The protozoan pathogen Giardia lamblia has been major cause of waterborne enteric disease. In this study, we tried to identify G. lamlbia of human infectious species and to detect viable C. lamblia in river water samples including three sites of Han River mainstream and an its creek using PCR and RT-PCR technique. The PCR/RT-PCR methods were performed by using giardin primer based on the giardin gene targeting ventral disk of Giardia. Sensitivity testing in the DNA/RNA extraction and PCR/RT-PCR amplification steps showed that it was possible to detect a single cyst of G. lamblia and viable G. lamblia. The PCR/RT-PCR methods were compared with immunofluorescence(IF) assay by analyzing 48 samples collected from the mainstream water and the creek water. The mean concentration of the total cysts were 6.3 cysts/10 L(arithmetic mean, n = 48) and the positive detection rate were 62.5%(30/48). And the mean concentration of the cysts excluding empty cysts were 4.5 cysts/10 L and the positive detection rate were 52.1%(25/48). It resulted that 24 of 48 samples included Giardia lamblia by PCR assay and 10 of 48 samples included viable G. lamblia by RT-PCR assay. It resulted that the PCR/RT-PCR technique would be available to river water samples with low concentration of Giardia cysts. And it could support the Korean protozoan standard method, which provides useful information for species and viability.
Purpose: This study presents clinical results of anterior cruciate ligament(ACL) augmentation using autogenous two-strand semitendinosus tendon in the treatment of partial ACL tears with preservation of residual portion. Materials and Methods: From January 2004 to June 2006, twenty two patients who had an ACL injury underwent ACL augmentation using autogenous two-strand semitendinosus tendon were enrolled in this study. We evaluated the clinical results with regard to clinical findings(range of motion, Lachman test, pivot shift test), standard knee scales(Lysholm, Modified Feagin Scoring System), and KT-1000 arthrometer testing. Results: At minimum one year postoperatively, there was no limitation of range of motion. Lachman and pivot shift tests were negative in all knees. On instrumented anterior laxity test by KT-1000 arthrometer, mean side to side difference was improved from 4.6mm preoperatively to 1.7mm postoperatively. Average Lysholm score was improved from 70 to 92. Ninety one percent of cases were rated as good or excellent in Modified Feagin Scoring System. Conclusion: Augmentation using autogenous two-strand semitendinosus tendon with preservation of residual portion of the ACL seems to be an acceptable method for restoring knee stability and proprioceptive function.
Presently, the performance level of the college student has received a great deal of attention, particularly in the area of professional education. The problem of under achievement, especially in areas dealing with science and technology, has gone through considerable investigation by numerous Psychologists and Educators. Thus for, however, they have not been successful in determining conclusively the non-intellectual factors involved. The maladjustment problem of these students has been a thorn in the side of nursing education for some time. This topic has been discussed among nursing educators without any tangible results. Furthermore, the fact that the number of students who withdraw from nursing education programs before graduation has increased. This represents a major problem for nursing education. This problem area had increased attention drawn to it on October 1957, when Russia successfully launched to first satellite "Sputnik" into space. Various studies seem to indicate that factors related to over achievement and under.achievement can be found in the motivation of the students. This study is aimed at testing 3 hypotheses which hopefully will lead to a better understanding of the learning activities of nursing students and to determine some of their nonintellectually personality traits. Hypothesis I: learning activities in nursing students and persona1ity are correlated, there will be significant differences in personality need structures between 4 classes of nursing collegiate students. Sub-hypothesis: There will also be significant differences in the degree of student satisfaction in her major subject. Hypothesis I: If there is a special personality need structure which is required for the clinical learning activity in general, then there will be significant differences between the personality need structures of under-achievers and overachiever. Hypothesis II : If each clinical nursing subject requires different personality need structures, then there will be differences in personality need structure between the different clinical groups Methodology: 1. Tool: A Korean translation and correction of the Edwards Personal Preference Schedule of U.S.A. was used. 2. Subject and Test Method: Subjects were nursing students at Yonsei University, who enrolled for Fall semester of 1971. The Researcher herself executed the test for the test for the class as a group, and the absent students were tested indidually. Out of 307 students, 293 students (95.4%) were reported. 3. Statistical treatment: The mean and standard deviation for each of the 15 personality need variables were computed and the value of T-test was obtained to determine the differences in the personality need structures between each group. Results: 1, For the first hypothesis, the personality need structures between each of the 4 classes and the reported degree of satisfaction in major subject showed significant differences. Therefore the hypothesis I was accepted. 2. The comparison of personality need structure of under-achievers and over-achievers in clinical fields showed significant differences. Thus hypothesis II was accepted. 3. Personality need structure between clinical subject groups showed significant differences therefore the 3rd hypothesis was accepted.

Digital X-ray imaging devices using a TFT based flat panel array has been used in medical field. But, recently, lots of research on the photon counting sensor has been reported. In this study, we evaluated the physical properties of the photoconductor by suggesting the standard and testing method for quantitative performance evaluation of photon counting x-ray imaging sensor. First, we measured the leakage current and the sensitivity of photon counting x-ray imaging sensor and we evaluated the characteristic of rising time for determining the signal shaping time. In addition, the set-up study was conducted on the basis of the IEC 62220-1-2 recommendations to define the number of incident photons per unit area. And the efficiency of the charge collection was also evaluated. As a result, the characteristic was measured as $200pA/mm^2$ of the leakage current, $7{\mu}C/cm^2R $ of the X-ray sensitivity, and $0.765{\mu}s$ of the rising time.
The emergence of next-generation sequencing technologies has lead to application of new computational and statistical methodologies that allow incorporating genetic information from entire genomes of many individuals composing the population. For example, using single-nucleotide polymorphisms (SNP) obtained from whole genome amplification platforms such as the Ilummina BovineSNP50 chip, many researchers are actively engaged in the genetic evaluation of cattle livestock using whole genome relationship analyses. In this study, we estimated the genomic relationship matrix (GRM) and compared it with one computed using a pedigree relationship matrix (PRM) using a population of Hanwoo. This project is a preliminary study that will eventually include future work on genomic selection and prediction. Data used in this study were obtained from 187 blood samples consisting of the progeny of 20 young bulls collected after parentage testing from the Hanwoo improvement center, National Agriculture Cooperative Federation as well as 103 blood samples from the progeny of 12 proven bulls collected from farms around the Kyong-buk area in South Korea. The data set was divided into two cases for analysis. In the first case missing genotypes were included. In the second case missing genotypes were excluded. The effect of missing genotypes on the accuracy of genomic relationship estimation was investigated. Estimation of relationships using genomic information was also carried out chromosome by chromosome for whole genomic SNP markers based on the regression method using allele frequencies across loci. The average correlation coefficient and standard deviation between relationships using pedigree information and chromosomal genomic information using data which was verified using a parentage test andeliminated missing genotypes was $0.81{\pm}0.04$ and their correlation coefficient when using whole genomic information was 0.98, which was higher. Variation in relationships between non-inbred half sibs was $0.22{\pm}0.17$ on chromosomal and $0.22{\pm}0.04$ on whole genomic SNP markers. The variations were larger and unusual values were observed when non-parentage test data were included. So, relationship matrix by genomic information can be useful for genetic evaluation of animal breeding.
All-ceramic restorations have had a more limited life expectancy than metal ceramic crowns be-cause of their lower strength. The relatively lower strength has limited the use of all-ceramic crowns to the areas where occlusal loads are lower Therefore many researches have been done to increase the strength of all-ceramic crowns. IPS Empress 2 is a new type of lithium disilicate glass-ceramic with enhanced physical characteristics which has been in use clinically since 1998. Previous researches reported that the flexural strength of all-ceramic material was greater than 300 MPa, and all-ceramic crowns can be used in staining or layering technique. The objective of this study was to investigate the influence of the thickness of IPS Empress 2 ceramic on fracture strength. Both staining technique and layering technique was investigated. Vita VMK was used as control. For all three groups, five specimens each of 0.8mm, 1.0mm, 1.4mm, 1.8mm, and 2.2mm thick-ness (a total of 75 specimens) were prepared. Control group : Vita VMK Porcelain specimens were prepared with dentine ceramic and liquid glazing was done. Group I : IPS Empress 2 were prepared with staining technique and stained twice and glazed once. Group II : IPS Empress 2 were prepared with layering technique and glazed after wash firing. The thickness and diameter of the specimen were measured and controlled after specimen preparation. Biaxial Flexure Test (ASTM Standard F394-78) was adopted as this test method produces results least affected by the edge condition of the specimens. Fracture strength was measured with Instron Universal Testing Machine. Conclusions are as follow : 1. The fracture strength was increase in order of control group, test group I, test group II. 2. Fracture strength of the group I (Empress 2 Staining) was 65.54 N in 0.8mm, 155.2 N in 1.0mm, 233.5 N in 1.4mm, 434.5 N in 1.8mm, and 600.1 N in 2.2mm. 3. Fracture strength of the group II (Empress 2 Layering) was 190.0 N in 0.8mm, 283.5 N in 1.0mm. 437.2 N in 1.4mm, 732.0 N in 1.8mm, and 1115.0 N in 2.2mm. 4. No statistical difference was found in flexural strengths according to thickness in a specified group(p>0.05).
Parial drainage characteristics of clayey silt with low plasticity from the west coast (Incheon and Hwaseong) was analyzed using CPTU based existing correlation equations and compulsory replacement method. Generally, the estimated $OCRs={\kappa}{\cdot}((q_t-{\sigma}_{vo})/{\sigma}^{\prime}_{vo})$ using Powell and Quartman(1988) were higher than those obtained by the oeodometer tests. These trends were noticeable for the layers containing a lot of silty and sand soils. The assessment of partial drainage conditions was performed through Schnaid et al. (2004)'s equation; it is based on plotting the normalized cone resistance, $Q_t$ versus the pore pressure parameter, $B_q$ in combination with the strength incremental ratio, $s_u/{\sigma}^{\prime}_{vo}$ to the CPTU data. It is evident that more than half of the data fall in the range where $B_q$ < 0.3, corresponding to the domain in which the partial drainage prevails when testing normally consolidated soils at a standard rate of penetration (2 cm/s). To estimate the replacement depth of clayey silt with low plasticity, back analysis was carried out to evaluate the internal friction angle based on where the design depths are equal to the checked depths using bearing capacity equation. The internal friction angels obtained from the back analysis tended to increase as the plasticity index decreases, which is ranged approximately from ${\varphi}^{\prime}=2^{\circ}$ to ${\varphi}^{\prime}=7^{\circ}$.

In this paper, we implemented the urine analysis system capable of measuring a qualitative and semi-quantitative and assay using strip. The analysis algorithm of urine analysis was adopted a fuzzy logic-based classifiers that was robust to external error factors such as temperature and electric power noises. The spectroscopic properties of 9 pads In a strip were studied to developing the urine analysis system was designed for robustnesss and stability. The urine analysis system was consisted of hardware and software. The hardware of the urine analysis system was based on one-chip microprocessor, and Its peripherals which composed of optic modulo, tray control, preamplifier, communication with PC, thermal printer and operating status indicator. The software of the urine analysis system was composed of system program and classification program. The system program did duty fort system control, data acquisition and data analysis. The classification program was composed of fuzzy inference engine and membership function generator. The membership function generator made triangular membership functions by statical method for quality control. Resulted data was transferred through serial cable to PC. The transferred data was arranged and saved be data acquisition program coded by C+ + language. The precision of urine analysis system and the stability of fuzzy classifier were evaluated by testing the standard urine samples. Experimental results showed a good stability states and a exact classification.
Diagnostic performance of polymerase chain reaction (PCR) for detecting Dirofilaria immitis in dogs was evaluated when no gold standard test was employed. An enzyme-linked immunosorbent assay test kit (SnapTM, IDEXX, USA) with unknown parameters was also employed. The sensitivity and specificity of the PCR from two-population model were estimated by using both maximum likelihood using expectation-maximization (EM) algorithm and Bayesian method, assuming conditional independence between the two tests. A total of 266 samples, 133 samples in each trial, were randomly retrieved from the heartworm database records during the year 2002-2004 in a university animal hospital. These data originated from the test results of military dogs which were brought for routine medical check-up or testing for heartworm infection. When combined 2 trials, sensitivity and specificity of the PCR was 96.4-96.7% and 97.6-98.8% in EM and 94.4-94.8% and 97.1-98% in Bayesian. There were no statistical differences between estimates. This finding indicates that the PCR assay could be useful screening tool for detecting heartworm antigen in dogs. This study was provided further evidences that Bayesian approach is an alternative approach to draw better inference about the performance of a new diagnostic test in case when either gold test is not available.

The objective of this study was to determine the effects of granular type of silicate fertilizer on watermelon growth, yield, and characteristics of soil in the greenhouse. Four different levels of silicate fertilizer, 0(control), 600, 1,200, $1,800kg\;ha^{-1}$ were applied for experiment. The silicate fertilizer was applied as a basal fertilization before transplanting watermelon. Compost and basal fertilizers were applied based on the standard fertilizer recommendation rate with soil testing. All of the recommended $P_2O_5$ and 50% of N and $K_2O$ were applied as a basal fertilization. The N and $K_2O$ as additional fertilization was split-applied twice by fertigation method. Watermelon (Citrullus lanatus Thunb.) cultivar was 'Sam-Bok-KKuol and main stem was from rootstock (bottle gourd: Lagenaria leucantha Standl.) 'Bul-Ro-Jang-Sang'. The watermelon was transplanted on April, 15. Soil chemical properties, such as soil pH, EC, available phosphate and exchangeable K, Mg, and available $SiO_2$ levels increased compared to the control, while EC was similar and the concentrations of soil organic matter decreased. Physical properties of soils, such as soil bulk density and porosity were not different among treatments. The growth characteristics of watermelon, such as stem diameter, fresh and dry weight of watermelon at harvest were thicker and heavier for silicate treatment than the control, while number of node was shorter than the control. Merchantable watermelon increased by 3-5% compared to the control and sugar content was 0.4 to $0.7^{\circ}Brix$ higher than the control. These results suggest that silicate fertilizer application in the greenhouse can improve some chemical properties of soils and watermelon stem diameter and dry weight, which are contributed to watermelon quality and marketable watermelon production.
