韓國 濟州島産 새뱅이의 一新亞種(甲殼綱, 長尾目, 새뱅이 科) (A New Subspecies of Caridina denticulata (Crustacea, Decapoda, Atyidae) from Jeju Island, Korea)

  • 1973년 7월 15일 제주도 서귀포 천지연 폭포 상류에서 채집된 Caridina denticulata가 신아종임을 확인하고 이를 학명으로 Caridina denticulata keunbaei, 한국말로 제주새뱅이라 명명하여 기재한다. 이 신아종은 다른 유연자들에 비해 이마뿔이 매우 짧다.

Characterization of Isolated Lactobacillus spp. And classification by RAPD-PCR Analysis

  • The genetic relationship of six Lactobacillus strains and five laboratory isolated form fermented milk were determined by a random amplified polymorphic DNA(RAPD)-Polymease chan reaction (PCR) method. With 42 random primers. the result were analyzed by using the NTSYS-PC software for phenetic analysis. it revealed that all tested bacteria were divided into three distinct clusters. The clusters implied three subgenuses existed for the genus Lactobacillus, which were previously proposed by Rogosa and Sharpe. From the results, it was also possible to determine that the isolated Lactobacillus strains from fermented milk were grouped into L. acidophilus or L. bulgaricus. Interestingly. the three tested L. casei strains were divided into different clusters implying different subgenuses, i.e., Thermobacterium (L. casei YIT 9018) and Streptobacterium(L. casei CHR. Hansen and L.casei ATCC 4646). According to the distance matrix generated by an UPGMA program, the isolated bacteria LT01 and LT02 were determined as a subspecies of L. bulgaricus. The HK01, HK02 and HK03 were very closely related to either L. acidophilus or L. case YIT 9018. Hence, RAPD-PCR appears to be a very practical method to determine the genetic relationships of the Lactobacillus species and to characterize the unknown Lactobacillus strains at the subspecies level.

한국산 천남성속 두루미천남성절의 1 신아종(천남성과), 거문천남성 (Arisaema thunbergii supsp. geomundoense S.C.Ko (Araceae), a new subspecies from Korea)

  • 다도해해상국립공원에 속한 거문도에서 천남성속, 두루미천남성절, 무늬천남성아절에 해당되는 1 신아종인 거문천남성(Arisaema thunbergii subsp. geomundoense S.C. Ko)을 기재 및 도해하였다. 화수부속체 기부의 비후부의 색깔이 진한 자색 또는 적자색인 것이 기본종인 무늬천남성(A. thunbergii supsp. thunbergii)과 다르고, 비후부에 주름이 있는 것이 일본에 자생하고 있는 비후부가 평활한 A. thunbergii supsp. urashima와 다르다.

Method of DNA Extraction from Pinus rigida Wood Pretreated with Sandpaper

  • Species identification of wood provides important information for archaeology, restoration of cultural assets, preventing illegal logging, and more. Wood species are usually identified based on their anatomical features with the use of a microscope. However, this method may not be able to distinguish between anatomically similar species or subspecies. To overcome this problem, wood species need to be identified at the molecular level using DNA sequencing. However, unlike living plant cells, wood is difficult to pulverize using a mortar, and DNA extraction from dried wood is challenging. To solve these problems, we propose a pretreatment method in which wood is pulverized using 60-grit sandpaper and hydrated with water for 2 days. Using this method, we were able to stably amplify the rpoB gene from the extracted DNA of Pinus rigida. In addition, sequence analysis of the rpoB gene revealed six single nucleotide polymorphisms (SNPs), which classified the rpoB sequences in the genus Pinus into five groups. Our data indicate that although these SNPs were not suitable for species identification, they can potentially be used to determine the origin of different wood subspecies or individual samples of wood.

  • 본 연구는 한국산 녹슬은방아벌레아과(방아벌레과)의 분류학적 연구를 위하여 1984년 4월부터 1986년 9월까지 경기도 수원근교와 남해안지역(해남, 완도, 목포, 남해), 강원 충북지역(춘천, 충주)과 제주도에서 채집한 표본과, 서울대학교 농과대학 및 농촌진흥청 농업기술연구소 소장표본을 분류, 동정하였다. 아울러 문헌상의 기록을 종합, 정리한 결과 2속 10종 1아종이 밝혀졌으며, 그중 3종 (Agrypnus setigera, A. miyamotoi, A. hypponicola)은 한국 미기록종으로 기재한다.

Bacillus thuringiensis를 이용한 버섯 파리(Lycoriella sp.)의 생물적 방제에 관한 연구 (A stydy on The Biological Control of Sciarid Fly(Lycoriella sp.) Using Bacillus thuringiensis)

  • Thirteen subspecies of Bacillus thuringiensis including B. t. israelensis, B. t. morrisoni PG-14 and B.t. darmstadoemsos known to be toxic to dipteran insects were treated on the mushroom (Flammulina velutipes) compost to estimate the biological control effect of a sciarid fly, Lycoriella sp. According to the results, it was found that there were no significant effects of the tested strains of B, thuringiensis on the control of Lycoriella sp. For further confirmation, larval gut juice of Lycoriella sp. and trypsin were respectively treated into the parasporal crystal proteins of three subspecies of B. t. israelensis, B. t. morrisoni PG-14, and B. t. darmstadiensis. The proteins were separated by SDS-PAGE. According to the results, the major parasporal crystal proteins were respectively produced by B. t. morrisoni as the amount of 52 kd, B. y. israelensis as 37kd and B. t. darmstadiensis as 39kd, but the activity of these proteins could not be unfortunately confirmed in this study.

동물원(動物園)의 야생동물(野生動物) 분변(糞便)에서 분리(分離)한 살모넬라균의 생물형(生物型), 혈청형(血淸型) 및 약제내성(藥劑耐性) (Bioserotype and drug resistance of Salmonella spp isolated from feces in zoo animals)

  • Feces samples, obtained from zoo animals around Seoul, were examined for the isolation of Salmonella species, bioserotype and drug resistance for the prevention and treatment of salmonellosis, Salmonella spp were isolated 19(4.7%) from 408 samples of zoo animals. The subspecies in 19 Salmonella were all subspecies 1. The serological identification of Salmonella isolated were 31.6% in Sal typhimurium, 26.3% in Sal hadar, 21.1% in Sal muenchen, 15.8% in Sal enteritidis and 5.3% in Sal ayinde. The antibiotic resistance of Salmonella isolated were 13(68.4%) strains. The multiple resistant patterns of antibiotics in Salmonella were 2 drugs- and 3 drugs-resistance 30.8% respectively. The transferred rate of resistance to recipients(E coli ML 1410 $NA^r$) in Salmonella was 38.5%.

무플론 요네병 발생 사례 (Paratuberculosis in mouflon (ovis musimon) : A case report)

  • A 2-years-old female domesticated mouflon with a clinical history of chronic diarrhea and emaciation was submitted to NVRQS. Grossly, there were severe thickening of small intestine wall and enlargement of mesenteric lymph nodes. Microscopically, severe granulomatous inflammation was found in small and large intestine, mesenteric lymph nodes, spleen and liver. By Ziehl-Neelsen stain, innumerable acid-fast rod bacteria were found in the cytoplasm of epitheloid and Langhans type giant cells present in these organs. By PCR assay with primer pair specific for Mycobacterium avium subspecies paratuberculosis(IS 900) with small intestine sample, strong positive reaction was detected, although the organism was not isolated from this organ. Based on the results of histopathology and PCR, we concluded that the case was a typical paratuberculosis in mouflon. As far as we know, this is the first case report of paratuberculosis in mouflon Korea.

  • 1. This study was made on the flora of bryophytes on Mt. Kwanak, which is situated ($37^{\circ}$30' N. Lat. $127^{\circ}$O'E. Long) to the south of Seoul area. 2. The bryophytic flora of this mountain consists of the following elements. 1) Holarctic elem, Musci………16 species (25.0%), Hepaticae………4 species (28.6%). 2) North Pacific elem. M.………1 species (1.56%), H………O. 3) Cosmopolitan elem. M………10 species (15.65%), H………1 species (7.1%). 4) East-Asiatic elem. M………16 species (25%), H.………4 species (28.6%). 5) Tropical elem. M.………3 species (4.68%), H.………3 species (21.4%). 6) Japane-Korean elem. M.………17 species (26.56%), H.………2 species (14.3%). 7) Endemic elem. M.………1 species (1.56%), H.………O. 3. In the list of bryophytes of Mt. Kwanak, 62 species, 1 variety and 1 subspecies belonging to 47 genera for Musci and 12 species 2 subspecies belonging to 10 genera for Hepaticae were enumerated. Of these the following 5 species are new to Korean flora. 1) Macromitrium japonicum D.M. 2) Entodon arenosus Okam. 3) E. okamurae Broth. 4) Gollania ruginosa (Mitt.) Broth. 5) Temnona birmensis (St.) Hatt.
