Studies on the Mulberry Sapling 1. Soil Characteristics of Nursery Garden Producing Nonsprouting Sapling. (뽕나무 접목묘에 관한 연구 I. 활착불량상묘 발생 묘포지의 토양특성에 관한 연구)

  • soil survey and chemical analysis on mulberry nursery garden were carried out to study the causes of the non-sprouting phenomenon occured seriously in Buy and Ogchun in 1983 and Sangju in 1984. In addition, 115 nursery garden soils taken from 21 sapling produsers in Chungbuk province were analyzed in 1984. The results were as follows, 1. Symptom of dead saplings was the highest in boron deficiency by 59% and the lowest in rot symptom. The rest of saplings by 34.9% were not detected any symptom. 2. The nursery gardens showen non-sprouting phonomenon were located along the stream. As the result, soil depth was shallow, around 20cm deep and subsoil composed with coarse sand and gravels. 3. Nursery soils were mostly strong acid, low in Ca and Mg content, especially B, whereas available phosphorus and potassium were abundant at some gardens, and deficient at others. 4. Application of red earth or borax to nursery garden, especially to paddy, increased sprouting rate. 5. Paddy nursery garden occupied by 52.8% of 60.8ha of total garden area in Chungbuk province. Ninty point five percent of the nursery garden was lower in pH than 6.5, 87.0% lower in K than 0.5me/100g, 40.8% lower in Ca than 6.5me/100g, 94.8% lower in Mg the 2.00me/100g 99.1% lower in B than 0.3ppm.

  • The experiment on sprouting and emergence properties of Eleocharis kuroguwai was conducted at laboratory and greenhouse conditions in 1993. Most of tubers had the apical dominance of buds at sprouting and had the competitive relationship for mesocotyl elongation. When pyrazosulfuron was applied on soil surface, all of apical buds were killed, but the first and second lateral buds survived to 50% and 90%, respectively, and the third lateral buds were almost at presprouted state. The tubers at 10-15cm deep soil emerged 1 week late and 35% low in emergence rate compared with the tubers at 2-5cm deep. As the plants emerged from the deep soil, the mesocotyl and the roots were distributed at deep soil more or less. Among the tubers buried at 10cm and 15cm deep soil, 25% and 30% respectively were not emergerd due to the suspension of mesocotyl elongation. Although the roots and shoots were removed from the tubers at the stage of 20cm high, all of mother tubers emerged again even 2 weeks late.

Elucidation of Cause of Cotyledon Black-Decay of Soybean Sprout by Bean Bug, Riptortus clavatus

  • Cotyledon Black Decay (CBD) on soybean sprout mimics the black spot due to microbial infection. CBD, not visible or predictable at seedlot state, for some reason, shows up exclusively on cotyledon of soybean sprout during sprouting process. Such an incidence rate fluctuated from 0.8 to 19.5% over three years from 2004. We suspected some pod-infecting anthracnose fungi and/or pod-blight pathogen, or pod-sucking bean bug, one of the major pests of soybean, might have involved, of which we ruled out fungal pathogen because it was preventable through heat treatment, a proven method for seedlot disinfestation. The healthy seeds artificially fed by bean bug for one to seven days were sprouted, and 6 to 41% of the soybean sprout revealed the CBD mimic to those occurred in soybean sprout from previous commercial seedlot screening experiments. This finding is the first report to confirm that bean bug damage to pod at $R_8$ stage is directly responsible for the CBD, which did not concur with any other deleterious effects on sprouting such as reduction in hypocotyls elongation and rooting except unsightly sprout quality. However, earlier feeding either at green pod or greenish yellow pod stage ($R_6$ -early $R_7$ stage) resulted in rather severe damages, which strikingly reduced hypocotyls growth to about one forth to about two third, as well as the reduction in rates of seed germination.

Dormancy Breaking by Hot-water Treatment of Lilium Oriental Hybrids Bulblets Regenerated in Vitro (온탕처리에 의한 오리엔탈나리 소인경의 휴면 타파)

  • The objective of this study was to investigate the effect of hot water and dipping times, and hot- PGR's solution on dormancy breaking of Lilium Oriental Hybrid 'Casa Blanca' and 'Siberia' bulblets regenerated in vitro. Hot-water treatments with distilled water were effective not sprouting rate but shortening of days to sprouting. While $100mg{\cdot}L^{-1}\;GA_{4+7}$ was effective in the dormancy breaking process of bulblets. The content of mitochondrial protein and fumarase activity were highest in bulblet of $100mg{\cdot}L^{-1}\;GA_{4+7}$ addition in hot water treatment. Therefore $GA_{4+7}$ addition in hot-water treatment seem to be the alternative dormancy breaking method to low-temperature treatment in Lilium Oriental Hybrid bulblet regenerated in vitro.

  • This experiment was conducted to clarify the effects of physiological properties in culture media on the proliferation and the acclimation of bulblets of several kinds of lilies cultured in vitro. Proliferation rate of bulblets from bulblet of Oriental hybrid ‘Marco Polo’, ‘Casa Blanca’ and Asiatic hybrid ‘Jolanda’ was higher in solid MS media compared with bulblets from scale Acclimation rate of regenerated bulblets was high in solid media which was pre-cultured in vitro, compared with liquid media. Sprouting rate of bulblets from bulblet was higher than bulblets from scale.

A New Wheat Variety, "Sukang" with Good Noodle Quality, Resistant to Winter Hardiness and Pre-harvest Sprouting (내한 내수발아성 제면용 밀 신품종 "수강밀")

  • "Sukang", a winter wheat (Triticum aestivum L.) cultivar was developed by the National Institute of Crop Science, RDA. It was derived from the cross "Suwon266" / "Asakaze" during 1994. "Sukang" was evaluated as "Iksan312" in Advanced Yield Trial Test in 2005. It was tested in the regional yield trial test between 2006 and 2008. "Sukang" is an awned, semi-dwarf and hard winter wheat, similar to "Keumkang" (check cultivar). The heading and maturing date of "Sukang" were similar to "Keumkang". Culm and spike length of "Sukang" were 90 cm and 8.1 cm, longer culm length and similar spike length compared to "Keumkang" (80 cm and 7.9 cm, respectively). "Sukang" had similar test weight (819 g/L) and lower 1,000-grain weight (40.2 g) than "Keumkang" (813 g/L and 44.9 g, respectively). "Sukang" showed resistance to winter hardiness and pre-harvest sprouting, which lower withering rate on the high ridge (4.5%) and rate of pre-harvest sprouting (0.2%) than "Keumkang" (21.9% and 30.4%, respectively). "Sukang" had lower flour yield (71.1%) and higher ash content (0.45%) than "Keumkang" (74.1% and 0.42%, respectively). "Sukang" showed lower lightness (89.13) and higher yellowness (10.93) in flour color than "Keumkang" (90.02 and 9.28, respectively). It showed higher protein content (12.8%) and gluten content (11.1%) and lower SDS-sedimentation volume (56.8 ml) and mixing time of mixograph (2.6 min) than "Keumkang" (11.9%, 10.2%, 62.3 ml and 4.7 min, respectively). Fermentation properties, amylose content and pasting properties of "Sukang" were similar to "Keumkang". "Sukang" showed different compositions in high molecular weight glutenin subunits (HMW-GS, $2^{\ast}$, 13+16, 2+12) and puroindolines (pina-1b/pinb-1a) compared to "Keumkang" ($2^{\ast}$, 7+8, 5+10 in HMW-GS and Pina-1a/Pinb-1b in puroindolines, respectively). "Sukang" showed lower hardness (4.53 N) and similar springiness and cohesiveness of cooked noodles (0.94 and 0.63) compared to "Keumkang" (4.65 N, 0.93 and 0.64, respectively). Average yield of "Sukang" in the regional adaptation yield trial was 5.34 MT/ha in upland and 4.72 MT/ha in paddy field, which was 4% and 1% lower than those of "Keumkang" (5.55 MT/ha and 4.77 MT/ha, respectively). "Sukang" would be suitable for the area above $-10^{\circ}C$ of daily minimum temperature in January in Korean peninsula.

Effect on Identification of Irradiated Wheat and Soybean by the Full-overlapped Gravitational Field Energy(FGFE) Treatment (중첩중력에너지가 방사선 조사된 밀과 대두의 판별특성에 미치는 영향)

  • The aim of this study was to investigate the changes in identification markers of irradiated foods after treatment of the full-overlapped gravitational field energy (FGFE). Wheat and soybean samples were irradiated at 0-5 kGy of Co-60 gamma energy, and analyzed for photostimulated and thermo luminescence characteristics (PSL and TL) and sprouting rate at 0 and 6th month after FGFE treatment. As a screening method for irradiated samples, PSL photon counts (PCs) for the non-irradiated samples appeared negative (<700 PCs), while irradiated samples gave positive (>5,000 PCs). But FGFE-treated irradiated samples appeared intermediate (700-5,000 PCs), showing decreased PCs during storage. The TL analysis on irradiated samples exhibited glow curve peaks in range of $150-200^{\circ}C$ and TL ratio ($TL_1/TL_2$) was also >0.1. Therefore, identification of irradiated samples was possible using thermoluminescence. But the glow curve range of FGFE-treated irradiated samples shifted from $150-200^{\circ}C$ to $180-230^{\circ}C$ and TL intensity was decreased 37-60% resulting from FGFE treatment. After 6 months of storage, all the samples showed a decrease in TL intensity, but identification was still possible. The sprouting rate of irradiated samples decreased by about 72%, whereas that of FGFE-treated irradiated samples showed by about 85%, as compared to non-irradiated samples. More detailed study is required to investigate sprouting phenomena for FGFE-treated samples.

Effect of Packaging Methods and Temperatures on the Shelf-life of Korean Yam(Dioscorea opposita Thumb.) during Marketing (포장방법 및 온도가 유통중 마의 품질에 미치는 영향)

  • The influence of packaging methods(0.03mmPE, 0.05mmPE, 0.1mmPVC, VACUUM) and marketing temperatures(2$0^{\circ}C$, 1$0^{\circ}C$) on the shelf-life and quality were investigated in yam(Dioscorea). Respiration, weight-loss and rotting-rate was reduced in polyethylene film packaging in the order of VACUUM, 0.1mmPVC, 0.05mmPE, 0.03mmPE and the effect was more outstanding at 1$0^{\circ}C$ than 2$0^{\circ}C$. The ethylene concentration of PVC packaging group was higher than that of other packaging methods, which was related with sprouting. firmness was higher in low-temperature than in room-temperature. PPO activity was lower in vacuum than in other packaging methods, which was associated with browning. The shelf-life of yam was the longest in vacuum Packaging, 3 weeks at 2$0^{\circ}C$ and more than 4 weeks at 1$0^{\circ}C$.

Effect of Application of Nitrogen and Phosphate Fertilizer on Yield and Storage of Onions (Allium CePa L.) (질소 및 인산 시비량이 양파의 수량 및 저장성에 미치는 영향)

  • This study was carried out investigate to optimum levels of nitrogen and phosphate fertilizer to get a higher yield and a quality of an onion(Allium Cepa L.) from 1993 to 1994. Three fertilizer levels were applied with 120, 240 and 360kg/ha of nitrogen and 100, 200 and 300kg/ha of P2O5 during the onion growing season. After harvesting the onion, it was storaged at given deposit. The growth and yield of the onion were better at the fertilizer levels of both 240 and 360kg/ha of nitrogen and of both 200 and 300kg/ha of P2O5 than at the level of 120kg/ha in nitrogen and 100kg/ha of P2O5. The content of total nitrogen and P2O5 in plant was decreased with reduced application levels of nitrogen and phosphate fertilizer. The rotting rate was higher at the levels of both 360kg/ha in nitrogen and 300kg/ha in P2O5 than the other treatment during the storage period. The sprouting rate tended to be high at 240kg/ha of nitrogen and 100kg/ha of P2O5.

Effects of Seed Decontamination Treatments on Germination of Red Radish Seeds during Presoaking (적무 새싹종자의 소독제 처리에 의한 발아 시 미생물 제어효과)

  • The antibacterial effects of seed decontamination during presoaking before sprouting as an intervention step for eliminating foodborne pathogens on red radish seeds were evaluated. The effect of seed decontamination on seed germination rate was also evaluated. Red radish seeds were inoculated (at a level of 3 to 4 log CFU/g) with Listeria monocytogenes ATCC 19111 and decontaminated with 20,000 ppm calcium hypochlorite, 50 and 100 ppm chlorinated water, acidic electrolyzed water, low-alkaline electrolyzed water, and ozonated water for 6 hours. The control seeds were immersed in distilled water. The germination rate was measured on each treatment for 48 hours. Treatments with 20,000 ppm calcium hypochlorite, acidic and low-alkaline electrolyzed water were more effective than treatments with chlorinated water and ozonated water. Immersion in 20,000 ppm calcium hypochlorite resulted in the largest microbial reduction (more than 3 logs). Treatments with acidic and low-alkaline electrolyzed water reduced APC by 3 logs and L. monocytogenes counts by 2 logs. After sprouting, APC and L. monocytogenes counts on seeds treated with 20,000 ppm calcium hypochlorite, acidic and low-alkaline electrolyzed water were significantly lower than the control. The germination rate ranged from 93.5% to 97.7% except for 20,000 ppm calcium hypochlorite (from 82.3% to 84.8%) after 48 hours. Although the treatments tested in this study will not eliminate L. monocytogenes on inoculated red radish seeds, the results show that rapid growth of surviving cells during sprouting could be prevented if red radish seeds are given a presoak treatment used in combination with a disinfectant treatment of irrigation water.
