Inhibitory Substance Produced by Aspergillus sp. on the Snake Venom Proteinase - Isolation of Microorganism and Biological Activities of the Inhibitor - (Aspergillus 속 균주가 생성되는 사독 Proteinase에 대한 저해물질 - 균의 분리 및 저해물질의 생물학적 작용상 -)

  • Aspergillus sp. (MK-24) producing a biological active substance that inhibited the venom proteinase activity was isolated from soil. The substance also inhibited the activity of trypsin and coagulation of blood, but did not inhibit papain, $\alpha$-chymotrypsin and pepsin. The substance was partially purified from culture filtrate by precipitaion with acetone, and by chromatography of DEAE-Sepadex A-50 column and Amberlite IRC-50 ion exchange. The inhibitory substance was stable in the wide pH range from 2.0 to 12.0 at 37$^{\circ}C$, but not stable at $65^{\circ}C$ in the alkaline pH. Only 12% of the activity was decreased by the heat treatment at 10$0^{\circ}C$ for two hours. The inhibition on venom proteinase (Agkistrodon bromohoffi brevicaudus) was a mixed type. The inhibitory activity depended on the preincubation time and completely depressed by cupric, zinc and cobalt ions. The inhibition on the venom proteinase was appeared strongly on casein but not on ovalbumin or hemoglobin as a substrate.

  • DA-5018은 여러 opiate수용체 실험에서 morphine 또는 naloxone 보다 10-100배 정도 낮은 친화력을 나타내었다. 기니픽 회장표본과 랫드 수정관표본의 전기자극 실험에서는 DA-5018의 의해 유도된 수축반응이 naloxone의 영향을 받지 앉았고, 랫드에서의 진통효과도 naloxone 전처치에 의해 차단되지 않았으므로, DA-5018은 opiate 수용체를 경유하지 앉는 것으로 생각된다. 또한, OA-5018 120$\mu$M에서 cyclooxygenase 생성을 50% 증가시켰고 mM 농도에서 5-lipooxygenase 합성을 약간 억제하였으므로, NSAID계 유사약물이 아님을 확인하였다. DA-5018은 기니픽 기관 표본에 대해 capsaicin과 동일하게 수축반응을 나타내었고, 이것은 capsazepine 전처리에 의해 억제되었다. 척수에서의 substance P 유리활성은 capsaicin보다 약 9배 강한 것으로 나타났다. 또한 랫드에 DA-5018 진통유효용량인 1mg/kg을 피하주사한 후 분리한 척수에서 capsaicin에 의한 substance P의 유리활성은 15분 후에 감소하였고 120분 후에는 회복되었다. 따라서 OA-5018의 진통작용에도 substance P의 고갈이 관여하는 것으로 생각된다. 이상의 결과로부터, DA-5018의 진통작용은 capsaicin수용체를 매개하는 것으로 사료된다.

Effects of substance P on mineralization markers and heme oxygenase-1 Expression in human immortalized periodontal ligament cells

  • Substance P (SP) is known to be expressed in the nerve fibers of dental pulp and periodontal tissues. It was recently reported that SP expression increased in response to orthodontic force. In the present study, we investigated the effect of SP on expression of mineralization markers and heme oxygenase-1 (HO-1) in human immortalized periodontal ligament (IPDL) cells. Cell viability was measured using a 3,4,5-dimethylthiazol-2-yl-2,5-diphenyl tetrazolium bromide (MTT) assay. The expression of mineralization markers, including alkaline phosphatase (ALP), osteonectin (ON) and bone sialoprotein (BSP), and heme oxygenase-1 (HO-1) was assessed by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. SP did not significantly change human IPDL cell viability, with the exception of the 24 hour treatment group. Treatment of human IPDL cells with $10^{-10}$ to $10^{-4}M$ SP upregulated mineralization marker and HO-1 expression in a time- and concentration-dependent manner. Our results suggest that SP may modulate osteoblastic cell differentiation of human IPDL cells through a mechanism involving HO-1 expression.

Immunohistochemical study of the endocrine cells of the gastrointestinal mucosa of the African clawed toad, Xenopus laevis (아프리카 발톱두꺼비의 소화관내분비세포에 관한 면역조직화학적 연구)

  • The gastrointestinal endocrine cells of the African clawed toad, Xenopus laevis have been investigated immunohistochemically using the avidin-biotin method. Seven antisera were tested and three endocrine cell types immunoreacted with antisera to neurotensin, GRP and substance P. A moderate number of neurotensin-immunoreactive cells were weakly reacted in the small intestine. GRP-immunoreactive cells were mainly situated among the upper portion in the fundic glands, and the basal portion in the pyloric glands. These cells were oval and round in shape. On the other hand, in the intestine they were thin spindly cells with the epithelium. Substance P-immunoreactive cells were observed in among intestinal epithelium. However, no secretin-, motilin-, M-Enk- and PYY-immunoreactive cells were found in the GIT of the African clawed toads.

  • The conformation of a neuropeptide, substance P (SP), in isotropic (q = 0.5) acidic bicelles was investigated using two-dimensional NMR techniques. By the nuclear Overhauser effect (NOE) cross peaks between SP and long-chain lipid molecules SP was probed to bind on the flat surface of the disc-like bicelles. Structural analysis of NMR data indicated that the helical conformation of SP extended to the C-terminal region of Leu10 as well as in the mid-region from Pro4 to Phe8. As compared with the conformations of SP bound on the sodium dodecylsulfate (SDS) or the dodecylphosphocholine (DPC) micelles with curved surfaces, the surface curvature of the membrane mimics was found to be one of the major factors inducing the biologically relevant conformation of SP. The negative surface charge of the membrane is also a key factor inducing both the binding of SP on the membrane and its biologically active structure.

Distribution of Substance P(SP) Immunoreactive Nerve Fibers in the Tracheal Submucosal Glands of Cats (고양이 기관점막하분비선에 있어서의 Substance P(SP)양성신경섬유의 분포)

  • Immunohistochemistry combined with electron microscopy was employed to investigate the distribution of substance P -immunoreactive(SP-IR) nerve fibers in the tracheal submucosal gland of cats. SP-IR nerve fibers were found to form network around the glands. Numerous varicosities were also detected within the basement membrane of the acini and secretory tubules. All the intraglandular varicosities showed close spatial contact with serous cells, mucous cells and myoepithelial cells. Our findings suggest that SP-induced mucus secretion from tracheal submucosal glands in cats may be caused not only by glandular contractile response of myoepithelial cells but also by direct stimulation to both serous and mucous cells.

Capsaicinoids-induced Neurotoxic Desensitization in Guinea Pig: Antinociception and Loss of Substance P-like Immunoreactivity from Peripheral Sensory Nerve Endings in Bronchi

  • Antinociceptive and desensitizing effects of systemically administered capsaicinoids (capsaicin and KR25018) were investigated in guinea pig. Nociceptive sensitivity to chemical stimulus was examined to test sensory function, and the content of substance P-like immunorractivity (SP-LI) in bronchi was determined as a peripheral marker of capsaicin-sensitive primary afferent neurons. Guinea pigs were pretreated s.c. with several doses of capsaicin (1,2.5,5, 10 mg/kg) or KR25018 (1, 2.5, 5, 10 mg/kg) one week prior to the experiments. Frequency of eye wiping was significantly decreased by capsaicin and KR25018 in a pretreatment dosedependent manner. In capsaicin- or KR25018-pretreated guinea pigs, there was a significant loss of SP-LI in bronchial tissue extracts. In summary, a newly synthesized capsaicin analogue H725018 exhibited antinociceptive effect against chemical stimulus in guinea pig, with comparable potency to capsaicin. This desensitizing activity of capsaicin or KR25018 might be related to the loss of SP-LI in peripheral afferent nerves.

Alterations of c-Fos mRNA Expression in Hypothalamic-Pituitary-Adrenal Axis and Various Brain Regions Induced by Intrathecal Single and Repeated Substance P Administrations in Mice

  • The effect of substance P (Sub P) injected intrathecally (I.t.) on c-fos mRNA expression in vari-ous tissues was examined in the present study. We found that a single administration of Sub P(0.5 nM) caused an increase of the c-fos mRNA level in the hypothalamic-pituitary-adrenal(HPA) axis, hippocampus, and spinal cord. The time-course study showed that c-fos mRNA level was maximal at 10 min and began to decrease 30 min after the Sub P injection in all tis-sues, and the Sub P-induced increase of the c-fos mRNA level was returned to the control level 1 h after the injection. The kinetics of the c-fos mRNA expression in mice that were repeatedly injected with Sub P (every 30 min interval up to 4 times) were different in the HPA axis, hippocampus, and spinal cord. The increased c-fos mRNA level in the hypothalamus and the spinal cord induced by I.t. injected Sub P remained at a high level. In the pituitary gland, adrenal gland, and hippocampus, the increased level of c-fos mRNA expression gradually returned to the control level during the repeated substance P injections up to 4 times. Our results suggest that spinally injected Sub P-induced pain stress increases c-fos mRNA expres-sion in the spinal cord, hippocampus, and HPA axis. In mice repeatedly injected with Sub P, the kinetics of c-fos mRNA appear to be different varied from tissue to tissue.

Separation of Chromophoric Substance from Madder Plant under Different Extraction and Analytical Conditions (염료추출 및 분석 조건에 따른 꼭두서니의 색소성분 분리 거동)

  • This research was aimed to establish the standard extraction and analytical procedures for examining the chromophoric substance in madder root with the ultimate goal of identifying the dyes in badly faded textiles of archaeological origin. The separation temperature of gas chromatography, pH and other extraction conditions were tested. The results were as follows: The suitable separation temperature for the GC cappillary column was 50∼305$^{\circ}C$, and methanol was a good GC solvent for both standard alizarin and madder extraction. The best extraction of madder was achieved by 90 min soaking in room temperature followed by filtration and the actual heat extraction procedure. The best pH for extracting alizarin was pH 3 and above pH 5 alizarin was not detectible. Only alizarin and no purpurin was found in the extraction of the currently used madder plant.

Isolation and Identification of Streptomyces sp. Producing Anti-vancomycin Resistant Staphylococcus aureus Substance (반코마이신 내성 Staphylococcus aureus 억제 물질 생산 Streptomyces sp.의 분리 및 동정)

  • An Actinomycetes producing an anti-VRSA (vancomycin-resistant Staphylococcus aureus) substance was isolated from soil. The cultural, morphological, physiological and phylogenetic analyses of an isolated strain were investigated for identification. Cultural characteristics based on ISP (International Streptomyces Project) were as follows: white aerial mycelium, yellow reverse side, and good growth on various medium. Also, the isolate did not produce the soluble pigment. Morphological characteristics were showed cylindrical spore chain and smooth spore surface by SEM (Scanning Electron Microscope). Physiological characteristics were showed LL-type by DAP isomer analysis and detected glycine, glutamic acid and alanine. A phylogenetic analysis of the 16S rDNA provided a clue that the isolated strain was actually a member of the genus Streptomyces, because the determined sequence exhibited a higher homology with Streptomyces echinatus. The isolate was identified to be a genus of Streptomyces sp.. The optimal culture conditions for the maximum production of anti-VRSA substance by Streptomyces sp. were attained in a culture medium composed of $2.0\%$ (w/v) glucose, and $0.4\%$ (w/v) yeast extract. The anti-VRSA substance was highly produced after 5 days of culture. Optimal pH and temperature conditions for the production of anti-VRSA substance were pH 7.0 and $28^{\circ}C$, respectively.
