Is $^{201}Tl-^{99m}Tc$-Subtraction Scan an Accurate Diagnostic Method to Detect Parathyroid Mass? ($^{201}Tl/^{99m}Tc$ 감영스캔으로 부갑상선종괴를 얼마나 찾을 수 있는가?)

  • We studied 65 patients with hypercalcemia who had heed performed $^{201}Tl/^{99m}Tc$ subtraction scan, 21 of them were confirmed as parathyroid tumor (19 adenoma, 2 carcinoma). The diagnostic sensitivity of $^{201}Tl/^{99m}Tc$ subfraction scan for detecting parathyroid mass was 90.5%, specificity was 97.6%, ultrasonography was 80.6%, 58.8%, respectively. The causes of two false negative cases were relatively small size ($1.5{\times}1{\times}0.8cm$) compared to remainig cases and poor thallium uptake due to cystic necrosis of parathyroid adenoma. The one false positive case was non-functioning thyroid nodule. $^{201}Tl/^{99m}Tc$ subtraction scan was simple, effective diagnostic tool and superior to ultrasonography for evaluating the parathyroid mass with high sensitivity and specificity.

Airborne Suspended Particulates Concentration and Cancer Risk Assessment of Polycyclic organic matter in Seoul (서울시 대기부유분진의 농도와 다환방향족 유기물질에 의한 발암 위해성)

  • Airborne suspended particulates were collected at Shinchon by a high volume cascade impactor from Sep. 1990 to Aug. 1991. Organic matter was extracted from particulates and fractionated by liquid-liquid extraction and thin layer chromatography. Substances in the PAHs and nitroarenes'subfraction of neutral fraction were determined by capillary gas chromatography. Based on unit risk estimates by multi-stage model of benzo[a]pyrene and the results of exposure estimates, cancer risk was assessed. The annual average concentration of total suspended particulates was 201.77g/$m^3$. The percentage of fine particulates was 57.40. The concentration of total suspended particulates showed seasonal variations and was high in winter and spring. The average concentration of extractable organic matter was 8.12g/$m^3$. In all, 21 PAHs were identified and quantified. The annual concentration of fluoranthene was 2.38ng/$m^3$, and that was the highest value of all PAHs. A carcinogenic compound, benzo[a]pyrene, was at a concentration of 1.84ng/$m^3$. All the 10 nitroarenes were also identified and quantified. The major nitroarene in the Shinchon area was 2,7-dinitrofluorene. The annual concentration of 1-nitropyrene was 1.56ng/$m^3$. Concentrations of PAHs and nitroarenes were high in winter and low in summer. The life time excess risk estimates of benzo[a]pyrene was calculated as 0.96 persons/a million population in this experiment. In the rank of relative potenties, carcinogenic effects of the other PAHs were calculated as 0.004-0.108 persons/a million population.

Inhibitory Mechanism of Blood Coagulation by the Anticoagulant Polysaccharide from Coriolus versicolor (구름버섯 기원 항응고성 다당류의 혈액응고 저해기작)

  • Inhibitory mechanism of the anticoagulant polysaccharide purified from the fruit body of Coriolus versicolor was investigated in this paper. The anticoagulant polysaccharide (CV-40-Va-1) was proposed to have functions of the inhibition of intrinsic pathway in the blood coagulation pathway together with the interuption of a human platelet aggregation induced by von Willebrand factor (vWF). CV-40-Va-I inhibited other factors of the coagulation cascade such as factor VIII, IX, and as well as thrombin. Especially, CV-40-Va-I inhibited the fibrin formation mediated by thromin, however the polysaccharide did not affect the fibrin formation directly but affected the anticoagulant activity through the activation of antithrombin III. The sulfation of the anticoagulant polysaccharide increased the anticoagulant activity, showing that the sulfate concentration of anticoagulant polysaccharide was important factor in the blood coagulation cascade. Low molecular weight subfraction (MW 1,000) obtained by partial hydrolysis of the CV-40-Va-1 generated potent antiplatelet activity, but showed decreased anticoagulant activity.

Inhibition of Klebsiella pneumoniae ATCC 13883 Cells by Hexane Extract of Halimeda discoidea (Decaisne) and the Identification of Its Potential Bioactive Compounds

  • The inhibitory effect of the Klebsiella pneumoniae ATCC 13883 strain caused by the hexane extract of Halimeda discoidea (Nor Afifah et al., 2010) was further evaluated by means of the microscopy view and its growth curves. The morphological changes of the K. pneumoniae ATCC 13883 cells were observed under the scanning electron microscope (SEM) and transmission electron microscope (TEM) after they were treated at minimum inhibitory concentration (MIC; 0.50 mg/ml) (Nor Afifah et al., 2010) for 12, 24, and 36 h. The results showed the severity of the morphological deteriorations experienced by the treated cells. The killing curve assay was performed for 48 h at three different extract concentrations (1/2 MIC, MIC, and 2 MIC). An increase in the extract concentration of up to 2 MIC value did significantly reduce the number of cells by approximately 1.9 $log_{10}$, as compared with the control. Identification of the potential compounds of the extract responsible for the antibacterial activity was carried out through the gas chromatography-mass spectrum (GC-MS) analysis of the active subfraction, and the compound E-15-heptadecenal was identified and suggested as the most potential antibacterial compound of this extract. The subsequent cellular degenerations showed by the data might well explain the inhibitory mechanisms of the suggested antibacterial compound. All of these inhibitory effects have further proven the presence of an antibacterial compound within H. discoidea that can inhibit the growth of K. pneumoniae ATCC 13883.

Antimicrobial activity of Gynura segetum's leaf extracts and its active fractions

  • $Gynura$ $segetum$ (Lour.) Merr. ($Compositae$) is a well recognized medicinal plant in Indonesia and Malaysia. It is believed to have an anticoagulant effect and is used in treating snake-bites, inflammations and other skin afflictions. This study was carried out to evaluate the antimicrobial effects of $Gynura$ $segetum$ leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. The antimicrobial activities of different solvent extracts of leaves of $Gynura$ $segetum$ were evaluated using the agar well-diffusion method. The Minimum Inhibitory Concentration (MIC) of the active subfractions was determined by the tube dilution method. Gas Chromatography-mass spectrometry (GC-MS) analysis was carried out to identify the chemical compositions of the active extracts. The ethyl acetate fraction and its subfraction E4 performed potent antimicrobial activities and fifteen known chemical constituents were identified by GCMS analysis as 4-vinylphenol, 1-tetradecene, phenol, 2,4-bis(1,1-dimethylethyl), 1-hexadecene, E-15-heptadecenal, hexadecanoic acid, 1,2-benzenedicarboxylic acid, dibutyl ester, 1-docosene, octadecanoic acid, 1-eicosene, cyclotetracosane, 1,2-benzenedicarboxylic acid, bis(2-ethylhexyl)ester, butanedioic acid, monomethyl ester, niacin and 4-hydroxy-benzoic acid. The results of this study suggested a connection between the antimicrobial activities and the chemical structures. The plant may be used as a potential source for antimicrobial agents.

Effect of Acidic Polysaccharide Components of Korean Ginseng on Lipolytic Action of Toxohormone-L and on Activity of Angiotensin Converting Enzyme (고려인삼중 다당체 성분이 암독소 호르몬-L의 지방분해 작용과 안지오텐신 변환효소의 활성에 미치는 영향)

  • This study was devised to observe in vitro, the inhibitory effects of acidic polysaccharide fractions from Korean red ginseng (KRG) and white ginseng (KWG) on the lipolytic action of loxohormone-L and on angiotensin converting enzyme (ACE, peptidyldipeptidase hydrolase, EC 3.4.15.1) . The crude acidic polysaccharides (CAP) extracted from main and lateral roots of KRG and KWG were separately purified through several procedures. The total inhibitory activities on the lipolytic action of toxohormone-L of CAP from main roots of KRG and KWG was higher than those of CAP from lateral roots of KRG and KWG, respectively, and that of CAP from main root of KRG was 3.1 times higher than that of CAP from main root of KWG. The specific activity of CAP from main root of KRG was measured as 5.40 units/mg, when one unit was defined as the amount giving 50% inhibition on toxohormone-L induced lipolysls. A subfraction named PG4 3 obtained by replanted chromatography on DEAE-TOYOPEARL 650M gave the specific activity of 24.4 units/mg. On the other hand, it was found that the total inhibitory activity on ACE of CAP from lateral root of KRG was the highest among the 4 kinds of CAP, but the specific activity of CAP from lateral root of KWG was the highest.

Addictive Effects of Ecklonia cava Methanol and its Solvent-soluble Extract Against Antibiotic-resistant Cutibacterium acnes Causing Acne Vulgaris (항생제 내성 Cutibacterium acnes에 대한 감태(Ecklonia cava) 추출물의 항균 시너지 효과)

  • Acne vulgaris is often associated with acne-related bacteria such as Cutibacterium acnes. In this study, we investigated the antibacterial effects of the methanol extract of the algae Ecklonia cava and its solvent-soluble extract against C. acnes. Among five solvent fractions, the ethyl acetate (EtOAc) fraction exhibited the strongest antibacterial activity against C. acnes. Furthermore, the EtOAc-soluble extract exhibited the highest total phenolic contents among the five solvent fractions tested. The EtOAc subfraction 07 (Fr. 07) extract showed the highest antibacterial effect against C. acnes and isolated C. acnes along with minimum inhibitory concentration (MIC) ranging from 32 to 64 ㎍/mL. Additionally, MICs of antibiotics against antibiotic-resistant C. acnes strains were substantially reduced when antibiotics were combined with Fr. 07, suggesting that Fr. 07 restore the antibacterial activity of the antibiotics. The fractional inhibitory concentration indices clearly revealed an additive synergistic effect of Fr. 07 with antibiotics. The results of the present study suggest a potential role for E. cava in the control of infections related to acne vulgaris.

Effects of Natural Product on the Inhibition of $5{\alpha}-Reductase$ Type 2 for the Development of Chemopreventive Agents in LNCaP Cells

  • The enzyme steroid $5{\alpha}-reductase$ is responsible for the conversion of testosterone into the most potent androgen dihydrotestosterone (DHT). In man, this steroid acts on a variety of androgen-responsive target tissues to mediate such diverse endocrine processes as male sexual differentiation in the fetus and prostatic growth in men. Androgen levels in the prostate may influence carcinogenesis in this organ. The use of a $5{\alpha}-reductase$ inhibitor, finasteride, in the chemoprevention of prostate cancer is being evaluated in a clinical trial and have been used successfully for treatment of benign prostatic hyperplasia. Therefore, for the discovery of $5{\alpha}-reductase$ type 2 inhibitors, we have evaluated the inhibitory effects of solvent fractionated extracts of natural products on $5{\alpha}-reductase$ type 2 activity. We have tested approximately 80 kinds of natural products after partition into n-hexane, ethyl acetate and aqueous layers from 100% methanol extracts of plants. The ethyl acetate fractions of Perilla sikokiana $(seed,\;IC_{50}\;:\;6.2\;ug/ml)$, Sophora flavescens $(root,\;IC_{50}\;:\;8.9\;ug/ml)$, and Angelica tenuissima $(root,\;IC_{50}\;:\;11.7\;ug/ml)$ revealed inhibitory effects on $5{\alpha}-reductase$ 2 activity in LNCaP cells. The effective ethyl acetate fractions of Perilla sikokiana, Sophora flavescens, Hydnocarpus anthelmintica, and Angelica tenuissima were subfractionated by column chromatography and tested. The subfractions $F4\;(IC_{50}\;:\;1.1\;ug/ml),\;F5\;(IC_{50}\;:\;2.0\;ug/ml),\;and\;F6\;(IC_{50}\;:\;5.8\;ug/ml)$ of the ethyl acetate fraction of Perilla sikokiana and the subfraction $F8\;(IC_{50}\;:\;5.3\;ug/ml)$ of the ethyl acetate fraction of Sophora flavescens displayed greater inhibition of $5{\alpha}-reductase$ type 2 than did finasteride in LNCaP cells. These active fractions are under the process of further sequential fractionation to find the effective pure compounds against $5{\alpha}-reductase$ 2 activity.

Cosmetic Activities of Nyasol from the Rhizomes of Anemarrhena asphodeloide (지모의 뿌리줄기로부터 분리된 nyasol의 미용효과)

  • The rhizomes of Anemarrhena asphodeloide was extracted with 100% ethanol and concentrated subfractions were separated with medium pressure liquid chromatography-based activity profiling. One compound was isolated from the subfraction 10 through the repeated preparative high performance liquid chromatography (prep-HPLC). According to physico-chemical and spectroscopic data including NMR and MS, the chemical structures of the compound was determined as nyasol (1). Nyasol was exhibited potent inhibitory activity for NO ($IC_{50}:12.5{\mu}g/mL$), tyrosinase ($IC_{50}:12.5{\mu}g/mL$), melanin contents ($IC_{50}:12.5{\mu}g/mL$), thymus and activation-regulated chemokine (TARC) production ($IC_{50}:6.25{\mu}g/mL$). As a result, nyasol has an excellent inflammation-dependent anti-whitening and TARC production activity. It could be used to a large range of functional cosmetics.

Antioxidant Activity of Grape Seed Ethanol Extract According to Serial Solvent Fractionation (포도종실 에탄올 추출물의 순차 용매 분획에 따른 항산화 활성)

  • Ethyl acetate and butanol fractions among the serial solvent fractions of grape seed ethanol extract contained the catechin at the levels of 35.7 mg/g and 20.2 mg/g, respectively However, the POV increasing patterns of two linoleic acid samples containing each solvent fraction were so similar that the difference in antioxidant activity by the catechin content of each solvent fraction could not be found. Each solvent fraction was fractionated on C18 cartridges into three subfractions which were mono-, dimers fraction (FI), oligomers fraction (FII) and polymers fraction (FIII) to examine the effect by the difference in degree of Polymerization of proanthocyanidin. The catechin contents of ethyl acetate subfractions (E-F) were in the order of E-FI (26.0 mg/g) > E-FII (18.6 mg/g) > E-FIII (13.7 mg/g) but the three subfractions showed nearly similar antioxidant activities, by the POV measurement at 1,000 ppm concentration. Also the catechin contents of butanol subfractions (B-F) were in the order of B-FI (35.3 mg/g) > B-FII (30.8 mg/g) > B-FIII (22.7 mg/g) but similar antioxidant activities were observed in all subfractions. In this study, similar antioxidant activities of each solvent subfraction in spite of different catechin contents inform that the degree of polymerization of proanthocyanidin as well as the total catechin content should be considered in quality control of grape seed extract produced for natural antioxidant.
