Quality Characteristics of Apple Vinegar by Agitated and Static Cultures (교반 및 정치배양에 따른 사과식초의 품질특성)

  • Quality characteristics of apple vinegar by using agitated and static cultures without any additive were compared. pH was reduced with passage of acetic acid fermentation time. Total acidity of the agitated culture vinegar (A) was 6.08% at the 8 day according to the progress of fermentation and that of the static culture vinegar (B) recorded 5.20% at the 60 day of fermentation. There was no significant difference in sugar content of (A) and (B). L value was lower in (B) than (A) but a and b values were higher in (B) than (A). Organic acid content of (A) was high only in acetic acid but malic, citric and succinic acids appeared high in (B). As free sugar, fructose and glucose were observed in both of them. In sensory examination results, (B) showed the highest taste and overall preferences. From all of these results traditional static culture are expected to be used to make high-quality vinegar.

  • Possibility of using microcarriers for the growth of a transformed human embryonic kidney cell line 293 was investigated. The cell grew well in a static culture such as T-flasks with medium of DME/F12 (3:1) mixture supplemented with 5% FBS, but it was most difficult to make the cells grow on microcarriers mainly due to the low attachment efficiency and poor spreading at initial stage of the culture. Consequently, 30-50% of the cells were lost upon inoculation into microcarrier suspension and significant fraction of the mirrocarrier became bald. The medium supplemented with the concentrated conditioned medium by hepatoma cell line HpG2 supported the active growth of the cells on microcarrier and the cells showed a very healthy and well spreading morphology. It was probable that some spreading and attachment factors of HpG2 conditioned medium were effective for 293 cells.

  • Longline type aquaculture facilities are being used for scallpop culture in 30 m of water 2.5 km off the coast of Joomoonjin, Kangwon-do. In this paper, the facilities are modeled by the cabele-buoy-weight system, subject to the nonlinear behaviors of the mooring lines and the effects of current. Its static configuration is shown as a solution of 3-D nonlinear static equation and Runge-Kutta $4^{th}$ method is employed.

Combined TGE-SGE Expression of Novel PAI-1-Resistant t-PA in CHO DG44 Cells Using Orbitally Shaking Disposable Bioreactors

  • An important modification of thrombolytic agents is resistance to plasminogen activator inhibitor-1 (PAI-1). In previous studies, a new truncated PAI-1-resistant variant was developed based on deletion of the first three domains in t-PA and the substitution of KHRR 128-131 amino acids with AAAA in the truncated t-PA. The novel variant expressed in a static culture system of Chinese Hamster Ovary (CHO) DG44 cells exhibited a higher resistance to PAI-1 when compared with the full-length commercial drug; Actylase. In the present study, the truncated-mutant protein was expressed in CHO DG44 cells in 50 ml orbital shaking bioreactors. The final yield of the truncated-mutant in the culture was 752 IU/ml, representing a 63% increase compared with the static culture system. Therefore, these results suggest that using the combined features of a transient and stable expression system is feasible for the production of novel recombinant proteins in the quantities needed for preclinical studies.

  • This paper discussed a transformer-less shunt static synchronous compensator (STATCOM) with consideration of the following aspects: fast compensation of the reactive power, harmonic cancelation and reducing the unbalancing of the 3-phase source side currents. The STATCOM control algorithm is based on the theory of instantaneous reactive power (P-Q theory). A self charging technique is proposed to regulate the dc capacitor voltage at a desired level with the use of a PI controller. In order to regulate the DC link voltage, an off-line Genetic Algorithm (GA) is used to tune the coefficients of the PI controller. This algorithm arranged these coefficients while considering the importance of three factors in the DC link voltage response: overshoot, settling time and rising time. For this investigation, the entire system including the STATCOM, network, harmonics and unbalancing load are simulated in MATLAB/SIMULINK. After that, a 35KVA STATCOM laboratory setup test including two parallel converter modules is designed and the control algorithm is executed on a TMS320F2812 controller platform.

Influence of hydrogel encapsulation during cryopreservation of ovarian tissues and impact of post-thawing in vitro culture systems in a research animal model

  • Objective: Using domestic cats as a biomedical research model for fertility preservation, the present study aimed to characterize the influences of ovarian tissue encapsulation in biodegradable hydrogel matrix (fibrinogen/thrombin) on resilience to cryopreservation, and static versus non-static culture systems following ovarian tissue encapsulation and cryopreservation on follicle quality. Methods: In experiment I, ovarian tissues (n=21 animals; 567 ovarian fragments) were assigned to controls or hydrogel encapsulation with 5 or 10 mg/mL fibrinogen (5 or 10 FG). Following cryopreservation (slow freezing or vitrification), follicle viability, morphology, density, and key protein phosphorylation were assessed. In experiment II (based on the findings from experiment I), ovarian tissues (n=10 animals; 270 ovarian fragments) were encapsulated with 10 FG, cryopreserved, and in vitro cultured under static or non-static systems for 7 days followed by similar follicle quality assessments. Results: In experiment I, the combination of 10 FG encapsulation/slow freezing led to greater post-thawed follicle quality than in the control group, as shown by follicle viability (66.9%±2.2% vs. 61.5%±3.1%), normal follicle morphology (62.2% ±2.1% vs. 55.2%±3.5%), and the relative band intensity of vascular endothelial growth factor protein phosphorylation (0.58±0.06 vs. 0.42±0.09). Experiment II demonstrated that hydrogel encapsulation promoted follicle survival and maintenance of follicle development regardless of the culture system when compared to fresh controls. Conclusion: These results provide a better understanding of the role of hydrogel encapsulation and culture systems in ovarian tissue cryopreservation and follicle quality outcomes using an animal model, paving the way for optimized approaches to human fertility preservation.

  • The purpose of this study was to compare the effects of internet fashion advertisement (Ad) formats according to university students' online lifestyle. Static banner, rich media, floating, shopping, and target advertisement were selected as stimuli and a self-administered questionnaire was used for data collection. SPSS PC (Ver. 16.0) was used for factor analysis, ANOVA, and Chi-square test. Factors of online lifestyle were economy, early adaption, cyberspace activity, sociability, innovation, and entertainment, and subjects were segmented into online activity (OA) retard group, OA mania group, hedonic early adapter group, and OA intermediate group. OA retard group was positive to a static banner Ad with intimacy, and OA mania group and OA intermediate group were positive to a static banner Ad with confidence, attention, and intimacy and rich media Ad and floating Ad with confidence and attention. Hedonic early adapter group was positive to a target Ad with attention and intimacy. Internet shopping mall managers should select internet Ad format after segmenting their customers according to OA lifestyle.

Effect of Chitosan Treatment for Improving Physical Properties of Taegwondo Uniform (시판 태권도복의 기능성 증진을 위한 키토산 처리 효과)

  • Taegwondo uniforms need powerful durability in fabrics against friction and shock because of higher exercising power. Therefore, in this study we developed more comfortable Taegwondo uniform for functional fabrics(deodorization rate, antibacterial activity and static charge). Chitosan is a deacetylated product of chitin, has widely been used in the biomedical sector, food industry, and textile industry. For the purpose of this study, we used the chitosan, which was congenial to the human body. Then, we compared the differences between the chitosan treated fabrics and non treated fabrics of Taegwondo uniform(100% polyester, 65% polyester/35% cotton). Chitosan was dissolved in 1% acetic acid. Also, the fabrics were washed using distilled water. Afterward, its treatment with chitosan was completed by padding the sample to its wet pick-up at $80{\pm}5%$ and by heating $150^{\circ}$ for 3 minutes. The chitosan treated Taegwondo uniform was improved on deodorization rate than non treated uniform. And the chitosan treated Taegwondo uniform was improved remarkably the antibacterial activity in all samples. In the further researches, the static charge was reduced in chitosan treatment uniform.

A study on Countermeasures by Detecting Trojan-type Downloader/Dropper Malicious Code

  • There are various ways to be infected with malicious code due to the increase in Internet use, such as the web, affiliate programs, P2P, illegal software, DNS alteration of routers, word processor vulnerabilities, spam mail, and storage media. In addition, malicious codes are produced more easily than before through automatic generation programs due to evasion technology according to the advancement of production technology. In the past, the propagation speed of malicious code was slow, the infection route was limited, and the propagation technology had a simple structure, so there was enough time to study countermeasures. However, current malicious codes have become very intelligent by absorbing technologies such as concealment technology and self-transformation, causing problems such as distributed denial of service attacks (DDoS), spam sending and personal information theft. The existing malware detection technique, which is a signature detection technique, cannot respond when it encounters a malicious code whose attack pattern has been changed or a new type of malicious code. In addition, it is difficult to perform static analysis on malicious code to which code obfuscation, encryption, and packing techniques are applied to make malicious code analysis difficult. Therefore, in this paper, a method to detect malicious code through dynamic analysis and static analysis using Trojan-type Downloader/Dropper malicious code was showed, and suggested to malicious code detection and countermeasures.

N-terminal amino acid sequence analysis of major manganese peroxidase (MnP3) produced by static culture of Pleurotus ostreatus (느타리 버섯균의 정치배양으로부터 생산되는 중요한 망간퍼옥시데이즈(MnP3)의 N-말단 아미노산 배열 분석)

  • Pleurotus ostreatus No.42, known as the ligninolytic basidiomycetes, showed production of MnP and Lac, but did not show any LiP acitivity in static culture, grown in GPYW liquid medium. Maximum production of MnP (80U/flask) was observed on day 11 of culturing in this medium. Chromatographic purification of MnP included the use of Sepharose CL-6B and Mono-Q. The major MnP isozyme purified by column chromatography was observed to be a 36.4 KDa (single band on SDS PAGE). The 19-amino acid sequence from the N-terminal was determined by protein sequencing to be ATCADGRTTANAACCVLFP. The N-terminal sequence of the major MnP isozyme of P. ostreatus No.42 was found to be the same as a previously reported sequence of an MnP3 isozyme from this fungus.
