Effect of glucose and nitrogen sources on production of gellan

  • The gellan was produced by Pseudomonas elodea ATCC 31461 under aerobic condition. Gellan provides various functionalities such as gelling, suspending, stabilizing, emulsifying and binding properties in aqueous systems. In this study, the effect of glucose concentration and nitrogen sources concentration, bacto peptone and $NH_4NO_3$, on the cell growth and the production of gellan were evaluated. The maximum specific yield (g gellan/g cell) of 2.22 was obtained at 1.0% of glucose. Bacto peptone increased cell growth. And nitrogen limitation was essential for higher production of gellan. The highest cell and gellan production were abtained at 0.5 g/${\ell}$ of bacto peptone without $NH_4NO_3$.

  • In an automotive spark ignition engine, it is important to form the proper mixture (air/fuel) on each driving condition for developing the stabilizing combustion and exhaust characteristics. Since most of supply fuel si attached on the inside wall of the intake manifold for unadequate nonuniformity of fuel distribution to each cylinder and mixture variation. Also it affects engine performance variation and causes noises and vibration. In this study, we verified the effect of the mixture variation which is caused by fuel liquid film in the intake manifold on combustion characteristics and engine performance.

  • In a premixed flame stabilized by a streamlined step, the flame structure and combustion characteristics were investigated to identify the effect of the pressure pulsation in a combustion air. A flame stabilizing limits, visualization, mean temperature, ion current and gas concentration (O$_{2}$, CO$_{2}$, CO, UHC) were measured. With the combustion air of higher pressure pulsation, the development of the mixing layer was fast and wide, the temperature and combustion intensity were higher at arbitrary section. But, the effect was notably decreased with X=150 mm downstream. And a first eddy formation from step edge was earlier. Thus, with the combustion air of higher pressure pulsation, high rate of heat generation was expected.

  • Employing the Phan-Thien tanner (PTT) fluids model, dynamic behavior of the non-isothermal melt spinning has been investigated. Subjects such as draw resonance instability, the effects of spinline cooling and of the fluid viscoelasticity on the spinning dynamics have been studied using the governing equations of the system. In particular, the draw resonance criterion based on the traveling times of various kinematic waves in the spinline has been confirmed, the reason why the spinline cooling is stabilizing is analyzed, and the effect of fluid viscoelasticity on the spinline stability is summarized. It is believed that the same method as in this study can be applied with equal ease to other extension deformation processes like film casting and film blowing.

Zeolite-Mediated Cation Exchange Enhances the Stability of mRNA during Cell-Free Protein Synthesis

  • The addition of zeolite particles enhances the stability of mRNA molecules in a cell-free protein synthesis system. When $20{\mu}g/{\mu}L$ of zeolite (Y5.4) is added to a reaction mixture of cell-free protein synthesis, a substantial increase in protein synthesis is observed. The stabilizing effect of zeolite is most dearly observed in an in vitro translation reaction directed by purified mRNA, as opposed to a coupled transcription and translation reaction. Upon the addition of zeolite in the in vitro translation reaction, the life span of the mRNA molecules is substantially extended, leading to an 80% increase in protein synthesis. The effect of zeolite upon the mRNA stability appears be strongly related to the cation exchange (potassium to sodium) reaction. Our results demonstrate the possibility of modifying this biological process using heterogeneous, non-biological substances in a cell-free protein synthesis system.

Rheological Behaviour of Emulsions Containing Polyacrylamide (폴리아크릴아마이드를 함유한 에멀젼의 유변학적 거동)

  • We have investigated the rheological behaviours of emulsions containing polymers. The polymers used in this study were polyacrylamide and carboxy vinyl polymer. The emulsion with polyacrylamide shows better prior preference than that with carboxy vinyl polymer on the skin texture. In the toxicity test, such as, LLNA cell toxicity, skin organ culture and human patch test, polyacrylamide was the appropriate materials for the cosmetics. The viscosity of polyacrylamide shows maximum value in neutral pH and good stability against temperature. The decreasing effect of viscosity against salt was not good enough but showed a better result than that of carboxy vinyl polymer. Two times the amount of polyacrylamide should be used for the formation of a similar viscosity of carboxy vinyl polymer. Yield value of polyacrylamide was low and the effect for the stabilizing emulsions was also low.

  • The shape of nozzle cross-section plays an important role in stabilizing electrospray jet. The angle of contact line is governed based on the famous Young-Laplace equation. Compared to a round nozzle that has a constant curvature along the orifice, the square nozzle has four square corner edges and four straight edges that hold the meniscus in a different manner and is of interest in this study. By utilizing both square and round capillary nozzle, we examine the effect of nozzle shape in electrohydrodynamic jetting. The ejections were recorded with a high speed camera and analyzed to examine the jetting repeatability based on dynamic movement of meniscus. The result suggests that if the corner edges are not sharp, then its effect on repeatability is also limited.

  • Polyurethane microcapsules were synthesized by interfacial polymerization in an aqueous poly(ethylene glycol) dispersion with ethylenediamine as chain extender of toluene diisocyanate in perfume oil using poly(vinyl alcohol) as the stabilizing agent. The effect of chemical structure on the average particle size and distributions, morphologies, and thermal properties to design microcapsules for the sustained release system was investigated. It came to be known that polyurethane microcapsules with ethylene diamine as chain extender had a rounder, more permeable and controlled release membranes. And the release test of polyurethane microcapsules with different soft segment content was done to certify the effect of long methylene chain. According to the higher molecular weight of polyether polyol, the release rate of microencapsulated disperse dye molecular was faster.

An Experimental Study on Breakup Mode of Epoxy Paint Discharging from a Fan Spray Nozzle (선형분무노즐로부터 분무되는 에폭시계 도료의 분열기구에 대한 실험적 연구)

  • In the present work, the breakup mechanism of highly viscous epoxy paints discharged from a fan spray nozzle was examined experimentally. The paints tested were non-Newtonian fluids, composed of epoxy resin, solid particles and other additives. The paint spray discharged from the nozzle was visualized and recorded using a digital camera with back illumination. Due to presence of the solid particles, perforation of liquid sheet was observed in most cases, even at low-Reynolds number conditions (Re < 15,000) where the aerodynamic-wave breakup mode is used to be dominant for pure liquids. However, with the increase of the particle concentration, the sheet became longer and the thickness at breakup became thinner to some extent. This is because, with higher concentration of solid particles, the stabilizing effect by the viscosity increase predominates over the destabilizing effect by perforation.

Effect of ethanolic extract of some anti-asthmatic herbs on clonidine and haloperidol-induced catalepsy in mice

  • The ethanolic extract of some medicinal plants having anti-asthmatic activity such as Solanum xanthocarpum, Curcuma longa, Glycyrrhiza glabra, Piper longum, A. vasica, A. lebbeck, and Tinospora cordifolia was evaluated for antihistaminic and anti-cataleptic activity. The aqueous solution of ethanolic extract of S. xanthocarpum and G. glabra potentiated histamine-induced tracheal chain contractions. Whereas, C. longa, P. longum, and T. cordifolia, and A. lebbeck were without any significant effect on histamine. Only A. vasica inhibited histamine-induced tracheal chain contraction. G. glabra per se produced contraction of the tracheal chain, which was blocked by pretreatment with atropine. Single dose of S. xanthocarpum potentiated clonidine-induced catalepsy but on repeated doses (once in a day for 3 days) inhibited catalepsy. Pretreatment with ethanolic extract of C. longa, P. longum, T. cordifolia inhibited catalepsy whereas G. glabra and A. lebbeck significantly potentiated clonidine-induced catalepsy. None of the extracts inhibited haloperidol-induced catalepsy. Thus the extracts having antihistaminic activity or mast cell stabilizing activity inhibited clonidineinduced catalepsy.
