Antitimor Activity of Some Phytobased Polysaccharides and their Effects on the Immune Function

  • Polysaccharide fractions were prepared from Ginseng root, Mori Radicis Cortex (M. R. C. ), Phellodendri Cortex (Ph. C. ), Sappan Wood (S. W. ) and Tigli Semen (T. S.). Water extract was also prepared from the mixture of ph. C., S. W. and T. S. Ginseng polysaccharide and water extract of the mixture showed marked antitumor activity against sarcoma 180. Ginseng polysaccharide showed a mild increasing effect on the number of circulating leucocytes and a marked increasing effect on the number leucocytes and a marked increasing effect on the number of plaque forming cells (PEC). Polysaccharides from ginsing root, S. W., Ph. C. + T. S. and water extract of the mixture showed dramatic inducing activities of periotoneal exudate cells (PEC), polymorphonuclear leucocytes (PMN) and macrophages. These results suggest the possibility that water extract of the mixture may have the lentinan like effect and ginseng polysaccharide may have stimulating effects on the general immune system.

Properties of BE0623 to serve as a growth factor of Bifidobacterium

  • Prebiotics are defined as "Nondigestible food ingredients that beneficially affect the host by selectively stimulating the growth and activity of bacteria in the intestine" and as defined improve host health. This study was carried out to investigate the effects of bifidobacteria (Bifidobacterium lactis BB12 and Bifidobacterium longum BB536) growth enhancer (BE0623) supplement as a prebiotic. The addition of BE0623, a growth promoting material for bifidobacteria, significantly increased bifidobacteria viable cells counts in fermented milk by about 45 to 75 times compared to the non-added control group. In addition, microscopic observation showed a significant effect on proliferation of bifidobacteria in fermented milk with added BE0623. The viable cell counts in bifidobacteria also increased roughly 102-fold compared to the control group (non-added BE0623) and was higher than that of commercial growth promoters. Each fraction obtained though the purification of BE0623 influenced the increase of bifidobacteria growth. Culturing bifidobacteria with a combination of fractions of BE0623 had a synergistic effect compared to culturing bifidobacteria with each fraction individually. When any of the fractions were not added, the effect of the growth enhancer on bifidobacteria was reduced. These results indicate that all fractions contain substances that promote the growth of bifidobacteria. Therefore, BE0623 is considered to be available as a growth promoting material for bifidobacterium.

Reduction of Interlukin-8 by Peptides from Digestive Enzyme Hydrolysis of Hen Egg Lysozyme

  • Lysozyme was treated with digestive enzymes and the production of interleukin 8 (IL-8) was measured in Caco-2 cell with the peptides from lysozyme upon stimulating with lipopolysaccharide (LPS) to investigate the overall anti-inflammatory activity of lysozyme when it is in digestive tracts. Lysozyme reduced IL-8 production, and the peptides from pepsin hydrolysis of lysozyme had the similar effect. The products of trypsin digestion of lysozyme had no effect on the reduction of IL-8 production while those of pepsin-trypsin hydrolysis did. The effectiveness of lowering IL-8 production was not different by time of the peptide addition. When Caco-2 cells were pre-incubated with peptides for 24 hr, the reduction effects were observed from the peptides from pepsin hydrolysis, indicating that some of the peptides are still remaining in the cells. Therefore, it can be concluded that the IL-8 reduction effect of lysozyme against LPS still remained even after the pepsin and trypsin hydrolysis.

Physiological Activities of Extracts of Cedrela sinensis leaves (참죽나무 잎 추출물의 생리활성)

  • The purpose of this study was to confirm the content of total polyphenol, antioxidative and immune activities of the extracts of Cedrela sinensis leaf. The content of total polyphenol of water extracts ranged from 46.5-59.6 mg/100 g, which was higher than other extracts using organic solvents such as EtOAc, $CH_2Cl_2$ and $C_6H_{14}$. The antioxidant activity of the water and organic solvents extracts showed 6-33% in terms of 2,2-diphenyl-picryl-hydrazyl (DPPH) scavenging activity. To analyze the immuno-stimulation activity of C. sinensis leaf extract, we investigated the effect of the extracts on NO synthesis which is important in host defense against bacterial infection. Hot water extracts significantly increased NO generation by RAW 264.7, macrophage cell line, while organic solvent extract has no significant effect on NO production. To further analyzed the anti-inflammatory effect of the extracts, we investigated the effects of the extracts on lipopolysaccharide(LPS)-induced NO generation. Organic solvent extracts of C. sinensis leaves showed strong inhibitory effect on NO production in LPS-stimulated RAW 264.7 cells. These results suggest that C. sinensis leaf extract may represent a useful immune stimulating agent and anti-inflammatory agent.

Efficacy of corifollitropin alfa followed by recombinant follicle-stimulating hormone in a gonadotropin-releasing hormone antagonist protocol for Korean women undergoing assisted reproduction

  • Objective: To evaluate the effect of a gonadotropin-releasing hormone (GnRH) antagonist protocol using corifollitropin alfa in women undergoing assisted reproduction. Methods: Six hundred and eighty-six in vitro fertilization-embryo transfer (IVF)/ intracytoplasmic sperm injection (ICSI) cycles were analyzed. In 113 cycles, folliculogenesis was induced with corifollitropin alfa and recombinant follicle stimulating hormone (rFSH), and premature luteinizing hormone (LH) surges were prevented with a GnRH antagonist. In the control group (573 cycles), premature LH surges were prevented with GnRH agonist injection from the midluteal phase of the preceding cycle, and ovarian stimulation was started with rFSH. The treatment duration, quality of oocytes and embryos, number of embryo transfer (ET) cancelled cycles, risk of ovarian hyperstimulation syndrome (OHSS), and the chemical pregnancy rate were evaluated in the two ovarian stimulation protocols. Results: There were no significant differences in age and infertility factors between treatment groups. The treatment duration was shorter in the corifollitropin alfa group than in the control group. Although not statistically significant, the mean numbers of matured (86.8% vs. 85.1%) and fertilized oocytes (84.2% vs. 83.1%), good embryos (62.4% vs. 60.3%), and chemical pregnancy rates (47.2% vs. 46.8%) were slightly higher in the corifollitropin alfa group than in the control group. In contrast, rates of ET cancelled cycles and the OHSS risk were slightly lower in the corifollitropin alfa group (6.2% and 2.7%) than in the control group (8.2% and 3.5%), although these differences were also not statistically significant. Conclusion: Although no significant differences were observed, the use of corifollitropin alfa seems to offer some advantages to patients because of its short treatment duration, safety, lower ET cancellation rate and reduced risk of OHSS.

Stimulation of Seed Germination of Korean Lawn grass (Zoysia japonica Steud.) by Sodium Hypochlorite Treatment (Sodium Hypochlorite 처리(處理)에 의한 한국잔디 종자(種子)의 발아촉진(發芽促進))

  • In order to obtain an easy and effective scarification method for stimulating the seed germination of Korean lawn grass (Zoysia japonica Steud.), sodium hypochlorite (SH), which is used as a commercial bleach and a seed disinfestant, was tested as a chemical scarifier instead of potassium hydroxide (KOH). 1. Scarification with 4% SH for 2 hours followed by 8-hour water-rinse stimulated seed germination more effectively compared to generalized scarification method by KOH treatment. 2. The degree of stimulating seed gemination was different by the various water-rinsing time after SH treatment, but the optimum rinsing time was about 4 to 8 hours. 3. GA treatment after SH scarification stimulated seed germination. However, the stimulation effect of GA was not showed in non-scarified control. 4. SH treatment significantly decreased the percentage of the infection of microflora in seed samples compared to KOH treatment. 5. Under practical seed scarification conditions for stimulating germination of Korean lawn grass seed. SH may have advantages over KOH with respect to ease of preparing, securing and handling.

Separation of Follicular Fluid Components Stimulating Sperm Migration with Chromatographic Paper, $=mu$RPC and Superose Columns (Chromatography용 Paper, $\mu$RPC Column 및 Superose Column을 이용한 정자의 이동을 자극하는 난포액 성분의 분리)

  • To efficiently separate a protein stimulating sperm swim-up migration and movement from follicular proteins, the effect of paper chromatography and liquid chromatography with reverse phase column and superose column on protein separation was examined. And the results obtained were as follows; 1. The band component that was separated with paper chromatography stimulated sperm migration and movement depending on its additional levels. Especially, band I component significantly increased sperm migration. But, all components of bands 1, 2 and 3 showed lower sperm migration and movement, compared to follicular fluid at the same additional level. 2. Among the components separated from follicular protein of 2~5mm follicles with reverse phase column ($\mu$RPC), components at retention time (RT) of 3.33, 7.00, 13.87, and 16.6A minutes stimulated sperm migration within a limited range. 3. All components separated from follicular protein of 10mm follicles with $\mu$RPC column didn't stimulate sperm migration and movement. 4. Among the components separated from follicular protein of 2~5m follicles with superose column, components at retention volume (RV) of 1.35 and 0.82 ml significantly stimulated sperm migration and movement. In conclusion, protein components stimulating sperm migration and movement were efficiently separated with superose column in Smart system. Especially, components of RV 1.35 and RV0.82 stimulated sperm swim-up separation.

Study on Immuno-stimulating Activity of ${\beta}$-Glucan Isolated from the Cell Wall of Yeast Mutant Saccharomyces cerevisiae IS2 (효모변이주 Saccharomyces cerevisiae IS2 세포벽 유래의 베타글루칸 면역활성능에 관한 연구)

  • Yeast cell wall mutant, Saccharomyces cerevisiae IS2 was screened by the NTG treatment of Saccharomyces cerevisiae KCTC 7911. The mutant was highly resistant to zymolase, which specifically degrades ${\beta}$-1,3-D-glucose chain of ${\beta}$-glucan and mechanical disruption by glass beads. These phenomena demonstrate that the yeast mutant has cell wall structure different from the wild-type. The ${\beta}$-glucan of yeast mutant and wild-type strains was recovered by sequential extraction with NaOH. The injection of ${\beta}$-glucan into the abdominal cavity of mouse resulted in an increase in the number of peritoneal immune cells, NO (nitric oxide) production, and phagocytic activity of macrophage. The number of immune cells was found to be $3.90{\times}10^6\;cells/10\;mL$ and $5.48{\times}10^6\;cells/10\;mL$ with the wild-type and mutant ${\beta}$-glucan, respectively. The effect on the NO production and phagocytic activity of mutant ${\beta}$-glucan were 1.69 and 1.43-fold higher than those of wild-type. These results indicate that the immuno-stimulating activity of alternated ${\beta}$-glucan from mutant yeast is higher than that of wild-type.

  • Estrus cycle of cow, season and follicldar stimulating treatment, having an effect on the number of follicle, are investigated for the oocyte recovery rate in ovum pick-up(OPU). The number of follicle aspirated and oocyte collected on the different days of estrus cycle(D 4~5, D 9~10 and D 14~15) were not significantly different among the groups. The higher number of viable oocytes were produced on Jan-May(79.0%) than Jun-Aug(33.3%) by OPU in cow. The number of follicle and aspirated oocyte in cows treated FSH or PMSG combined with bovine somatotropin(bST) were 1.2~1.5 times higher than in cows treated alone follicular stimulating hormone(FSH) or pregnant mare serum gonadotropin(PMSG). In conclusion, OPU can be repeatedly practiced 2 or 3 times in an estrus cycle. In addition, the high environmental temperature is not good for ovarian function of cow and the bST co-treatment with FSH or PMSG is increasing the number of aspiratable follicle.

Effect and Mechanism of Glutamine on Productive Performance and Egg Quality of Laying Hens

  • This study was conducted to investigate the effects of dietary glutamine (Gln) on the productive performance and egg quality of laying hens. A total of four hundred Lingnan Yellow laying hens aged 34 weeks were randomly assigned into four groups (100 laying hens/group), and fed, respectively, with diets supplemented with 0% (control group), 0.2%, 0.4%, and 0.8% Gln during the 6-week feeding period. The results were as follows. First, the productivity of laying hens fed with 0.8% Gln in diet was significantly increased (p<0.05); however, the egg quality (egg weight, yolk weight, shell weight, egg shape index, shell thickness, shell density, shell breaking strength, yolk color, yolk index, and Haugh unit) was not affected compared with that of the control group (p>0.05). Second, luteinizing hormone (LH) (p<0.01), follicle stimulating hormone (FSH) (p<0.01), triiodothyronine ($T_3$), and tetraiodothyronine ($T_4$) contents (p<0.05) in blood of laying hens fed with 0.8% Gln in diets were also significantly improved, and greater improvement in the duodenum and oviduct structure was observed in that treatment group. This study indicated for the first time that diets with 0.8% Gln were able to increase the productive performance of laying hens through stimulating hormone secretion and better development of both the duodenum and oviduct structure in laying hens.
