A Study on Approximate Analysis of Steel Deck Bridges with Guss Asphalt Using Influence Line (영향선을 이용한 강상판 교량의 구스 아스팔트 포장에 대한 근사해석 연구)

  • In this study, steel deck bridges are chosen as analytic model to show the structural behaviors generated by high temperature of pavement and to formulate the simplified approximate analysis of thermal effects. In general, the thermal effect is changed by the material property of pavements and environmental temperature as well as shape, size and boundary conditions of bridge. Specially, this effect is the representative initial stress problem dependent on time. The thermal effect, however, does not depend on time and thermal effect is regarded as initial load in this study. After these thermal loading is modelled as moving loads, influence lines of reactions of shoes are calculated and the successive pavement steps with arbitrary segments are determined to minimize the thermal effect of shoes by influence line.

Antioxidant Potential of Cinnamomum cassia Ethanolic Extract: Identification Of Compounds (계피 에탄올 추출물의 유효성분 분석 및 항산화 효능 평가)

  • Objectives : Natural products containing bioactive compounds with high antioxidant activity are potentially important sources that can contribute to the improvement of various diseases. Therefore, the aim of this study was to investigate phenolic compounds of Cinnamomum cassia (C. cassia) ethanolic extract (CCEE). And then we evaluated the antioxidant effect. Methods : We used liquid chromatography with tandem mass spectrometry (LC-MS/MS) to identify the compounds in CCEE. LC-MS/MS was performed in positive ion mode using Shimadzu, Nexera HPLC system and IDA TOF mass system. Solvent A was distilled water and solvent B was acetonitrile as mobile phase. The analysis was performed at a flow rate of 0.5 ml/min, column temperature of 35 ℃ and wavelength of 284 nm. The antioxidant effect of CCEE was analyzed using DPPH (2,2-diphenyl-2-picrylhydrazyl free radical) and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)). In addition, total phenolics and total flavonoids contents were measured to determine antioxidant effects. Results : Analysis using LC-MS/MS identified four compounds: Coumarin, Trans-cinnamaldehyde, Trans-cinnamic acid, and 2-Methoxycinnamaldehyde. Free radicals decreased in a concentration-dependent manner starting from 10 ㎍/ml of CCEE, and decreased to a level similar to Ascorbic acid (AA) from a concentration of 60 ㎍/ml onwards. Conclusions : Based on the findings, CCEE exhibits strong antioxidant activity as evidenced by the presence of Coumarin, Trans-cinnamaldehyde, Trans-cinnamic acid, and 2-Methoxycinnamaldehyde. Consequently, this study suggests that CCEE can serve as an important source of natural antioxidants and can be efficiently used in the management of oxidative stress diseases.

The Induction of ROS-dependent Autophagy by Particulate Matter 2.5 and Hydrogen Peroxide in Human Lung Epithelial A549 Cells (미세먼지와 산화적 스트레스에 의한 인간 폐 상피 A549 세포에의 ROS 의존적 자가포식 유도)

  • Recently, interest in the harmful factors of particulate matter (PM), a major component of air pollution, has been increasing. In particular, PM2.5 with a diameter of less than 2.5 ㎛ is well known to induce oxidative stress accompanied by autophagy in human lung epithelial cells. However, studies on whether PM2.5 increases autophagy under oxidative stress and whether this process is reactive oxygen species (ROS)-dependent are insufficient. Therefore, in this study, we investigated whether PM2.5 promotes autophagy through the generation of ROS in human alveolar epithelial A594 cells. According to our results, cells co-treated with PM2.5 and hydrogen peroxide (H2O2) showed a lower cell viability than cells treated with each alone, which was associated with increased total and mitochondrial ROS production. The co-treatment of PM2.5 and H2O2 also increased autophagy induction, which was confirmed through Cyto-ID staining, and the expression of autophagy biomarker proteins increased. However, when ROS generation was artificially blocked by N-acetyl-L-cysteine pretreatment, the reduction in cell viability and induction of autophagy by PM2.5 and H2O2 co-treatment were markedly attenuated. Therefore, the present results suggest that PM2.5-induced ROS generation may play a critical role in autophagy induction in A549 cells.

A Rheological Study on Creep Behavior of Clays (점토(粘土)의 Creep 거동(擧動)에 관한 유변학적(流變學的) 연구(研究))

  • Most clays under sustained load exhibit time-dependent deformation because of creep movement of soil particles and many investigators have attempted to relate their findings to the creep behavior of natural ground and to the long-term stability of slopes. Since the creep behavior of clays may assume a variety of forms depending on such factors as soil plasticity, activity and water content, it is difficult and complicated to analyse the creep behavior of clays. Rheological models composed of linear springs in combination with linear or nonlinear dashpots and sliders, are generally used for the mathematical description of the time-dependent behavior of soils. Most rheological models, however, have been proposed to simulate the behavior of secondary compression for saturated clays and few definitive data exist that can evaluate the behavior of non-saturated clays under the action of sustained stress. The clays change gradually from a solid state through plastic state to a liquid state with increasing water content, therefore, the rheological models also change. On the other hand, creep is time-dependent, and also the effect of thixotropy is time-function. Consequently, there may be certain correlations between creep behavior and the effects of thixotropy in compacted clays. In addition, the states of clay depend on water content and hence the height of the specimen under drained conditions. Futhermore, based on present and past studies, because immediate elastic deformation occurs instantly after the pressure increment without time-delayed behavior, the factor representing immediate elastic deformations in the rheological model is necessary. The investigation described in this paper, based on rheological model, is designed to identify the immediate elastic deformations and the effects of thixotropy and height of clay specimens with varing water content and stress level on creep deformations. For these purposes, the uniaxial drain-type creep tests were performed. Test results and data for three compacted clays have shown that a linear top spring is needed to account for immediate elastic deformations in the rheological model, and at lower water content below the visco-plastic limit, the effects of thixotropy and height of clay specimens can be represented by the proposed rheological model not considering the effects. Therefore, the rheological model does not necessitate the other factors representing these effects. On the other hand, at water content higher than the visco-plastic limit, although the state behavior of clays is visco-plastic or viscous flow at the beginning of the test, the state behavior, in the case of the lower height sample, does not represent the same behavior during the process of the test, because of rapid drainage. In these cases, the rheological model does not coincide with the model in the case of the higher specimens.

Partial Purification of OsCPK11 from Rice Seedlings and Its Biochemical Characterization (벼 유식물에서 OsCPK11의 부분 정제 및 생화학적 특성 규명)

  • Calcium is one of the important secondary signaling molecules in plant cells. Calcium-dependent protein kinases (CDPK)-the sensor proteins of Ca2+ and phosphorylating enzymes-are the most abundant serine/threonine kinases in plant cells. They convert and transmit signals in response to various stimuli, resulting in specific responses in plants. In rice, 31 CDPK gene families have been identified, which are mainly involved in plant growth and development and are known to play roles in response to various stress conditions. However, little is known about the biochemical characteristics of CDPK proteins. In this study, OsCPK11-a CDPK in rice-was partially purified, and its biochemical characteristics were found. Partially purified OsCPK11 from rice seedlings was obtained by three-step column chromatography that involved anion exchange chromatography consisting of DEAE, hydrophobic interaction chromatography consisting of phenyl-Sepharose, and gel filtration chromatography consisting of Sephacryl-200HR. An in vitro kinase assay using partially purified OsCPK11 was also performed. This partially purified OsCPK11 had a molecular weight of 54 kDa and showed a strong hydrophobic interaction with the hydrophobic resin. In vitro kinase assay showed that the OsCPK11 also had Ca2+-dependent autophosphorylation activity. The OsCPK11 phosphorylated histone III-S, and the optimum pH for its kinase activity was found to be 7.5~8.0. The native OsCPK11 shared several biochemical characteristics with recombinant OsCPK11 studied previously, and both had Ca2+-dependent autophosphorylation activity and favored histone III-S as a substrate for kinase activity, which also had a Ca2+-dependence.

Optimization of sterilization conditions for the production of retorted steamed egg using response surface methodology (반응표면분석을 이용한 레토르트 계란찜의 살균조건 최적화)

  • The purpose of this study was to determine the optimum sterilization conditions for the production of retorted steamed egg using response surface methodology. Sterilization processes for eighteen conditions using varying sterilization temperature ($X_1$), time ($X_2$), and method ($X_3$) as the independent variables were carried out through a $3^2{\times}2$ experimental factorial design. Quality evaluations after sterilization included measurements of $F_0$ value ($Y_1$), peak stress ($Y_2$), pH ($Y_3$), color value ($Y_{4-6}$), and organoleptic test [preference for appearance ($Y_7$), overall acceptability ($Y_8$), and preference for texture ($Y_9$) and egg taste ($Y_{10}$)]. Dependent variables ($Y_{1-10}$) of eighteen conditions were more affected by temperature and time than by the sterilization method. Eight factors were selected among the dependent variables as significant factors related to the quality of the steamed egg. Finally, by establishing an optimum range of each dependent variable and contour analysis, the optimum sterilization conditions for the production of steamed egg were determined to be $120^{\circ}C$ for 25 min using a 2-step sterilization process.

Effects of Solvent Extracts from Dried Beet (Beta vulgaris) on Antioxidant in Cell Systems and Growth of Human Cancer Cell Lines (건조 비트(Beta vulgaris) 추출물의 Cell System에서 항산화 및 항암 효과)

  • The inhibitory effects of solvent extracts from dried beet (Beta vulgaris) on $H_2O_2$-induced oxidative stress in cell systems and on the growth of cancer cell lines (HT-29 human colon cancer and AGS human gastric adenocarcinoma cells) were investigated. Inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts on the growth of HT-29 and AGS cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was more significant on the growth of AGS cells and A+M extracts had a higher inhibitory effect compared to MeOH extracts. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of both cancer cells (p<0.05). Among fractions, hexane and 85% aq. methanol fractions showed higher inhibitory effects. In order to determine the protective effect on $H_2O_2$-induced oxidative stress, DCHF-DA (dichlorodihydrofluorescin diacetate) assay was conducted. The A+M and MeOH extracts of dried beet appeared to significantly reduce the levels of intracellular (ROS) with dose responses. Among the fractions, 85% methanol fractions showed a higher protective effect on production of lipid peroxides. These results indicate that the intake of dried beet may improve oxidative stress in cell and reduce cancer risk.

Activity and Survival of the Natural Bacteria under the Stressed Conditions Detected by Bioluminescent Phenotype (스트레스 하의 자연세균의 활성 및 생존의 발광표현형을 이용한 탐지)

  • To investigate whether the introduced genetic marker is useful to detect the survivalship and activity of the natural bacteria under the stressed conditions, one Gram-negative isolate, KP964 was transformed to the luminous phenotype by transferring luxAB gene. Under the starvation-stress this luminous bacterial culturability (determined by colony-forming-units [CFU] on agar plate) decreased rapidly below the detection limit by 37 days, while its total cell number (determined by AODC) remained almost the same as its initial inocular size. At that time period, the viable cell number was estimated to be 1400 times higher than its CFU number. The bioiuminescence (determined by relative light units [RLU]) produced under the same condition was also monitored and found to decrease more rapidly than the culturability by 5-fold. Under the other stresses, e.g., osmotic shocks, acid shock, and exposure to toxic chemicals, this bacterial strain did not show the reliable correlation between CFU and RLU. These results might not suggest the direct estimation of bioiuminescence from the stressed bacteria be an index of both the survivalship and its activity. However, when the stressed bacterial cells were incubated under the favorable condition by relieving from the existing stress, the potential bioiuminescence (the lag periods before the increase of bioiuminescence, the increase rates of bioiuminescence, and the maximal levels of bioiuminescence) was shown to be highly dependent upon the strengths of the stresses exposed to the bacterial cells. Therefore, analysis of the potential bioiuminescence from the stressed bacteria revealed good relationships with survival as well as activity.

Intramuscular Administration of Zinc Metallothionein to Preslaughter Stressed Pigs Improves Anti-oxidative Status and Pork Quality

  • This study was conducted to determine the effects of exogenous zinc-metallothionein (Zn-MT) on anti-oxidative function and pork quality. After feeding a corn-soybean meal-based diet for two weeks, 48 pigs ($Duroc{\times}Landrace{\times}Chinese\;Black Pig$) were assigned randomly to four groups. Pigs in Group 1 were maintained under non-stress conditions, whereas pigs in Groups 2, 3 and 4 were aggressively handled for 25 min to produce stress. Pigs in Groups 1, 2, 3, and 4 received intramuscular administration of saline (control group; CON), 0 (negative control group; NCON), 0.8 (low dose group; LOW), and 1.6 (high dose group; HIGH) mg rabbit liver Zn-MT per kg body weight, respectively. Pigs were slaughtered at 3 and 6 h post-injection. Zn-MT treatment increased (p<0.05) the activities of superoxide dismutase (SOD) and glutathione-peroxidase (GSH-PX) while decreasing the concentration of malondialdehyde (MDA) in liver. These responses were greater (p<0.05) at 6 h than at 3 h post Zn-MT injection. Zn-MT treatment increased (p<0.05) hepatic SOD mRNA levels in a time and dose-dependent manner and decreased (p<0.05) serum glutamate-pyruvate transaminase and lactate dehydrogenase activities (indicators of tissue integrity). Zn-MT administration decreased (p<0.05) lactate concentration and increased (p<0.05) pH and water-holding capacity in the longissimus thorasis meat. Collectively, our results indicate that intramuscular administration of Zn-MT to pre-slaughter stressed pigs improved tissue anti-oxidative ability and meat quality.

An Experimental Study on Time Dependency of Strain for Saturated Clay (포화점토(飽和粘土)의 변형(變形)에 있어서 시간의존성(時間依存性)에 관한 실험적(實驗的) 연구(研究))

  • This paper is concerned with the strain characteristics of the time effect on the remoulded saturated day sampled from the downstream of the Yeongsan river, and the constitutive equation that can generally explain time-dependent behaviors of norma1ly consolidated clay. This paper examines whether or not the afore-said constitutive equation can be applied to the remoulded Mooan-clay. Throughout this study, the conclusions obtained are as follows. 1. Throughout the isotropic consolidation test for 7 days and the isotropic relaxation test, the existence of the static and dynamic yielding surfaces is confirmed respectively. 2. The characteristics of time effect of the deformation, namely, the existence of a unique stress-strain-time relation, is conformed from the experimental result on the Mooan-clay. 3. The prodictions of the stress path and the strain on the Cam-clay theory is not consistent with those observed during the experiments. 4. Constitutive equation(2-3-12) obtained by applying Cam-clay theory to Perzyna's elastic-viscoplasticity theory can explain the behavior of pore water pressure during isotropic stress relaxation, concerned with time dependency under undrained condition. The equation can also explain the results of the undrained triaxial compression test for the clay with different strain rate under the same or different consolidation history. 5. This constitutive equation has eight material parameters which can be determined from triaxial compression tests.
