Expression of colSR Genes Increased in the rpf Mutants of Xanthomonas oryzae pv. oryzae KACC10859

  • The rpf genes and $colS_{XOO1207}/colR_{XOO1208}$ were known to require for virulence of Xanthomonas oryzae pv. oryzae (Xoo). In Xoo KACC10331 genome, two more colS/colR genes, $colS_{XOO3534}$ (raxH)/$colR_{XOO3535}$ (raxR) and $colS_{XOO3762}/colR_{XOO3763}$ were annotated. The $colS_{XOO3534}/colR_{XOO3535}$ were known to control AvrXa21 activity and functions of $colS_{XOO3762}/colR_{XOO3763}$ were unknown in Xoo. To characterize the relationship between rpf and colS/colR genes, expression of colS/colR genes in Rpf mutants of Xoo were analyzed with quantitative reverse transcription PCR (qRT-PCR). Expressions of all three colS/colR genes increased in the rpfF mutant in which DSF synthesis is defective. Expression of $colS_{XOO1207}/col-R_{XOO1208}$, $colS_{XOO3534}/colR_{XOO3535}$ and $colS_{XOO3762}/colR_{XOO3763}$ increased 2, 2-7, 3-13 folds respectively. Expression of $colS_{XOO3534}$ and $colS_{XOO3762}$ also increased 2-4 folds in the rpfG mutant in which the signal from DSF is no longer transferred to down-stream. Expression of the other colS/colR genes was not significantly changed in the rpfG mutant compared to the wild type. Since RpfF and RpfG are responsible for DSF synthesis and signal transfer from DSF to down-stream to regulate virulence gene expression, these results suggest that the DSF and DSF-mediated signal regulate negatively three colS/colR genes in Xoo.

Velocity Measurement of Stream Water Surface Using Microwave (전자파를 이용한 하천수 표면유속 측정)

  • Applying microwave, a velocity measurement system has been developed in order to measure the velocity of stream water surface. It's main purpose is the measurement for high velocity of flood water. It is under the developing stage of experimental measurement system. The microwave surface velocity meter uses Doppler effects of microwave. It consists of a radio frequency(RF) part and that of signal processing. Thr RF part has the function of microwave oscillation, reception of reflected wave, and determination of Doppler frequency, etc. Signal processing designates amplification, fast Fourier transform, etx. Various measuring experiments were performed at bridges and a spillway of Taechong re-regulation dam with the microwave velocity meter. Verification test was also made through water tank of ship model test at Research Institute of Ships and Ocean Engineering. It shows 4% error inherent in A/D converter and additional several percentage errors from measurement circumstance. The measuring ranges are from 0.5 to 3.5 m/s. The result shows good linear relationship between carriage velocity and measured velocity, thus proves usefulness as a measuring instrument for flood water velocity. The instrument requires overall re-engineering procedure and number of data should be accumulated and analyzed to treat wind effects and random fluctuations of water surface.

Denitrification Potential and Denitrifier Abundance in Downstream of Dams in Temperate Streams (온대지역 댐 하류의 탈질화 능력과 탈질화균 분포)

  • Various studies have been conducted to investigate effects of dams on river ecosystems, but less information is available regarding damming impacts on downstream denitrification. We measured denitrification enzyme activity (potential denitrification rate) and denitrifier abundances (using nirS, nirK, and nosZ as markers) in dammed headstreams of the Nakdong River in South Korea. Sediments in Phragmites-dominated riparian areas and in-stream areas across streams (dammed vs. reference) with different streambed materials (gravel and sand) were sampled occasionally. We hypothesized that (i) the higher available N and C contents in sediments downstream of dams foster larger denitrifier communities than in the reference system and (ii) differences in potential denitrification rates across the systems correspond with denitrifier abundances. Despite 30 years of different hydrological management with dams and greater inorganic N and DOC contents in sediments downstream of dams, compared to the references, abundances of denitrifier communities and potential denitrification rates within the whole sediment were not significantly different across the systems. However, nirS and nosZ denitrifier abundances and potential denitrification rates were considerably increased in specific sediments downstream of dams (gravelly riparian and sandy in-stream) with regard to flooding events and seasonal temperature variation. nirK was not amplified in all sediments. Canonical correspondence analyses (CCA) revealed that the relationship between abundances of denitrifier communities and nutrient availabilities and potential denitrification rates was a weak one.

Changes in the Riverbed Landforms Due to the Artificial Regulation of Water Level in the Yeongsan River (인위적인 보 수위조절로 인한 영산강 하도 지형 변화)

  • A river bed which is submerged in water at high flow and becomes part of the river at low flow, serves as a bridge between the river and the land. The channel bar creates a unique ecosystem with vegetation adapted to the particular environment and the water pool forms a wetland that plays a very important role in the environment. To evaluate anthropogenic impacts on the river bed in the Middle Yeongsangang River, the fluvial landforms in the stream channel were analyzed using multi-temporal remotely-sensed images. In the aerial photograph of 2005 taken before the construction of the large weirs, oxbow lakes, mid-channel bars, point bars, and natural wetlands between the artificial levees were identified. Multiple bars divided the flow of stream water to cause the braided pattern in a particular section. After the construction of the Seungchon weir, aerial photographs of 2013 and 2015 revealed that most of the fluvial landforms disappeared due to the dredging of its riverbed and water level control(maintenance at 7.5El.m). Sentinel-2 images were analyzed to identify differences between before and after the opening of weir gate. Change detection was performed with the near infrared and shortwave infrared spectral bands to effectively distinguish water surfaces from land. As a result, water surface area of the main stream of the Yeongsangang River decreased by 40% from 1.144km2 to 0.692km2. A large mid-channel bar that has been deposited upstream of the weir was exposed during low water levels, which shows the obvious influence of weir on the river bed. Newly formed unvegetated point bars that were deposited on the inside of a meander bend were identified from the remotely sensed images. As the maintenance period of the weir gate opening was extended, various habitats were created by creating pools and riffles around the channel bars. Considering the ecological and hydrological functions of the river bed, it is expected that the increase in bar areas through weir gate opening will reduce the artificial interference effect of the weir.

Fibroin Enhances Insulin Sensitivity and Reverses Insulin Resistance in 3T3-L1 Adipocytes

  • Type 2 diabetes is characterized by hyperglycemia and hyperinsulinemia, features of insulin resistance. In vivo treatment of ob/ob mice with hydrolyzed fibroin reverses these pathological attributes (6). To explore the mechanism underlying this effect, we have used the 3T3-Ll adipocytes as a cell type which would represent the periphery, in vivo. Exposure of 3T3-Ll adipocytes to chronic insulin leads to the a 50% loss of insulin-stimulated glucose uptake. Chronic exposure to fibroin blocked, in part, the response to chronic insulin but also increased the sensitivity of control cells to the acute action of insulin. The later effect was most robust at physiological concentrations of insulin. Fibroin did not prevent the insulin-induced down-regulation of the insulin receptor or the tyrosine kinase activity associated with the receptor. Further, fibroin had no affect on the loss in activity of the insulin-sensitive down-stream kinase, Akt. Interestingly, fibroin accelerated glucose metabolism and glycogen turnover independent of insulin action. In addition, fibroin up-regulated GLUT1 which increased its expression at the cell surface and caused the redistribution of GLUT4 to the plasma membrane. Together, these later effects would lead to an improvement in hyperglycemia in vivo which would in turn reduce the need for insulin.

  • The global regulator, GacS (global antibiotic and cyanide sensor kinase), was required for the increased resistance to hydrogen peroxide occurring as cultures of the rhizobacterium, P. chlororaphis O6, matured. Specific stationary-phase peroxidase and catalase isozymes were absent in the GacS mutant, whereas a manganese-superoxide dismutase isozyme was expressed earlier and to a great extent than wild type. In the wild type cell, transcript accumulation of rpoS was higher in late logarithmic-phase cells than cells from mid logarithmic- or stationary-phase. Transcripts from rpoS in the GacS mutant were reduced in each of these growth phases compared to the wild type expression. The down stream sequence from rpoS lacked sequences encoding a small RNA, rsmZ, found in other pseudomonads and implicated in control of genes activated by the GacS system. These findings suggest that GacS-mediated regulation of RpoS plays role in control of oxidative stress in P. chlororaphis O6 by as yet an unknown mechanism.

  • In this paper, we analyzed various noise in the video stream system that have largely influence on EMI noise. We presented the remedy for these various noises and then designed the wireless video streaming system for a vehicle. To minimize EMI noise, we derived the improvement of noise characteristic from impedance matching, new design of the inner layer of the PCB line design and new design of high-speed data Interfaces. As a result, the final system showed 40[dBuV/m] and 47[dBuV/m] dB in the each regulation band.

Analysis of Problems and Improvement of Environmental Impact Assessment in Social-Economic Items Based on 19 Major Large Scale Development Projects (주요개발사업의 환경영향평가서 분석을 통한 사회경제항목평가의 문제점과 개선방안)

  • Assessment in Socio-economic items should be used to estimate social influence when policies and projects were introduced. To estimate current situation of socio-economic items 19 Environmental Impact Statement (EIS) of large scale projects were analyzed. The projects were selected based on magnitude of social impact when the projects were implemented. Environmental Impact Statement was categorized into line projects(road construction, railroad construction, stream development, etc), and surface projects(energy development, wetland reclamation, recreation and sport development, and housing and residential development, etc) thus being chosen for 19 projects in each categories. This report was based on the analysis of 7 items in socio-economic environmental items(i.e., population, residents, industry, public facilities, education, transport and historical monument). Most EIS did not follow the regulation suggested by Ministry of Environment, and only current circumstances were briefly described. Indifference of in-depth analysis of socioeconomic environmental items would influence the process of social and environment impact negatively in the midst of construction of National Projects such as Outer Circle Seoul Highways, Saemankeum Reclamation Projects, etc. This abrupt halt of construction was mostly based on a lack of public hearing or public participation. Socio-economic items are also very much lacking in quantitative method and strengthening socio-economic environmental items is needed via checklist or matrix that brings decision-makers better ideas objectively.

Analysis of Micropollutants Present in Raw Water Supplied for the Several Drinking Water Treatment Plants in Seoul (서울시 정수장 유입 원수내 미량유해물질의 조사)

  • This study investigated the micropollutants present in raw water supplied for the several drinking water treatment plants in Seoul. The target sample waters were collected from the several sites, such as Jayang (JY), Kuui (KI), Paldang (PD) and Kangbuk (KB) at the Han-River stream. The analytical method used in this study enable us to detect about 300 kinds of chemicals commonly found in surface water at ppt level by GC-ion trap MS. In this study, the consideration on the analytic results focused on the four hazardous organics, such as benzenes, phenols, phthalates and pesticides. The numbers of each detected micropollutant were 1~8 kinds for benzenes, 1~7 kinds for phenols, 5~7 kinds for phthalates and 1~9 kinds for pesticides. For the pesticides, the higher concentration was detected in the water samples collected from PD and KB adjacent to the farming area, and at June and July, which is the busy farming season. The total concentrations of each micropollutants detected at all the sites were significantly lower than those of drinking water regulation in Korea as well as other advanced countries. However, the frequently detected micropollutants requires the steady and precise monitoring for the effective management of drinking water source.

Makorin 1 Regulates Developmental Timing in Drosophila

  • The central mechanisms coordinating growth and sexual maturation are well conserved across invertebrates and vertebrates. Although mutations in the gene encoding makorin RING finger protein 3 (mkrn3) are associated with central precocious puberty in humans, a causal relationship has not been elucidated. Here, we examined the role of mkrn1, a Drosophila ortholog of mammalian makorin genes, in the regulation of developmental timing. Loss of MKRN1 in $mkrn1^{exS}$ prolonged the $3^{rd}$ instar stage and delayed the onset of pupariation, resulting in bigger size pupae. MKRN1 was expressed in the prothoracic gland, where the steroid hormone ecdysone is produced. Furthermore, $mkrn1^{exS}$ larvae exhibited reduced mRNA levels of phantom, which encodes ecdysone-synthesizing enzyme and E74, which is a down-stream target of ecdysone. Collectively, these results indicate that MKRN1 fine-tunes developmental timing and sexual maturation by affecting ecdysone synthesis in Drosophila. Moreover, our study supports the notion that malfunction of makorin gene family member, mkrn3 dysregulates the timing of puberty in mammals.
