Analysis of Productivity in Rice Plant -IV. Sink-filling rate and sink-source relation (벼의 생산력 분석(分析) -IV. 수기(受器)의 충전속도(充塡速度)와 수기(受器)-급기(給器) 관계(關係))

  • Sink (grain)-filling rates of IR667 line (Suwon 213 and 214) and local leading varieties (Jinheung and Paldal) were investigated in relation to sink-source ratio, sink-source distance, forms of photosynthates, and weather factors. The results are as follows. 1. IR667 line have higher maximum filling rate (g. grain/day. ear) and shorter duration to reach maximum filling rate than local varieties. The curve pattern of sink-filling rate was high and steep type in IR667 line and low and broad type in local varieties. 2. Weather factors seem to give little effect on the curve pattern of sink-filling rate. 3. The functional sink-source distance (actual sink-source distance multiplied by the contribution rate of leaf for grain production) was shorter in upper leaves (flag and 2nd leaf) for IR667 line, the upper leaf-dependent type in grain production, and was longer in lower leaves (3rd and 4th) for local varieties, the lower leaf-dependent type. Specially short first internode from top may contribute to the upper leaf depencency of IR667 line. 4. According to free sugar-starch ratio (sugar/starch) in the culm and leaf sheath IR667 line could be classified as the high sugar type and the local varieties as the high starch type. The ratio of transportable form (sugar) to non transportable photosynthate (starch) seems to relate with sink-filling rate. And high sugar type is expected to have higher efficiency for grain production in view of energy balance. 5. A hypothesis that the higher in the uniformity within the series of productive structure the more efficient in grain production is proposed and discussed in relation to productivity. 6. According to the pattern of percent nutrient retention of each leaf blade IR667 showed the central retention type and Jinheung showed the apical retention type and each retention type appears to be a cause of each canopy conservation pattern. 7. From the content and percent distribution of nutrient in various organ IR667 could be classified as the leaf sheath dominant type and Jinheung as the leaf blade dominant type. 8. The fact that the greater the percent translocation of nutrient into grain the greater the percent nutrient retention in leaf blade was held between nitrogen and phosphorus within a variety and between varieties within a nutrient (N, P or K).

  • The objective of this study was to examine the changes of physical properties by additions of different kinds of stabilizers milk proteins concentration, when stored at 4$^{\circ}C$ or 20$^{\circ}C$ for yoghurt. the results were summarized as follows: 1. Addition of 2% carboxyl methyl cellulose and carrageenan, gelation 0.4%, pectin and starch 0.6%, and carrageenan & pectin 0.8% in the manufacture of yoghurt increased the viscosity, water-holding capacity and protein hydration of yoghurt. 2. Addition of 3% skim milk powder, Ca-caseinate or Na-caseinate 0.6% increased the viscosity, water-holding capacity and protein hydration of yoghurt. 3. Twenty five percent of evaporation of milk promoted to build up the optimal structure of the micelles of yoghurt and improved viscosity, water-holding capacity and protein hydration of yoghurt. 4. Addition of stabilizers to yoghurt showed an increase of viscosity, water-holding capacity and protein hydration when compared with non-addition of stabilizers to yoghurt at 4$^{\circ}C$, 20$^{\circ}C$ storage for 12hrs, 96hrs followed by the decrease of it.

Fruit Quality of 'Tsugaru' Apples Influenced by Meteorological Elements (사과 '쓰가루' 품종의 과실 품질에 영향을 미치는 기후요인)

  • An analytical study was conducted in 2002 to observe the relationship between meteorological elements and the fruit quality of 'Tsugaru' apples at 8 orchards. The higher average air temperature in August made a higher content of soluble solids, but showed a lower anthocyanin content. As the average air temperature from April through August increased, flesh firmness decreased; and as the maximum air temperature from April through August increased, Hunter a value also decreased. Additionally, it was observed that the cytohistological characteristics of 'Tsugaru' apples were correlated with the average air temperature during the growing season. As average air temperature during the growing season increased, the epidermal layer of the fruit skin became thinner, starch density in the flesh decreased, intercellular space was larger, and tissue structure became looser.

Purification, Characterization and Chemical Modification of the Xylanase from Alkali-tolerant Bacillus sp. YA-14

  • The xylanase from alkali-tolerant Bacillus sp. YA-14 was purified to homogeneity by CM-cellulose, Sephadex G-50, and hydroxyapatite column chromatographies. The molecular weight of the purified enzyme was estimated to be 20, 000 Da by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The purified enzyme slightly hydrolyzed carboxymethyl cellulose and Avicel, but did not hydrolyze soluble starch, dextran, pullulan, and ${\rho}-nitrophenyl-{\beta}$-D-xylopyranoside. The maximum degree of hydrolysis by enzyme for birchwood xylan and oat spelts xylan were 47 and 40%, respectively. The Michaelis constants for birchwood xylan and oat spelts xylan were calculated to be 3.03 mg/ml and 5.0 mg/ml, respectively. The activity of the xylanase was inhibited reversibly by $HgCl_2$, and showed competitive inhibition by N-bromosuccinimide, which probably indicates the involvement of tryptophan residue in the active center of the enzyme. The Xylanase was identified to be xylose-producing endo-type xylanase and did not show the enzymatic activities which cleave the branch point of the xylan structure.

  • We have developed a new process for the production of new structure oligosaccharides using the mixed-culture fementation of Lipomyces starkeyi KSM22 and leuconostoc mesenteroides B-512FMCM.L.starkeyi KSM22 produces a novel DXAMase(an enzyme containing both dextranase and amylase activities). It hydrolyzes the soluble starch and dextran. The hydrolyzates were used as acceptors for dextransucrase of L.mesenteroides to synthesize the new oligosaccharides(NOS). In fermentation, as the concentration of sucrose was increased from 9%(w/v) to 15%(w/v), the yields of dextran(sum of dextran I, MW=66kD, and dextran II, MW=21kD) was increased from 12.7% to 42.5%, and NOS was increased from 3.9% to 5.2% of the theoretical, respectively. The NOS of dp(degree of polymerization) 5 and over was increased from 33.1% to 58.3% of the total NOS. The NOS showed heat resistant up to 12$0^{\circ}C$ and was stable at pHs ranged from 2 to 6. The NOS decreased the pH changes in the culture of S. mutans, and also showed inhibitory effects on the growth of S. aureus or S. typhimurium.

Genetic Study of Mating System of Sasa borealis in Korea (한국내 조릿대의 교배계에 대한 유전학적 연구)

  • Sasa borealis Makino is distributed in East Asia such as Korea, Fushun in China, and Japan. Especially the species is only found in the high altitude (above 600 m) at mountain of cold regions including The Korean Peninsula. The level of genetic diversity and population structure of this species was surveyed using starch gel electrophoresis at putative five enzyme loci from three natural populations in Korea. Results from twelve loci indicated that genetic diversity was low. In addition, analysis of fixation indices revealed a substantial heterozygosity deficiency in some populations and loci. The reasons for the deficit of heterozygosity may be partly considered inbreeding, small population sizes, and mating of demes. S. borealis can reproduce extensively by producing rhizomes and potentially by sexually produced seeds. Rhizomes are generally long, prostate stems rooting at the nodes.

  • Electro-Rheological(ER) fluids undergo a phase-change when subjected to an external electic field, and this phase-change typically manifests itself as a many-order-of-magnitude change in the rheological behavior. This phenomenon permits the global stiffness and energy- dissipation properties of the beam structures to be tuned in order to synthesize the desired vibration characteristics. This paper reports on a proof-of-concept experimental investigation focussed on evaluation the vibration properties of hollow cantilevered beams filled with an ER fluid. and consequently deriving an empirical model for predicting field-dependent vibration characteristics. A hydrous-based ER fluid consisting of corn starch and silicone oil is employed. The beams are considered to be uniform viscoelastic materials and modelled as a viscously-damped harmonic oscillator. Natural frequency, damping ratio and elastic modulus are evaluated with respect to the electric field and compared among three different beams: two types of different volume fraction of ER fluid and one type of different particle concentration of ER fluid by weight. Transient and forced vibration responses are examined in time domain to demonstrate the validity of the proposed empirical model and to evaluate the feasibility of using the ERfluid as an actuator in a closed-loop control system.

Light and Electron Microscopic Observations on Erythrolobus coxiae gen.et sp.nov. (Porphyridiophyceae, Rhodophyta) from Texas U.S.A.

  • Low molecular weight carbohydrates, phycobilin pigments and cell structure using light and transmission electron microscopy were used to describe a new genus of unicellular red algae, Erythrolobus coxiae (Porphyridiales, Porphyrideophyceae, Rhodophyta). The nucleus of Erythrolobus is located at the cell periphery and the pyrenoid, enclosed by a cytoplasmic starch sheath, is in the cell center. The pyrenoid matrix contains branched tubular thylakoids and four or more chloroplast lobes extend from the pyrenoid along the cell periphery. A peripheral encircling thylakoid is absent. The Golgi apparatus faces outward at the cell periphery and is always associated with a mitochondrion. Porphyridium and Flintiella, the other members of the Porphyrideophyceae, also lack a peripheral encircling thylakoid and have an ER-mitochondria-Golgi association. The low molecular weight carbohydrates digeneaside and floridoside are present, unlike both Porphyridium and Flintiella, which have only floridoside. The phycobilin pigments B-phycoerythrin, R-phycocyanin and allophycocyanin are present, similar to Porphyridium purpureum. The cells have a slow gliding motility without changing shape and do not require substrate contact. The ultrastructural features are unique to members of the Porphyrideophyceae and recent molecular analyses clearly establish the validity of this new red algal class and the genus Erythrolobus.

Effect of Processing Method on Change of Water Soluble Dietary Fiber of Fagopyrum tataricum

  • Seed, stem and sprout of F. tataricum were separately milled using the ultra fine mill under the same condition to investigate the effect of roasting or extruding on the particle size, microstructure and water solubility of dietary fiber. The mean particle size of MR (roasting) is increased in stem and sprout, and that of ME (extruding) is increased in seed, compared to that of control. The microscopic views of seed show that control has the spherical shape but ME the larger and irregular shape, and those of stem and sprout show that control has the needle like shape but ME more rounded shape. Water solubility index of ME is much higher than that of control or MR in seed, stem and sprout. It shows that seed, stem and sprout are damaged more in extruding than in roasting, and the starch and cell wall structure must be destroyed to change the water insoluble dietary fiber into the water soluble dietary fiber.

Textural Changes of Glutinous Rice Cakes during Storage (찹쌀떡의 저장중 텍스쳐 변화)

  • Textural changes of glutinous rice cakes during storage at $4^{\circ}C$ were evaluated by ${\beta}$-amylase digestibility and hardness. Some physical properties of starch including X-ray diffraction, swelling power, water holding capacity and gelatinization temperature were investigated. Changes in hardness were inversely related to the enzyme digestibility. The initial hardness for Tongil glutinous rice cake is much higher than that for Traditional one. Both parameters were changed rapidly during one and two days of storage for Tongil and Traditional glutinous rice cakes, respectively. These results implied that the differences in the strach structure might have significant influences on the texture of rice cakes. Sensory evalution revealed that hardness of the rice cakes was highly significant to the storage time and rice variety.
