In this study, investigated the general characteristics and diagnostic methods types of streptococcosis among various fish disease pathogens that caused a lot of economic damaged to aquaculture fish based on the previous research paper. Streptococcosis infection of fish is considered a reemerging disease affecting a variety of wild and cultured fish throughout the world. Calssifiacation of Gram positive cocci based on DNA-DNA hybridization coupled with 16S sequencing has shown that at least five different species are considered of significance as fish pathogens: Lactococcus garvieae, L. piscium, Streptococcus iniae, S. agalactiae, S. paruberis, Vagococcus salmoninarum. Symptoms of infection with streptococcosis disease such as body color change, eyeball abnormality, gill discoloration, bleeding, abdominal distension, swelling of the kidney and spleen. In addition, it usually occurs from June to October when the water temperature rise a lot of fish death. Currently, 16S rRNA, 16S-23S rRNA intergenic spacer region (ISR), Random Amplified polymorphic DNA (RAPD), Ribotyion (RT), Loop-mediated isothermal amplification (LAMP) are among the methods for diagnosing streptococcosis. Among them, the LAMP method, which is high applicable to the aquaculture farm has attracted the spotlight, but due to problems such as confirmation of results. This seems to minimize the economic loss of streptococcosis which complements the problem so that it can be easily used from the diagnosis to the results confirmation.
This review is about current Streptococcosis of cultured olive flounder, Paralichthys olivaceus, in South Korea. The main bacterial species are Streptococcus iniae, S. parauberis and Lactococcus garvieae. The occurrence trend has been arranged with published data and seed bank (NIFS) data that was examined with deposited strains and the number from 2002 to 2017. S. iniae and L. garvieae were collected until 2012 and 2002, respectively, but not collected any more after that year. In recent years, the dominant strain of Streptococcosis of cultured olive flounder is S. parauberis. The phenotypic and genetic characteristics of standard strains and isolated strains were examined with published paper. Notably, we describe about distinguishing gross signs and histological signs of 3 types of Streptococcosis in this report. White turbidity of eye, hemorrhage, enteritis and ascites are main signs of S. iniae infection. Darkness of body surface, hemorrhage in somatic muscle and pericarditis are predominant in occasion S. parauberis infection. In occasion of L. garvieae infection, common signs of Streptococcosis are revealed, but these are weak tendency compared with the previous two cases.
This study was performed for the identification of Streptococcus sp. from cultured flounders (Paralichthys olivaceus) showing streptococcosis in the Jeju island. We isolated 10 strains of Streptococcus iniae from the cultured olive flounders with streptococcosis. Isolated strains were identified in S. iniae since they have formed the expected band through performing PCR assay using specific primers, Sin-1 (5'-CTAGAGTACACATGTACT(AGCT)AAG-3') and Sin-2 (5'-GGATTTTCCACTCCCATTAC-3'). In addition to 16S-23S rRNA intergenic spacers (ISR), operon structure of isolated strains showed that all strains had three 16S-23S rRNA ISR band patterns. The 16S-23S rRNA ISR sequence of isolated strains showed 96% sequence identity with S. iniae (GenBank accession number AF 048773). This paper is the first report that S. iniae is associated with streptococcosis of Olive flounder in Korea.
Two infectious species of Streptococcosis pathogens were detected by multiplex PCR assay. Detection rates of Streptococcus iniae and S. parauberis could reach 44.9% and 55.1% respectively for one year during 2004 to 2005 in Jeju island. These findings showed that S. parauberis strains were important pathogen with streptococcosis of olive flounder in Jeju island. In the present study we have investigated the interspecific relationship of all Jeju area of S. parauberis by RAPD analysis. Represent strains divided to four groups by RAPD fingerprints. Though, serological research of S. parauberis strains in Jeju island not exist yet. These strains doing the serological evolution.
Streptococcosis of olive flounder(Paralichthys olivaceus) is an important bacterial disease in Jeju island. In this study, we investigated monthly infection pattern of this disease in different size of the flounder fish. Even though the disease occurred throughout the year, the infection ratio was relatively higher in the months with warm water season. The infection was more prevalent in adult flounder over 30 cm total length compare to these of small size fish. Two infectious species of streptococcosis pathogens were detected by multiplex PCR assay. Detection ratios of Streptococcus iniae and S. parauberis reached up to 46% and 54%, respectively, from June 2003 to May 2005 in Jeju island. S. iniae occurred intensively from September to October, whereas S. parauberis reported from March to May. S. iniae and S. parauberis infections of cultured flounder share some common features, but clinical findings showed considerable differences between two diseases. Distended abdomen, protruded anus and ascitic fluid in the peritoneal cavity are evident lesions detected in S. iniae infection, whereas, flounders infected by S. parauberis showed prominent lesions such as darkened surface and haemorrhaging in the non-ocular side. Both streptococcosis pathogens were sensitive to antibiotics, such as ampicillin and amoxicillin. However, S. iniae strains were more sensitive to doxycycline, erythromycin and oxytetracycline than S. parauberis strains.
The potential of Streptococcus iniae glyceraldehyde-3-phosphate dehydrogenase (GAPDH) as an antigen for a subunit vaccine was investigated using a zebrafish model. The recombinant S. iniae GAPDH was purified using His-tag column chromatography, and antisera against the recombinant GAPDH (rGAPDH) were produced by intraperitoneal immunization of rats. By immunization with S. iniae rGAPDH, the survival rates of zebrafish against an S. iniae challenge increased, suggesting that GAPDH would be an antigen capable of inducing protective immune responses in fish. Furthermore, we demonstrated using Western blotting, that the antisera against rGAPDH of S. iniae had cross-reactivity with GAPDH from Streptococcus parauberis and Lactococcus garviae, which are also culprits of streptococcosis in cultured fish in Korea. These results suggest that S. iniae GAPDH may be used as an antigen for the development of a subunit vaccine against streptococcosis caused by diverse cocci in cultured fish.

To identify and quantify the effects of a combination of dietary $1{\times}10^8CFU/g$ Lactococcus lactis subsp. lactis I2 ($LI_2$) and 0.1% ${\beta}$-glucooligosaccharides (BGO) on the growth and immunity of olive flounder (Paralichthys olivaceus), a feeding experiment was conducted. Flounder ($14{\pm}0.5g$) were divided into two groups and fed control and synbiotic feeds for 8 weeks. Investigations were carried out on growth and feed utilization, innate immunity, serum biochemical parameters, intestinal lactic acid bacterial (LAB) viability, microvillus length, and changes in the expression levels of genes encoding pro-inflammatory cytokines (tumor necrosis factor $[TNF]-{\alpha}$, interleukin $[IL]-1{\beta}$, and IL-6). Results demonstrated the synbiotic diet had significantly better (p < 0.05) responses in terms of weight gain and specific growth rate, three innate immune parameters (respiratory burst, serum lysozyme, and superoxide dismutase), intestinal LAB viability, and the relative $TNF-{\alpha}$ expression level (p < 0.05). Moreover, after challenge with Streptococcus iniae ($1{\times}10^8CFU/ml$), the synbiotically fed group exhibited significantly higher (p < 0.05) protection against streptococcosis, validating the observed changes in immune parameters and induction of the cytokine-encoding gene. Therefore, according to the results of the present study, synbiotic feed ($LI_2+BGO$) increased growth, modulated innate immune parameters and protected olive flounder against streptococcosis.

The monitoring was performed to survey the mortalities and medications occurred in the inland aquaculture farms of olive flounder in South Korea from May to October, 2012. Both of the indirect inquiry for entire inland farms and the sample survey for selected farms were carried out. The aquatic organism disease inspectors, who have the national licenses for the diagnosis and prevention of aquatic organism diseases and have close relationship with the farms, investigated the rates and causes of mortalities according to the standard manual. The cumulative mortalities rate by the indirect inquiry on 565 farms, was calculated to 27.18%, and the mortalities rate by infectious diseases was 22.64%. Otherwise, the mortalities rate by sample survey on 60 farms was 25.50%, 19.33% of them were caused by infectious diseases. The high mortality rates were recorded by scuticociliatosis, non-infectious loss, streptococcosis, VHS, artificial eliminations, vibriosis and gliding bacterial disease. Streptococcosis and non-infectious mortality caused to serious loss in productivity and economy of the farms, because of their outbreaks in the flounder groups over 600 g. The monitoring of medications in the selected farms revealed that formalin for the treatment of external parasites, such as scuticociliates, was the most commonly used drugs in the farms. As the antibiotic medications, amoxicillin and florfenicol for streptococcosis, and oxytetracycline and neomycin expecting wide antibacterial spectrum, were frequently prescribed.
Streptococcosis caused by Streptococcus parauberis is a very important disease in farmed olive flounder Paralichthys olivaceus. For most fish pathogens, including S. parauberis, there are no analytical criteria to distinguish antibioticsusceptible strains from antibiotic-resistant strains. In this study, epidemiological cut-off ($CO_{WT}$) values were generated to classify 75 strains of S. parauberis isolated from 1999 to 2018 as wild type (WT) and non-wild type (NWT) using disc diffusion data and normalized resistance interpretation (NRI) analysis. The susceptibility of the isolates to 16 antibiotics was evaluated using CLSI guideline M42-A. The wild-type cut-off values for amoxicillin, erythromycin, oxytetracycline, and florfenicol for S. parauberis were ${\geq}35$, 31, 28, and 27 mm, respectively. The NWT ratios of S. parauberis strains to treatment with GEN, FFC, ENR, SXT, EFT, VAN, and CHL were 17% or less, indicating that these antibiotics may be used to treat streptococcosis caused by S. parauberis. For recent S. parauberis isolates, the NWT ratios for AMX, ERY, OTC and FFC are much higher than for strains isolated from 1999-2007. The $CO_{WT}$ data from this study will assist aquatic animal disease professionals in prescribing appropriate antibiotics for the treatment of streptococcosis caused by S. parauberis, which will help reduce the misuse and abuse of antibiotics in the aquaculture sector.

During the period from 2003 to 2020, a total of 470 Streptococcus species were isolated from farmed flounder in Jeju. Out of 470 isolates, 92 (19.6%) isolates and 378 isolates (80.4%) were identified as Streptococcus iniae and S. parauberis, respectively by multiplex PCR assay. During that period, the percentage of S. iniae decreased from 56.9% in 2003 to 0.0% in 2020 whereas that of S. parauberis increased from 43.1% in 2003 to 100% in 2020. In the PCR assay for serotyping, the isolated S. parauberis showed 3 subserotypes, Ia (34.9%), Ib/Ic (46.3%) and II (18.8%). In 2003 and 2004, serotype II was dominant at 59.1% and 50.0% of isolation rates, however between 2005 and 2009, subserotype Ib/Ic was dominant (57.6%, 86.0%, 84.6%, 57.9%, and 83.3%). After 2010, except for 2015, subserotype Ia was the most dominant one. In the last 3 years (2018 to 2020), subserotype Ia was most abundant (70%), followed by subserotype Ib/Ic (16-30%). Serotype II was not isolated in 2018, but in 2019 and 2020, it showed an increased tendency to 3.4% and 16.7%, respectively. It is believed that continuous monitoring is necessary for research on counter-measures against streptococcosis of flounder.
