Alpha-Synuclein Inclusion Formation in Human Oligodendrocytes

  • Multiple system atrophy (MSA) is a neurodegenerative disease characterized by presence of α-synuclein-positive inclusions in the cytoplasm of oligodendrocytes. These glial cytoplasmic inclusions (GCIs) are considered an integral part of the pathogenesis of MSA, leading to demyelination and neuronal demise. What is most puzzling in the research fields of GCIs is the origin of α-synuclein aggregates in GCIs, since adult oligodendrocytes do not express high levels of α-synuclein. The most recent leading hypothesis is that GCIs form via transfer and accumulation of α-synuclein from neurons to oligodendrocytes. However, studies regarding this subject are limited due to the absence of proper human cell models, to demonstrate the entry and accumulation of neuronal α-synuclein in human oligodendrocytes. Here, we generated mature human oligodendrocytes that can take up neuronderived α-synuclein and form GCI-like inclusions. Mature human oligodendrocytes are derived from neural stem cells via "oligosphere" formation and then into oligodendrocytes, treating the cells with the proper differentiation factors at each step. In the final cell preparations, oligodendrocytes consist of the majority population, while some astrocytes and unidentified stem cell-like cells were present as well. When these cells were exposed to α-synuclein proteins secreted from neuron-like human neuroblastoma cells, oligodendrocytes developed perinuclear inclusion bodies with α-synuclein immunoreactivity, resembling GCIs, while the stem cell-like cells showed α-synuclein-positive, scattered puncta in the cytoplasm. In conclusion, we have established a human oligodendrocyte model for the study of GCI formation, and the characterization and use of this model might pave the way for understanding the pathogenesis of MSA.

CD166 promotes the cancer stem-like properties of primary epithelial ovarian cancer cells

  • Cancer stem cells (CSCs) or tumor-initiating cells are thought to play critical roles in tumorigenesis, metastasis, drug resistance, and tumor recurrence. For the diagnosis and targeted therapy of CSCs, the molecular identity of biomarkers or therapeutic targets for CSCs needs to be clarified. In this study, we identified CD166 as a novel marker expressed in the sphere-forming CSC population of A2780 epithelial ovarian cancer cells and primary ovarian cancer cells. The CD166+ cells isolated from A2780 cells and primary ovarian cancer cells highly expressed CSC markers, including ALDH1a1, OCT4, and SOX2, and ABC transporters, which are implicated in the drug resistance of CSCs. The CD166+ cells exhibited enhanced CSC-like properties, such as increased sphere-forming ability, cell migration and adhesion abilities, resistance to conventional anticancer drugs, and high tumorigenic potential in a xenograft mouse model. Knockdown of CD166 expression in the sphere-forming ovarian CSCs abrogated their CSC-like properties. Moreover, silencing of CD166 expression in the sphere-forming CSCs suppressed the phosphorylation of focal adhesion kinase, paxillin, and SRC. These results suggest that CD166 plays a key role in the regulation of CSC-like properties and focal adhesion kinase signaling in ovarian cancer.

멸종위기 수종 석곡(Dendrobium moniliforme)의 Protocorm-like Body 형성 및 기내증식 (In vitro Propagation and Protocorm-like Body Formation of Endangered Species, Dendrobium moniliforme)

  • This experiment was conducted to establish the in vitro propagation of Dendrobium moniliforme through protocorm like body (PLB) induction from the culture of leaf, stem and root explants. Frequency of PLB formation from root explants was higher than those of leaf and stem explants. PLB's proliferation in MS medium including 1.0 mg/L NAA was better than 1.0 mg/L IBA. Frequency of PLB's proliferation on medium with various concentrations of BA, Kinetin, Tidiazuron (0. 1.0, 3.0 mg/L) and NAA (0, 1.0 mg/L) was tested. The maximun proliferation of PLB's was obtained on MS medium supplemented with 3.0 mg/L BA and 1.0 mg/L NAA after 6 weeks of culture. Addition of 200 mg/L activated charcoal (AC) and 30 g/L sucrose was effective on PLB proliferation for D. moniliforme.

Supplementation of retinoic acid alone in MSC culture medium induced germ cell-like cell differentiation

  • Background: Germ cells undergo towards male or female pathways to produce spermatozoa or oocyte respectively which is essential for sexual reproduction. Mesenchymal stem cells (MSCs) have the potential of trans-differentiation to the multiple cell lineages. Methods: Herein, rat MSCs were isolated from bone marrow and characterized by their morphological features, expression of MSC surface markers, and in vitro differentiation capability. Results: Thereafter, we induced these cells only by retinoic acid supplementation in MSC medium and, could able to show that bone marrow derived MSCs are capable to trans-differentiate into male germ cell-like cells in vitro. We characterized these cells by morphological changes, the expressions of germ cell specific markers by immunophenotyping and molecular biology tools. Further, we quantified these differentiated cells. Conclusions: This study suggests that only Retinoic acid in culture medium could induce bone marrow MSCs to differentiate germ cell-like cells in vitro. This basic method of germ cell generation might be helpful in the prospective applications of this technology.

Monitoring the Differentiation and Migration Patterns of Neural Cells Derived from Human Embryonic Stem Cells Using a Microfluidic Culture System

  • Microfluidics can provide unique experimental tools to visualize the development of neural structures within a microscale device, which is followed by guidance of neurite growth in the axonal isolation compartment. We utilized microfluidics technology to monitor the differentiation and migration of neural cells derived from human embryonic stem cells (hESCs). We co-cultured hESCs with PA6 stromal cells, and isolated neural rosette-like structures, which subsequently formed neurospheres in suspension culture. Tuj1-positive neural cells, but not nestin-positive neural precursor cells (NPCs), were able to enter the microfluidics grooves (microchannels), suggesting that neural cell-migratory capacity was dependent upon neuronal differentiation stage. We also showed that bundles of axons formed and extended into the microchannels. Taken together, these results demonstrated that microfluidics technology can provide useful tools to study neurite outgrowth and axon guidance of neural cells, which are derived from human embryonic stem cells.

다형성 교모세포종의 항생제 내성 종양 줄기세포 (Chemotherapeutic Drug Resistant Cancer Stem-like Cells of Glioma)

  • 다형성 교모세포종은 뇌종양 가운데 가장 빈번하게 발병하는 악성종양이다. 다형성 교모세포종에 종양 줄기세포가 존재한다는 보고가 있음에도 불구하고, 항암제 내성과 종양 줄기세포 사이의 상호 연관성에 관한 연구는 아직 미비한 실정이다. 본 연구에서 다형성 교모세포종 세포주 A172 및 뇌종양 환자로부터 확립한 GBM2에 1,3-bis(2 -chloroethyl)-1-nitrosourea (BiCNU)를 처리시 극소량의 세포군만이 생존하며, 이들 생존 세포군은 BiCNU 재처리에 내성을 나타내는 것으로 조사되었다. 또한 이 다형성 교모세포종 유래 BiCNU-내성세포군의 Erk 및 Akt 인산화 활성이 증가되었으며, CD133 줄기세포 표지인자를 발현하는 세포가 다량 존재하였다. 이와 아울러, 다형성 교모세포종 유래 BiCNU-내성세포를 severe combined immuno-deficient (SCID) mouse brain에 이식하였을 때 암이 형성되는 것을 관찰할 수 있었다. 이와 같은 결과는 다형성 교모세포종 유래 BiCNU-내성세포가 종양줄기세포의 능력을 가지는 것으로 생각된다. 따라서 이상의 결과는 다형성 교모세포종에 존재하는 종양줄기세포가 항암제 내성에 관여 한다는 중요한 단서를 제공해줄 수 있을 것으로 사료된다.

Clinical utilization of cord blood over human health: experience of stem cell transplantation and cell therapy using cord blood in Korea

  • Cord blood (CB) has been used as an important and ethical source for hematopoietic stem cell transplantation (SCT) as well as cell therapy by manufacturing mesenchymal stem cell, induced pleuripotential stem cell or just isolating mononuclear cell from CB. Recently, the application of cell-based therapy using CB has expanded its clinical utility, particularly, by using autologous CB in children with refractory diseases. For these purposes, CB has been stored worldwide since mid-1990. In this review, I would like to briefly present the historical development of clinical uses of CB in the fields of SCT and cell therapy, particularly to review the experiences in Korea. Furthermore, I would touch the recent banking status of CB.

Effect of Plant Growth Regulators on Clonal Production through Basal Stem Explant Cultures of a Phalaenopsis Hybrid

  • This study was conducted to develop the clonal propagation technique through in vitro culture using basal stem explants in Phalaenopsis hybrid grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when basal stem explants were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 ml/L coconut water, 1 mg/L NAA, 5 mg/L 2iP and 2.5g/L gel rite. PLBs transferred to Hyponex medium were regenerated to plantlets. Plantlets transferred to plastic pots containing spagnum moss were developed and successfully acclimatized under greenhouse. The flower was bloomingly opened in plants regenerated from basal stem explants. The flower was not different from both mother plant and plant induced through clonal propagation of Phalaenopsis hybrid.

Skeletal myogenic differentiation of human periodontal ligament stromal cells isolated from orthodontically extracted premolars

  • Objective: To investigate the stem cell-like characteristics of human periodontal ligament (PDL) stromal cells outgrown from orthodontically extracted premolars and to evaluate the potential for myogenic differentiation. Methods: PDL stromal cells were obtained from extracted premolars by using the outgrowth method. Cell morphological features, self-replication capability, and the presence of cell-surface markers, along with osteogenic, adipogenic, and chondrogenic differentiation, were confirmed. In addition, myogenic differentiation was induced by the use of 5-aza-2'-deoxycytidine (5-Aza) for DNA demethylation. Results: PDL stromal cells showed growth patterns and morphological features similar to those of fibroblasts. In contrast, the proliferation rates of premolar PDL stromal cells were similar to those of bone marrow and adipogenic stem cells. PDL stromal cells expressed surface markers of human mesenchymal stem cells (i.e., CD90 and CD105), but not those of hematopoietic stem cells (i.e., CD31 and CD34). PDL stromal cells were differentiated into osteogenic, adipogenic, and chondrogenic lineages. Myotube structures were induced in PDL stromal cells after 5-Aza pretreatment, but not in the absence of 5-Aza pretreatment. Conclusions: PDL stromal cells isolated from extracted premolars can potentially be a good source of postnatal stem cells for oromaxillofacial regeneration in bone and muscle.
