거베라 기내 엽병조직으로부터 Callus 유도 및 식물체 재분화 (Callus induction and plant regeneration from in vitro cultured petiole of 3 Gerbera cultivars.)

  • 본 연구는 거베라의 신품종을 육성하여 식물체 재분화 조건을 찾기 위하여 경남농업기술원 화훼육종연구소에서 육성한 화이트데이, 송송이 그리고 러브송 등 3품종을 이용하여 기내배양묘의 엽병으로부터 callus 유도조건 및 식물체 재분화에 미치는 몇 가지 요인을 구명하기 위해 실시되었다. Callus의 유도에 있어서는 3품종 중 '화이트데이'가 다른 품종보다 callus형성이 양호하였다. Callus의 유도를 위한 가장 효과적인 식물생장조절물질의 조건은 MS 기본배지에 NAA 0.1 mg/L+TDZ 0.5 mg/L의 조건에서 양호하였다. 식물체 재분화를 위한 가장 효과적인 식물생장조절물질의 조건은 MS 기본배지에 IAA 1.0 mg/L+BA 1.0 mg/L+Zeatin 0.1 mg/L의 혼용조건에서 양호한 경향이었다. 배양 식물체 재분화를 위한 엽병의 적정 계대배양 기간은 계대배양 후 32일이 경과된 엽병에서 양호한 경향이었다. 또한 식물체 재분화를 위한 적정 MS 배지의 농도는 1/2MS에서 식물체의 분화가 양호하였다.

Characteristics of Extreme Sports Participants' Lifestyles and Sportswear Benefits Sought -A Comparison between Participants of Extreme and Traditional Sports-

  • Extreme sports refers to sports activities with a high level of inherent danger, such as extreme skiing, snow-boarding, mountain biking, motocross bicycling, aggressive in-line skating, wake-boarding, and paragliding. This study investigates the characteristics of extreme sports participants as group identities through an examination of the differences between extreme sports participants and traditional sports groups in lifestyle and sportswear benefits sought. We composed a total 108-item lifestyle profiles characterized by six lifestyle dimensions (dress, diet, home, recreation, consumption, and sense of values); in addition, we developed 32 items for sportswear benefits sought. The surveys targeted 422 sports participants living in Korea (216 men and 206 women, aged 15-23): 119 extreme sports participants and 303 traditional sports participants. All items were evaluated on a 5-point Likert scale, and SPSS 17.0. was used for data analysis. Exploratory factor analysis was conducted to confirm the substructure of each lifestyle dimension and sportswear benefits sought. The results of the factor analysis on each lifestyle dimension are as follows: 5 factors in dress dimension, 4 in diet, 3 in home, 6 in recreation, 5 in consumption, and 6 in sense of values. Six factors of sportswear benefit sought were identified. A t-test revealed that there were significant differences between the two groups in lifestyle dimensions and sportswear benefits sought. This study reveals that the lifestyle characteristics of extreme sports participants are more likely than traditional sports group to be more self-expressive, more distinctive, more fashion-oriented, more conspicuous, and more brand-oriented, while preferring more active sports, more dynamic leisure, and a more sensible life. Another finding is that extreme sports participants seek the distinctive, conspicuous, and latest sportswear benefits.

멜론(Cucumis melo L.) 유묘 절편으로부터 고빈도의 체세포배발생과 식물체 재분화 (High Frequency Somatic Embryogenesis and Plant Regeneration in Seedling Explant Cultures of Melon (Cucumis melo L.))

  • 멜론의 배발생 캘러스의 유도와 식물체 재분화 시스템을 확립하기 위해서 자엽과 배축의 절편을 여러 농도의 2,4-D 와 BA가 첨가된 MS 기본배지에 치상하여 관찰한 결과, 초기배양에서는 4 mg/L 2,4-D와 1 mg/L BA 그리고 2 mg/L 2,4-D+O.1 mg/L BA가 첨가된 배지에서 연한 노란색의 배발생 캘러스가 형성되었고, 위의 조건에서 형성된 캘러스로부터 배발생 캘러스는 0.5와 1 mg/L 2,4-D가 첨가된 동일배지에서 약 75%가 형성되었다. 그리고 생장조절제를 첨가하지 않은 기본배지에서 대부분의 체세포배는 식물체로 재분화되었으며, 두개의 자엽을 갖춘 정상적인 체세포배보다 비정상적인 나팔형의 자엽을 가진 것이 3배 높은 빈도로 나타났다. 이러한 체세포배들은 MS 기본배지에서 식물체로 발아되었으며 생육상에서 생장하여 견실한 열매를 맺었다.

Thidiazuron 처리에 의한 히어리나무의 기내번식 (Micorpropagation of Corylopsis coreana by Thidiazuron Treatment)

  • This study was conducted to evaluate the effect of thidazuron(TDZ) on shoot proliferation and growth from axillary buds of 20-years-old Corylopsis coreana. Shoots proliferation was effectively achieved on WPM(Woody Plant Medium) supplemented with 0.03∼0.1mg/L TDZ. The highest shoot number(6.5$\pm$0.7) was obtained on 0.1mg/L TDZ treatment. On the TDZ medium shoots formed as clusters less than 1cm in height and therefore needed to subculture on GA$_{3}$ containing medium to induce elongation. In consecutive cultures, phenolic compounds were excreted at the proximal part of the explants and inhibited growth of the explants. Growth inhibition by the compounds was overcome using liquid and paper bridge culture system. About 60% of the elongated shoots rooted on half- strength MS medium containing IBA. Generally, IBA was mire effective on in vitro rooting than NAA with optimal range of 0.5mg/L to 1.0mg/L. Rooted plantlets were transferred in an artificial soil(vermculite) and acclimatized in high humidity greenhouse condition. Survival rate differed greatly depending on rooting types of the explants. Two types of rooting were observed. The first type was direct rooting from the explants. The second type was callus formation followed by rooting from the callus. The explants showing the 1st type rooting survived can be multiplicated in vitro by TDZ treatment followed by elongation with GA$_{3}$ and rooting with IBA.

어류 병원성 세균 Edwardsiella tarda의 형질전환 및 재조합 ghost 세균에서의 E-lysis 유전자의 전사 발현 특징 (Transformation of Edwardsiella tarda and Transcriptional Characteristics of E-lysis Gene in Recombinant Bacterial Ghosts)

  • 어류 에드워드감염증에 대한 예방 재조합 ghost 백신을 개발하기 위한 연구의 일환으로 어류 병원성 세균인 Edwardsiella trada를 대상으로 플라스미드 형질전환을 실시하고 형질전환 안정성을 평가하였으며, 형질 도입된 재조합 ghost 세균의 E-lysis 유전자 발현을 분석하였다. E. tarda를 대상으로 한 ghost 유도는 대장균에 비해 상대적으로 장시간의 반응 시간이 요구되며 lysis의 개시가 지연된 점을 고려 시 발현된 E protein의 용해 능력 또는 E-gene의 전사발현 양이 E. tarda에서 다소 약화되는 것으로 나타났다. 그러나 대장균에 비해 ghost 유도 속도가 다소 낮음에도 불구하고 반응이 완성되었을 시점에서의 E. tarda의 ghost 효율은 대장균과 전혀 차이가 없이 99.99% 이상의 유도효율을 나타내었다.

Visual Representation in Glam Style under the Influence of Andy Warhol

  • The purpose of this study is to illuminate the relations between Pop Art and pop music, which formed a serious coupling loop in an identical cultural background. Also this study intends to set up a position of visualized sexual identity as it Points out the conceptional characteristics of glam, a subculture outside mainstream, which includes the matters of sexual minorities such as homosexuality and bisexuality under the influence of the aesthetic and philosophical composition by Andy Warhol. In conjunction herewith, we explore the visual representation of glam style focusing on the influence of Warhol. The classification and explanation about the visual representation of glam style under the influence of Warhol are practised by distinguishing as denotative representation and connotative representation. For the denotative representation shown in the glum style, first, the discordant images were put together by bricolage, then adapted into the new dramatic symbol of youth. Second, through a visually androgynous style a subversion of sex for symbols of sexuality and gender was represented. Third, the factitiveness as a weird display and fallacy is shown from boisterous make-up and unisex sかling in theatricality and put-ons, featuring artificiality, assemblage and unnaturality. And the connaotative representation shown in glum inglam style, first, glam style implies its experimental nature which attempts to break down boundaries between masculinity and femininity, homosexuality and heterosex-uality. Second, the bricolage in sequin and other discordant elements have connotative meanings as sensuality and excessiveness. Third, mixing various style as sexual play shows ironic visual images, in accordance with Superior Theory and Discord Theory.

프리프합(Prep-hop) 패션의 디자인 특성 (The Design Characteristics of Prep-hop Fashion)

  • This research aims to analyze the prep-hop fashion, which is an original look created through the convergence of the hip-hop and preppy look, and identify the design characteristics of the prep-hop fashion. The style mixes the fashion of what was traditionally a lower-class look with the upper class. The design characteristics are as follows: First, the prep-hop fashion expresses slim, casual and sophisticated look with items and silhouettes. The items are categorized into jackets, shirts and pants. The prep-hop jackets provides a casual look to its wearer unlike the jackets of preppy or conservative looks. Slim suits show different meanings and features depending on the changes in the time and culture despite the continued use of the items. Second, the colors of the prep-hop fashion include overall basic colors used in the preppy look and hip-hop fashion. The aesthetic values of the colors express pleasant color images different from the conservative and resistant images expressed in the preppy and hip-hop style before the convergence, respectively. Third, the materials and patterns are expressed in a unique way to fulfill the emotional needs that are considered vital to contemporary fashion. The materials in the prep-hop fashion extend the ranges of usage by using the materials with unexpectedness broken out of general range of usage. While the use of the patterns is one of the methods to demonstrate that the prep-hop fashion is created by the convergence between hip-hop fashion and preppy look, the meaning concludes pleasantness and casualness different from the past. Fourth, the prep-hop fashion is mainly composed of hip-hop fashion accessories, which express the authenticity of the hip-hop fashion to protect the non-mainstream characteristics. The design characteristics of the prep-hop fashion pursue the individuality of the contemporary fashion, which lacks originality due to commercialization. The development of the prep-hop fashion, as a new form that combined contrasting genres and trends, is expected to develop more publicly.

Binary Vector System을 이용한 당근 (Daucus carota) 세포의 형질전환 (Transformation of Carrot (Daucus carota) Cells Using Binary Vector System)

  • 고등식물의 형질전환용 유전자운반체로써 가장 많이 사용되고 있는 Ti-plasmid를 이용해서 당근세포를 형질 전환시키기 위한 연구의 일환으로 Agrobacterium spp를 helper로 이용하여 NPT II gene를 함유하고 있는 binary vector GA472를 당근세포에 삽입시켜 kanamycin에 대해 저항성을 나타내는 세포주를 선발하고자 본 연구를 수행하였다. 국내 토양에서 선발한 A.tumefaciens 2종과 disarmes된 PC2760 그리고 hypervirulent균주인 A281에 tri-parental mating 방법에 의해서 binary vector인 pGA472을 도입하여 transconjungants인 A. tumefaciens c-23-1/pGA472,K29-1/pGA472, PC2760/PGA472 그리고 A281/pGA472를 획득하였다. Transconjungants는 plasmid의 분리, 정제방법에 의해서 추출한 후 0.7% agarose gel 상에서 관찰해 본 결과 4균주 공히 NTPII gene이 삽입된 pGA472와 Ti-plasmid를 함유하고 있는 것을 확인 하였다. 확인된 conjugant와 당근정상조직을 동시배양방법에 의해서 형질전환을 유도한 후 정상조직은 전혀 생존이 되지않은 kanamycin에 대해서 저항을 나타내는 callus를 선발할 수 있었다.

An easy and efficient protocol in the production of pflp transgenic banana against Fusarium wilt

  • This study describes an efficient protocol for Agrobacterium tumefaciens-mediated transformation of two subgroups of genotype AAA bananas (Musa acuminata cv. Pei Chiao and Musa acuminata cv. Gros Michel). Instead of using suspension cells, cauliflower-like bud clumps, also known as multiple bud clumps (MBC), were induced from sucker buds on MS medium containing $N^6$-Benzylaminopurine (BA), Thidiazuron (TDZ), and Paclobutrazol (PP333). Bud slices were co-cultivated with A. tumefaciens C58C1 or EHA105 that carry a plasmid containing Arabidopsis root-type ferredoxin gene (Atfd3) and a plant ferredoxin-like protein (pflp) gene, respectively. These two strains showed differences in transformation efficiency. The EHA105 strain was more sensitive in Pei Chiao, 51.3% bud slices were pflp-transformed, and 12.6% slices were Atfd3-transformed. Gros Michel was susceptible to C58C1 and the transformation efficiency is 4.4% for pflp and 13.1% for Atfd3. Additionally, gene integration of the putative pflp was confirmed by Southern blot. Resulting from the pathogen inoculation assay, we found that the pflp transgenic banana exhibited resistance to Fusarium oxysporum f. sp. cubense tropical race 4. This protocol is highly advantageous to banana cultivars that have difficulties in setting up suspension cultures for the purpose of quality improvement through genetic transformation. In addition, this protocol would save at least 6 months in obtaining explants for transformation and reduce labor for weekly subculture in embryogenic cell suspension culture systems.

Metabolic engineering of Lilium ${\times}$ formolongi using multiple genes of the carotenoid biosynthesis pathway

  • Lilium ${\times}$ formolongi was genetically engineered by Agrobacterium-mediated transformation with the plasmid pCrtZW-N8idi-crtEBIY, which contains seven enzyme genes under the regulation of the CaMV 35S promoter. In the transformants, ketocarotenoids were detected in both calli and leaves, which showed a strong orange color. In transgenic calli, the total amount of carotenoids [133.3 ${\mu}g/g$ fresh weight (FW)] was 26.1-fold higher than in wild-type calli. The chlorophyll content and photosynthetic efficiency in transgenic orange plantlets were significantly lowered; however, after several months of subculture, they had turned into plantlets with green leaves that showed significant increases in chlorophyll and photosynthetic efficiency. The total carotenoid contents in leaves of transgenic orange and green plantlets were quantified at 102.9 and 135.2 ${\mu}g/g$ FW, respectively, corresponding to 5.6- and 7.4-fold increases over the levels in the wild-type. Ketocarotenoids such as echinenone, canthaxanthin, 3'-hydroxyechinenone, 3-hydroxyechinenone, and astaxanthin were detected in both transgenic calli and orange leaves. A significant change in the type and composition of ketocarotenoids was observed during the transition from orange transgenic plantlets to green plantlets. Although 3'-hydroxyechinenone, 3-hydroxyechinenone, astaxanthin, and adonirubin were absent, and echinenone and canthaxanthin were present at lower levels, interestingly, the upregulation of carotenoid biosynthesis led to an increase in the total carotenoid concentration (+31.4%) in leaves of the transgenic green plantlets.
