Profiling of Volatile Components Using Gas Chromatography-Mass Spectrometry in Commercial Pine Needle (Pinus densiflora S. and Z.) Powder

  • Volatile components in commercial pine needle (Pinus densiflora S. and Z.) powder were extracted using simultaneous steam distillation and a solvent extraction (SDE) apparatus, and were analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 230 compounds divided into 13 groups were identified, which included alcohols (42), ketones (39), aldehydes (32), terpenes (30), alkenes (17), esters (14), furans (14), benzenes (10), alkanes (8), napthalenes (7), acids (6), miscellaneous compounds (6), and phenols (5). Among the 230 compounds identified, 96 compounds were positively confirmed and quantified, and the rest of the compounds were tentatively identified. The major volatile components identified at relatively high levels were dodecanoic acid, hexadecanoic acid, hexanal, benzaldehyde, (Z)-3-hexen-1-ol, 1-penten-3-one, limonene, and $\beta$-caryophyllene oxide. Among the groups, terpenes accounted for 60.18% of the total concentration of all the volatile components. Some volatile components might account for the unique aroma and the biological activity of the sample.

Antioxidant Activities of Aroma Extracts in Commercially Available Red Wines in Korea

  • The antioxidant activities of aroma extracts from commercially available red wines in Korea were evaluated. The aroma extracts of the red wines were extracted by simultaneous steam distillation. Antioxidant activity was measured by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity and ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid) radical cation scavenging activity. DPPH radical scavenging activity of the aroma extracts in the red wines increased with increases in the amount of wine used for aroma extraction. Antioxidant activities of domestic wine 1, imported wine 7, and imported wine 12 were 97.16, 96.72 and 94.52%/20 mL wine by DPPH assay and 7.09, 8.07 and 7.28 mg ascorbic acid equivalents per mL wine by ABTS assay, respectively. This study demonstrates potent antioxidant activities of the aroma extracts of commercially available red wines in Korea.

  • To develop useful antifungal agents from essential oils in Korean plant resources, the activities of Thymus quinquecostatus and T. quinquecostatus var. japonica were evaluated against ten pathogenic fungi. Their results were compared with those of T. vulgaris, which is native to Europe.The essential oils of the tested Thymus species were obtained by steam distillation using a simultaneous steam distillation-extraction apparatus. The above ground parts of plants cultivated in the herbal garden of Duksung Women"s University were used. (omitted)

  • 야생식용 산채류인 이고들빼기와 개비름의 휘발성 풍미성분을 수증기 증류법으로 추출하여 GC와 GC-GMS 조합에 의하여 분석하였다. 이고들빼기에서는 alcohols류 10종, esters류 2종, aldehydes류 6종, ketones류 5종, hydrocarbons류 3충, acids류 2종 및 기타 3종의 총 31성분이 확인되었다. 개비름에서는 alcohols류 12종, esters류 4종, aldehydes류 5종, ketones류 9종, hydrocarbons류 7종, acids류 3종, phenols 2종 및 기타 6종으로 총 48성분이 확인되었다.

  • 수증기 증류, 헥산 및 초임계 추출법에 의하여 추출된 고수 및 계피 정유성분을 통해 복숭아혹진딧물, 점박이응애 및 배추좀나방에 대한 살충효과를 검정하였다. 고수의 수증기 증류, 헥산 및 초임계 추출 정유성분의 살충 및 살비활성 결과, 수증기 증류 추출물은 복숭아혹진딧물 성충에 대해 $30.59{\mu}g/cm^2$ 로 우수한 활성을 나타내었다. 계피의 수증기 증류, 헥산 및 초임계 추출법 정유성분의 살충 및 살비활성 결과, 수증기 증류 및 헥산 추출물은 점박이응애 성충에 대해 각각 5.96, $4.64{\mu}g/cm^2$ 의 우수한 활성을 나타내었다. 또한 초임계 추출에서 복숭아혹진딧물 성충에 대해 $6.50{\mu}g/cm^2$의 활성을 나타내었다. 본 연구결과는 고수와 계피의 수증기 증류, 헥산 및 초임계 추출 정유성분을 이용한 친환경 방제 가능성을 보여준다.

추출방법에 따른 편백 정유의 향기 성분 비교 및 아토피 개선에의 응용 (Comparison of Volatile Compounds of Chamaecyparis obtusa Essential Oil and its Application on the Improvement of Atopic Dermatitis)

  • Volatile flavor compounds of Chamaecyparis obtuse essential oil were extracted by simultaneous steam distillation extraction (SDE) and supercritical fluid extraction (SFE) and analyzed by GC-MS. A total of 48 and 50 components were identified in essential oil by SDE and SFE, respectively. Monoterpenes, oxygenated monoterpenes, sesquiterpenes, oxygenated sesquiterpenes, and diterpenes in essential oil by SDE were 37.24, 10.9, 9.61, 0.22, and 0.22%, respectively. In the case of SFE, they were 19.1, 23.3, 22.66, 1.31, and 10.57%, respectively. Antioxidant activities were increased with the increase of essential oil up to $80{\mu}L/mL$, irrespective of extraction method. Especially, when the essential oil concentration extracted by SDE was increased from 20 to $80{\mu}L/mL$, the antioxidant activity was increased from 10.5 to 55.1%. However, over $80{\mu}L/mL$ of essential oil, an equilibrium state was maintained. In the case of essential oil extracted by SFE, it was decreased compared to that of SDE. For the improvement of atopic dermatitis, various cosmetics such as an ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE were tested. About over 90% was useful for the improvement of atopic dermatitis after 4 weeks of clinical trial targeting 40 female adults. These results demonstrate that ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE could be used in functional cosmetics.

  • 최근 문제 해충인 갈색날개매미충에 대하여 수증기 증류, 용매 및 초임계 추출법에 의하여 추출된 길초근 정유성분을 가지고 생물활성을 검정하여 방제 가능성을 확인하였다. 길초근의 수증기 증류, 용매 및 초임계 추출 정유성분의 살충활성 결과, 수증기 증류 추출물은 갈색날개매미충 성충에 대해 $1,040{\mu}L/mL$ 로 가장 뛰어한 활성을 나타냈고, 약충에 대하여 $2,370{\mu}L/mL$ 의 살충 활성을 나타내었다. 추출방법별 수율은 0.67%로 다른 방법에 비하여 다소 낮았지만 추출 시간과 비용대비 효과를 감안하였을 때 길초근의 수증기 증류 추출물이 가장 방제 효율이 높다고 여겨진다. 이번의 연구결과는 길초근의 정유성분의 갈색 날개매미충에 대한 방제 가능성을 보여준다.

Exreaction of Thiosulfinates from Garlic Using Gas-in-Liquid-Dispersion

  • Extraction yields of thiosulfinates from garlic were studied by using gas-in-liquis-dispersion(GLD) system to maxmize the thiosulfinate extraction. Extracted thiosulfinates were spectrophotometically quantified using synthesized dially1 thiosulfinate. The conditions for maximum extraction were optimized by response surface methodology. The volatile components extracted by various methods, such as hexane extraction, simultaneous steam distillation and GLD system, were compared by using gas chromatography. The results indicated that the thiosulfinate yield was increased by incresaing temperature and nitrogen gas flow rate, while the effects of bubble sizes on thiosulfinate extraction were not significant at the ranges tested. Application of GLD system resulted in extraction of more volatile components than other extraction methods, Therfore, it was suggested that GLD system was one of the efficient extraction methods among the ever introduced ones, for thiosulfinate extraciton.

초임계유체 추출과 SDE에 의한 솔 추출물의 휘발성분 비교 (A Comparison of Volatile Compounds in Pine Extracts Obtained by Supercritical Fluid Extraction with Those by Simultaneous Steam Distillation and Solvent Extraction)

  • 본 연구에서 행하여진 온도$(35,\;55^{\circ}C)$ 및 압력(100, 300bar) 범위에서 초임계 추출된 솔 추출물의 수율은 300bar, $35^{\circ}C$와 300 bar, $55^{\circ}C$의 경우 각각 5.0%와 5.3%이었으며 100 bar, $35^{\circ}C$와 100 bar, $55^{\circ}C$에서 각각 3.7% 및 2.8%이었다. 300 bar에서는 wax류 등의 추출로 인하여 얻어진 추출물들은 상온에서 고체 상태이며 노란 색이었고, 100 bar에서 얻어진 추출물은 모두 액체 상태이고 무색에 가까웠다. 관능검사 결과 100 bar, $35^{\circ}C$에서의 초임계 추출물이 SDE 추출물보다 1% 유의수준에서 우수함이 판명되었다. 그러나 100 bar, $35^{\circ}C$에서의 추출물과 300 bar, $55^{\circ}C$ 추출물 간에는 5% 유의수준에서 차이가 없었다. 가장 많이 함유되어 있는 휘발성 향기 성분은 limonene으로써 $32.6{\sim}43.4%$를 차지하였고 ${\beta}-pinene(10.8{\sim}18.5%),\;{\beta}-myrcene(11.5{\sim}17.3%),\;{\alpha}-pinene(5.3{\sim}11.7%)$의 순이었다. 한편 향의 품질 특성에 결정적인 역할을 하는 것으로 알려진 함산소 terpene의 경우 100 bar, $35^{\circ}C$의 경우가 가장 많아서 3.6%를 차지하였으며 300 bar, $35^{\circ}C$의 경우에도 3.4%로서 300 bar, $55^{\circ}C$의 2.5%, 100 bar. $55^{\circ}C$의 1.8%보다 많았다. SDE 추출물의 경우는 6.7%를 차지하여 초임계 추출물보다도 훨씬 많은 양을 나타내었다.

초임계 이산화탄소를 이용한 유자 과피로부터 향기성분의 추출

  • 유자과피의 향기성분 중 대부분을 차지하는 limonene은 추출물의 함량을 결정하는데 많은 영향을 준다. 일반적으로 Citrus속의 과실류의 향기 성분들 중 가장 많이 함유되어 있는 limonene의 경우 초임계 이산화탄소에 대한 용해도는 압력 140${\sim}$150 bar, 온도 $35{\sim}40^{\circ}C$일 때 최대이고 유자 또한 다량의 limonene을 함유한 과실류로서 초임계 이산화탄소 압력 2000 psi와 온도 $40^{\circ}C$에서 향기성분 중 limonene의 추출율이 향상되어 함량이 786.6088 mg/g으로 최대로 나타났다.
