Monascus sp. MK2 균주가 생산하는 홍국색소의 안정성 (Stability of Monascus Pigment Produced by Monascus sp. MK2)

  • 본 연구에서는 Monascus sp. MK2가 생산하는 홍국색소의 안정성에 대하여 조사한 결과에서 광 안정성에서는 암 조건>자외선>실내(형광)>태양광 조건 순서로 안정하였으며, 온도에 대한 안정성은 온도가 증가하고 저장시간에 길어짐에 따라 홍국색소의 감소율 또한 비례적으로 증가하였고, pH에 대한 안정성은 전체적인 pH 영역에서 비교적 안정하였으며, 유기산 첨가에 대한 홍국색소의 안정성은 높은 것으로 생각되며, 합성 항산화제와 천연물 추출물 첨가시에는 안정성이 증대되는 것으로 조사되었다.

홍국의 황색색소 Monascin에 대한 안정성 연구 (Stability of Monascin Pigment Isolated from Monascus purpureus)

  • The stability of monascin yellow pigment isolated from Monascus purpureus was determined over a period of storage for the wide range of pH, various metal ions and antioxidants. The absorption maximum of monascin pigment was 385 nm. Monascin pigment was more stable in acid solutions than in alkaline (pH 9 and pH 11) during storage period. It was also observed the reduction of absorption was occur after 3 days storage. The stability of monascin pigment was not changed by adding the various metal ions of the concentration of 10-4 M, however, it was unstable by adding the Zn2+, Al3+ and Fe3+ of 103- M concentration. The antioxidants. BHA, BHT, cysteine and L-ascorbic acid, have no effects on the stability of monascin yellow pigment. Thus, it may be concluded that the monascin pigment is stable and useful food additives as the natural colorant except for the alkaline food and food containing the Zn2+, Al3+ and Fe3+.

떫은감 Carotenoid의 색소 안정성 (The Stability of Carotenoid Pigments in Astringent Persimmon(Diospyros kaki) Consumed in Korea)

  • Studies on extraction and color stability of carotenoids from astringent persimmon(Diospyros kaki) were performed to provide the basic information for the utilization of persimmon pigment as a new source of natural food colorant. The major carotenoids in astringent persimmon were beta-cryptoxanthin, lycopene and beta-carotene. Beta-cryptoxanthin was the first major pigment and lycopene the next. Total carotenoid content in persimmon calculated as beta-carotene equivalent was 107.4 $\mu\textrm{g}$/g. The data indicated that the astringent persimmon was a good source of carotenoid pigment. The physical and chemical stabilities of carotenoid from astringent persimmon were investigated at various conditions of temperature, pH and antioxidant. The effects of oxygen and light on the stability of carotenoid pigment has been investigated. The factors that cause the discoloration were visible light, temperature above 60, pH below 5 and oxygen. Especially, the carotenoid pigment was very sensitive to light and oxygen. Carotenoid stability was much improved with increasing nitrogen concentration in the atmosphere. Persimmon pigments have been found to be most stable at 5∼10. And tocopherol was the most effective inhibitor of the pigment discoloration.

자색고구마로부터 붉은 색소 농축물의 추출 및 그 색소 분말의 제조 특성 (Extraction of Natural Red Color Pigment Concentrate and Manufacturing Characteristics of Pigment Powder from Purple-Fleshed Sweet Potato)

  • Pigment concentrates with violet-red color and sweet taste were obtained from purple-fleshed sweet potato(PFSP) using ethyl alcohol and water. Extract from general potato(GP) were used as a control. The relative stability of PFSP pigment concentrate(PFSPPC) in a storage test over 15 days was confirmed in the order of dark > fluorescence > sun-light irradiation. The relative stability of GP pigment concentrate(GPPC) in a storage test over 15 days was confirmed in the order of sun-light > fluorescence > dark storage. The RRP of PFSPPC was higher than that of GPPC, but the color strength of GPPC was 1/2 that of PFSPPC. Treatment of PFSPPC with aluminum potassium sulfate(0.2~0.3%, w/w) best improved its stability. The improved RRPs of PFSPPC were 45.16~47.31% in sun light irradiation, 55.91~60.22% in fluorescence irradiation, and 76.34~75.97% in dark storage conditions. In substituting aluminum potassium sulfate for chitosan, an amount of 0.2~0.3%(w/w) was suitable, giving similar results in improving pigment stability for all concentrates tested. Also, freeze-dried PFSPPC powder was manufactured as a substitute for dextrin, and also as a substitute for chitosan to the extent of 0.25%(w/w). The results of storage stabilite for freeze-dried PFSPPC and GPPC powder over 15 days, irradiation were, PRRs of 74.47~89.36% and 61.54~76.92%, respectively. The stability improving effect of freeze dried PFSPPC powder was confirmed by the results of storage experiments at various conditions. The use of freeze-dried PFSPPC powder was therefore confirmed to be an effective treatment for general foods.

Zooshikella sp. 17TA 색소 추출물의 물리화학적 안정성과 항균활성 (The Physicochemical Stabilities and Antimicrobial Activities of Pigment Extracts from Zooshikella sp. 17TA)

  • In this study, the stability of the extracted natural pigments against light, temperature, pH, metal ions, and antimicrobial activity were evaluated in marine bacteria Zooshikella sp. 17TA. The pigment of the strain used in the study was red with maximum absorption at a wavelength of 541 nm. The stability of the pigment was evaluated by measuring the absorbance while preserving for 15 days and examining the retention rate. After 15 days of irradiation, the pigment of this bacterium showed 98% retention in the dark and 91% retention in the temperature range of -20℃ ~ 30℃. When the pH was in the range 4-7, the retention was about 80%, and the retention rate was higher than 85% for all kinds of metal ions except for CuCl2, ZnCl2, and KCl. The bacterial pigments showed high stability under the given irradiated pH, temperature, and metal ion conditions and had shown activity against gram-positive strains. These results suggest that this highly conserved microbial pigment can be applied to the food industry.

선인장 열매로부터 추출된 점질물 및 색소의 기능성 (Functional Properties of Mucilage and Pigment Extracted from Opuntia ficus-indica)

  • Functional properties of mucilage and pigment extracted from Opuntia ficus-indica var. saboten were determined at various temperatures, pHs and alcohol concentrations. The crude mucilage extracted from pickly pear showed pH 4.2, 0.14% total acidity and 8.1% total soluble solid content(w/w, wet basis). Polysaccharide was purified from mucilage extract by isopropanol precipitation. Intrinsic viscosity of polysaccharide was 18.1dl/g and decreased with increasing KCl concentration. Relative viscosity and color stability of mucilage extract were determined with capillary viscometer and spectrophotometer at 534nm, respectively. In additions of 1~20%(v/v) ethanol, the red pigment of mucilage extract was very stable, but relative viscosity, increased gradually. For heating above 7$0^{\circ}C$, the stability of red pigment decreased drastically, but rheological property of mucilage was not changed. During storage, the red pigment was extremely unstable at above pH 8.3. At both pH 3.0 and pH 4.2, the red pigment was the most stable at 4$^{\circ}C$ for 18 days. In the case of storage at 37$^{\circ}C$, pigment of mucilage extract at pH 3.0 was destroyed more quickly than that at pH 4.2. Natural mucilage extract(pH 4.2) showed the good stability of red pigment at 3$0^{\circ}C$ for 10 days.

멍게 껍질(Ascidian shell)로부터 추출한 천연색소의 안정성에 대한 연구 (Studies on the Stability of Natural Pigment Extracted from Ascidian shell)

  • 본 연구는 멍게 껍질을 100.0 % 에탄올 용매로 추출한 후 DMSO로 희석하여 색소안정성을 평가하였다. pH에 따른 흡광도와 색차계 값을 측정한 결과 pH 7.0에서 흡광도와 색도의 ${\pm}a$값이 가장 안정하게 나타났다. pH 3.0에서 흡광도의 감소가 나타났고 색도의 ${\pm}a$값이 감소하여 멍게 껍질 색소는 pH가 중성에 가까울수록 변색방지의 효과가 있을 것으로 판단된다. 항산화제 첨가시 색소의 흡광도가 증가하였으며 ${\alpha}-tocopherol$과 Glutathione이 가장 우수한 효과를 나타내었다.

Characteristics of Red Pigment from Marine Bacterium Utilizing Colloidal Chitin

  • Studies on extraction of red pigment was performed to provide the basic information for the utilization of red pigment as s new source of natural food colorant. A bacterium isolated from marine resources were carried out the test for excretion of red pigment. One strain of a marine bacterium, KSR-97 showed a high production of red pigment on the medium of colloidal chitin, peptone-yeast extract with minerals. In physicochemical and sensory properties in aqueous solution of red pigment was investigated at various condition of pH, temperature, concentration of ethanol and stability of storage. Potent antioxidative of red pigment was separated by thin layer chromatograpy, silica gel chromatography and reverse high performance liquid chromatography using ODS hypersil column.

검정콩 종피 색소추출액의 안정성 (Stability of Black Soybean Pigment Extract)

  • 한국산 검정콩 종피의 천연색소로의 이용가능성을 알아보기 위하여 온도, 당류, 유기산류, 비타민 C 그리고 금속 이온의 영향을 살펴보았다. 검정콩 색소의 열안정성은 다른 안토시아닌 계열의 색소와 비교할 때 유사하거나 더 안정한 것으로 나타났다. 당류의 영향에서는 타 색소와는 달리 농색화 현상은 일어나지 않았고 저장성에서는 모두 포도당에서 양호하였다. 유기산에서는 모든 첨가구에서 색의 강도가 증가하였으며 비타민 C 첨가의 경우 안토시아닌 색소에 부정적인 영향을 나타내었고 금속이온의 영향에서는 $Cu^{2+}$$Zn^{2+}$ 이온에서 안정하였으며 $Mn^{2+}$에서는 불안정하였다. 이상의 결과를 종합해 볼 때 검정콩 종피색소는 pH 3에서 다른 색소와 비교하여 상당히 안정한 색소임을 알 수 있었다.

해양세균 Pseudoa/teromonas psicida TA20의 색소 추출물의 물리화학적 안정성과 생리활성 (The Physicochemical Stabilities and Biological Activities of Pigment Extract from Marine Bacterium Pseudoalteromonas psicida TA20)

  • 해양세균 Pseudoalteromonas psicida TA20의 ethanol 색소 추출물에 대한 안정성과 기능성을 검토한 결과, 본 해양세균 색소는 pH 4.0에서 pH 8.0의 조건과 $40^{\circ}C$ 이하에서 매우 안정하였으며, $25^{\circ}C$ 14일간 90% 이상의 잔존율을 나타내어 빛에 대한 안정성이 매우 우수한 것으로 나타났다. 금속이온의 경우 $Al^{3+}$$Cu^{2+}$를 제외한 실험된 모든 금속이온에 대하여 매우 안정하였으며 특히, 다른 색소추출물들에 비해 $Fe^{2+}$에 대한 안정성이 높은 것으로 나타났다. 해양세균 색소추출물에 대한 생리활성 실험 결과, free radical 소거 활성 (3495 ${\mu}g/ml$)이 나타났으며, 색소농도 10 ${\mu}g/ml$일 때 44%의 인체세포 DNA 손상 회복능이 있음을 보여 항산화능이 우수함을 알 수 있었다. 또한 주요 식품유해세균에 대하여 항균활성을 나타내었다. 따라서 본 해양세균 색소는 항산화능과 항균활성을 갖는 식품 색소로의 적용을 검토할 수 있을 것으로 기대된다.
