Spring Shoot Damage and Cold Hardiness of Grape in Different Varieties and Phenological Stages (봄철 포도 신초 저온 피해 양상과 품종별 전엽기 내한성 비교)

  • Grapes are one of the most important fruit trees both domestically and globally. Recently, changes in plant phenology and frequent low temperatures due to climate change are increasing the possibility of damage to grape shoots in spring, which is a serious threat to grape production. This study was conducted to investigated the severity of shoots damage and the change of free sugar content in the plant organs by phenological stage, especially, from germination to leafing period. Furthermore, in order to compare the cold hardiness among grape varieties including 'Campbell Early', 'Kyoho' and 'Shine Muscat' widely grown in Korea, lethal temperature (LT50) and free sugar content by grape variety were analyzed. Shoot damage by low temperatures continued to increase as the phenological stage progressed gradually, from the bud burst to the fourth leafing stage. On the other hand, the free sugar content of each organ except leaves continued to decrease, showing pattern to similar to cold hardiness. This indicates a close relationship between free sugar content and cold hardiness. In terms of cold hardiness comparison among grape varieties, 'Shine Muscat' showed the highest cold resistance in the leafing stage with the lowest LT50 and the highest total free sugar content. Next was 'Kyoho' and 'Campbell Early'. There are clear differences in cold hardiness depending on the variety. However, it is not the same at all growth stage. It may change according to phenological stage and influenced by free sugar content at that time.

  • Ebola virus disease is a lethal viral hemorrhagic fever that has been boiling in sub-Saharan Africa since 1970s. Last year, The Ebola virus epidemic that has spread not only mainly in West Africa, but also in locals such as USA, Europe and the Antipodes via infected travelers, was brought up. Human-to-human transmission of Ebola virus disease is known only through direct contact with the blood, secretions, tissues or other bodily fluids, including saliva. Although there has not been reported infection cases in the dental healthcare settings, the fact that the infection of the Ebola virus may be made from human secretions such as saliva suggests that there is a high risk of infection for the Ebola virus of dental healthcare workers. Therefore, it is important dental healthcare workers to identify infection-suspected patients through the oral findings for infection prevention. This article will review the oral signs and symptoms of Ebola virus disease and discuss the pathogenesis, treatment and prevention. Furthermore, Infection control guidelines for oral healthcare workers are also proposed.

Stanniocalcin 2 enhances mesenchymal stem cell survival by suppressing oxidative stress

  • To overcome the disadvantages of stem cell-based cell therapy like low cell survival at the disease site, we used stanniocalcin 2 (STC2), a family of secreted glycoprotein hormones that function to inhibit apoptosis and oxidative damage and to induce proliferation. STC2 gene was transfected into two kinds of stem cells to prolong cell survival and protect the cells from the damage by oxidative stress. The stem cells expressing STC2 exhibited increased cell viability and improved cell survival as well as elevated expression of the pluripotency and self-renewal markers (Oct4 and Nanog) under sub-lethal oxidative conditions. Up-regulation of CDK2 and CDK4 and down-regulation of cell cycle inhibitors p16 and p21 were observed after the delivery of STC2. Furthermore, STC2 transduction activated pAKT and pERK 1/2 signal pathways. Taken together, the STC2 can be used to enhance cell survival and maintain long-term stemness in therapeutic use of stem cells.

  • Sebaceous carcinomas of the eyelids are uncommon but lethal tumors. Lesions are usually seen in the elderly, predominantly women. The meibomian glands of the tarsus are the most frequent site of origin. Less commonly, the tumor arises in other sebaceous glands, e.g., the gland of Zeis, eyebrow or caruncle. Regardless of the location, sebaceous malignancies must be considered aggressive neoplasms with a potential for regional and distant metastasis. Diagnosis may be difficult, given the low incidence and inconsistencies in histopathologic classification. Treatment requires wide surgical excision with removal of involved regional lymph nodes and exenteration is reserved for those patients with orbital involvement or diffuse intraepithelial neoplasia. Opinions are divided regarding the use of postoperative irradiation or chemotherapy. Recently we experienced 46-year-old male patient with a 12-month history of painless, firm nodule and conjunctivitis due to sebaceous carcinoma of the left upper eyelid. After surgery, serial sections of the entire conjunctiva and eyelids showed a positive cut margin in medial and lateral border. We report herein this patient that supports irradiation as the postoperative treatment of these tumors in selected patients with a review of literatures.

  • This experiment was carried out to obtain basic informations on the development in sterile-male technique of rice stem borer. The species considered is Chile suppressalis Walker. Different developmental stages of the insect namely; larval, pupae and adult were irradiated with $Co^{60}$ gamma ray. The results obtained were as follows: 1) A 1:1 sex ratio in the pupal stage was observed. 2) In the pupal stage sub-lethal dosages$(LD_{50})$ were 26 KR and 26.7 KR for female and male, respectively. 3) The total number of egg masses per female increased with the increasing gamma ray dosages and average number of eggs produced per female decreased in the cross between normal female and irradiated male. 4) The sterile dosage $(SD_{90})$ of the gamma ray irradiation showed 23 KR in the male. 5) The pupal stage is most convenient developmental stage for irradiation.

Utilization of Google Earth for Distribution Mapping of Cholangiocarcinoma: a Case Study in Satuek District, Buriram, Thailand

  • Background: Cholangiocarcinoma (CCA) is a serious public health problem in the Northeast of Thailand. CCA is considered to be an incurable and rapidly lethal disease. Knowledge of the distribution of CCA patients is necessary for management strategies. Objectives: This study aimed to utilize the Geographic Information System and Google $Earth^{TM}$ for distribution mapping of cholangiocarcinoma in Satuek District, Buriram, Thailand, during a 5-year period (2008-2012). Materials and Methods: In this retrospective study data were collected and reviewed from the OPD cards, definitive cases of CCA were patients who were treated in Satuek hospital and were diagnosed with CCA or ICD-10 code C22.1. CCA cases were used to analyze and calculate with ArcGIS 9.2, all of data were imported into Google Earth using the online web page www.earthpoint.us. Data were displayed at village points. Results: A total of 53 cases were diagnosed and identified as CCA. The incidence was 53.57 per 100,000 population (65.5 for males and 30.8 for females) and the majority of CCA cases were in stages IV and IIA. The average age was 67 years old. The highest attack rate was observed in Thung Wang sub-district (161.4 per 100,000 population). The map display at village points for CCA patients based on Google Earth gave a clear visual deistribution. Conclusions: CCA is still a major problem in Satuek district, Buriram province of Thailand. The Google Earth production process is very simple and easy to learn. It is suitable for the use in further development of CCA management strategies.

Study of Single-dose Toxicity of Guseonwangdo-go Glucose Intramuscular Injection in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze single-dose intramuscular toxicity of Guseonwangdo-go glucose pharmacopuncture. Methods: Eighty six-week-old Sprague-Dawley rats were divided into two large groups of forty rats; Guseonwangdo-go glucose 5% and Guseonwangdo-go glucose 20% groups. Each group was sub-divided into four smaller groups of five males and five females, with the following dosages of pharmacopuncture being administered by intramuscular (IM) injection in each group: group 1 (G1, control group): 1.0 mL of normal saline solution, group 2 (G2, low-dose group): 0.1 mL, group 3 (G3, mid-dose group): 0.5 mL, and group 4 (G4, high-dose group): 1.0 mL. Results: No mortalities or clinical signs were observed in any group. Also, no significant changes in body weights or in hematological/biochemical analyses were observed between the control and the experimental groups during necropsy or histopathology. Conclusion: The above findings suggest that the lethal dose of Guseonwangdo-go glucose 5% and 20% pharmacopuncture administered via IM injection is more than 1.0 mL per animal in both male and female rats. Further studies on the repeated-dose toxicity of Guseonwangdo-go glucose should be conducted to yield more concrete data.

  • Accumulation of cadmium (Cd) was investigated in organs (gill, liver, kidney, intestine) of olive flounder (Paralichthys olivaceus) exposed to sub-lethal concentration (5, 10, 50, 100 $\mu$g/L) of Cd for 30 days. Cd exposure resulted in an increased Cd accumulation in liver of flounder for exposure periods and Cd concentration, and Cd accumulation increased linearly with the exposure time. At 20 days of Cd exposure. the order of Cd accumulation in organs was Bill>intestine>liver>kidney and after 30 days of exposure, Cd accumulation in organs was intestine>liver>gill>idney. An inverse relationship was observed between the accumulation factor (AS) and the exposure level, but AE shows an increase with exposure time.

Hygienic Studies on Inactive Substances of Sterigmatocystin by Artificial Gastric Juice (인공(人工) 위액(胃液)에 의한 Sterigmatocystin의 실활물(失活物)에 관한 위생화학적(衛生化學的) 연구(硏究))

  • Sterigmatocystin bears a close structural relationship to aflatoxin $B_1$ and is a carcinogenic compound that has been shown to affect various species of experimental animals. Reaction and toxicity of sterigmatocystin in the artificial gastric juice were investigated. Sterigmatocystin was degraded in artificial gastric juice and extracted by the method of A.O.A.C. After cleaned up by silica gel column chromatography, this substance was detected and characterized by thin layer chromatography, UV, IR and mass spectra. It showed $R{\mathcal{f}}$ 0.4 and brick-red color by TLC. Especially, in the mass spectrum of it, fragment peak at m/e 327 was due to the loss of the $-CH_3$ and $-H_2O$, fragment peak at m/e 341 was due to the loss of the $H_2O$ and $-H^+$, and fragment peak at m/e 239 was due to the loss of the 2-chloro-tetrahydrofuran and methyl group from the parent molecule. Therefore, a degraded substance of sterigmatocystin reacted in artificial gastric juice (Sub. K) was estimated with additional formation of hydrochloric acid. In four-day-old chicken embryos, the mean lethal dose $(LD_{50})$ was $140\;{\mu}g/egg$, and 90 to 100% of the embryos were killed with 1 mg/egg. This $LD_{50}$ $140\;{\mu}g/egg$ compared with an $LD_{50}$ $14.69\;{\mu}g/egg$ for sterigmatocystin (acute toxicity) showed the substance to be much less toxic than sterigmatocystin.

Mosquito Larvicidal Activity of Synechocystis PCC6803 Transformed with the cry11Aa gene to Culex tritaeniorhynchus and Anopheles sinensis (Cry11Aa 유전자로 형질전환된 Synechocystis PCC6803의 작은빨간집모기와 중국얼룩날개모기 유충에 대한 살충효과)

  • Bacillus thuringiensis produces crystal proteins toxic to medically and agriculturally important pests during sporulation. To improve the activity of insecticidal crystal protein in applying to mosquito larval control, an expression vector, pSyn4D harboring the mosquitocidal cry11Aa gene under control of psbA promoter of Amaranthus hybridus was constructed. This expression vector was transformed into Synechocystis PCC6803 and a transformant, Tr2C was selected with kanamycin. The mosquitocidal cry11Aa gene was stably integrated Into genomic DNA of Tr2C in PCR detection using cry11Aa-specific primers. The transformant expressed 72-kDa Cry11Aa protein and median lethal time (LT$\sub$50/) was approximately 2.1 days for Culex tritaeniorhynchus larvae and 0.7 day for Anopheles sinensis larvae, respectively. These results suggest this transformant can be used for mosquito larval control as a biological control agent.
