Isolation of Microorganisms and Development of Microbial Augmentation for Treatment of Industrial Wastewater containing Ammonium Nitrogen (암모니아성 질소함유 산업폐수처리를 위한 미생물의 분리 및 복합미생물제제의 개발)

  • For effective treatment of wastewater containing ammonium nitrogen (NH4-N), AT2, AT9, and AT12 strains, having high total organic carbon (TOC) removal capability, and FN47, possessing excellent ammonia nitrogen removal capability present in the activated sludge in the aeration tank of food wastewater treatment plants, were isolated and identified. The cells of these isolated strains were used for microbial augmentation with FIW-1 in the defatted rice bran as a medium to treat industrial wastewater. The investigation of the cultural characteristics of these isolated strains in the aeration tank showed that the affinities for substrate of the isolated strains were extremely high, of which AT12 (Alcaligenes sp. AT12) was the highest among the isolated strains. Ammonium nitrogen removal efficiency in the food wastewater was 71% in the isolated strain FN47 (Microbacterium sp. FN47) treatment group. When only activated sludge was added in the lab scale pilot using food wastewater during continuous culture experiment, the TOC removal efficiency was 63%. Meanwhile, the removal efficiency of 92% was obtained when the microbial augmentation FIW-1 for wastewater treatment was applied. In addition, the chemical oxygen demand (COD) level from the effluent wherein microbial augmentation FIW-1 was input for the initial three days in the wastewater treatment site experiment showed a treatment rate of about 43%, which was increased to 62% after an elapse of 5 days.

  • This study was performed to investigate the effects of microbial augmentation on the biological treatment of paper mill wastewater. Three bacteria (KN11, KN13, KN27) capable of degrading aromatic compounds and a bacterial strain (GT21) producing an extracellular cellulase were isolated from soil and wastewater by selective enrichment culture. Through morphological, physiological, and biochemical taxonomies, isolated strains of KN11, KN13, KN27, and GT21 were identified as Acinetobacter sp., Neisseria sp., Bacillus sp., and Pseudomonas sp. and named Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, respectively. For analysis of non-biodegradable and chemical oxygen demand (COD)-increasing matter in a paper mill wastewater, we utilized GC/MS to detect aromatic compounds and their derivatives containing several substituted functional groups. The microbial augmentation, J30 formulated with the mixture of bacteria including Acinetobacter sp. KN11, Neisseria sp. KN13, Bacillus sp. KN27, and Pseudomonas sp. GT21, was used for the treatment of paper mill wastewater. The optimum temperature and pH for COD removal of the microbial augmentation, J30, were $30^{\circ}C$ and 7.5, respectively. For evaluation of the industrial applicability of the microbial augmentation, J30 in the pilot test, treatment efficiency was examined using paper mill wastewater. The microbial augmentation, J30, showed a COD removal rate of 87%. On the basis of the above results, we designed the wastewater treatment process of the activated sludge system.

Antifungal Activity of Bacillus sp. GJ-1 Against Phytophthora capsici (Bacillus sp. GJ-1의 Phytophthora capsici에 대한 항진균활성)

  • Phytophthora capsici is one of major limiting factors in production of pepper and other important crops worldwide by causing foliage blight and rot on fruit and root. Increased demand for the replacement of fungicides has led to searching a promising strategy to control the fungal diseases. To meet eco-friendly agriculture practice, we isolated microorganisms and assessed their beneficial effects on plant health and disease control efficacy. A total of 360 bacterial strains were isolated from rhizosphere soil of healthy pepper plants, and categorized to 5 representative isolates based on colony morphology. Among the 5 bacterial strains (GJ-1, GJ-4, GJ-5, GJ-11, GJ-12), three bacterial strains (GJ-1, GJ-11, GJ-12) presented antifungal activity against P. capsici in an fungal inhibition assay. In phosphate solubilization and siderophore production, the strain GJ-1 was more effective than others. The strain GJ-1 was identified as Bacillus sp. using 16S rDNA analysis. Bacillus sp. GJ-1 was also found to be effective in inhibiting other plant pathogenic fungi, including Rhizoctonia solani, Pythium ultimum and Fusarium solani. Therefore, the Bacillus sp. GJ-1 can serve as a biological control agent against fungal plant pathogens.

Study of the Behavior of Concrete Slab Track on Earthwork According to the Variation of Train Axle Load and Speed (열차하중, 속도변화에 따른 토공상 콘크리트 슬래브궤도의 거동특성연구)

  • In recent high speed rail way, the operating speed of train is enhanced and the introduction of EMU train vehicles is increased. In addition, as expected the demand of the concrete slab track and the trend of design cross-section reduction, the clear behavior of evaluation of internal slab layer is demanded about the variation of design load and speed. The purpose of this study is to evaluate and identify the mechanical behavior pattern of concrete slab track and track-road bed with the variation of axle load and train speed. To this end, the behavior of TCL and HSB was evaluated in according to the variation of axle load and speed. And the analysis results and the data measured TCL strain sensor, which was embedded in TCL slab under installation on Honam high speed railway, was analyzed. The analysis result shows that the strain are increasing in according to the speed-up of train, and line regression was obtained from measured data. Analysis data of the state of bonding condition of slab layer and measured data was analyzed. It is conducted that the TCL layer stress of HEMU 430X, which of axle load, is lighter was similar to the stress of KTX-Honam, the standard deviation of measured stress is dramatically increased.

  • While the recent high demand for mega-tall buildings has led to the development of high-performance and high-strength steels, the requirements for architectural-structure-performance steel have been raised as engineers recognize the potential damage that an earthquake can wreak on a tall building. A 600MPa-class steel has emerged to meet such need, and many studies are currently exploring its practical applications on civil engineering works and mega-tall buildings. The available data on the horizontal-force behaviors of structures built with such new steel, however, are still insufficient. There is an urgent need to look into its design data, especially its toughness, and to compare the plastic strain ratios of column-to-beam connections using high-strength steel and regular steel. One of the first studies on the behavior of a column-to-beam connection using 600MPa-class steel (SM570 TMC), this thesis analyzes such steel's structural performance by conducting a structural test on seismic resistance on a full-scale column-to-beam welded connection with non-scallop and recommended-scallop details. Compared with the previous studies on SM490, this thesis evaluates the weldability of SM570 TMC and presents the latter's seismic design data for use in testing its practical application.

A Study on the Fatigue Characteristics and Life Prediction of the Tire Sidewall Rubber (타이어 사이드월 고무의 피로특성 및 수명예측에 관한 연구)

  • In the case of the UHP (Ultra high performance) tire that the demand has increased rapidly, compared with the commonly used tire, severe deformation has been observed because of the low aspect ratio. When repeated deformations are applied to the sidewall rubber, accumulated fatigue damage may cause fatigue failure. Thus, the evaluation of the durability of the tire sidewall rubber has become a very important issue to prevent accidents that occur while the vehicle is running. However, the research and design criteria for the durability performance of the tire sidewall rubber hardly exist. In this study, we suggest a lifetime prediction formula using strain energy density obtained by performing tensile tests and fatigue tests on two different kinds of the tire sidewall compounds. Additionally, the applicability of our findings for low fuel consumption tires was reviewed by converting the fatigue life of the sidewall rubber into the expected mileage of the tire.

Investigating the use of wollastonite micro fiber in yielding SCC

  • Self compacting concrete (SCC) has good flowability, passability and segregation resistance because of voluminous cementitious material & high coarse aggregate to fine aggregate ratio, and high free water availability. But these factors make it highly susceptible to shrinkage. Fibers are known to reduce shrinkage in concrete mixes. Until now for conserving cement, only pozzolanic materials are admixed in concrete to yield a SCC. Hence, this study compares the use of wollastonite micro fiber (WMF), a cheap pozzolanic easily processed raw mineral fiber, and flyash in yielding economical SCC for rigid pavement. Microsilica was used as a complimentary material with both admixtures. Since WMF has large surface area ($827m^2/kg$), is acicular in nature; therefore its use in yielding SCC was dubious. Binary and ternary mixes were constituted for WMF and flyash, respectively. Paste mixes were tested for compatibility with superplasticizer and trials were performed on a normal concrete mix of flexural strength 4.5 MPa to yield SCC. Flexural strength test and restrained shrinkage test were performed on those mixes, which qualified self compacting criteria. Results revealed that WMF admixed pastes have high water demand, and comparable setting times to flyash mixes. Workability tests showed that 20% WMF with microsilica (5-7.5%) is efficient enough in achieving SCC and higher flexural strength than normal concrete at 90 days. Also, stress rate due to shrinkage was lesser and time duration for final strain was higher in WMF admixed SCC which encourages its use in yielding a SCC than pozzolanic materials.

The Effect of Storage Period and Temperature on Egg Quality in Commercial Eggs

  • Consumers demand fresh and high-quality eggs. Egg quality may be represented by shell color, shell weight, egg weight, shell thickness, shell density, albumen height, yolk color, albumen pH and viscosity. Various factors such as strain, age of hen, storage temperature, humidity, the presence of $CO_2$ and storage time affect egg quality. Therefore, we investigated the effects of storage time and temperature on egg quality to define the freshness of Korean market eggs. A total of 1,800 eggs were used for this experiment and were separated into 45 treatments with 40 eggs in each. The treatments were consisted of 15 storage periods (2 d to 30 d) and 3 storage temperatures ($2^{\circ}C$, $12^{\circ}C$, $25^{\circ}C$). Each egg was weighed and broken, and the height of the thick albumen, Haugh units (HU), egg shell color and yolk color were measured by a QCM+system. We also observed the physiochemical properties of eggs such as yolk pH, albumen pH and albumen viscosity. The egg weight, shell weight, albumen height, HU and albumen viscosity significantly decreased with increasing storage time and temperature. However, the albumen and yolk pH significantly increased with increasing storage period and temperature. The interaction effects between the storage period and temperature were significant for shell weight, shell density, egg weight, albumen height, HU, yolk color, yolk pH, albumen pH and albumen viscosity. In the analysis of the correlation with egg quality, the storage temperature exhibited a higher correlation coefficient than the storage period. In conclusion, storage time and temperature are the major factors affecting egg quality, but the storage temperature is a more sensitive determinant of egg quality deterioration compared with the storage period.

This research examines the moment resis tance-type jointing method using structural laminated timber and H-section stel aiming at development of the two-direction frame for lar ge 9 mm and 12 mm) of the H section. Therefore, we conducted the experiment with bending test of the joints to investigate the s tifnes, strength, strain distributions of laminated timber an d of the flange of the H section, and failure paterns. As shown in the results, t he joints with a flange thicknes of 9 mm and 12 m have superi or strength with a flange thicknes of 9 mm and 12 mm were very large, whic h confirmed the high level of energy absorption of such structure s.

Performance of innovative composite buckling-restrained fuse for concentrically braced frames under cyclic loading

  • Concentrically Braced Frames (CBFs) are commonly used in the construction of steel structures because of their ease of implementation, rigidity, low lateral displacement, and cost-effectiveness. However, the principal disadvantage of this kind of braced frame is the inability to provide deformation capacity (ductility) and buckling of bracing elements before yielding. This paper aims to present a novel Composite Buckling Restrained Fuse (CBRF) to be utilized as a bracing segment in concentrically braced frames that allows higher ductility and removes premature buckling. The proposed CBRF with relatively small dimensions is an enhancement on the Reduced Length Buckling Restrained Braces (RL-BRBs), consists of steel core and additional tensile elements embedded in a concrete encasement. Employing tensile elements in this composite fuse with a new configuration enhances the energy dissipation efficiency and removes the tensile strength limitations that exist in bracing elements that contain RL-BRBs. Here, the optimal length of the CBRF is computed by considering the anticipated strain demand and the low-cyclic fatigue life of the core under standard loading protocol. An experimental program is conducted to explore the seismic behavior of the suggested CBRF compare with an RL-BRB specimen under gradually increased cyclic loading. Moreover, Hysteretic responses of the specimens are evaluated to calculate the design parameters such as energy dissipation potential, strength adjustment factors, and equivalent viscous damping. The findings show that the suggested fuse possess a ductile behavior with high energy absorption and sufficient resistance and a reasonably stable hysteresis response under compression and tension.
