Effects of the repetitive tasting of different blending types of coffee on teeth stain during home bleaching (자가 미백 처치 기간 중 커피 종류별 적용이 치아 착색에 미치는 영향)

  • Objectives : The purpose of this study was to examine effect of the application of different types of coffee on teeth stain during home bleaching. Methods : Twenty five premolars that were extracted within the past month and healthy without any signs of dental caries or restorations were collected and divided into five groups. The teeth of a control group were bleached everyday for two weeks, and the teeth of four experimental groups were bleached everyday for two weeks and then deposited respectively in four different types of coffee solution: brewed coffee, black coffee, coffee with sugar, and coffee with sugar and cream. Afterwards, the color of the teeth of the four experimental groups was checked by a shade pilot. Results : As a result, there were significant differences in the change of lightness and color according to the blending type with the lapse of time during home bleaching. Concerning changes in lightness and hue, there were the largest differences in the control group and the experimental group deposited in coffee with sugar and cream, followed by coffee with sugar, black coffee and brewed coffee. When the extent of change in lightness and hue was investigated after home bleaching, the experimental group deposited in coffee with sugar and cream was stained the least, and the experimental group deposited in brewed coffee was stained the most. Conclusions : In conclusion, the intake of coffee that is the cause of stain should be reduced during bleaching, and in case of having a cup of coffee, it's advisable to have coffee with cream.

  • Discoloration sensitivities of woods grown in this country haven't reported yet. Therefore we examined discoloration sensitivities of domestic wood specimens to iron (0.1 %, $FeCl_3.6H_2O$), alkali (pH 12.0, NaOH). acid (pH 1.0, $C_2H_2O_4$) and exposing to sunlight (40 hrs), Thirty-six hardwood species were collected and examined. All specimens were prepared from heartwoods of the collected species. But the specimens of 4 Betula species were divided into sapwoods and heartwoods. By iron stain, the color differences (${\Delta}E$) of 21 wood specimens including one Betula sapwood showed above 12.0, which means strong discoloration sensitivities, and of 3 specimens including one Betula sapwood showed below 2.5, which means weak discolorations. The most strong iron discoloration species was Jungkukgulpi-namu (Pterocarya stenoptera). By alkali stain, the color differences (${\Delta}E$) of 3 wood specimens showed above 9.0, which means strong discoloration sensitivities, and of 18 wood specimens including 4 Berula sapwoods showed below 2.5, which means weak discolorations. By acid stain, the color differences (${\Delta}E$) of 6 wood specimens showed above 10.0 which means strong discoloration sensitivities, and of 12 wood specimens including one Betula sapwoods showed below 2.5, which means weak discolorations. By exposing to sunlight, the color differences (${\Delta}E$) of 31 wood specimens including one Betula sapwoods showed below 6.5, which means, strong discoloration sensitivities, and of only one specimens showed below 2.5, which means weak discoloration. The most strong discoloration species by exposing to sunlight was Guirung-namu (Prunus padus). In general, it was shown that hardwoods grown in Korea were most subject to change of color by exposing to sunlight and next were by iron stain. Domestic hardwoods showed some differences in discoloration sensitivities from domestic softwoods previously reported.

  • Spermatozoa viability can be assessed by microscopy, flow cytometry, and other methods using fluorescent stain. Flow cytometry can be used to examine the morphological and functional characteristics of spermatozoa in a short time. The purpose of this study was to compare the viability of cryopreserved spermatozoa in Jeju black cattle by two dual fluorescent stain methods. Semen of Jeju black cattle raised in Subtropical Livestock Research Institute, National Institute of Animal Science, RDA were collected with artificial vaginal technique. Sperm was diluted with $Triladyl^{(R)}$-egg yolk diluent and then was performed cryopreservation. There was no significant difference in viability of spermatozoa according to the two dual fluorescent stain methods. However, when the distribution of spermatozoa according to the staining method was compared, the spermatozoa group stained with 6-CFDA/PI was more clearly distinguished than the spermatozoa group stained with calcein AM/PI.

A report of six unrecorded radiation-resistant bacterial species isolated from soil in Korea in 2018

  • Six bacterial strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 assigned to the phylum Proteobacteria, Firmicutes, and Actinobacteria were isolated from soil samples in Korea. The Cohnella species, strain 18JY42-3 was Gram-stain-positive, short rod-shaped and beige-colored. The Methylobacterium species, strains 18SH and 18JY76-11 were Gram-stain-negative, short rod-shaped and pink-colored. The Microterricola species, strain 17J11-11 was Gram-stain-positive, short rod-shaped and yellow-colored. The Paenarthrobacter species, strains 18JY14-14 and 18JY15-11 were Gram-stain-positive, short rod-shaped and white-colored. Phylogenetic analysis based on 16S rRNA gene sequence showed that strains 18JY42-3, 18SH, 18JY76-11, 17J11-11, 18JY14-14, and 18JY15-11 were most closely related Cohnella rhizosphaerae (MH497628; 98.8%), Methylobacterium goesingense (MH497632; 99.1%), Methylobacterium populi (MH497635; 99.9%), Microterricolagilva (MH504108; 98.4%), Paenarthrobacter nicotinovorans (MH497641; 100%), and Paenarthrobacter nitroguajacolicus (MH497646; 99.2%), respectively. All the six unrecorded strains showed resistance to UV radiation. This is the first report of these six species in Korea.

Study on the Combustion Characteristics of Tulip Tree (Liriodendron tulipifera) for Use as Interior Building Materials

  • In this study, the combustion characteristics of the Tulip tree, which is the representative broad-leaved afforestation tree in Korea, were analyzed. The flame retardant performance of the Tulip tree was analyzed by analyzing combustion characteristics on a total of three test samples; flame retardant treated, both flame retardant and oil stain-treated, and untreated. Then the flame retardance grade was classified for each of them. According to the result, test samples showed the strongest flame retardance were in order of flame retardant treated (C), both flame retardant and oil stain-treated (B), and untreated (A). As a result of analyzing the total heat emission and maximum heat emission rates, which is the evaluation standard for interior materials of Korean domestic buildings, test samples with flame retardant treat or flame retardant and oil stain treat were qualified for the flame-retardant standard. Both flame retardant and oil stain-treated samples showed higher total heat release (THR) and heat release rate compared to flame retardant-treated samples as the oil causes combustion with oxygen. On the other hand, they didn't qualify the THR in Quasi-non-combustible standards. To determine the correlation between the physical and combustion characteristics of wood, the combustion characteristics of other diffuse porous wood species, with which the Tulip tree is affiliated were analyzed, and noticed that the characteristic correlates with the density and quantity of wood. The results of this study are expected to be used as basic information on the combustion characteristics of the Tulip tree.

Stain Fungi and Discoloration Control on Rubberwood (Hevea brasiliensis Muell. Arg.) by Vacuum-Pressure Treatment with Catechin from Gambir (Uncaria gambir Roxb.)

  • Recently, the morphological and molecular features of five stain fungi infecting rubberwood (Hevea brasiliensis), namely Paecilomyces maximus, Paecilomyces formosus, Penicillium crustosum, Paecilomyces lecythidis and Aspergillus chevalieri, have been studied. Prior to this study, the authors revealed that catechin from gambir (Uncaria gambir) could inhibit the growth of the white-rot fungus Schizophyllum commune, and it was important to determine the bioactivity of the aforementioned agent against A. chevalieri. The efficacy of the biocidal agent was examined using a laboratory wooden block test. Rubberwood blocks, 8 mm in thickness, 20 mm in width, and 30 mm in length, were impregnated with catechin solution at concentrations of 6%, 9%, 12%, and 15% (w/v) using the vacuum-pressure method, and their bioactivity was monitored over three weeks through visual and scanning electron microscope assessment of fungal growth as well as the discoloration intensity of the wood samples. The results showed that catechin treatment increased the resistance of wood samples to A. chevalieri. Overall, the higher the catechin concentration, the lower the fungal growth. The lowest fungal growth was observed in the wood samples treated with 12% and 15% catechin (score of 0), demonstrating no discoloration. In contrast, the fungal growth score of the untreated wood samples reached 4, indicating severe discoloration. Catechins appear to be adequate biofungicides against stain fungi in rubberwood.

Opportunistic Parasites among Immunosuppressed Children in Minia District, Egypt

  • A total of 450 stool samples were collected from inpatient and outpatient clinics of Pediatric Department, Minia University Hospital, Minia District, Egypt. Two groups of patients were studied, including 200 immunosuppressed and 250 immunocompetent children. Stool samples were subjected to wet saline and iodine mounts. A concentration technique (formol-ether sedimentation method) was carried out for stool samples diagnosed negative by wet saline and iodine mounts. Samples were stained by 2 different methods; acid fast stain (modified Ziehl-Neelsen stain) and Giemsa stain. Total 188 cases (94%) were diagnosed positive for parasitic infections among immunosuppressed children, whereas 150 cases (60%) were positive in immunocompetent children ($P$<0.0001). The most common protozoan infection in immunosuppressed group was $Cryptosporidium$ $parvum$ (60.2%), followed by $Blastocystis$ $hominis$ (12.1%), $Isospora$ $belli$ (9.7%), and $Cyclospora$ $caytenensis$ (7.8%). On the other hand, $Entamoeba$ $histolytica$ (24.6%) and $Giardia$ $lamblia$ (17.6%) were more common than other protozoans in immunocompetent children.

  • Objectives : The electroacupuncture was generally has been used in oriental medicine doctors. In recent years, there have been a few studies about safety and stability of acupuncture needle in itself, but then again research of acupuncture needle safety and stability on applying electroacupuncture have been insignificant. Therefore we investigated the safety and change in mechanical characteristic of acupuncture needle in electroacupuncture therapy. Methods : We observed mechanical characteristic change by SEM(Scanning Electron Microscope) and EDX(Energy Dispersive X-ray Spectroscopy), evaluated the hardness by vickers hardness tester. We used MTT assay and cell stain to study about biocompatibility of electroacupuncture. Results : In this study, any corrosion of material, alternation of elements, and change of hardness were not observed in surface analysis using SEM and EDX. In cytotoxity evaluation using MTT assay and cell stain, cell survival rate was low when practicing the electroacupuncture for more than 3 hours. Conclusions : Change of mechanical property was not observed based on the test results using surface analysis and hardness estimation by the electroacupuncture. And considering the biocompatibility, electroacupuncture was thought to be safe in an hour based on cytotoxity evaluation using MTT assay and cell stain.

  • The cultured stain fungus isolated firstly from discolored wood of Pinus rigida was inoculated on wood blocks to investigate the invading pathway of the fungus. The inoculated fungus rapidly colonized in the ray parenchyma and moved radially along them. In process of time the fungus penetrated tracheids where it started to move into vertical direction. After 90 days the fungus colonized all over the wood tissue, therefore some cell lumens were filled with full of hyphae. In addition methanol extracts did not affect the invading pathway of the stain fungus.

Morphological Changes of Radiation-Induced Atretic Follicles in Mouse Ovary

  • This study was undertaken to investigate the morphological changes between normal and atretic follicle after gamma irradiation and treatment of follicle stimulating hormone (FSH). The ovaries of each group of treated immature mice were prepared the paraffin sections after 0, 6, 12, and 24 hours (hrs) of those treatment. Hematoxylin-eosin (HE) stain, reticulin stain, and terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick end labeling (TUNEL) immunohistochemical stain were performed on the each paraffin sections. As the results of HE staining, the condensed nuclei of oocytes were observed in the atretic primordial follicles, on the other hand the condensations of granulosa cell nuclei were prominent in the atretic primary, preantral, and antral follicles. Only the granulosa cells of atretic follicle were stained specifically with TUNEL staining but not stained in the theca cells, which suggested granulosa cells degenerated through apoptosis. In the reticulin staining, the basement membranes of atretic follicle which was stained weakly showed irregular structure and detachment from the follicles. The ratio of normal to atretic follicle in control and FSH treated group was about 33% but this ratio increased rapidly over 90% in the 6, 12, and 24 hrs group after the irradiation. It could be suggested that the gamma irradiation is the useful tool far the induction of follicle atresia and immunohistochemical staining methods are essential in the study of follicle atresia.
