Protective Effects of Cinnamomi Ramulus Herbal Acupuncture on $\beta$-cell Damage of Streptozotocin-induced Diabetic Rat (계지약침(桂枝藥鍼)이 Streptozotocin 유도 당뇨 흰쥐의 췌장세포 손상에 미치는 보호 효과)

  • Objectives : For evaluation of preventive and anti-diabetic activities of Cinnamomi ramulus(CR) herbal acupuncture on pancreatic islet damage in streptozotocin(STZ)-induced diabetic rat. Methods : CR herbal acupuncture was performed at Bisu($BL_{20}$) for 3 weeks subcutaneously starting1 week before STZ i.p. injection. SD rats were divided into four groups(n=10 for each group); 1) NC group, non-treated normal control group, 2) STZ group, STZ administered control group, 3) CR125 group, CR(125mg/kg) + STZ administered group, and 4) CR250 group, CR(250mg/kg) + STZ administered group. Results : Both of CR250 and CR125 groups showed increase in insulin secretion and decrease in the level of serum triglyceride and non-esterified fatty acid in a dose-dependent manner compared to the STZ group. Only CR250 group showed decrease in the levels of glucose and total cholesterol compared to the STZ group. CR herbal acupuncture prevents $\beta$-cell damage of pancreatic islet, showing round figure on the sections of the pancreas. In the pancreatic cells, expressions of iNOS, JNK-2, P-JNK-1/2 and ERK-1/2 were decreased compared to the STZ group. CR herbal acupuncture solution did not show any cytotoxicity by MTS assay and inhibited expressions of iNOS and COX-2 in the STZ-induced diabetic rats. Conclusions : Therefore, we suggest that CR herbal acupuncture may act as a prophylactic as well as a therapeutic modality for diabetes mellitus.

  • The aim of the present study was to investigate the effect of Palmiwon on the type 1-prediabetic models induced by streptozotocin (STZ) in RINm5F cells and HIT-T15 cells. Palmiwon increased the cell proliferation and insulin release when pre- and post-treated for the STZ-exposed pancreatic beta cells. The cell proliferation and insulin release of these beta cells were measured by $^3$H-thymidine uptake and RIA, respectively. We also analyzed nutrients such as sugars, fatty acid and amino acids and minerals containing in Palmiwon using by gas chromatography, amino acid analyzer and AA spectrometer, respectively. Palmiwon seems to have protective and recovery properties on the prediabetic model in cellular level, which were ascribe to various nutrients and minerals containing in Palmiwon. From these results, it could be suggested that Palmiwon may be available as preventive and therapeutic prescription of type 1 diabetes mellitus.

Anti-Diabetic Effects of Mori Folium Extract on High-Fat Diet and Streptozotocin-Induced Type II Diabetes Mellitus in Mice (고지방식이와 STZ 유도 제2형 당뇨병 마우스에서 상엽 추출물의 항당뇨 효과)

  • Objectives : The present study was designed to investigate the anti-diabetic effects of Mori Folium (Morus alba L. of Moraceae) extract (MFE) on high fat diet (HFD) and streptozotocin (STZ)-induced type II diabetes mellitus in mice. Methods : The mice (C57BL/6J) were fed HFD for 8 weeks and then was induced with a single injection of STZ (75 mg/kg). The diabetic mice were divided into four groups [(STD, HFD, HFD + MFE and HFD + quercetin (QUR)] and administered with MFE or OUR for 4 weeks. Fasting blood glucose, lipid profile (triglycerides and cholesterol etc.), glutamic oxaloacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), insulin and leptin were measured every 2 weeks. Results : Body weight gain was lower in the MFE and QUR groups than HFD group. The fasting blood glucose was lower in the MFE and QUR groups. Oral glucose and insulin tolerance were decreased in the MFE and QUR groups. The levels of serum total cholesterol, triglycerides, and LDL cholesterol were reduced in the MFE and QUR groups. The HDL cholesterol was much higher in the MFE and QUR groups than HFD group. The levels of GOT, GPT and atherogenic index were decreased in the MFE and QUR groups. The serum insulin and leptin concentrations were reduced in the MFE and QUR groups. Conclusions : These results showed that MFE could decrease blood glucose level and lead to an amelioration in dyslipidemia states on HFD/STZ-induced type II diabetes mellitus in mice.

Association between extract from fruit of Opuntia ficus-indica and streptozotocin-induced diabetic rats

  • Opuntia ficus-indica has traditionally been used in prevention and treatment of various diseases such as diabetes mellitus. The current study was performed to determine whether Opuntia ficus-indica is associated with diabetes. Diabetic rat models were induced with streptozotocin (STZ). This study divided rats into 1 day (short-term) and 4 consecutive weeks (long-terms) of daily administration. These groups were subdivided into four groups each other for assessment of blood glucose level as follows: Group 1, untreated rats given distilled water; Group 2, untreated rats given Opuntia ficus-indica; Group 3, STZ-induced diabetic rats given distilled water; Group 4, STZ-induced diabetic rats given Opuntia ficus-indica. Blood glucose level was measured for one day and four weeks. In addition, serum markers of alanine aminotransferase (ALT), aspartate transaminase (AST), cholesterol, and creatinine were determined, and total protein triglycerides were measured at four weeks. Blood glucose level was highest in both groups (Group 3 and Group 4) at 30 minutes and two weeks and gradually decreased in a time-dependent manner. The difference in blood glucose among the four groups was significant (p < 0.05). Additionally, the levels of ALT, AST and triglycerides were significantly decreased by Opuntia ficus-indica.

Antioxidant Effects of Sesamin and Sesamolin in Streptozotocin-induced Diabetes Mellitus Rat (Streptozotocin유발 당뇨쥐에서 세사민과 세사몰린의 항산화효과)

  • Oxidative stress is believed to play an important role in the development of vascular complications associated with diabetes mellitus. We examined the antioxidative effect of sesamin and sesamolin on the preventing the development of insulin dependent diabetes mellitus using streptozotocin-induced Spraque Dawley diabetic rats. From 48 hours after injection of streptozotocin (60 mg/kg body weight), a portion of diabetic rats were fed with 0.2% sesamin and sesamolin containing diet for 3 weeks. There were significant differences of blood glucose and kidney weight between diabetic ports and control. Sesamin and sesamolin increased glutathione-S-transferase activity in kidney. The concentration of the thiobarbituric acid-reactive substances in the serum, liver, and kidney of diabetic rats administered sesamin and sesamolin decreased significantly as compared with that of the non-treated diabetic group. Dietary sesamin and sesamolin suppressed the oxidative stress in the diabetic rats. These results demonstrated that sesamin and sesamolin are potential and effective antioxidants that can protect the complications associated with diabetes.

Effects of QZ-16 on blood glucose and lipids in Streptozotocin induced diabetic rats

  • The present study was designed to investigate the hypoglycaemic and hypolipidaemic activities of Qurs-e-Ziabetus 16 (QZ-16) in Streptozotocin (STZ) induced diabetic rats. QZ-16, a polypharmaceutical herbomineral formulation developed on the principles of Unani medicine is used for non-insulin dependent diabetes mellitus (NIDDM). The elevated levels of fasting blood glucose, serum levels of cholesterol, triglycerides and urea observed in rats treated with STZ (55 mg/kg body wt.) were significantly reduced by the treatment of QZ-16 (240 mg/kg, p.o.) and gliclazide (30 mg/kg, p.o.). The reduced HDL cholesterol levels were also increased by the QZ-16 and gliclazide treatments in the STZ induced diabetic rats. These data show that QZ-16 has hypoglycaemic, hypolipidaemic properties in STZ induced diabetic rats.

The Effect of BuOH Fraction of Polygonatum odoratum with Selenium on Blood Glucose Level and Lipid Peroxidation in Streptozotocin Induced Diabetic Rats (둥굴레 분획물과 Selenium이 Streptozotocin 유발 당뇨 흰쥐의 혈당수준과 지질과산화에 미치는 영향)

  • The purpose of this study was to investigate the effect of butanol(BuOH) fraction of Polygonatum odoratum with selenium tr-eatment on blood glucose levels and lipid peroxidations in streptozotocin(STZ) induced diabetric rats. Male Sprague-Dawly rats weighing(180-200g) were divided into five groups : normal STZ-control and three expreimental groups(P, odoratum group P, odo-Se group and Se group) Diabetes mellitus was induced by injection STZ in the tail vein at the dose of 45mg/kg B.W The BuOH fraction of Polygonatum odoratum(500mg/kg. B,W) given orally administered for 14 days. The Se treated group were fed a AIN-76 recommendation diet mixed with Na2Seo3(2mg/kg diet). Diabetic rats showed the lower weight gain compared to the normal rats. the plasma glucose levels of the P. odo-Se group were significantly lower than the other experimental groups. The plasma cholesterol levels were higher in STZ-control and Se groups compared toP.odoratum and P. odo-Se groups and HDL-cholesterol levels were increased in the diabetic experimental groups fed on BuOH fraction of P. odoratum with Se supplementation. The liver and muscle glycogen levels were not significantly differ among all groups. The plasma free fatty acid levels were lower in diabetic experimental groups fed on BuOh fraction of P. odoratum or Se sup-plementation than STZ-control and Se groups. Diabetics rats showed the higher levels of triglyceride in plasma andlower levels in liver compared with the normal group. Supplementation with Se decreased significantly the liver triglyceride level. The MDA levels in liver and kidney were significantly reduced in all the experimental groups. In conclusion administration of BuOH fraction of Polygonatuum odoratum with selenium supplementation reduced blood glucose levels and peroxdative tissue damage in STZ induced diabetic rats showing the possibility of preventiave and therapeutic use of the wild edible plant to the diabetes mellitus.

  • The hypoglycemic effects of butanol(BuOH) fraction of Polygonatum odoratum with vitamin E in streptozotocin(STZ)-induced diabetic rats were investigated. Sprague-Dawley rats weighing 200-230g were devided into five groups, and four groups induced diabetes mellitus by the STZ injection(45mg/kg b.w.) into the tail vein : Normal, diabetic-control, and three diabetic experimental groups(p. odoratum group, P. od-vit. E group and Vit. E group). All groups were fed on a AIN-76 diet, and the experimental groups were orally administered with the BuOH fraction of Polygonatum odoratum(500mg/kg b.w.) and vitamin E(10mg/kg b.w.) for 14 days. The body weight, diet intake and organ weights were monitored. The plasma levels of glucose, insulin, cholesterol, triglyceride, HDL-cholesterol and aspartate and alanine aminotransferase activities were analyzed. The levels of glycogen in liver and muscle, cholesterol in liver were determined. The malondialdehyde(MDA) levels in liver, kidney and lung were assayed. The body weight loss was seen in P. odoratum group, P. od-vit. E group, Vit. E group and diabetic control group, while the loss in P. odoratum group was much less than that in the diabetic control group. The plasma glucose levels were significantly lowered in P. odoratum group compared to diabetic control group. The plasma insulin levels were noticeably higher in P. odoratum and Vit. E groups. The rats in P. odoratum and P. od-vit. E group showed higher liver glycogen levels than in the diabetic control group. The MDA levels in liver, kidney and lung were also significantly reduced in P. od-vit. E and Vit. E groups compared to the diabetic control group. The results suggest that the administration of BuOH fraction of Polygonatum odoratum along with vitamin E reduced blood glucose levels and peroxidative tissue damage in STZ-induced diabetic rats, showing the possibility of preventive and therapeutic use of the wild edible plant to the diabetes mellitus. The Effects of Several Herbal Medicine Concentrated Solution on the Hyperglycemic Mice Induced with Streptozotocin (수종의 한약재가 Streptozotocin으로 유발된 고혈당 생쥐에 미치는 영향)

  • Objective : This study has been carried out to understand the effect of several herbal medicine concentrated solution on the hyperglycemic mice Induced with streptozotocin(STZ). Methods : The 60 mg/kg of STZ injection into mice twice by 24 h interval and then 120 mg/kg of STZ injection again 3 days after the earlier injection. Control group was subjected to natural recovery, however, treated groups were fed 0.2 ml of several herbal medicine-concentrated solution (PA (x2, several herbal medicine-concentrated solution 1 group); PB (x4 several herbal medicine-concentrated solution 2 group) daily for 6 weeks. Result : The weight of PA was higher than that of control, but weight of PB was lower than control. The blood level of control increased continuously, reaching to 350mg/dL after 6 weeks, however, PA and PB showed a fast reduction of blood glucose. In blood glucose tolerance test, PA and PB showed better resistance than control. The GOT level in significantly(p<0.05) decreased in PA and PB compared with control group. The BUN and creatinine levels are significantly(p<0.01) decreased in PA compared with control group. Feeding of several herbal medicine-concentrated solution in a concentration of PA had an efficient effect on regeneration or recovery of Langerhans islet and ${\beta}-cell$ damaged by STZ. More Langerhans islet and high insulin-immunohistochemical resistance were observed in PA compared with control, but they were higher in PB than in PA. The number of Langerhans islet ${\beta}-cell$ and Langerhans islet. Conclusions : The result from the six weeks of observation demonstrates that the several herbal medicine concentrated solution have a positive effect of lowering the level of blood sugar and they increased insulin concentration. They have an effect for recovery of pancreas tissue and recovery of kidney, liver function from a diabetes mellitus.

  • The hypoglycemic effects of four edible plants (Angelicae tenuissimae (A. ten.), Pleurospermum kamtschaticum (P. kam.), Adenophora remotiflora (A. rem.) and Zanthoxylum schinifolium (Z. sch.)) in streptozotocin (STZ) -induced diabetic rats were investigated. Sprague-Dawley male rats weighing 190-230 g were induced diabetes mellitus by the STZ injection (45 mg/kg) into the tail vein and were divided into six groups ; normal, STZ-control and four edible plant groups (A. ten., P kam., A. rem. and Z. sch. groups). Normal and STZ-control groups were fed a AIN-93 diet and four groups of STZ-induced diabetic rats were fed one of each experimental diets containing 10% of the edible plant powder for 4 weeks. Diabetic rats showed the lower weight gain compared to the normal rats. In experimental groups except P. kam., AST activities were close to normal. A. ten. group were lowered ALT activities slightly. The plasma glucose levels of the diabetic experimental groups were significantly decreased at 4th week. The plasma insulin levels in diabetic experimental groups were not significantly different compared to the STZ-control group. The liver glycogen levels in STZ injected rats were significantly lower in compared to the normal rats. However no significant differences were found in response experimental plants intake in diabetic rats. The muscle glycogen were not significantly different among all the groups.
