Identifying carbapenem-resistant Enterobacteriaceae (CRE) is necessary to prevent nosocomial CRE infection outbreaks. Here, a rapid identification method with reduced enrichment time was developed without compromising accuracy. A total of 49 rectal swabs requested for CRE screening at the Department of Diagnostic Medicine at Hospital B in Busan, Korea, were included in this study. Specimens were inoculated on MacConkey and CHROMID Carba media either directly or following enrichment for 3, 6, and 24 h in 100 μl trypticase soy broth containing an ertapenem disk. The enriched cultures were further inoculated on CHROMID Carba or MacConkey media containing an ertapenem disk. In total, 19 CRE and 5 carbapenem-intermediate Enterobacteriaceae isolates were obtained from the 49 swabs. Among the 19 CRE isolates, carbapenemase-producing Enterobacteriaceae constituted 13 strains. Moreover, of the 19 CRE isolates, 16 (81.25%) and 17 (88.24%) were identified from the direct cultures on MacConkey and CHROMID Carba media, respectively. After 3 h of enrichment, the proportions of the CRE identified in the media were: MacConkey medium, 16/19 (81.25%); CHROMID Carba medium, 17/19 (88.24%); and MacConkey medium containing an ertapenem disk, 17/19 (88.24%). The detection rates after 6 h of enrichment were the same for all three media (19/19 strains, 100%), whereas those after 24 h of enrichment were 21, 22, and 24 strains, respectively, but included false positives. These findings suggest that a 6-h enrichment before inoculation on the CHROMID Carba medium is optimal for the rapid and accurate detection of CRE in clinical samples.
In the present study, we demonstrate that the growth of salt-stressed pepper plants is improved by inoculation with plant growth-promoting rhizobacteria (PGPR). Three PGPR strains (Microbacterium oleivorans KNUC7074, Brevibacterium iodinum KNUC7183, and Rhizobium massiliae KNUC7586) were isolated from the rhizosphere of pepper plants growing in saline soil, and pepper plants inoculated with these PGPR strains exhibited significantly greater plant height, fresh weight, dry weight, and total chlorophyll content than non-inoculated plants. In addition, salt-stressed pepper plants that were inoculated with B. iodinum KNUC7183 and R. massiliae KNUC7586 possessed significantly different total soluble sugar and proline contents from non-inoculated controls, and the activity of several antioxidant enzymes (ascorbate peroxidase, guaiacol peroxidase, and catalase) was also elevated in PGPR-treated plants under salt stress. Overall, these results suggest that the inoculation of pepper plants with M. oleivorans KNUC7074, B. iodinum KNUC7183, and R. massiliae KNUC7586 can alleviate the harmful effects of salt stress on plant growth.
The potential ability of Button mushroom compost (BMC) to solubilize heavy metals was estimated with metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strains in BMC were isolated for investigating the mobilization of metals in soil or plant by the strains and identified according to 16S rRNA gene sequence analysis. When metal solubilization potential of BMC was assessed in a batch experiment, the BMC was found to be capable of solubilizing metals in the presence of metals (Co, Pb and Zn) and the results showed that inoculation of BMC could increase the concentrations of water soluble Co, Pb and Cd by 35, 25 and 45% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 27, 25 and 28% respectively in Co, Pb and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction of Co, Pb and Zn from contaminated soils.
Lactobacillus pentosus K1-23 was selected from among 25 lactic acid bacterial strains owing to its high inhibitory activity against several pathogenic bacteria, including Escherichia coli, Salmonella typhimurium, S. gallinarum, Staphylococcus aureus, Pseudomonas aeruginosa, Clostridium perfringens, and Listeria monocytogenes. Additionally, among 13 strains of Aureobasidium spp., A. pullulans NRRL 58012 was shown to produce the highest amount of ${\beta}$-glucan ($15.45{\pm}0.07%$) and was selected. Next, the optimal conditions for a solid-phase mixed culture with these two different microorganisms (one bacterium and one yeast) were determined. The optimal inoculum sizes for L. pentosus and A. pullulans were 1% and 5%, respectively. The appropriate inoculation time for L. pentosus K1-23 was 3 days after the inoculation of A. pullulans to initiate fermentation. The addition of 0.5% corn steep powder and 0.1% $FeSO_4$ to the basal medium resulted in the increased production of lactic acid bacterial cells and ${\beta}$-glucan. The following optimal conditions for solid-phase mixed culture were also statistically determined by using the response surface method: $37.84^{\circ}C$, pH 5.25, moisture content of 60.82%, and culture time of 6.08 days for L. pentosus; and $24.11^{\circ}C$, pH 5.65, moisture content of 60.08%, and culture time of 5.71 days for A. pullulans. Using the predicted optimal conditions, the experimental production values of L. pentosus cells and ${\beta}$-glucan were $3.15{\pm}0.10{\times}10^8CFU/g$ and $13.41{\pm}0.04%$, respectively. This mixed culture may function as a highly efficient antibiotic substitute based on the combined action of its anti-pathogenic bacterial and immune-enhancing activities.
A total of eight strains of avian reoviruses were isolated from chickens with arthritis or stunted growth. The isolations were made from broilers or broiler breeders under 12 weeks of age. The viruses had a typical morphology of reoviruses with double capsid layers and 81nm of diameter. In agar gel precipitation tests, the isolates reacted with antisera prepared against S-1133 or R-1 strains of avian reoviruses and cross reacted with S-1133 antigen. They did not agglutinated RBC's from day-old chicks, adult chickens, guinea pigs, and horses. The isolates showed strong resistance against the treatments of chloroform, IUdR, and heat, When infectivities of the viruses were titrated in cell cultures of chicken embryo fibroblast, chicken embryo liver, and Vero cells, similar end points reached four to five days after inoculation, regardless of tell types and virus inoculation time, either inoculated simultaneously at the time of cell seeding or on confluency. Mean times of mortality of chicken embryos inoculated with the isolates via the chorioallantoic membrane ranged from 54 to 59 hours and that of S-1133 strain was 73 hours.
Objective: This study identified the major lactic acid bacteria (LAB) strains from different fermented total mixed rations (FTMRs) via metataxonomic analysis and evaluated the ability of their standard strain as ensiling inoculants for corn stover silage. Methods: The bacterial composition of eight FTMRs were analyzed by 16S rDNA sequencing. Corn stover was ensiled without LAB inoculation (control) or with 1×106 cfu/g LAB standard strain (Lactobacillus vaginalis, Lactobacillus reuteri, Lactobacillus helveticus, or Lactobacillus paralimentarius) selected from the FTMRs or 10 g/t commercial silage inoculant (CSI) around 25℃ for 56 days. For each inoculation, a portion of the silage was sampled to analyze ensiling characteristics at time intervals of 0, 1, 3, 7, 14, 28, and 56 days, gas production (GP), microbial crude protein and volatile fatty acids as the measurements of rumen fermentation characteristics were evaluated in vitro with the silages of 56 days after 72 h incubation. Results: Lactobacillus covered >85% relative abundance of all FTMRs, in which L. pontis, L. vaginalis, L. reuteri, L. helveticus, and L. paralimentarius showed >4% in specific FTMRs. CSI, L. helveticus, and L. paralimentarius accelerated the decline of silage pH. Silage inoculated with L. paralimentarius and CSI produced more lactic acid the early 14 days. Silage inoculated with L. paralimentarius produced less acetic acid and butyric acid. For the in vitro rumen fermentation, silage inoculated with CSI produced more potential GP, isobutyric acid, and isovaleric acid; silage inoculated with L. helveticus produced more potential GP and isovaleric acid, silage inoculated with L. paralimentarius or L. reuteri produced more potential GP only. Conclusion: The standard strain L. paralimentarius (DSM 13238) is a promising ensiling inoculant for corn stover silage. The findings provide clues on strategies to select LAB to improve the quality of silage.
28 porcine enteroviruses were isolated from 86 pig-feces of 9 swine farms located in south region, Chung-buk, from March to September 1990. Physico-chemical properties and pathogenicity of isolates were investigated. Results obtained throughout experiments are summarized as follows. According to the age, weanlings(40-90 days), sucklings(10-30 days) and adult pigs(6 months over) showed the isolation rate of 67%. 8% and 4%, respectively. By physico-chemical tests, YD-90/22, YD-90/43 and YD-90/64 strains were found to be ether, chloroform and PH stable. Nucleic acid test suggests the virus to have a DNA genome. Most of the Isolates were not evident of hemagglutinin using erythrocytes from various mammalian & avian. 22 strains among the isolates were shown CPE type I and the remainders were CPE type II. 3 strains among isolates of CPE type I strains were neutralized with high titers to serotype 2 antiserum. In the study on virus growth curve in PK-l5 cells, YD-90/22, YD-90/43 and YD-90/64 strains showed the maximum infectivity titers($10^{6.0}-l0^{6.5} TCID({50}ml$) at 4days post inoculation(PI). When 30 day-old commercial piglets were inoculated only intraoral route with the YD-90/22 strain at $10^{6.0} TCID_{50}ml,$ piglets not showed the symptoms. But piglets inoculated by intramuscle route, intraoral and intramuscle route after pretreat with dexamethasone(2.5mg /kg) for 5 days were shown the symptoms of anorexia, diarrhea, pyrexia and ataxia at 4th-6th days PI. The viral reisolation in the virus-inoculated piglets was examined from feces. The viruses were recovered intermittently from 2nd to 16th day PI and at 4th-6th day PI, all piglets excreted viruses.

Strains of Toxoplasma gondii in Brazil are highly genetically diverse compared to strains from North America and Europe. Dogs are epidemiologically important because they act as sentinels for T. gondii infections in humans and are good indicators of environmental contamination. The aim of this study was to isolate and genetically characterize T. gondii strains from tissues of naturally infected Brazilian dogs. For this study, 21 blood samples were collected from dogs at the Zoonosis Control Centers of $Ilh{\acute{e}}us$ and Itabuna cities, Bahia, Brazil. The sera were examined for T. gondii antibodies using the indirect hemagglutination test. Brains and hearts of seropositive dogs were bioassayed in mice to isolate and characterize T. gondii parasites by PCR-RFLP using 10 genetic markers (SAG1, newSAG2, SAG3, BTUB, c22-8, c29-2, GRA6, PK1, APICO, and L358). However, T. gondii was isolated from only 4 (57.1%) dogs, designated TgDgBr6, 13, 17, and 21. All strains were virulent, causing clinical changes (rough hair coat, lethargy, and abdominal distention) and the death of all mice within 8-20 days after inoculation. Genetic analysis of these 4 T. gondii isolates revealed 4 distinct genotypes with different clonal lineage combinations (types I, II, and III) and 2 atypical alleles. Using PCR-RFLP with several markers, this study contributes to evaluations of the genetic diversity of strains circulating in Brazil.
Bacterial leaf blight is a major disease of eucalypt, especially under nursery conditions. Different bacterial species have been associated with the disease in several countries, and despite its importance worldwide, it is not clear to date whether similar disease symptoms are caused by the same or by different etiological agents. In this study, 43 bacterial strains were isolated from blighted eucalypt leaves collected in different geographic areas of Brazil and inoculated onto a susceptible eucalypt clone. Polyphasic taxonomy, including morphological, physiological, biochemical, molecular, and pathogenicity tests showed that only certain strains of Xanthomonas axonopodis caused symptoms of the disease. Strains varied in their aggressiveness, but no correlation with geographic origin was observed. MLSA-based phylogenetic analysis using concatenated dnaK, fyuA, gyrB and rpoD gene sequences allocated the strains in a well-defined clade, corresponding to Rademarker's group RG 9.6. Inoculation of nineteen plant species belonging to seven botanical families with representative strain LPF 602 showed it to be pathogenic only on Eucalyptus spp, and Corymbia spp. Based on distinct biochemical and pathogenic characteristics that differentiate the eucalypt strains from other pathovars of the X. axonopodis species, here we propose their allocation into the new pathovar X. axonopodis pv. eucalyptorum pv. nov.
The necessity to develop economical and eco-friendly technologies is steadily increasing. Plant growth promoting rhizomicrobial strains PGPR are a group of microorganisms that actively colonize plant roots and increase plant growth and yield. Pot experiments were used to investigate the potential of some rhizobacterial strains to enhance the Brassica rapa growth. Microbial strains were successfully isolated from the rhizosphere of Panax ginseng and characterized based on its morphological and plant growth promotion characters. Surface disinfected seeds of Wisconsin Fast B. rapa were inoculated with the selected PGPR microorganisms. The different pots treatments were inoculated by its corresponding PGPR ($10^7cfu\;mL^{-1}$) and incubated in the growth chamber at $25^{\circ}C$ and 65% RH, the light period was adjusted to 24 hours (day). NPK chemical fertilizer and trade product (EMRO, USA) of effective microorganisms as well as un-inoculated control were used for comparison. Plants harvested in 40 days were found to have significant increase in leaf chlorophyll units and plant height and also in dry weight of root and shoot in the inoculated seedlings. Root and shoot length and also leaf surface area significantly were increased by bacterial inoculation in sterile soil. The study suggests that Rhodobacter capsulatus and Azotobacter chroococcum are beneficial for B. rapa growth as they enhance growth and induced IAA production and phosphorus solubilization. This study presents some rhizomicrobial strains that significantly promoted growth of Wisconsin Fast Plant B. rapa in pot experiment under different soil conditions.
