Isolation of vibrio species from cultured flounders (paralichthys olivaceus) with uicers and ascites in the southern coast of Korea during the winter season (동절기 한국 남해안의 궤양증 및 복수증 양식 넙치로부터 vibrio종의 분리)

  • this study was performed for the purpose of surveying the distribution and characteristics of the Vibrio species in the diseased farm flounders (Paralichthys olivaceus) during the winter season in Korea. Samples were collected along the southern coast in the Sinam, Keoje Island, and Teosu areas from November '90 to Feburary '91. Nine species of Vibrio were identified as V. tubiashii (85 strains), V. damsela (78 strains), V. anguillarum (21 strains), V. compbelli (9 strains), V. fluvialis (9 strain), V. costicola (1 strain), V. alginolyticus (1 strain), V. gazogenes (1 strain), V. marinus (1 strain), and unidentified 3 groups of bacteria (154 strains) which may have related with V. tubiashii were isolated. Eleven kinds of discs (BBL Co.) were used for antibiotic susceptibility test and 116 strains were selected for the experiment. Most bacteria were powerfully inhibited their growth by 4 antibiotics such as cephalothin, chloramphenicol, colistin, and tetracycline, but they were poorly inhibited by 4 antibiotics such as carbenicillin, kanamycin, penicillin G, and streptomycin.

Clinical Study of Empyema Thoracis: a review of 64 cases (농흉의 임상적 고찰64례 보고)

  • A Clinical analysis of 64 patients of thoracic empyema was done who received surgical intervention at Dept. of Thoracic Surgery of the Chosun University Hospital in the period of 3 years from September 1976 to October 1979. Following was the results: 1. Seven cases [10.9%] were under the age of 15 years, 16 cases [25%] was between 15-30 years and 41 cases [64.1%] was above the age of 30 years. A proportion of children and adult was 1:8. 2. Male and female ratio was 3:1. Right and left side pleural cavity ratio was 2.4:1. 3. Predisposing factors were pneumonia [35.9%] and pulmonary tuberculosis [28.1%]. 4. Most frequently encountered symptoms were dyspnea, cough, chest pain and fever in order. 5. Etiologic organisms were confirmed in 39 cases [86.7%] which requested in 45 cases. Staphylococcal infections were 11 cases and streptococcal, pneumococcal pseudomonas infection was infected in order. 6. Pneumothorax was associated with empyema on 21 cases [32.8%]; among those 13 cases [61.9%] were tuberculous in nature. 7. Sensitivity test was revealed that Minocin was most very sensitive drug, and next Erythromycin, Gentamycin and Penbrex in order. But most resistant drugs were Penicillin, Kanamycin, Streptomycin and Tetracycline in order. 8. Treatments were combined with antibiotics therapy and several surgical procedures for empyema. 26 cases [40.6%] were treated with closed thoracotomy drainage, 17 cases [26.6%] with open thoracotomy tube drainage and 9 cases decortication and 9 cases thoracoplasty. 9. 2 death cases occurred in 64 cases of thoracic empyema, and 79.7% cases were discharged with recovery and improvement.

lacZ- and aph-Based Reporter Vectors for In Vivo Expression Technology

  • Three vectors, pSG1, 2, and 3, which facilitate in vivo expression technology (IVET) in Gram-negative bacteria, were developed. Vectors pSG1and 2 are derivatives of ColE1, and pSG3 is a derivative of an R6K replicon. These vectors contain oriT sites that allow mobilization when the RK2 Tra functions are provided in trans. These vectors contain promoterless lacZ (pl-lacZ) and promoterless aph (pl-aph) transcriptionally fused together, which allow qualitative and quantitative measurements of the expression of genes in the genome of bacterial cells. pSG1 and 3 contain gentamicin-resistance genes, and pSG2 carries a streptomycin-/spectinomycin-resistance gene, allowing for selection of recombinants generated by a single crossover between a library fragment cloned into a pSG vector and the identical region in the genome of a bacterial species from which the library fragment originated. These vectors were successfully applied to the generation of random fusions at high rates in the genomes of four representative Gram-negative bacteria. In addition, the expression level of ${\beta}-galactosidase$ and the degree of resistance to kanamycin in cells with fusions generated by these vectors were found to be linearly correlated, proving that these vectors can be used for IVET.

Evaluating the role of vaccine to combat peste des petits ruminants outbreaks in endemic disease situation

  • Among the main intimidation to the sheep and goat population, PPR outbreaks are causing huge losses especially in endemic areas. During recent times, six outbreaks of PPR were confirmed at semi-organized goat farms/herds in various regions of Punjab province and Islamabad capital territory (ICT), Pakistan. The disease started after introduction of new animals at these farms with no history of previous PPR vaccination. The clinical signs appeared affecting respiratory and enteric systems and spread quickly. Disease caused mortality of 10-20% and morbidity of 20-40% within a time period of four weeks. Morbidity and mortality rates were 30.38% (86/283) and 15.55% (44/283), respectively. Three treatment regimes were executed to demonstrate the role of vaccination during outbreak at these farms. First was to use only the broad spectrum antibiotics (Penicillin & Streptomycin and/or Trimethoprim and Sulfadiazine) at two farms (Texilla and Attock). Second treatment regime was to use the same broad spectrum antibiotic along with extensive fluid therapy (Farms at ICT-1 and ICT-2). The third regime was to use of broad spectrum antibiotic plus fluid therapy along with vaccinating the herd against PPR during first week of outbreak (ICT-3 and ICT-4). The third scheme of treatment gave the better results as there was no mortality in third week post-outbreak. Therefore, it is suggested to give proper importance to PPR vaccination along with conventional symptomatic treatment when dealing the PPR outbreaks in endemic disease conditions.

Serotypes and Sensitivity Distribution of Pseudomonas aeruginosa (임상재료에서 분리된 녹농균의 혈청형과 약제 감수성과의 관계)

  • The correlation between the serotypes and sensitivity distribution of Pseudomonas aeruginosa was studied with stock strains of department, strains isolated from various clinical sources and strains of haspital environment of the past two years. Two hundred and fifty seven strains were typable and 38 strains were untypable out of 295 clinical sorces. Serotype B was most provalent(27.6%) followed by serotype G(24.9%), serotype E(23.3%) and serotype A(7.8%) among 257 typable strains. However, no serotype J,L and M were observed. Serotype B,G,E and A were isolated from pus, sputum, wound, burn site and urine. Serotypes E were most frequently isolated from nasal discharge and serotypes K were isolated from pus. There were no apparent differences in sensitivity distribution of streptomycin, carbenicillin and ampicillin between the clinical and The environmental origin. However, the strains of environmental origin were found to be relatively more susceptable to tetracycline, gentamicin and fradiomycin than the strains of clinical sources in high concentration. The strains of clinical sources of serotype B,G,E and A showed different resistant patterns to all antimicrobial agents except for carbenicillin. Serotype E showed the highest percentage resistance followed by serotype G,B and A.

Antioxidant and Anti-Inflammatory Effect of Probiotic Lactobacillus plantarum KU15149 Derived from Korean Homemade Diced-Radish Kimchi

  • Lactobacillus plantarum KU15149 was demonstrated to have probiotic behavior and functions, including antioxidant and anti-inflammatory activity. L. plantarum KU15149 obtained from homemade diced-radish kimchi has a high survival rate under artificial gastric acid (pH 2.5, 0.3% pepsin) and bile salt (0.3% oxgall) conditions. However, L. plantarum KU15149 did not produce β-glucuronidase, which is known to be a carcinogenic enzyme with resistance to several antibiotics, such as gentamycin, kanamycin, streptomycin, tetracycline, and ciprofloxacin. L. plantarum KU15149 strongly adhered to HT-29 cells and had high antioxidant activity in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging and β-carotene bleaching assays. L. plantarum KU15149 also exhibited a pronounced inhibition of nitric oxide (NO) production, along with expression of nitric oxide synthase (iNOS) and cyclooxygenase -2 (COX-2) as well as pro-inflammatory cytokines, such as TNF-α, IL-1β, and IL-6, when RAW 264.7 cells were stimulated with LPS. Therefore, L. plantarum KU15149 exhibited pharmaceutical functionality as a potential probiotic.

Screening and Characterization of Pro biotic Lactic Acid Bacteria Isolated from Korean Fermented Foods

  • To examine their potential as probiotics, acid and bile tolerance, antibiotics resistance, adhesion capacity to Caco-2 and HT-29, and antibacterial activity, of LAB isolated from Korean fermented foods such. as dongchimi, kimchi, Meju, and doenjang were assayed against foodborne pathogenic bacteria. DC 55, DC 136, DC 222, KC 21, KC 24, KC 34, KC 43, KC 117, MJ 54, MJ 301, SP 33, and SP 170 strains were resistant to acid and bile conditions. In particular, DC 55, DC 136, KC 24, KC 43, and MJ 301 strains were highly resistant to higher than 20 ${\mu}g/ml$ concentrations of vancomycin, streptomycin sulfate, or amoxicillin, whereas, DC 222, KC 21, KC 34, KC 117, MJ 54, and SP 33 strains were susceptible to lower than 2 ${\mu}g/ml$ concentrations of those antibiotics. The adhesion to HT-29 and Caco-2 cells varied with the strains tested in a strain-dependent manner. The highest level of adhesion was observed with DC 55, KC 21, KC 24, and MJ 301 strains, having higher than 50% of adhesion to HT-29 or Caco-2 cells. In addition, Staphylococcus aureus was the most sensitive to KC 21, showing an inhibition of about 70%, and the antibacterial activity of KC 21 against S. aureus resulted most likely from both organic acids and bacteriocin. Based on its phenotypic characteristics and utilization of various sugars, the KC 21 strain was identified as Lactobacillus plantarum.

A Broad-Host-Range Promoter-Probe Vector, pKU20, and Its Use in Promoter Cloning and Expression of Bacillus thuringiensis Crystal Protein Gene in Pseudomonas putida

  • We have constructed a promoter-probe vector pKU20 using pKT230, a derivative of broad-host-range plsmid RSF1010, as a base. The pKU20 contains structural gene for aminoglycoside phos-photransferase (aph), without promoter, and a multiple cloning site upstream the aph. Using this vector, a 412base pairs (bp) PstI fragment showing strong promoter activity both in Escherichia coli LE392 and Pseudomonas putida KCTC1644 has been cloned from Pseudomonas fluorescens chromosomal DNA on the basis of streptomycin resistance. The nucleotide sequence of the 412 bp fragment has been determined and the putative - 35 and -10 region was observed. Insecticidal protein gene of Bacillus thuringiensis subsp. kurstaki HD-73 inserted on downstream of the promoterlike DNA fragment was efficiently expressed in E. coli and P. putida. The toxin protein was efficiently synthesized in an insoluble form in both strains.

Phage typing and pulsed-field gel electrophoresis of Salmonella typhimurium and S enteritidis isolated from domestic animals in Gyeongbuk province (경북지역 가축에서 분리된 Salmonella typhimurium과 S enteritidis의 phage typing 및 pulsed-field gel electrophoresis)

  • Forty-five Salmonella typhimurium isolates were encountered 8 phage types in which DT197 and U302 were the predominant types. The DT104 type which was first found from pig in Korea, and was resistant to chloramphenicol, streptomycin, sulfamethoxazole/trimethoprim, tetracycline, gentamicin and nalidixic acid. Twenty-two S enteritidis isolates were encountered 5 phage types in which PT4 were the representative (predominant). S enteritidis isolates were susceptible to all antimicrobial agents. As a result of PFGE analysis for S typhimurium and S enteritidis, PFGE patterns was better than phage typing in discriminating of strains. PFGE patterns were not in accord with phage type even though some strain had the same phage types.

Isolation and antimicrobial susceptibility test of Pasteurella multocida from respiratory disorder piglets (호흡기증세 자돈으로부터 파스튜렐라 속균 분리 및 약제감수성시험)

  • This study were carried out to investigate isolation of Pasteurella multocida from respiratory disorder piglets, to examine the biochemical properties and antimicrobial susceptibility. The results were summarized as follows; P multocida was isolated from 31($10.3\%$) of the 302 respiratory disorder or growing piglets of $4{\sim}10$ week olds. The majority of biochemical and cultural properties of the P multocida isolates were identical to those of the standard strains. The isolates were highly susceptible to norfloxacin($100.0\%$), enrofloxacin($96.8\%$) and ampicillin($87.1\%$), but resistant to streptomycin ($77.4\%$), penicillin($61.2\%$) and neomycin($54.8\%$).
