Purification and Identification of Squalene Synthase Inhibitor Isolated from Fermented Soybean Paste

  • Squalene synthase (SQS) inhibitors were screened from various plants and food extracts. Effective SQS inhibitor was purified from fermented soybean paste using ethanol extraction, HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, and crystallization. Through UV spectrometry, $^1H$ NMR, $^{13}C$ NMR, and mass spectrometry, SQS inhibitor was identified as daidzein with molecular mass of 254 and molecular formula of $C_{15}H_{10}O_4$. Daidzein showed $IC_{50}$ value of 50 nmol/L against SQS, confirming its potential as therapeutic agent for hypercholesterolemia.

Effects of Squalene on the Rat Liver Treated with a Anticancer Agent (Squalene이 항암제를 투여한 흰쥐의 간에 미치는 효과)

  • This paper aims to probe the effect of SQ in the rat liver which pretreated with CP was examined by transmission electron microscope. In the A group, the difference between the normal and the treated groups were not detected at 24 hours, but the few mitochondria were expanded at the 72 hours. In the B group, the cisternae of rough-surfaced endoplasmic reticulum were partially destructed and attached ribosomes were remarkably decreased at 24 hours. A number of the mitochondria were dilated and increased in number, the filamentous materials also detected at 72 hours. These results suggest that SQ is not only concerned with construction of the membrane of the cell organelles but also decreased the cellular toxicity in the hepatic cells.

Major Chemical Constituents of Supercritical Carbon Dioxide Extract of Pandanus odorus Leaves

  • Supercritical carbon dioxide was used as a solvent in the extraction of freeze-dried Pandanus odorus leaves. Analysis of the extract with GC-MS showed that Pandanus odorus leaves contain nutritional constituents such as ${\alpha}-tocopherol$ (Vitamin E) and squalene. The contents of ${\alpha}-tocopherol$ and squalene extracted from freeze-dried ground Pandanus odorus leaves at pressures ranging from 80 to $200\;kg\;cm^{-2}$ and temperatures between 40 to $80^{\circ}C$ were $134{\sim}300\;ppm$ and $750{\sim}1,200\;ppm$ respectively. The highest yield was obtained at $200\;kg\;cm^{-2}$ and $40^{\circ}C$. Other major components identified were hexadecanoic acid, 9,12,15-octadecatrien-1-ol, campesterol, stigmasterol and ${\beta}-sitosterol$.

Pre- and Post-Germination Changes in Pharmaceutical Compounds of Germinated Brown Rice (현미 종류별 발아현미의 발아 전 후 생리활성물질 함량의 변화)

  • Recent consumption of germinated brown rice is increasing due to its health-beneficial effects. To clarify germination-dependent changes in functional compound contents, 5 different types of (ordinary, glutinous, green-kerneled, red-colored and black-colored) rices were selected and their pre- and post-germination content changes in vitamin E (${\alpha}-$, ${\beta}-$, ${\gamma}-tocopherols$ and ${\alpha}-$, ${\gamma}-$, ${\delta}-tocotrienols$), squalene, and phytosterols (campesterol, stigmasterol, sitosterol) were evaluated. In the case of vitamin E isomer contents, germination generally resulted in decreasing tendency around 10% in most isomers as well as in total tocopherol and tocotrienol contents in all types of rices. In the case of squalene, significant increase could be observed in glutinous rice (by 31%), while other types of rices exhibited no changes or slight reduction by germination. Regarding stigmasterol contents, all tested types of germinated brown rice exhibited significant increment by 19 to 27% compared to nongerminated cases, except for green colored-rice which showed statistically not significant 5% increment. No changes or slight reductions, however, could be observed in campesterol and sitosterol, as well as in total phytosterol contents. As a conclusion, most of tested pharmaceutical compounds exhibited statistically not significant changes except for stigmasterol which were increased by the germination process.

Molecular Cloning and Characterization of the Yew Gene Encoding Squalene Synthase from Taxus cuspidata

  • The enzyme squalene synthase (EC 2.5.1.21) catalyzes a reductive dimerization of two farnesyl diphosphate (FPP) molecules into squalene, a key precursor for the sterol and triterpene biosynthesis. A full-length cDNA encoding squalene synthase (designated as TcSqS) was isolated from Taxus cuspidata, a kind of important medicinal plants producing potent anti-cancer drug, taxol. The full-length cDNA of TcSqS was 1765 bp and contained a 1230 bp open reading frame (ORF) encoding a polypeptide of 409 amino acids. Bioinformatic analysis revealed that the deduced TcSqS protein had high similarity with other plant squalene synthases and a predicted crystal structure similar to other class I isoprenoid biosynthetic enzymes. Southern blot analysis revealed that there was one copy of TcSqS gene in the genome of T. cuspidata. Semi-quantitative RT-PCR analysis and northern blotting analysis showed that TcSqS expressed constitutively in all tested tissues, with the highest expression in roots. The promoter region of TcSqS was also isolated by genomic walking and analysis showed that several cis-acting elements were present in the promoter region. The results of treatment experiments by different signaling components including methyl-jasmonate, salicylic acid and gibberellin revealed that the TcSqS expression level of treated cells had a prominent diversity to that of control, which was consistent with the prediction results of TcSqS promoter region in the PlantCARE database.

Effects of Squalene on Renal Toxicity Induced by Lead Acetate in Proximal Tubules of the Mice (생쥐 근위세뇨관에서 납에 의한 신장 독성에 대한 스쿠알렌의 효과)

  • To investigate the effect of the squalene against the lead toxicity and recovery of renal failure. Healthy male ICR mice were used for experiment. The activity of nitric oxide (NO) was observed after the intraperitoneal injection in mice. The ultrastructural changes of the kidney were observed after the intraperitoneal injection of lead acetate in mice. The experimental groups were divided into three groups. Group 1 was normal mouse. Group 2 was not treatment with squalene after intraperitoneal contamination of lead acetate (30 mg/kg). And, Group 3 was injected squalene (180 mg/kg) after intraperitoneal contamination of lead acetate. All groups were used to 10 mice. The results were as follow: In the case of the group 2, swelling of the outer membrane and destruction of the inner membrane (cristae) of the mitochondria, dripping of the ribosomes from the rough endoplasmic reticulum were happened at 24 hours and 48 hour. These were gradually reparied after 72 hours. In the case of the group 3, damages of the mitochondria and the rough endoplasmic reticulum were showed less than the group 2 at 24 hours. Especially, after 48 hours, these were almost same as the group 1. In the case of group 2, the level of NO was decreased. However, In the case of group 3, the level of NO was increased more than normal as well as repaired the decreased NO level by Pb (P<0.05). It was concluded that the squalene was the protective and recovery effects for the toxicity of the lead in the renal proximal tubules.

Effects of Squalene on The Epidermal Growth Factor (EGF) Expression and Histological Changes by Glycerol-Induced Acute Renal Failure in Mice (Glycerol-유도 급성신부전에서 표피성장인자 발현 및 조직학적 변화에 관한 스쿠알렌의 효과)

  • Kidney had recovery functions against toxicants, ischemia, reperfusion-induced damage, acute-renal failure (ARF). Urinary epidermal growth factor (EGF) is produced by the juxtaglomerular apparatus. Kidney accumulates or excretes the EGF. In case of renal diseases, excreted EGF was decreased. The aim of this study is to evaluate the effects squalene (SQ) on the prevention of experimental acute renal failure induced by glycerol. In case of in vitro study, we investigated the expression of EGF by RT-PCR. After the proximal tubular cells was isolated, glycerol (1, 2, 4 mM) or glycerol plus squalene (0.1, 0.05 or 0.1%) was added. In case of in vivo study, we investigated the changes of BUN, creatine, and ultrastructure. Experimental groups were divided into four groups. Group 1 was normal mouse. Group 2 was injected with SQ only (180 mg/kg). Group 3 was not treated with squalene after intraperitoneal contamination of glycerol (50%, 8 ml/kg). And, Group 4 was treated with squalene (180 mg/kg) after intraperitoneal contamination of glycerol (50%, 8 ml/kg). All groups were used to 7 mice. In the results, we investigated the glycerol induced renal failure. The expression of EGF mRNA was decreased in renal proximal tubules when treated with only glycerol. SQ increased the mRNA expression of EGF in renal proximal tubules. SQ also quickly recovered the levels of BUN and creatine compared with those of mice treated with only glycerol (P<0.01). In case of ultrastructure, group 3 had heavily damaged mitochondria, but, mitochondria in group 4 had evidences of the recovery. It was concluded that SQ had the recovery effects for the glycerol-induced acute renal failure.

  • The natural full-fat rice bran is reported to contain 8.4 to 14.7 wt % Lipids, but the amount and composition of bran depend on the type of rice, quality of paddy, pretreatments to paddy such as parboiling, type of milling system employed, and the degree of polishing. These lipids are usually mixtures of several class fatty acids containing palmitic acid, linolenic acid, linoleic acid, oleic acid, stearic acid, tocopherol, squalene, etc. In this study the oil rich essential fatty acid (EFA) including squalene was extracted from the domestic brown rice bran using supercritical fluid extraction (SFE) and cosolvent induced SFE process, respectively. And the extracts were analyzed with GC-MSD. The extracted amount of rice bran oil was dependent upon the operating pressure and temperature, and the fatty acid composition of oil was varied with the reduced density (${\rho}_{\gamma}$) of supercritical carbon dioxide. About 70~80% of rice bran oil was extracted in 4hrs. The cosolvent induced SFE process shortened the total extraction time, extracted greater amount of oil than SFE process. Especially squalene which was not found in solvent extract phase was identified in SFE and cosolvent induced SFE process.

Healing Effects of Squalene on the Epidermis in Burned Mouse (생쥐의 화상에 미치는 Squalene의 치료 효능)

  • The purpose of this experiment was to know the healing effects of saualene on the burned mouses. The experimental groups were divided into seven groups: the control group, the burned groups, and 50 treated groups after burned. All groups except the control group burned with second degree on a dorsoanterior part. All groups that treated with 50 adapted three drops of pure squalene at 10 seconds interval; one time a day. The histological and ultrastructural changes during 10 days after burned were observed by light and electron microscope. Under the light microscope, all parts of epidermal layer of the burned groups were wounded deeper than the 50 treated groups. At 6 and 10 days groups with 50 treated, especially 10 days, the basal layer was greatly differentiated, and the prickle cells in the spinous layer were greatly increased in number. Under the electron microscope, the cell divisions of basal layer in all groups that treated with 50 were more activated and rapidly regenerated than the all burned groups. Especially, some fantastic results obtained from the 10 days group with SQ treated; much thicker spinous layer than the control group, many prickle cells, fine intercellular bridges, and healthy basal layer contained melanocytes. These results suggest that squalene may active the epidermis growth factor (EGF), acts as scavenger, and provide sources of energy in the membrane system. The results of this experiment consider that squalene has specific effects for burn healing and regeneration.

  • Introduce of gene connected with disease and transformation system of ginseng, Squalene systhase(PSS) gene cloned from and disease resistant gene were carried out for expression and transformation of plant using Agrobacterium. PSS of 35S-35S-AMV-PSS-Tnos, has been constructed which were mobilized into Agrobacterium tumefaciens strain MP 90 disarmed Ti-plasmid. PSS gene were introduced into the binary vector pRD 400. The transgenic ginseg plants were propagated using repetitive secondary embryogenesis and introduced NPTII and PSS genes of the transgenic ginseng were successfully indentified by the PCR and survival test on the medium.
