We have previously established a system for plant regeneration through somatic embryogenesis and Agrobacterium-mediated transformation of Korean ginseng. In this study to produce a fungus-resistant plant, we introduced a bean chitinase gene into ginseng using the transformation system. A binary vector pChi/748 was constructed by introducing the bean basic chitinase gene into EcoRI site of pGA748 which carries the CaMV 35S promoter governing the introduced gene and neomycin phosphotransferase II(NPT-II)gene as a positive selection marker. Cotyledonary explants were cocultured with A. tumefaciens strain LBA4404 harboring the binary vertor pChi/748 for 48 h, and transferred to MS medium supplemented with l mg/L2,4-D,0.1mg/L kinetin, 100 mg/L kanamycin, and 500mg/L carbenicillin. Kanamycin-resistant calli were formed on the cut surface of cotyledonary explants after one month of culture, and subsequently they gave rise to somatic embryos. Upon transfer onto medium containing 1 mg/L each of BA and GA$_3$, most of them converted to plantlets after 5 weeks of culture. The genomic DNA of eight kanamycin-resistant regenerants was subjected to polymerase chain reaction (PCR) using two specific 21-mer oligonucleotides derived from the chitinase gene. PCR-Southern blot analysis confirmed that the chitinase gene was incorporated into six out of the eight regenerants..

The main purpose of this study is to evaluate the potential of producing bioethanol from yellow poplar ($Liriodendron$$tulipifera$) wood chips by oxalic acid pretreatment and to examine the pretreatment conditions by response surface methodology (RSM). Based on $2^3$ factorial design, adjusted variables were reaction temperature ($^{\circ}C$), residence time (min), and acid loading (g/g), and a series of distinct 15 experimental conditions was organized with duplication at central point (total 16 performances). After pretreatment, simultaneous saccharification and fermentation (SSF) was subjected on solid fraction with yeast strain $Pichia$$stipitis$. Maximum ethanol yields of the most samples were measured at 72 hours and applied to RSM as a dependent variable. 9.7 g/${\ell}$ of ethanol was produced from the solid pretreated at $180^{\circ}C$ for 40 min with 0.013 g/g of oxalic acid loading. According to the response surface methodology, it was determined that the temperature is the most governing factor via statistic analysis.
A bacterium isolated from contaminated white ginseng was indentified by using API kit and electron microscope. The isolate was determined as rod shaped bacterium having 0.6-1.0 ${\mu}{\textrm}{m}$ in diameter and 1.2-3.0 ${\mu}{\textrm}{m}$ in length. It had motility by flagellum. The isolate had $\beta$-galactosidase, arginine dihydrolase and omithin decarboxylase. It used citrate as sole carbon source but not produced H$_2$S. It also fermented glucose, manitol, sorbitol, rhamnose, sucrose, melibiose, arabinose and amygdalin. The isolate was identified as Enterobacter sp by the above API kit analysis and electron microscopy observation. Ginseng saponin was added to culture of Enterobacter sp. in order to investigate saponin's influence on its growth. The strain was incubated at 38$^{\circ}C$ for 3 days after addition of 0.05, 0.5, 2.0 and 4.0% (w/v) of saponin, respectively and the growth rates were investigated. The relative bacterial growth rates showed 75.0, 37.5, 7.5 and 0.5%, respectively, when compared with 100% of saponin non-added group. These results suggest that the growth of Enterobacter sp. is inhibited by saponin with the concentration dependency.
Since the study of Lee et al.(1994) there have been some case studies on the set-up effect of driven piles in Korea country. However, comprehensive examination on the analyses of the set-up effect with various testing data has not been carried out. In particular, the analysis of the influence of soil type and pile shape on the set-up effect has not been reported. It is necessary to analyse the test results of production piles in order to apply the set-up effect of driven piles for the field engineering. In this study some test piling and analyses were performed to give basic information to the piling design as well as the research on the set-up effect in sandy soils. The analyses on the set-up effect were performed with the monitoring data obtained from the high-strain dynamic loading tests. It was shown that the set-up effect of driven piles was not only affected by soil type but also by soil formation history It turned out that the set-up effect in sandy soils was considerable one that should not be ignored in the field, and that the bearing capacity increase of pile is mainly caused by the increase of shaft resistance. It was shown that the set-up effect of closed pile was larger than that of opened pile in clayey soils, while the set-up effect of opened pile was larger than that of closed pile in sandy soils.
The resonant column testing determines the shear modulus and material damping factor dependent on the shear strain magnitude, based on the wave-propagation theory. The determination of the dynamic soil properties requires the theoretical formulation of the dynamic behavior of the resonant column testing system. One of the theoretical formulations is the use of the wave equation for the soil specimen in the resonant column testing device. Wood, Richart and Hall derived the wave equation by assuming the linear elastic soil, and didn't take the material damping into consideration. Hardin incorporated the viscoelastic damping of soil in the wave equation, but he had to assume the material damping factor for the determination of the shear modulus. For the better theoretical formulation of the resonant column testing, this study derived a new wave equation to include the viscosity of soil, and proposed an approach for the solution. Also, in this study, the equation of motion for the testing system, which is another approach of the theoretical formulation of the resonant column testing, was also derived. The equation of motion leads to the better understanding of the resonant column testing, which includes the dynamic magnification factor and the phase angle of the response. For the verification of the proposed equation of motion for the resonant column testing, the finite element analysis was performed for the resonant column testing. The comparison of the dynamic magnification factors and the phase angles far the system response were performed.
This paper presents an experimental study of the plugging effect on the capacity of open-ended piles installed in sandy soil. Full-scale tests, including dynamic and static axial-compression load tests, were carried out on three instrumented piles with different diameters (508.0, 711.2 and 914.4 mm). To measure the outer and inner shaft resistances acting on the piles, a double-walled system was utilized with instrumented strain gauges on the outside and inside walls of the pile. The results of field tests show that the inner shaft resistance was mostly mobilized at the location between the pile tip and 18-34% of the total plug length. It was found that the soil plugging in the lower portion has influence on the inner shaft resistance. In addition, it can be also demonstrated that the ratio of inner shaft resistance plus annulus load resistance to total resistance decreased with increasing pile diameters. The results of these tests show that the relationship between the degree of plugging and pile diameter is clearly established. Direct observations of the soil plugs were made and used to quantify both the plug length ratio (PLR) and the incremental filling ratio (IFR). Based on this result, it was found that the N value of the standard penetration test (SPT) is highly correlated with the IFR.
Background: Cancer is a major threat to the public health whether in developed or in developing countries. As the most common primary malignant tumor, the morbidity and mortality rate of lung cancer continues to rise in recent ten years worldwide. Chemotherapy is one of the main methods in the treatment of lung cancer, but this is hampered by chemotherapy drug resistance, especially MDR. As a component of the 60S large ribosomal subunit, ribosomal protein L39-L gene was reported to be expressed specifically in the human testis and human cancer samples of various tissue origins. Materials and Methods: Total RNA of cultured drug-resistant and susceptible A549 cells was isolated, and real time quantitative RT-PCR were used to indicate the transcribe difference between amycin resistant and susceptible strain of A549 cells. Viability assay were used to show the amycin resistance difference in RPL39-L transfected A549 cell line than control vector and null-transfected A549 cell line. Results: The ribosomal protein L39-L transcription level was 8.2 times higher in drug-resistant human lung cancer A549 cell line than in susceptible A549 cell line by quantitative RT-PCR analysis. The ribosomal protein L39-L transfected cells showed enhanced drug resistance compared to plasmid vector-transfected or null-transfected cells as determined by methyl tritiated thymidine (3H-TdR) incorporation. Conclusions and Implications for Practice: The ribosomal protein L39-L gene may have effects on the drug resistance mechanism of lung cancer A549 cells.

In the present paper, the effects of the thickness and width of a curved wide-plate, the crack length, and the strain hardening exponent on the crack-tip constraint of the curved wide-plate were investigated. To accomplish this, detailed three-dimensional elastic-plastic finite element (FE) analyses were performed considering various geometric and material variables. The material was characterized by the Ramberg-Osgood relationship, and the Q-stress was employed as a crack-tip constraint parameter. Based on the present FE results, the variations in the Q-stress of the curved wide-plate with the geometric variables and material properties were evaluated. This revealed that the effect of out-of-plane constraint conditions on the crack-tip constraint was closely related to the in-plane constraint conditions, and out-of-plane constraint conditions affected the crack-tip constraint more than in-plane constraint conditions.
A detailed understanding of the mechanical behaviors for crushed coal rocks after grouting is a key for construction in the broken zones of mining engineering. In this research, experiments of grouting into the crushed coal rock using independently developed test equipment for solving the problem of sampling of crushed coal rocks have been carried out. The application of uniaxial compression was used to approximately simulate the ground stress in real engineering. In combination with the analysis of crack evolution and failure modes for the grouted specimens, the influences of different crushed degrees of coal rock (CDCR) and solidified grout strength (SGS) on the mechanical behavior of grouted specimens under uniaxial compression were investigated. The research demonstrated that first, the UCS of grouted specimens decreased with the decrease in the CDCR at constant SGS (except for the SGS of 12.3 MPa). However, the UCS of grouted specimens for constant CDCR increased when the SGS increased; optimum solidification strengths for grouts between 19.3 and 23.0 MPa were obtained. The elastic moduli of the grouted specimens with different CDCR generally increased with increasing SGS, and the peak axial strain showed a slightly nonlinear decrease with increasing SGS. The supporting effect of the skeleton structure produced by the solidified grouts was increasingly obvious with increasing CDCR and SGS. The possible evolution of internal cracks for the grouted specimens was classified into three stages: (1) cracks initiating along the interfaces between the coal blocks and solidified grouts; (2) cracks initiating and propagating in coal blocks; and (3) cracks continually propagating successively in the interfaces, the coal blocks, and the solidified grouts near the coal blocks. Finally, after the propagation and coalescence of internal cracks through the entire specimens, there were two main failure modes for the failed grouted specimens. These modes included the inclined shear failure occurring in the more crushed coal rock and the splitting failure occurring in the less crushed coal rock. Both modes were different from the single failure mode along the fissure for the fractured coal rock after grouting solidification. However, compared to the brittle failure of intact coal rock, grouting into the different crushed degree coal rocks resulted in ductile deformation after the peak strength for the grouted specimens was attained.

Background: Recent studies have shown that the nontuberculosis mycobacterium (NTM) recovery rate in clinical cultures has increased within Korea. However, another study conducted by a secondary hospital within Daegu reported different results. Therefore, the purpose of this study is to understand and evaluate the microbiological distribution and clinical features of NTM in Daegu. Methods: A retrospective study was conducted on 11,672 respiratory specimens undergoing acid fast bacilli (AFB) culture from 6,685 subjects who visited Yeungnam University Respiratory Center from January 2012 to December 2013. Results: Of the 11,672 specimens undergoing AFB culture, 1,310 specimens (11.2%) showed positive results. Of these specimens, NTM was recovered from 587 specimens, showing a recovery rate of 44.8%. Identification test for NTM was performed on 191 subjects; the results were as follows: M. avium-intracellulare complex (MAC) 123 (64.4%), M. abscessus 20 (10.5%), M. kansasii 12 (6.3%), and 33 other NTM germ strains. Of the 382 subjects with NTM, 167 were diagnosed with pulmonary NTM disease (43.7%), however virulence differed depending on NTM strain. Multivariate analysis showed that nodular bronchiectasis, the nodules, and finding consistent with cavity under imaging study were statistically significant for triggering pulmonary NTM disease. AFB culture showing MAC and M. abscessus was statistically significant as well. Positive predictive value for NTM polymerase chain reaction (NTM-PCR) was 88.6%. Conclusion: Results for NTM recovery rate within the Daegu area were similar to those for the Seoul metropolitan area. We can assume that NTM infection is increasing in our community, therefore AFB-positive subjects (1) should undergo NTM-PCR, (2) should have their culture results checked for differentiation of mycobacterium tuberculosis complex (MTB) from NTM, and (3) undergo NTM identification test to confirm its type. Administration of treatment with the above results should be helpful in improving the patients' prognosis.
