While single strand DNAs have been widely used for the scaffold of brightly fluorescent silver nanoclusters (Ag NCs), double strand DNAs have not been as successful. Herein, we report a novel synthetic approach for bright Ag NCs using branched double strand DNAs as the scaffolds for synthesis. X-shaped DNA (X-DNA) and Y-shaped DNA (Y-DNA) effectively stabilized Ag NCs, and both X-DNA and Y-DNA resulted in brightly fluorescent Ag NCs. The concentration and molar ratio of silver and DNA were found important for the fluorescence efficiency. The brightest Ag NC with the photoluminescence quantum efficiency of 19.8% was obtained for the reaction condition of 10 ${\mu}M$ X-DNA, 70 ${\mu}M$ silver, and the reaction time of 48 h. The fluorescence lifetime was about 2 ns for the Ag NCs and was also slightly dependent on the synthetic condition. Addition of Cu ions at the Ag NC preparations resulted in the quenching of Ag NC fluorescence, which was different to the brightening cases of single strand DNA stabilized Ag NCs.

This paper presents results of one-year monitoring on prestressing force of a 7-wire steel post-tensioning strand which is installed in a UHPC(ultra high performance concrete) bridge with 11.0 m long, 5.0 m wide, and 0.6 m high by using a FBG-encapsulated 7-wire steel strand. The initial prestressing forces and the prestress changes during a vehicle load test were measured using the FBG-encapsulated strand. The results show that the FBG-encapsulated 7-wire strand is very effective for monitoring the prestress forces even the change in the tension force is very small. Additionally, it was indicated that selection of the thermal expansion coefficient which is used for the temperature correction shall be carefully carried out.

Recently, a lot of interest has been shown in structural maintenance managements of civil infrastructures. Many researchers have been conducted on various maintenance techniques and repair materials. Among other fiber materials the carbon fiber materials are especially focused on maintenance management of Highway Bridges. Extensive work has been done on Carbon Fiber Sheet (CFS). Nevertheless, Carbon Fiber Strand Sheet (CFSS) is a newly developed material, on which limited work has been done until now. Therefore, in this study bonding the CFSS to RC slab specimen and fatigue resistance evaluation has been conducted. The results demonstrated an increase of 25.3 times more reinforcement of RC slab compared to non-reinforced RC slab. Moreover, compared to CFS-bonded RC slab, The CFSS-bonded RC slab showed 1.2 times greater reinforcement.
Composites with various layer constructions involving the ratio of S/P were produced from wood strands(S) and particles(P) (Cryptomeria japonica D.Don) to measure various Young's moduli and anisotropy by ultrasonics. As a result, static Young's moduli of composites were almost same as dynamic Young's moduli obtained from natural frequency. However, these were smaller than those evaluated from ultrasonic wave propagation velocity. The differences between propagation velocity of the parallel(${\parallel}$) and perpendicular(${\bot}$) in-plane direction resulted in a tendency of anisotropy. The tendency of anisotropy was larger in three-layer constructions than in seven-layer constructions. The differences of strand surface layers showed larger values than those of particle surface layers. Also, composites with higher weight ratio of strand had a tendency to propagate rapidly in each direction. In contrast to these results, the propagation velocity in the thickness direction had a tendency to propagate rapidly in composites with particle surface layers and the lower weight ratio of strand.

RNA-RNA interactions and the dynamic RNA conformations are important regulators in virus replication in several RNA virus systems and may also involved in the regulation of many important virus life cycle phases, including translation, replication, assembly, and switches in these important stages. The 5' non-translated region of Potato virus X(PVX) contains multiple cis-acting elements that facilitate various viral processes. It has previously been proposed that RNA-RNA interactions between various RNA elements present in PVX RNA genome are required for PVX RNA accumulation(Hu et al., 2007; Kim and Hemenway, 1999). This model was based on the potential base-pairing between conserved sequence elements at the upstream of subgenomic RNAs(sgRNAs) and at the 5' and 3' end of RNA genome. We now provide more evidence that RNA-RNA base-pairing between elements present at the 5' end and upstream of each sgRNA is required for efficient replication of genomic and subgenomic plus-strand RNA accumulation. Site-directed mutations introduced at the 5' end of plus-strand RNA replication defective mutant(${\Delta}12$) increasing base-pairing possibility with conserved sequence elements located upstream of each sgRNAs restored genomic and subgenomic plus-strand RNA accumulation and caused symptom development in inoculated Nicotiana benthamiana plants. Serial passage of a deletion mutant(${\Delta}8$) caused more severe symptoms and restored wild type sequences and thus retained possible RNA-RNA base-pairing. Altogether, these results indicate that the RNA element located at the 5' end of PVX genome involved in RNA-RNA interactions and play a key role in high-level accumulation of plus-strand RNA in vivo.

It is growing the application of the removal ground anchor with tension force for earth retaining constructions in the downtown. Nowadays, we can find the compression dispersion anchor on many site. But, it is occur some probelems in behabior of anchors because of impossible to tense p.c strand uniformly with existing equipment due to different length of p.c strand. So we tried to tense each p.c strand uniformly with auto back equipment in-situ test. This study compared and analyzed apply to elastic theory in-situ test results of an existing equipment with those of auto back equipment. As a result of the test, It has been proved that differences of tension force in the existing equipment increases with increasing the number of p.c strand. This can cause an ultimate failure of the concentrated p.c strand and a shear failure of ground. So it has been proved that auto back equipment is necessary.
During meiosis, exchange of DNA segments occurs between paired homologous chromosomes in order to produce recombinant chromosomes, helping to increase genetic diversity within a species. This genetic exchange process is tightly controlled by the eukaryotic RecA homologs Rad51 and Dmc1, which are involved in strand exchange of meiotic recombination, with Rad51 participating specifically in mitotic recombination. Meiotic recombination requires an interaction between homologous chromosomes to repair programmed double-strand breaks (DSBs). In this study, we investigated the budding yeast meiosis-specific proteins Hop2 and Sae3, which function in the Dmc1-dependent pathway. This pathway mediates the homology searching and strand invasion processes. Mek1 kinase participates in switching meiotic recombination from sister bias to homolog bias after DSB formation. In the absence of Hop2 and Sae3, DSBs were produced normally, but showed defects in the DSB-to-single-end invasion transition mediated by Dmc1 and auxiliary factors, and mutant strains failed to complete proper chromosome segregation. However, in the absence of Mek1 kinase activity, Rad51-dependent recombination progressed via sister bias in the $hop2{\Delta}$ or $sae3{\Delta}$ mutants, even in the presence of Dmc1. Thus, Hop2 and Sae3 actively modulate Dmc1-dependent recombination, effectively progressing homolog bias, a process requiring Mek1 kinase activation.

Fludarabine, a chain terminating anti-cancer drug, is a purine analogue that causes DNA strand breaks in normal cells. In this study, we determined if A. melanocarpa and Korean red ginseng extract mixture reduce cytotoxicity of fludarabine. Treatment of HaCaT cells with $10{\mu}M$ of fludarabine for 24 hours decreased cell viability and increased DNA strand breaks. Treatment of A. melanocarpa and Korean red ginseng extract mixture for 24 hours increased cell viability as compared with single extract treatment. The protective effect of these extracts on cell activity increased in a concentration-dependent manner. DNA strand breaks induced by fludarabine decreased as concentration of extract mixture increased. p-H2AX level, a marker of DNA strand breakage, decreased depending on the concentration of extract mixture. The effect of mixed extract of A. melanocarpa and Korean red ginseng on DNA damage is due to the anti-oxidative effect of A. melanocarpa and signal transmission through glucocorticoid receptor upon binding of saponin of Korean red ginseng.

Background The management of flexor tendon injuries has evolved in recent years through industrial improvements in suture materials, refinements of repair methods, and early rehabilitation protocols. However, there is no consensus on the ideal suture material and technique. This study was conducted to compare the tensile strength, repair time, and characteristics of 4-strand cruciate, modified Kessler, and 4-strand horizontal intrafiber barbed sutures for flexor tenorrhaphy with a 12-mm suture purchase length in an animal model. Methods The right third deep flexors of 60 adult Leghorn chicken feet were isolated and repaired with a 12-mm suture purchase length. The tendons were randomly assigned to three groups of equal number (n=20 each). Groups 1 and 2 received 4-strand cruciate and modified Kessler repair with conventional suture materials, respectively. A 4-strand horizontal intrafiber barbed suture technique was used in group 3. The repaired tendons were biomechanically tested for tensile strength, 2-mm gap resistance, and mode of failure. Repair times were also recorded. Results The maximum tensile strength until failure was $44.6{\pm}4.3N$ in group 1, $35.7{\pm}5.2N$ in group 2, and $56.7{\pm}17.3N$ in group 3. The barbed sutures were superior to the other sutures in terms of the load needed for 2-mm gap formation (P<0.05). Furthermore, the barbed sutures showed the shortest repair time (P<0.05). Conclusions This study found that 4-strand horizontal intrafiber barbed suture repair with a 12-mm purchase length in a chicken flexor tendon injury model showed promising biomechanical properties and took less time to perform than other options.
This paper deals with an experimental and numerical study of a mono-strand wedge anchor head mechanism. First, the experimental program is presented and monitored data such as wedge slippage, anchor deflection and strain distributions along external peripheral surfaces of the anchor head are presented and discussed. In accordance with the experimental set up, these data concern only the global behaviour of the mechanism and cannot provide valuable information such as internal stress-strains distributions, stress concentrations and percentage of yielded volume. Therefore, the second part of this paper deals with the development of an efficient numerical finite element model capable of providing mechanism of the core information. The numerical model which includes all kinematics/material/contact non-linearities is first calibrated using experimental data. Subsequently, a numerical study of the anchorage mechanism is performed and its behaviour is compared to the behaviour of a slightly geometrically modified mechanism where the external diameter has been increased by 5 mm. Finally, different topics influencing the anchorage mechanism behaviour are addressed such as lubrication and wedge shape.
