Novel Function of Cytokinin: A Signaling Molecule for Promotion of Antibiotic Production in Streptomycetes

  • Cytokinin has been known to act as a plant hormone to promote cell division and function in diverse processes in plant growth and development. Besides being produced in plants, it is also produced by various bacteria and fungi; however, its ecological significance is still unclear. In this report, we present an interesting finding that transzeatin riboside (tZR), a naturally occurring cytokinin compound, increased antibiotic production in many different streptomycetes, including Streptomyces coelicolor Ml3O, S. pristinaespiralis ATCC 25486, S. violaceoruber Tu22, S. anfibioticus ATCC l1891, and S. griseus IFO 13350. In vitro plate assays showed that the addition of 100 $\mu$M tZR increased the growth inhibition of Pseudomonas syringae pv. syringae, a plant pathogen, by S. griseus, a streptomycin producer. We suggest that cytokinin could act as a signaling molecule for antibiotic production in streptomycetes, a group of rhizosphere bacteria.

Antimicrobial-resistance Profiles and Virulence Genes of Vibrio parahaemolyticus Isolated from Seawater in the Wando Area (완도해역 해수에서 분리한 장염비브리오(Vibrio parahaemolyticus)의 항균제 내성 및 병원성 유전자의 특징)

  • Sixty-seven Vibrio parahaemolyticus isolates from surface seawater from the Wando area, on the southern coast of Korea, were analyzed for their susceptibility to 15 different antimicrobials and the presence of virulence genes. According to the disk diffusion susceptibility test, all of the strains studied were resistant to ampicillin and oxacillin, while decreasing percentages were resistant to vancomycin (64.2%), streptomycin (56.7%), amikacin (31.3%), kanamycin (22.3%), cephalothin (20.9%), erythromycin (10.4%), ciprofloxacin (4.5%), and tetracycline (3.0%). All of the strains were susceptible to five antimicrobials: chloramphenicol, gentamycin, nalidixic acid, sulfamethoxazole/trimethoprim, and trimethoprim. Fifty-nine isolates (88.1%) were resistant to three or more classes of antimicrobial and defined as multidrug resistant, and two strains were resistant to seven antimicrobial agents. The minimum inhibitory concentration (MIC) of the 67 V. parahaemolyticus isolates to ampicillin and oxacillin ranged from 512-2,048 and $64-512{\mu}g/mL$, respectively. All 67 isolates were also examined for the presence of the tdh and trh virulence genes using the polymerase chain reaction (PCR). However, no isolates possessed either tdh or trh. The VPA0477 (${\beta}$-lactamase) gene, present in all of the tested strains, was validated as a new specific marker gene in PCR assays for the accurate detection and identification of V. parahaemolyticus.

Identification of Lactobacillus ruminus SPM0211 Isolated from Healthy Koreans and Its Antimicrobial Activity against Some Pathogens

  • The intestinal microbiota are important to the host with regard to resistance they impart against bacterial infections and their involvement in mediating metabolic functions. Lactic acid producing bacteria such as Lactobacillus play an important physiological role in these matters. The aim of the present study was to isolate Lactobacillus sp. that inhibits enteric pathogens. Initially, 17 isolates from healthy Koreans were collected on Lactobacillus selective medium. Resistance of the isolates to antibiotics including rifampicin, streptomycin, clindamycin and vancomycin was measured. One of the isolate was identified as Lactobacillus ruminus on the basis of bacterial cell morphology, cultural characteristic and biochemical characteristics, 16S rRNA sequence analysis and PCR-RAPD. Antimicrobial activity of the bacterium against Vancomycin Intermediate Resistant Staphylococcus aureus (VISA) and Vancomycin-Resistant Enterococci (VRE) was measured. About $10^4$ cells of VISA or VRE were mixed with 1, 5, and 9 mL of L. ruminus SPM 0211 and the final volume was adjusted to 10 mL with brain heart infusion (BHI) broth. The cell suspension was incubated for 3, 6, 9, and 24 h, serially diluted and then plated on BHI agar plates. As numbers of L. ruminus SPM 0211 were increased, viable cell count of VISA and VRE decreased. The strongest antimicrobial activity of SPM 0211 was observed after 9 h incubation in any mixture, almost completely inhibiting the growth of these two bacteria. The results suggest that the freshly isolated L. ruminus SPM 0211 may be used as a pro-biotic microbe that prevents the colonization of enteric pathogens and can thereby promote good gastrointestinal health.

Isolation and Antimicrobial Drug Susceptibility of Mastitic Pathogens from Dairy Cattles with Clinical Mastitis in Gyeongnam South Area (경남 남부지방에서 임상형 유방염의 원인균 분리 및 약제 감수성 시험)

  • The present study was conducted to investigate isolation and antimicrobial drug susceptibility of clinical mastitic milking total 610 (897 quaters) dairy cattles from 36 dairy farms in Gyeongnam south area (Cosung, Masan) during the period from March 1999 to August 2002. The results obtained were summerized as follows . 1. Incidence of bacterial infection in four quaters was showed that right anterior quarter was 178(19.8%), right posterior quarter was 292(32.6%), left anterior quarter was 148 (16.2%), and left posterior quarter was 279 (31.1%), respectively. Isolation rate of posterior two quarters was higher 2 times than anterior two quarters. 2. Incidence of double infections of 897 clinical mastitic milk were showed that single infection was 549 (61.2%), double infection was 167(18.6%), triple infection was 9(1%) and no isolation was 172(19.2%). 3. Isolation of infected etiologic bacteria was showed that Streptococci spp., was 267(31%), Staphylococci spp., was 267(41%), S aureus, was 48(5.6%), G(-) bacillus was 126(5.6%), and Corynebactrium spy. was 52(6%), respectively, from total 861 samples. 4. The results of antimicrobial drug susceptibility of all isolates were showed that Streptococci spp., Staphylococci spp., S aureus, (G)(-) bacillus, and Corynebactrium spp. were susceptible to cefuroxime, cefoperazone, amoxicillin, cefazolin, ampicilin, penicillin, gentamicin, erythromycin, cloxacilin, ciprofloxacin, sulfamethoxasole/trimetoprim, teteracyclin, and norfloxacin (> 70%), but some of them were resistant to neomycin, streptomycin colistin, and cephalothin(> 60). 5. The results of drug susceptibility obtained from each farms had different susceptibility, even though, etiological microorganisms were same in each farms.

Observations on Livestock Wastewater in Taegu Area (대구지역 축산폐수에 대하여)

  • To estimate pollution status of livestock wastewater on four piggeries and one abattoir in Taegu area, physicochemical water analysis such as pH, suspended solid(SS), biochemical oxygen demand(BOD) and chemical oxygen demand(COD), and bacteriological examinations such as number of total viable cells and number of coliform with or without antibiotic resistance were carried out. The results obtained were as follows : The pH values of raw sewage ranged from 9.0 to 7.2 that of the effluent treated was lowed to 5.6~7.7. The SS values of raw sewage ranged from 5,275ppm to 120ppm and those of the efflunet decreased to 162~30ppm. The BOD values of raw sewage ranged 6,200ppm to 120ppm and those of the effluent treated decreased to 111 ~80ppm. The COD values of raw sewage ranged from 5,725ppm to 298ppm and those of the effluent decreased to 137~76ppm. The total viable cells of raw sewage ranged from $8.5{\times}11^{11}$/ml to $9.9{\times}10^7$/ml, those of the effluent decreased to $5.6{\times}10^6{\sim}4.2{\times}10^8/ml.$ The total coliforms of raw sewage ranged from $5.5{\times}10^9$/ml to $1.3{\times}10^5$/ml, those of the effluent decreased to $3.6{\times}10^4ml{\sim}9.0{\times}10^6$/ml. The incidence of streptomycin resistant coliforms was the highest(1.8~66.7%), and followed by tetracycline(1.7~64%), kanamycin(9.3~50.l%), ampicillin(0.06~45.5%) and chloramphenicol(14.3~33.5%) to total coliforms of raw sewage. The incidence of antibiotic resistant coliforms of raw sewage in farms ranged from 3.4~66.7% and that of abattoir's was 0.06% to 14.3%. Antibiotic resistant coliform counts of raw sewage ranged from 1.3$\times$10$^{8}$ /ml to 3.9$\times$10$^3$/ml, those of the effluent decreased to $3.0{\times}10^1{\sim}2.3{\times}10^5/ml$.

Analysis of antimicrobial resistance and PFGE patterns of Salmonella spp. isolated from chickens at slaughterhouse in Incheon area (인천지역 닭 도축장에서 분리된 Salmonella spp.의 항생제 내성 및 PFGE 패턴분석)

  • Salmonella spp. are the important pathogens both economically and clinically in animals as well as human. Some of them have highly zoonotic potentials even though they are asymptomatic in animals. Therefore, the prevalence of Salmonella spp. in animals is highly concerned for human health. The present study was carried out to investigate the prevalence, antimicrobial resistance and PFGE patterns of Salmonella spp. isolated from chickens at slaughterhouse in Incheon area. The overall isolation rate of Salmonella spp. from cloaca and cecum specimens was 7.3 % (37/510). Thirty seven isolates of Salmonella spp. were identified to 5 serotypes; S. Enteritidis, S. Newport, S. Typhimurium, S. Gallinarum, and S. Derby with prevalence of 46.0%, 40.5%, 8.1%, 2.7%, and 2.7%, respectively. Resistance to nalidixic acid was found in 97.3% of Salmonella spp. isolated, followed by streptomycin (16.2%), tetracycline (16.2%), ampicillin (5.4%). Only 6 isolates (16.2%) showed resistance to more than two antimicrobials. In PFGE analysis of chicken and human isolates with Xba I, S. Enteritidis isolates from chicken showed very high similarity over 82.8% and also the similarity was very high in the comparison with human isolates. However, the higher similarity (100%) was observed among chicken isolates of S. Typhimurium. These results suggest the close genetic relatedness of Salmonella spp. isolated from chickens with human.

Studies on Salmonella Isolated from Cattle (우(牛) 유래(由來)의 Salmonella속균(屬菌)에 대하여)

  • This paper deals with the isolation of Salmonella on 3 herds during the period from May 1984 to May 1985. Isolated Salmonella were examined for serotypes, adhesive fimbriae, antibiotic susceptibility and detection of R plasmid. The results were as follows; Of total 1505 cattle, 24 Salmonella were isolated from 18 cattle (1.2%) and their serotypes were S. enteritidis (9 strains), S. derby(4 strains), S. infantis (1 strain), $C_1$ group(8 strains), $C_2$ group(1 strain) and untypable(1 strain). Twelve strains of Salmonella were isolated from 227 cattle with diarrhea and their serotypes were S. enteritidis(4 strains), S. derby(3 strains), S. infantis(1 strain), $C_1$ group(3 strains) and untypable(1 strain). The isolation rate of Salmonella in cattle varied from 1.6 to 0.9% in 3 herds, it was higher in summer and autumn and lower with the age. Of total 24 strains, 23 were adhesive type 1 fimbriae. Antibiotic susceptibility test of Salmonella isolated was performed by the agar dilution method, using 9 antibiotics as follows: ampicillin(Am), chloramphenicol(Cm), gentamicin(Gm), kanamycin(Km), nalidixic acid(Na), rifampicin(Rf), streptomycin(Sm), sulfadimethoxine(Su) and tetracycline(Tc). All the strains tested were susceptible to Am, Cm, Gm, Km, Na, Rf and Tc. Of total 24 strains, 23(95.8%) were resistant to Su and 14(58.3%) to Sm. Of the 23 resistant Salmonella strains, all the strains were found to carry R plasmid. Among them, two strains which had the R plasmid conferring SmSu resistance was transferable at $25^{\circ}C$ only.

Antimicrobial resistance and distribution of tetracycline resistance determinants in Escherichia coli isolated from aquatic birds (수생조류에서 분리한 대장균의 항균제 내성 및 Tetracycline 내성인자의 분포)

  • One hundred and sixty nine Escherichia (E.) coli strains isolated from fecal samples of aquatic birds in Geumho river basin and Dalseong park were tested by agar dilution method to determine their susceptibility patterns to 14 antimicrobial agents. The distribution of tetracycline resistance determinants (tetA, tetB, tetC, tetD and tetE) were also examined by PCR in 76 tetracycline-resistant ($TC^r$) E. coli isolates. The high resistance was observed in tetracycline, cephalothin and ampicillin (45.0~36.7%). Resistance of E. coli isolates derived from Dalseong park to tetracycline, cephalothin, ampicillin and streptomycin (65.7~44.8%) were significantly higher than those isolated from Geumho river basin (31.4~14.7%). About seventy percent (70.4%) of the strains isolated were resistant to one or more drugs tested. Thirty (39.5%) of 76 $TC^r$ E. coli isolates which were resistant to one or more drugs transferred all or a part of their resistance patterns to the recipient strain of E.coli J53 by conjugation. All of $TC^r$ E. coli isolates contained at least one or more of 5 tet genes examined. The most common genes found in these isolates were tetA (60.6%) and followed by tetB (7.9%) and tetC (1.3%). However, tetD and tetE were not found in any of the isolates tested. Twenty one (27.6%) of $TC^r$ E. coli isolates had two determinants, tetA/tetB (20 strains), tetA/tetC (1 strain). And two strains (2.6%) contained three determinants (tetA/tetB/tetC).

Antimicrobial resistance and transfer of R plasmid of pathogenic Eseherichia coli isolated from poultry in Korea (가금 유래 병원성 대장균의 항균제 내성 및 R plasmid 전달 양상)

  • Antimicrobial drugs are widely used in poultry industry as growth promoters or to control infectious diseases. However, this practice is reported to have caused high resistance to antimicrobial drugs in normal chicken flora and pathogens. Antimicrobial resistance to Escherichia coli (E. coli) from chicken has been mainly reported in normal flora, but rare in pathogenic organism in Korea, recently. Therefore, this study was conducted to investigate prevalence of antimicrobials resistance, transfer of R plasmid, and association between antimicrobial drug resistance and O serotype of 203 pathogenic E. coli from poultry in Korea during the period from April 2003 to December 2005. These isolates showed a high resistance to tetracycline (Tc, 93.6%), nalidixic acid (Na, 92.6%), streptomycin (Sm, 81.8%), ampicillin (Ap, 77.3%), ciprofloxacin (Ci, 70.9%), sulfisoxazole (Su, 66.5%), and trimethoprim (Tp, 58.1%). Two hundred-one (99.0%) of the isolates were resistant to one or more drugs. They showed 57 different resistant patterns, and the most prevalent resistant pattern among them was Tc, Sin, Su, Ap, Tp, Ci, Na. Sixty-eight (33.8%) of the isolates transferred all or a part of their antimicrobial resistant pattern to the recipient strain by R plasmid. The most common antimicrobial resistant pattern was Tc, Sm, Su, Ap, Tp, Ci, Na in serotype O78, O88 and O15, respectively. These results exhibit high individual and multiple resistance to antimicrobials of pathogenic E. coli from poultry in Korea. They also suggest the needs for surveillance to monitor antimicrobial resistance in pathogenic bacteria that can be potentially transmitted to humans from food animals and to regulate the abuse of antimicrobials on food-producing animals in Korea.

Laboratory Measurements on the Uptake of Carbon Monoxide by Soils (토양의 일산화탄소 제거에 관한 연구)

  • The consumption of atmospheric carbon monoxide by soil was measured under laboratory conditions in different types of soils. Laboratory experiments were performed with humus containing high proportion of organic matter, roadside soils, and humus and roadside soils previously exposed to high concentration of CO by reusing in the experiment. CO concentrations in the 18.2 l-reaction vessel were varied from 2,000 ppm to 24,000ppm to estimate the effectiveness of CO consumption at high level of CO. The uptake of CO by soil was measured by gas chromatography using a TCD detector. The control experiments conducted along with the soil experiments evidently indicated that the potting soil is responsible for CO consumption. Humus showed much higher CO uptake rates compared with the soil taken from roadside. The humus reused in the experiment showed somewhat higher rates(15%) of uptake than the fresh one. The soil's ability to remove CO from the test atmosphere reached a maximum near the CO concentration of 13,000 ppm in the range of $9,000~24,000ppm$. The addition of streptomycin did not influence the removal capacity of soil significantly, whereas 10% saline solution remarkably prevented CO uptake of the humus sample.
