EFFECTS OF A CYTOTOXIC SUBSTANCE, PANAXYTRIOL FROM PANAX GINSENG C.A. MEYER ON THE IMMUNE RESPONSES IN NORMAL MICE

  • The effects of panaxytrion as known to be a cytotoxic substance isolated from Panax ginseng on the immune responses were examined. The i.p. administration of panaxytriol to normal mice for 6 consecutive days as doses of 5, 10 and 20 mg/kg suppressed the increase of body weight dose-dependently but did not affect the weight ratio of immunoorgans to body weight, No significant changes were observed in the humoral immune responses as measured by Arthus reaction and plaque forming cells and in the cellular immune response as measured by delayed hypersensitivity as well as phagocytic activity of reticuloendotherial system. These results suggested that panaxytriol, a cytotoxic substance to cancer cells, has no detrimental effects on the immune function in normal mice.

무당개구리 (Bombina orientalis) 피부의 초기 상처치유반응 (Early Healing Responses of the Skin Wounds in the Korean Fire-Bellied Toad, Bombina orientalis)

  • Early wound healing responses in the skin of the Korean fire bellied toad, Bombina orientalis, were examined using histochemical , scanning and transmission electron microscopical techniques. Cutaneous wounds were induced by sharp razor blade , and maintained in special cages for up to 3 days. By this treatment whole parts of epithelium and dermis were profoundly affected. After injury, retraction of damaged surface of the skin was observed. At 3 hours after injury, poly-band was form by mucous substance. The wound skin consisted of mucous gland secreted mucous substance which flows into wound cavity and the wound area was sealed by mucous substance which form thick layer at 6 hours. It appeared that natural disposition showed acid and neutral mucous substance by reaction of PAS and Alcian blue (pH 2.5) staining. After formation of the poly-band, epithelial cells were formed by migration of regenerating epithelial cells. These cells moved over the wound surface by migration at 12 hours after wounding. Poly-band was much increased in thickness and migrating cells were completely formed wound closure at 24 hours post-wounding. After 72 hours regenerated epithelial layer consists of four or five cell layers which are similar to clear, granular, and spinous layer.

간호대 학생과 일반대 학생이 지각하는 간호사 이미지 (Perceived image of nurses on major and non-major students)

  • Purpose: This study was conducted to investigate the perceived image of nurses on major and non-major students. Methods: The subjects were 120 nursing school students and 120 other university students in J province. The data was collected by using structured questionnaire. T-test, ANOVA were performed by SPSS/PC 20.0. Results: The score for image of nurses was as follows. special image($3.86{\pm}0.51$), traditional image($3.71{\pm}0.53$), individual image($3.53{\pm}0.85$), social image($3.43{\pm}0.58$), and grand mean($3.66{\pm}0.48$). Perceived image of nurses was significant difference between students of a major and non-major(t=6.390, p<.001). Conclusion: It is needed to monitoring of media for the improvement the perceived image of nurses and also nurses have to try to make an individual role model.

Effect of Vitamin E and Zinc Supplementation on Energy Metabolites, Lipid Peroxidation, and Milk Production in Peripartum Sahiwal Cows

  • The study was conducted to evaluate the effect of vitamin E and zinc supplementation on energy metabolites, lipid peroxidation, and milk production in peripartum Sahiwal cows. For this, thirty-two pregnant dry Sahiwal cows were selected at sixty days prepartum and divided into four groups viz control, $T_1$, $T_2$, and $T_3$ of eight each. Group $T_1$ were supplemented with zinc at 60 ppm/d/cow, group $T_2$ were supplemented with vitamin E at 1,000 IU/d/cow and group $T_3$ were supplemented with combination of vitamin E at 1,000 IU/d/cow and zinc at 60 ppm/d/cow during d 60 prepartum to d 90 postpartum. Blood samples were collected on d -60, -45, -30, -15, -7, -3, 0, 3, 7, 15, 30, 45, 60, 90, and 120 with respect to day of parturition and analysed for glucose, non esterified fatty acid, and thiobarbituric acid reactive substance. Body condition score was maintained significantly better (p<0.05) in $T_3$ than in the control, $T_1$ and $T_2$ groups. Overall glucose level was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Levels of nonesterified fatty acid, and thiobarbituric acid reactive substance were lower (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. Milk yield was higher (p<0.05) in $T_3$ than control, $T_1$, and $T_2$ groups. In conclusion, the present study indicated that the supplementation of vitamin E and zinc in peripartum Sahiwal cows enhanced milk production by reducing negative energy balance.

PPAR-$\gamma$ 조절을 통한 여택통기탕(麗澤通氣湯)의 항알레르기와 항염증 효과 (The Antiallergic Effects of Yeotaectonggi-tang (YTT) on the Mice Model with Allergic Rhinitis by Regulation of PPAR-$\gamma$)

  • Objectives : The aim of this study is to investigate the antiallergic effects of YTT on allergic rhinitis by regulation of peroxisome proliferator-activated receptor gamma (PPAR-$\gamma$). Methods : The thirty rats were divided into three groups : normal group, control (allergic rhinitis elicited) group, sample (Yeotaectonggi-tang treated after allergic rhinitis elicitation) group. To induce allergic rhinitis in control group and sample group, rats were sensitized intraperitoneally with 0.1% ovalbumin(OVA) solution 3 times at intervals of 1 week. Then intranasal sensitization was performed by diffusing 0.1% ovalbumin(OVA) solution 3 times at intervals of 2 days. After that time rats in sample group were oral administration treated by YTT for 7days. The change of the amounts of eosinophil, substance P, MIP-2, PPAR-$\gamma$, IL-4, iNOS were observed in each group. we used the statistical method of ANOVA test(p<0.05). Results : The number of eosinophil in sample group noticeably decreased than control group. And the decrease of substance P and MIP-2 positive reaction were observed in mucosa. YTT inhibited IL-4 and iNOS production, mucus secretion, activation of mast cells and fibrosis remodeling by regulation of PPAR-$\gamma$ activation. Conclusion : According to the above results, it is considered that YTT mitigated mucosa damage on the mice model with allergic rhinitis by regulation of PPAR-$\gamma$.

Monascus sp. MK2 균주가 생산하는 홍국색소의 안정성 (Stability of Monascus Pigment Produced by Monascus sp. MK2)

  • 본 연구에서는 Monascus sp. MK2가 생산하는 홍국색소의 안정성에 대하여 조사한 결과에서 광 안정성에서는 암 조건>자외선>실내(형광)>태양광 조건 순서로 안정하였으며, 온도에 대한 안정성은 온도가 증가하고 저장시간에 길어짐에 따라 홍국색소의 감소율 또한 비례적으로 증가하였고, pH에 대한 안정성은 전체적인 pH 영역에서 비교적 안정하였으며, 유기산 첨가에 대한 홍국색소의 안정성은 높은 것으로 생각되며, 합성 항산화제와 천연물 추출물 첨가시에는 안정성이 증대되는 것으로 조사되었다.

Differences in toughness and aging potential of longissimus lumborum muscles between Hanwoo cow, bull and steer

  • Thirty Hanwoo cattle including bulls, cows, and steers (n = 10 each) were slaughtered and investigated for carcass traits (weight, meat color, fat color, yield index, maturity, marbling score, back-fat thickness, and firmness) and meat quality. The meat quality such as: pH, color, cooking loss, fatty acid, thiobarbituric acid reactive substance, warner-bratzler shear force, tensile tests, and texture profiles were analyzed on longissimus lumborum (LL) muscles of the carcasses at different aging times (3 d and 21 d). The results showed that steers and cows had higher back-fat thickness and marbling score, and a lower firmness (p < 0.001) than bulls. Bulls exhibited a lower meat quality indicating by higher cooking loss, thiobarbituric acid reactive substance content, warner-bratzler shear force and tensile test values (p < 0.01). Regarding the sensory property, the bull meat also had higher hardness, and lower tenderness, juiciness and flavor scores than the cow or steer meat (p < 0.01). Additionally, the bull meat had a higher polyunsaturated fatty acid and a lower monounsaturated fatty acid contents (p < 0.01). With increased aging time, the meat tenderness was improved in all the genders. Taken together, the present study demonstrated that the gender and aging time affected the carcass traits, fatty acid and sensory quality of beef. Postmortem aging could improve the meat tenderness of all genders especially bulls.

Rahnella aquatilis AY2000균주가 생산하는 항효모성 물질의 이화학적 특성 (Physicochemical Properties of an Anti-Yeast Substance Produced by Rahnella aquatilis Strain AY2000)

  • Rahnella aquatilis AY2000 균주는 항효모성 물질(AYS)을 생산하지만, AYS가 저장 중에 활성이 감소되는 경향이 있었다. 따라서 본 연구에서는 AYS활성이 저하되는 원인을 알아보기 위하여 AYS에 다양한 이화학적인 처리를 행하였다. 그 결과 열처리가 AYS의 활성을 감소시키는 하나의 인자이며, AYS를 침전시키는 유기용매인 methanol의 사용 또한 AYS의 활성을 감소시키는 원인이었다. 또한 $\beta$-mercaptoethanol 과 dithiothreitol 등 thiol화합물 역시 AYS의 활성을 감소시키는 인자로 작용하였으나, pH, EDTA나 NaCl은 AYS의 활성저하를 가져오지 않았다. 한편, 정제과정에서 AYS의 조성분 중 다당류와 미지의 물질(230 nm)을 DEAE-cellulose 이온교환수지로 분리시키면 AYS의 활성이 완전히 소실되지만, AYS를 Sephacryl S-400 gel 여과를 행하여 이들 성분이 분리되지 않은 상태에서는 그 활성이 잘 유지되었다. 본 실험에 사용된 AYS의 MIC는 S. cerevisiae 대해 $7.8-15.6{\mu}g/mL$ 범위로 측정된다.

Lactobacillus salivarius LCH1230으로부터 생산된 Listeria 균 억제물질의 특성 (Characterization of Anti-Listerial Substance Produced by Lactobacillus salivarius LCH1227)

  • 전통발효식품으로부터 L. monocytogenes ATCC 19114에 강한 항균활성을 보이는 균주를 분리하였다. 분리된 균주의 형태학적 및 생화학적 특성과 16S rRNA 염기서열 분석을 통해 Lactobacillus salivarius LCH1227으로 동정되었다. L. salivarius LCH1227이 분비하는 항균물질은 L. monocytogenes 뿐만 아니라 vancomycin-resistant Enterococcus faecalis, Streptococcus agalactiae, Bacillus cereus, Lactobacillus fermentum 등과 같은 그람양성 세균에 대해 폭 넓은 항균 활성을 나타내었다. L. salivarius LCH1227의 생육에 따른 항균 활성을 측정한 결과, 생장정체기 초기에 최대 활성(300 AU/mL)을 나타내었고, 생장정체기 이후부터 활성이 점차 감소하였다. 항균물질의 활성은 pH 2-12 구간에서 비교적 안정하였으며, 열처리에 의해서도 활성이 소실되지 않아 열에 안정한 물질임을 알 수 있었다. L. salivarius LCH1227이 생산하는 항균물질은 단백질 분해효소 처리 후에 활성이 실활됨으로써 단백질성 물질인 것으로 추정되었으며, Tricine-SDS-PAGE 실험을 통하여 항균물질의 분자량은 약 5 kDa 정도임을 알 수 있었다.

Biofilm 생성이 토양흡착 나프탈렌 분해에 미치는 영향 (Effect of Biofilm Formation on Soil Sorbed Naphthalene Degradation)

  • 나프탈렌 분해균주인 Pseudomonas aeruginosa CZ6을 오염된 토양에서 분리하였으며 분리된 균주는 결정상태의 나프탈렌에 부착하고 그 주변에 extracellular polymeric substance를 분비하는 특성을 가졌다. LB, YM과 MSM 배지를 사용하여 배지의 종류에 따른 biofilm 생성량을 측정한 결과, LB 배지에서 biofilm이 가장 많이 생성되는 것으로 나타났다. 나프탈렌을 기질로 한 배양조건에서 균주는 기질의 농도 영향을 크게 받지 않고 0.10% 기질, 150 rpm 조건에서 최적 배양조건은 $30^{\circ}C$, pH 7로 나타났다. 두 가지 토양에서 배지의 종류에 따른 나프탈렌의 분해특성을 관찰한 결과 초기에는 MSM 배지에서 나프탈렌이 가장 많이 분해가 되었다. 그러나 생물이용성이 제한을 받는 조건에서 LB 배지의 나프탈렌이 가장 빨리 제거가 되었다. 이런 결과는 biofilm의 형성과 extracellular polymeric substance 생성이 토양에 흡착된 잔류 나프탈렌의 생물학적 이용성을 향상시키기 때문인 것으로 사료된다.
