Effect of High Voltage Pulsed Electric Fields on Extraction of Purple Sweet Potato Pigment (자색 고구마 색소의 추출에 미치는 고전압 펄스 전기장의 영향)

  • High voltage pulsed electric fields (PEF) is a promising technology for the nonthermal extraction of effective components from biological materials. Plant cells were ruptured with PEF at ambient or refrigerated temperature for a short treatment time of second or microsecond. Treatments of coarsely ground purple sweet potato (PSP) with PEF(30 kV/cm, 500 Hz) resulted in maximum extraction yield of 65% as compared with 45-50% for control. An increase in electric field strength (from 10kV/cm to 35kV/cm) and frequency (from 100Hz to 500Hz) resulted in increased amount of extracted pigments, but treatment time is not affected on pigment extraction. Starch granules were not detected and large intracellular spaces were visible between the cells on light and scanning election microscopy of PEF treated PSP. This result suggests that PEF have potential to use on extraction of pigments from plant cells.

Browning and Pungent Taste Reduction Techniques in Onion Extract (양파추출물의 갈변 및 매운맛 억제기술)

  • The onion extractions of MIX treatment (5% $\beta$ cyclodextrin+1% calcium chloride+1% soluble starch mixture solution) using hot water (100${^{\circ}C}$ and 80${^{\circ}C}$) and ultrasonic treatment (25${^{\circ}C}$) incresed L values and decreased a and b values apart from the extraction methods. Extent of the browning reaction as caused by the MIX treatment (0.093) following 100${^{\circ}C}$ water extraction resulted in as low as 31% O.D. level, as compared to the control(0.296). Analysis of the pyruvic acid showed that the control had higher content of pyruric acid than MIX-treated samples. The MIX treatment had lower intensities and higher preferences of browning color and pungency taste compared to the control. The total and coliform microbial counts increased continuously during storage period, while the MIX treatment reduced the number of viable cells. Finally, it was concluded that the MIX treatment was highly effective in suppressing the undesirable browning color and pungent taste after processing, and the microbes increment during storage.

Analysis of Kernel Hardness of Korean Wheat Cultivars

  • To investigate kernel hardness, a compression test which is widely used to measure the hardness of individual kernels as a physical testing method was made simultaneously with the measurement of friabilin (15KDa) which is strongly associated with kernel hardness and was recently developed as a biochemical marker for evaluating kernel hardness in 79 Korean wheat varieties and experimental lines. With the scattered diagram based on the principal component analysis from the parameters of the compression test, 79 Korean wheat varieties were classified into three groups based on the principal component analysis. Since conventional methods required large amount of flour samples for analysis of friabilin due to the relatively small amount of friabilin in wheat kernels, those methods had limitations for quality prediction in wheat breeding programs. An extraction of friabilin from the starch of a single kernel through cesium chloride gradient centrifugation was successful in this experiment. Among 79 Korean wheat varieties and experimental lines 50 lines (63.3%) exhibited a friabilin band and 29 lines (36.7%) did not show a friabilin band. In this study, lines that contained high maximum force and the lower ratio of minimum force to maximum force showed the absence of the friabilin band. Identification of friabilin, which is the product of a major gene, could be applied in the screening procedures of kernel hardness. The single kernel analysis system for friabilin was found to be an easy, simple and effective screening method for early generation materials in a wheat breeding program for quality improvement.

Properties of Hot-water Extracts and Extract-gels of Starches for Mook (묵제조용 전분에서 분리한 열수추출물의 이화학적성질과 추출물겔의 특성)

  • In order to know the effects of hot-water extracts of starches on gel formation and gel properties, some physicochemical properties and molecular distribution of the extracts were measured. Also, after acid-hydrolysis of starch gels and extract gels, extent of hydrolysis and $\overline{D.P}._n$, and molecular distribution of hydrolysis-residue were measured. Extraction ratio of grain starches (buckwheat, corn, rice, and wheat) were $4.0{\sim}6.6%$. The ratio of acorn and mungbean starches were 11.7 and 13.5%, respectively. Iodine affinity of the extracts was $17.2{\sim}17.7%$ in starches using for Mook, SM(acorn, buckwheat, and mungbean), $13.7{\sim}14.9%$ in starches not using for Mook, SNM(corn, rice, and wheat). Blue value was also higher in SM than in SNM. $\overline{D.P}._n$. of extracts of SM were bigger than those of SNM. In molecular distribution experiment, amylopectin fraction was not found from the extracts except rice starch extract. The extent of acid hydrolysis of the starch gels were $70{\sim}84%$ after 60 days, and the extent of the extract gels were $22{\sim}35%$. The extent of hydrolysis of starch gels and extract gels made with SM were lower than those of SNM.

Potential Feeding Value of Deoiled Rice Bran by Japanese Quails. 1. The Metabolisable Energy Content

  • The apparent metabolisable energy (AME) and N-corrected ME (AMEn) of deoiled rice bran (DORB) were determined with adult quails at 6 and 10- week of age. The DORB obtained from two types of extraction process, Batch (DORB-B) and the Continuous (DORB-C), was each included in a practical type of the reference diet at 20 or 40% level. The analysed crude protein, ether extract, total ash, calcium, phosphorus, glucose and starch content of DORB-B and DORB-C were found at 19.0, 0.79, 17.05, 0.11, 1.92, 2.3, 11.22, and 15.02, 1.56, 13.0, 0.40, 2.76, 2.16, 19.0, respectively. The level of inclusion of DORB in diet appeared to exert a significant effect on the AME and AMEn values. When bioassayed at 20% inclusion level the DORB was found to have a significantly (p<0.01) lower value than that obtained at 40% inclusion level. However, no significant effect of age of quails on the AME values of DORB was evident. The ME bioassays with quails gave significantly (p<0.01) higher AME values for DORB-C than DORB-B thereby indicating that the continuous system of solvent extraction of rice bran is superior to the batch system from this point of view. The AME value of DORB predicted from its chemical composition also revealed that the DORB-C contained approximately 15% more energy than that in DORB-B.

  • A number of content-based image search methods have been proposed to this point. Each of these systems uses different image data and generates different data depending on the extraction method of different characteristics that the search capabilities of each system cannot be compared and assessed. In particular, there is a problem of applying the identical image data onto the contents based image search system on the web that cannot be compared and assessed. To resolve such a problem, the XML-based FEMAL is hereby presented for extracting data of characteristics generated from specific search system in a way that can be recognized from other starch system. In the experiment using FEMAL, the extract data for characteristics is mutually communicated and integrated and the comparison assessment of search capability is seemed to be available.

Effect of feeding raw potato starch on the composition dynamics of the piglet intestinal microbiome

  • Objective: Raw potato starch (RPS) is resistant to digestion, escapes absorption, and is metabolized by intestinal microflora in the large intestine and acts as their energy source. In this study, we compared the effect of different concentrations of RPS on the intestinal bacterial community of weaned piglets. Methods: Male weaned piglets (25-days-old, 7.03±0.49 kg) were either fed a corn/soybean-based control diet (CON, n = 6) or two treatment diets supplemented with 5% RPS (RPS5, n = 4) or 10% RPS (RPS10, n = 4) for 20 days and their fecal samples were collected. The day 0 and 20 samples were analyzed using a 16S rRNA gene sequencing technology, followed by total genomic DNA extraction, library construction, and high-throughput sequencing. After statistical analysis, five phyla and 45 genera accounting for over 0.5% of the reads in any of the three groups were further analyzed. Furthermore, short-chain fatty acids (SCFAs) in the day 20 fecal samples were analyzed using gas chromatography. Results: Significant changes were not observed in the bacterial composition at the phylum level even after 20 d post feeding (dpf); however, the abundance of Intestinimonas and Barnesiella decreased in both RPS treatment groups compared to the CON group. Consumption of 5% RPS increased the abundance of Roseburia (p<0.05) and decreased the abundance of Clostridium (p<0.01) and Mediterraneibacter (p< 0.05). In contrast, consumption of 10% RPS increased the abundance of Olsenella (p<0.05) and decreased the abundance of Campylobacter (p<0.05), Kineothrix (p<0.05), Paraprevotella (p<0.05), and Vallitalea (p<0.05). Additionally, acetate (p<0.01), butyrate (p<0.05), valerate (p = 0.01), and total SCFAs (p = 0.01) were upregulated in the RPS5 treatment group Conclusion: Feeding 5% RPS altered bacterial community composition and promoted gut health in weaned piglets. Thus, resistant starch as a feed additive may prevent diarrhea in piglets during weaning.

  • The implications of incorporating deoiled rice bran (DORB) in diet on the utilization of certain nutrients had been examined through balance studies made with 6- and 10- week-old Japanese quails. The stuffs evaluated in this study had their origin from two different types of processes- a batch extraction (DORB-B) and another continuous process (DORB-C), and each included at 10, 20 or 30% level in an iso-nitrogenous and iso-caloric diet. On chemical analyses, the DORB-B was found to contain crude protein, 19.0; ether extract, 0.79; total ash, 17.05; Ca, 0.11; P, 1.92; glucose, 2.35 and starch 11.22 g/100 g day matter. Corresponding values for DORB-C were 15.02, 1.56, 13.0, 0.40, 2.76, 2.16 and 19.0, respectively. The data on nutrient balances with quails fed diets containing the two-types of DORB suggested no significant (p>0.05) differences in per cent retention of diet dry matter (DM), organic matter (OM), ether extract (EE), gross energy (GE), nitrogen (N), crude fiber (CF), calcium (Ca) or phosphorus (P) content attributable to the extraction process. The age of the birds also appeared to have no significant (p>0.05) effect on nutrient utilization by them. However, the level of inclusion of DORB had a significant (p<0.05) effect on nutrient utilization by quails in that the retention of dietary N, EE, CF, Ca and P was better at lower than at higher levels. It is concluded that the DORB, irrespective of extraction process, can safely be employed upto 20% level in diet of adult Japanese quails without noticeable interference in utilizing the major dietary nutrients.

Efficient Extraction of Bioethanol from Freshwater Cyanobacteria Using Supercritical Fluid Pretreatment

  • For the production of ethanol from freshwater cyanobacteria, a new pretreatment method using supercritical fluid was introduced. In this study, it was found that the supercritical fluid could penetrate inside the cell wall and help to liberate starch from cyanobacterial cells which resulted in the increase of the efficiency of ethanol production. For Microcystis aeruginosa, supercritical fluid pretreatment increased the amount of ethanol produced from cyanobacteria from 1.53 g/L to 2.66 g/L. For Anabaena variabilis, the amount of ethanol was increased from 1.25 g/L to 2.28 g/L. With use of supercritical fluid pretreatment, the efficiency of the process to obtain higher ethanol yields from freshwater cyanobacteria was improved upto 80%. The optimum temperature and pressure conditions for supercritical fluid pretreatment were determined as the temperature of $40^{\circ}C$ and the pressure of 120 atm. This study demonstrates the feasibility of using supercritical fluid pretreatment for ethanol production using freshwater cyanobacteria.

Isolation and Optimal Culture Conditions of Prodigiosin-like Pigment Produced by Zooshikella sp. JE-34 (Zooshikella sp. JE-34가 생산하는 Prodigiosin계 색소의 분리 및 최적 배양조건)

  • A bacterial strain, JE-34, producing a high concentration of red pigment was isolated from a sediment in East China Sea. It was identified as Zooshikella sp. JE-34 based on the 16S rRNA gene sequence analysis. The red pigment was purified by solvent extraction and HPLC was identified as prodigiosin-like compound. Nutritional and cultural conditions were optimized for the production of prodigiosin-like pigment in the flask level. Optimal culture conditions were at initial medium pH $6.0{\sim}7.0$, $30^{\circ}C$ and 4 days incubation. For carbon and, nitrogen sources were soluble starch and malt extract.
