The aim of the present study was to update the calibration that is used for the measurement of the total nitrogen content in the rice plant samples by using the visible and near infrared spectrum. Before the equation merge, correlation coefficient of calibration equation for nitrogen content on each rice parts was 0.945 (Leaf), 0.928 (Stem), and 0.864 (Whole plant), respectively. In the calibration models created by each part in the rice plant under the various regression method, the calibration model for the leaf was recorded with relatively high accuracy. Among of those, the calibration equation developed by Partial least squares (PLS) method was more accurate than the Multiple linear regression (MLR) method. The calibration equation was sensitive based on variety and location variations. However, we have merged and enlarged various of the samples that made not only to measure the nitrogen content more accurately, but also later sampling populations became more diversified. After merging, $R^2$ value becomes more accurate and significantly to 0.950 (L.), 0.974 (S.), 0.940 (W.). Also, after removal of outlier, R2 values increased into 0.998, 0.995, and 0.997. In view of the results so far achieved, Standard error of prediction (SEP) and SEP (C) were reduced in the stem and whole plant. Biases were reduced in the leaf, stem as well as whole plant. Slopes were high in the stem. Standard deviation reduced in the stem but $R^2$ was high in the stem and whole plant. Result was indicated that calibration equation make update, and updating robust calibration equation from merge function and multi-variate calibration.
The automatic irrigation system using the stem diameter monitoring and the transpiration model for the determination, respectively, of irrigation timing and amount was designed and evaluated for its applicability in pot and field culture of greenhouse tomato. In the pot culture condition, the yield and quality of greenhouse tomato were improved when irrigation was practiced based on the stem diameter monitoring and the transpiration model as compared to the irrigation practice based on soil moisture monitoring. However, the effects were not significant in the field culture condition.

The sympathetic nervous system (SNS) or neurotransmitters in the bone marrow microenvironment has been known to regulate hematopoietic stem cell (HSC) functions such as self-renewal, proliferation and differentiation. However, the specific role of neuropeptide Y (NPY) in this process remains relatively unexplored. In this study, we demonstrated that NPY deficient mice have significantly reduced HSC numbers and impaired bone marrow regeneration due to apoptotic destruction of SNS fibers and/or endothelial cells. Moreover, NPY treatment prevented bone marrow impairments in a mouse model of chemotherapy-induced SNS injury, while conditional knockout mice lacking the Y1 receptor in macrophages did not restore bone marrow dysfunction in spite of NPY injection. Transforming growth factor-beta (TGF-β) secreted by NPY-mediated Y1 receptor stimulation in macrophages plays a key role in neuroprotection and HSC survival in the bone marrow. Therefore, this study reveals a new role of NPY in bone marrow HSC microenvironment, and provides an insight into the therapeutic application of this neuropeptide.
Embryonic stem cells (ESCs), representing a population of undifferentiated pluripotent cells with both self-renewal and multilineage differentiation characteristics, are capable of spontaneous differentiation into cardiomyocytes. The present study sought to define the kinetic characterization of lactate dehydrogenase (LDH) and creatine kinase (CK) of ESC- and neonatal-derived cardiomyocytes. Spontaneously differentiated cardiomyocytes from embryoid bodies (EBs) derived from mouse ESC line (Royan B1) and neonatal cardiomyocytes were dispersed in a buffer solution. Enzymes were extracted by sonication and centrifugation for kinetic evaluation of LDH and CK with spectrophotometric methods. While a comparison between the kinetic properties of the LDH and CK of both groups revealed not only different Michaelis constants and optimum temperatures for LDH but also different Michaelis constants and optimum pH for CK, the pH profile of LDH and optimum temperature of CK were similar. In defining some kinetic properties of cardiac metabolic enzymes of ESC-derived cardiomyocytes, our results are expected to further facilitate the use of ESCs as an experimental model.
Sprouty (Spry) genes encode inhibitors of the receptor tyrosine kinase signaling cascade, which plays important roles in stem cells. However, the role of Spry4 in the stemness of embryonic stem cells has not been fully elucidated. Here, we used mouse embryonic stem cells (mESCs) as a model system to investigate the role of Spry4 in the stem cells. Suppression of Spry4 expression results in the decreases of cell proliferation, EB formation and stemness marker expression, suggesting that Spry4 activity is associated with stemness of mESCs. Teratoma assay showed that the cartilage maturation was facilitated in Spry4 knocked down mESCs. Our results suggest that Spry4 is an important regulator of the stemness and differentiation of mESCs.
Recently, a new port reserves deep water depth for safe navigation and mooring, following the trend of larger ship building. Larger port facilities include long and huge breakwaters, and mainly adopt vertical type considering low construction cost. A vertical breakwater creates stem waves combining inclined incident waves and reflected waves, and this causes maneuvering difficulty to the passing vessels, and erosion of shoreline with additional damages to berthing facilities. Thus, in this study, the researchers have investigated the response of stem waves at the vertical breakwater near the entrance channel and applied numerical models, which are commonly used for the analysis of wave response at the harbor design. The basic equation composing models here adopted both the linear parabolic approximation adding the nonlinear dispersion relationship and nonlinear parabolic approximation adding a linear dispersion relationship. To analyze the applicability of both models, the research compared the numerical results with the existing hydraulic model results. The gap of serial breakwaters and aligned angles caused more complicated stem wave generation and secondary stem wave was found through the breakwater gap. Those analyzed results should be applied to ship handling simulation studies at the approaching channels, along with the mooring test.
This study, a new concept design of the stem and aims to determine the suitability of various carbon/PEEK composite designs for artificial hip joints. Shear stress tested with alternative materials of the Ti-based stem for artificial hip joints. In addition, FEA is conducted according to the fiber ply orientation and the load condition for carbon/PEEK composites. The stem shape of two types was designed through the shape normal of the femur. Multidirectional load cases were used for each FEA model. In the case of general shape, the results show that the stress of ply orientation case II was lower than for cases I and III. On the other hand, in the case of the curved shape, ply orientation case I was lowest. In the case of the Ti stem, the stress of the curved shape was 18% lower than the general shape.

Research and national standards, such as the Next Generation Science Standards (NGSS) in the United States, promote the development and implementation of K-12 interdisciplinary curricula integrating the disciplines of science, technology, engineering, mathematics, and computer science (STEM+CS). However, little research has explored how teachers provide epistemic support in interdisciplinary contexts or the factors that inform teachers' epistemic support in STEM+CS activities. The goal of this paper is to articulate how interdisciplinary instruction complicates epistemic knowledge and resources needed for teachers' instructional decision-making. Toward these ends, this paper builds upon existing models of teachers' instructional decision-making in individual STEM+CS disciplines to highlight specific challenges and opportunities of interdisciplinary approaches on classroom epistemic supports. First, we offer considerations as to how teachers can provide epistemic support for students to engage in disciplinary practices across mathematics, science, engineering, and computer science. We then support these considerations using examples from our studies in elementary classrooms using integrated STEM+CS curriculum materials. We focus on an elementary school context, as elementary teachers necessarily integrate disciplines as part of their teaching practice when enacting NGSS-aligned curricula. Further, we argue that as STEM+CS interdisciplinary curricula in the form of NGSS-aligned, project-based units become more prevalent in elementary settings, careful attention and support needs to be given to help teachers not only engage their students in disciplinary practices across STEM+CS disciplines, but also to understand why and how these disciplinary practices should be used. Implications include recommendations for the design of professional learning experiences and curriculum materials.

Inhibition of an increase of osteoclasts has become the most important treatment for osteoporosis. The CXCR4 antagonist, AMD3100, plays an important role in the mobilization of osteoclast precursors within bone marrow (BM). However, the actual therapeutic impact of AMD3100 in osteoporosis has not yet been ascertained. Here we demonstrate the therapeutic effect of AMD3100 in the treatment of ovariectomy-induced osteoporosis in mice. We found that treatment with AMD3100 resulted in direct induction of release of SDF-1 from BM to blood and mobilization of hematopoietic stem/progenitor cells (HSPCs) in an osteoporosis model. AMD3100 prevented bone density loss after ovariectomy by mobilization of HSPCs, suggesting a therapeutic strategy to reduce the number of osteoclasts on bone surfaces. These findings support the hypothesis that treatment with AMD3100 can result in efficient mobilization of HSPCs into blood through direct blockade of the SDF-1/CXCR4 interaction in BM and can be considered as a potential new therapeutic intervention for osteoporosis.

The shortage of transplantable kidneys has many efforts to regenerate bioartificial kidneys using transgenic animals and diverse kinds of scaffolds which are important tools for cell seeding. However, there are many limitations for clinical applications so far. Recently, decellularized bioscaffolds using animal organs come into spotlight because of its many superior advantages. In current study, we produced decellularized kidney bioscaffolds of pig which is an attractive animal as a clinical model for human. We decellularized pig kidneys with 1% SDS detergent solution using peristaltic pump systems for 12h. After decellularization process, the kidney bioscaffolds preserved intact 3D morphology including glomerular structure and almost DNA from pig was entirely removed. In addition, this process could preserve micro vascular network which is necessary for cell survival. Although, additional studies for recellularization and transplantation should be required, the decellular vascularized kidney bioscaffolds might have many potentials for kidney regeneration.
