Hypoglycemic and Antioxidative Effects of Dietary Sea-Tangle Extracts Supplementation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 다시마 추출물 첨가식이의 항당뇨 및 항산화 효과)

  • The purpose of this study was to investigate the effect of dietary sea-tangle extracts on blood glucose levels, serum lipid levels, thiobarbituric acid reactive substance (TBARS) and glutathione enzymes in diabetic rats treated with streptozotocin (STZ) Four groups of rats (Sprague-Dawley male rats, 180 - 200g) were consisted of normal rats fed control diet (C), diabetic rats fed control diet (CD), normal rats fed sea-tangl extracts diet (E), and diabetic rats fed sea-tangle extracts diet (ED). Diabetes was induced by single injection of streptozotocin (60 mg/kg B.W.). After 7 weeks, rats were sacrificed, serum glucose, serum total cholesterol, triglyceride levels and glutathione enzymes were measured. Urine was significantly higher in CD and ED groups than those of others (p < 0.05). Levels of amylase, calcium, uric acid, hemoglobin, cholesterol and low density lipoprotein (LDL)-cholesterol were different among four groups. But high density cholesterol (HDL)-cholesterol of ED group was significantly higher (p < 0.05) than other groups (C and E group) And the weekly change of serum glucose was decreased in the 3th,4th and 5th weeks. But serum triglyceride (TG) of diabetic rats fed sea-tangle extracts diet (ED) was lower than diabetic rats fed control diet (CD). Activity of hepatic microsomal G6Pase was significantly increased CD and ED groups higher than C and E group, but kidney was decreased ED group. Hepateic glutathione S-transferase (GST) of CD and ED group were significantly lower than C and E group (p<0.05), glutathione peroxidase (GPX) of E and ED group were significantly higher than C and CD group (p<0.05), glutathione reductase (GR) activities of ED group was significantly lower than other groups, malondialdehyde (MDA) of ED was lower than E and CD group, but kidney was increased significant in ED group compared to liver. These results suggested that dietary sea-tangle extracts reduce .hepatic disorders such as oxidant than kidney. In conclusion, dietary sea-tangle extracts groups reduced blood TG and hepatic MDA levels in STZ-induced diabetic rats.

Hypoglycemic Action of the Fat Soluble Fraction of Panax Ginseng C.A. Meyer in Streptozotocin Induced Diabetic Rats (Streptozotocin 유발 당뇨병 쥐에서의 인삼 지용성분획의 혈당 강하작용)

  • This study was made to understand a hypoglycemic action of the fat soluble fraction of Panax ginseng C.A. Meyer in streptozotocin induced diabetic rats by determining the activities of several enzymes related to carbohydrate and lipid metabolism as well as several blood component levels such as glucose and ketone bodies, and non-esterified fatty acids. Albino rats (Sprague Dawley, 170-200g, 3) were injected once with 70mg streptozotocinhg body weight intraperitoneally and fed with ordinary diet for 7 days, and then the fat soluble fraction (5 mg~20 mg/day/rat) was injected intraperitoneally once a day for three days to rats having high blood glucose level over 340 mg/100ml. After a final injection of the fat soluble fraction, rats u.ere starved for 16 hours followed by the analysis of blood serum and liver enzymes. It was found that increased levels of glucose, ketone bodies and free fatty acids in streptozotocin induced rats were decreased appreciably by administration of the fat soluble fraction. However, the amount of administered fat soluble fraction did not show any significantly different hypoglycemic action. Decreased activities of glucokinase, phosphofructokinase, pyruvate kinase, 6-phosphogluconate dehydrogenase and acetyl CoA carboxylase of the liver of streptozotocin induced diabetic rats were greatly modified suggesting that a hypoglycemic action of the fat soluble fraction was also appreciable as ginseng saponin fraction. We also compared a hypoglycemic action of the fat soluble fraction prepared from American ginseng and Chinese ginseng with that of Korean pain ginseng. 핀o significant difference of the hypoglycemic activity was observed between the above ginseng fat soluble fractions, suggesting that a study of the fat soluble fraction might be one of the most interesting subjects relating to diabetic hyperglycemia in the near future.

Phospholipase $A_{2}$ Activity and Lipid Peroxidation in Liver Microsome of Streptozotocin Induced Diabetic Rats (당뇨쥐의 간 Microsome에서 Phopholipase A_{2} 활성과 지질과산화)

  • The purpose of this study was to investigate phospholipase $A_{2}$ activity and lipid peroxidation I streptozotocin induced diabetic rats. Sprague-Dawley male rats weighting-Dawley male rats weighting 300$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group. Diabetes was induced by intravenous injection of 55mg/kg of STZ in sodium citrate buffer(pH 4.3). Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in DM group. Phosphatidylcholine hydrolysis in liver was not significantly different between two groups, whereas phosphatidylethanolamine hydrolysis in liver was increased by 69% in DM group comparing with that of normal group. Liver microsomal phospholipase $A_{2}$ activity and level of TBARS was increased by 91%, 109% in DM group compared with that of normal group, respectively. The present results indicate that phospholipase $A_{2}$ activity is specific to PE hydrolysis, leading to lipid peroxidation process in STZ induced diabetic rats.

L-Carnitine Administration Improves Lipid Metabolism in Styeptozotocin-Induced Diabetic Rat

  • The purpose of this study was to investigate the effects of L-carnitine administration on lipid metabolism in streptozotocin-induced diabetes. Diabetes was induced by a single intraperitoneal injection of streptozotocin (50 mg/kg b.w.) and was confirmed by determination of urinary glucose secretion. Diabetic rats in the three L-carnitine treated groups were given L-carnitine, 50(D5O), 100(D100) and 200 (D200) mg/kg body weight, by subcutaneously every other day for four weeks, while animals in normal (N) and diabetic (DM) groups for control received saline by the same method. The daily weight gain was not different between normal and diabetic rats, but daily dietary intake was significantly higher in diabetic rats than in normal rat. Diabetic rats had a significantly lower carnitine concentration in both serum and liver compared to normal rats. Total carnitine concentration in serum was increased dose dependently upon carnitine administration, but statistic significance was shown only in D200 group. Diabetic rats had significantly higher serum triglyceride and cholesterol concentrations compared to normal rats. However there were no significant differences in liver L-carnitine administration to diabetic rats significantly decreased serum triglyceride but not cholesterol concentrations. In liver, triglyceride and cholesterol concentrations were not attired by L-carnitine administration. These results indicated that streptozotocin induced-diabetic rats have decreased carnitine and increased lipid concentrations compared with normal rats. Also it indicated that L-carnitine administration has an effect on the normalization of serum triglyceride concentrations in diabetic rats.

Antioxidative Effect of Polygoni radix in Streptozotocin-Induced Diabetic Rats (하수오 추출물 투여가 Streptozotocin으로 유발된 당뇨 흰쥐의 항산화작용에 미치는 효과)

  • This study was done to investigate the antioxidative effect of polygoni radix in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The ethanol extract of Polygoni radix was orally administrated once a day for 7 days. The contents of Total cholesterol, triglyceride (TG) were significantly decreased in Polygoni radix treated STZ-sample group compared with to the those of STZ-control group. The hepatic cytosolic activities of glutathione-s-transferase (GST), superoxide dismutase (SOD) were significantly increased, Also the content of glutathione (GSH) was incresaed in Polygoni radix treated STZ-sample group compared with to the those of STZ-control group. but the content of malondialdehyde (MDA) and the hepatic cytosolic activity of Catalase (CAT) were decreaed but not statistical significant. These results indicated that ethanol extract of polygoni radix was effective for the antidxidative in the STZ-induced diabetic rats.

Hypoglycemic and Hepatoprotective Effects of Betaine on Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐의 베타인 첨가에 의한 항당뇨 및 간보호 효과)

  • This study was to investigate the effect of betaine on the hypoglycemia and hepatoprotection of streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing around 280 g were randomly assigned to the three experimental groups: a healthy normal group and two groups with STZ-induced diabetes and fed either control diet or betaine diet. Betaine given to the STZ-diabetic rats had significant effect in lowering the serum glucose concentrations compared to the STZ-diabetic rats. The alanine aminotransferase (AST) and aspartate aminotransferase (ALT) activities and triglyceride contents in serum were dramatically higher in the STZ-diabetic rats, but these increases in relation to diabetes also decreased in the STZ-diabetic rats fed betaine. However, the total-cholesterol concentration in the STZ-diabetic rats was even increased by betaine. The morphology of the pancreatic islets in the normal rats showed a typical round form, but most of the islets in the STZ-diabetic rats showed severe morphological alterations by being markedly destroyed. However, the islet morphology of STZ-diabetic rats given betaine mostly maintained a normal rounded appearance. The present study strongly suggests that the administration of betaine showed a moderate hypoglycemic effect by protecting the pancreatic beta-cells by morphological examination from STZ-induced destruction.

Effects of Fractions from Methanol Extract of Commelina communis on Blood Glucose Levels and Energy Metabolism in Streptozotocin-Induced Diabetic Rats (닭의장풀 분획물이 당뇨 유발 흰쥐의 혈당과 에너지원 조성에 미치는 영향)

  • Methanol extracts of Commelina communis, a Korean wild edible plant showing hypoglycemic effect on diebetic rats were fractionated systematically by hexane, chloroform, butanol, and water, and these four fractions were administered orally to streptozotocin-induced diabetic rats to evaluate the hypoglycemic effect. The butanol and water fractions of Commelina communis lowered blood glucose levels of diebetic rats at a dose of 500 mg/kg/day. Administration of each fraction except that of chloroform increased the plasma protein levels. Liver protein levels were significantly higher in the butanol- and water fraction-fed groups. The results suggest that the butanol fraction of Commelina communis has hypoglycemic effect on diabetic rats and the intake of Commelina communis may be beneficial for the management of diabetes mellitus.

Effect of Oligonol, a Low Molecular Weight Polyphenol Derived from Lychee on Oxidative Stress-Related Hepatic Damage in Streptozotocin-Induced Diabetic Rats (리치 저분자 폴리페놀인 Oligonol의 Streptozotocin 투여 당뇨 쥐에서 산화적 스트레스와 관련된 간 손상에 미치는 영향)

  • This study was conducted to examine whether or not oligonol, a low molecular weight polyphenol derived from lychee fruit, has an ameliorative effect on diabetes-induced oxidative stress-related hepatic damage in streptozotocin (STZ)-induced diabetic rats. Oligonol (10 or 20 mg/kg body weight; O10 or O20, respectively) was orally administered every day for 10 days to STZ-induced diabetic rats, and its effects were compared to vehicle-treated diabetic (Veh) and non-diabetic rats. Administration of 20 mg/kg of oligonol significantly decreased liver weight compared with the Veh group (P<0.05). Elevated levels of hepatic glucose, reactive oxygen species, peroxynitrite, and lipid peroxidation were detected in diabetic vehicle rats, whereas oligonol treatment significantly attenuated these levels (P<0.05). In diabetic vehicle rats, hepatic antioxidant enzyme protein levels decreased, whereas oligonol treatment showed significant elevated results. For inflammation-related protein expression, oligonol-treated groups showed insignificant reduction. Oligonol improved expression of proapoptotic protein caspase-3 in the liver of diabetic rats (P<0.05). In conclusion, these results provide important evidence that oligonol exhibits an inhibitory effect on oxidative stress and apoptosis-related protein expression as well as a hepato-protective effect against the development of diabetic complications in STZ-induced type 1 diabetic rats.

The Effect of Each Fraction of Methanol Extract of Alisma canaliculatum on Blood Glucose Levels and Lipid Metabolism in Streptozotocin Induced Diabetic Rats (택사 분획물의 투여가 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질대사에 미치는 영향)

  • Diabetes mellitus was induced in male Sprague-Dawley rats weighing 200-245g by injection of streptozotocin(STZ) into the tail vein at a dose of 45mg/kg and were divided into seven groups ; normal, diabetic control, and five experimental groups(fractions of hexane, chloroform(CHCl$_3$), ethylacetate(EtOAc), butanol(BuOH) and water($H_2O$)). The rats of all groups were fed on AIN-93 diet and the five experimental groups were orally administered each fraction for 14 days. The body weight and diet intake were monitored daily. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol, free fatty acid and aspartate and alanine aminotransferase activities were analyzed. Diabetic rats showed the lower weight gain compared to the normal rats. The plasma glucose levels of the CHCl$_3$, and $H_2O$ fraction groups were significantly lower than the other experimental groups. The plasma insulin level of the CHCl$_3$ fraction group was much higher than that of diabetic control group. The plasma cholesterol levels were increased in all the experimental groups. The groups of hexane, BuOH and H2() fractions showed the lower plasma triglyceride levels compared to diabetic control group. The plasma free fatty acid levels were not significantly different in all groups. HDL-cholesterol levels were definitely higher in hexane, CHCl$_3$ and EtOAc fraction groups than that of diabetic control group. In conclusion, administration of CHCl$_3$ and $H_2O$ fractions of methanol extract of Alisma canaliculatum exhibited hypoglycemic effects in STZ induced diabetic rats, showing the possibility of therapeutic use of Alisma canaliculatum to the diabetes mellitus.

  • Objectives : The Present study was carried out to investigate the preventive effect of Anti-diabetic prescription 1(AD-1) on streptozotocin(STZ)-induced diabetic nephropathy. Methods : AD-1 consists of eleven herbs that have an effect on diabetes mellitus. AD-1 was given to rats with the combination of oral administration and herbal-acupuncture stimulation. The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with AD-1 treatment. Experimental diabetes was induced by the injection of STZ(60mg/kg) to e rat via the peritoneum. The effect of AD-1 on STZ-induced diabetic nephropathy was observed by measuring the serum level of creatinine and BUN. Urine secretion of albumin for 24 hours and urine level of glucose measures too. Anti-oxidative stress of AD-1 administration in living body was estimated by measuring lipid peroxide and GSH content in cortex of kidneys. Results : STZ induced increase of serum creatinine, BUN and albumin secretion were lowered by AD-1 treatment. Conclusion : The AD-1 treatment showed protective effect on rat diabetic nephropathy model, and action mechanism of the effect was thought to be concerned with anti-oxidative stress.
