean powders and fermented soybean powders were shown to have intestinal mucous membrane permeability improvement effects. This effects could be applicable for autoimmune diseases, chronic inflammatory diseases and so additional studies are expected in the future. From the results of conducting DPPH, Polyphenol measurement, Fermented soybean may be useful as potential antioxidant.
