Spores of Trichoderma harzianum TCC 52445 for control of Rhlzocronia stem canker on potato were immobilized in various natural matrix, and germination rate and some rheological properties of the spore-matrix complex were Investigated. Germination rate of gelatin gel-spore complex and potato starch gel-spore complex were 2.8% and 2.9%, respectively, but hardness and cohesiveness of the gelstin gel-spoke complex were better than those of potato starch-spore complex. The hardness and cohesiveness were increased when the spores were immobilized in hybrid gelatin gel made by mixing several different types of natural matrix, but decreased their germination rate. Addition of corn steep liquor (1%) as spore nutrient in gelatin gel-spore complex was helpful for increasing the germination rate.
The effects of some chemicals for the germinative stimulation of Clostridium chauvoei spore studied. The results obtained were as follows: 1. Cooked meat medium (CMM) was superior to Liver-liver bouillon (LLB) for cultivation of the organisms. 2. Heating the organisms at $70^{\circ}C$ for 30 minutes prior to cultivation in CMM stimulated the germination rate. 3. The addition of 0.5mM L-alanine to CMM was found to be most effective for the rapid germination of the spores. 4. The addition of 0.2 mM D-alanine to CMM inhibited the germination of the spores even if the spores were heat treated. 5. The addition of 0.1mM manganese retarded the germination time, but the final germination rate was greater with 0.1mM manganese than with 0.5mM L-alanine. 6. The addition of both 0.5mM L-alanine and 0.1mM manganese stimulated the germination rate to 100 per cent. 7. The germination rate was greater with the addition of germination stimulants than without any stimulants. The germination rate was approximately 6 per cent greater with prior heat treatment than without heat treatment. 8. The optimum time for the harvest of vegetative forms of the organisms was 15 hours after cultivation in the media which contain suitable germination stimulants.
Initiation of spore germination in filamentous fungi such as Aspergillus nidulans and Botrytis cinerea requires the presence of nutrients. In this study, involvement of sugar sensing machinery was suggested in the germination of A. nidulans spores. Germination did not occur when the spores of A. nidulans were incubated in distilled water, whereas they were successfully germinated in the presence of 5% glucose with a germination rate of over 98% after 6hr incubation. Similar results were obtained when the spores were incubated in the presence of various sugars such as fructose, sucrose, and starch. Interestingly, spore germination was not observed in the presence of D-arabinose, whereas L-arabinose could induce germination as determined by the formation of germ tubes, indicating the presence of sugar sensing machinery that distinguish between the enantiomers of sugars. This inference was further supported by a decrease in germination rate (less than 25%) upon treatment of spores with trypsin. Subsequent MALDI-TOF mass spectrometry analysis of the surface proteins of spores identified ten proteins among which eight were involved in sugar metabolism. Taken together, our results suggest that spore germination in A. nidulans is initiated by the interaction of sugars with sugar binding proteins on the surface of spores.

A study was carried out to explore the method for the germination of the spore of Ganoderma. Three strains of Ganoderma were examined using 2 different media such as PDA and LBA at three different temperatures. As it has been known the germination rate was very low under all the conditions examined. G. lucidum ASI 7004 showed 0.02% germination on both media at $25^{\circ}C$ and $30^{\circ}C$ respectively. The germination rate of ASI 7091 was 0.05% on PDA medium at $30^{\circ}C$. The germination rate of G. oregonense ASI 7067 was 0.67% on PDA at $35^{\circ}C$. LBA medium was found to be inadequate for the germination of Ganderma species in this study. In a consequent study, four mating types of G. oregonense such as $A_1B_1,\;A_1B_2,\;A_2B_1\;and\;A_2B_2$ were identified.

This study was conducted to determine the optimal conditions of growth medium, temperature, and light quality for efficient propagation of Lygodium japonicum spores. The rate of spore germination and prothalium development was high in Knop and 1/8MS and 1/4MS media, which had low mineral content; in particular, the germination rate exceeded 74%, and the germinated spores developed into heart-shaped prothallia. However, in Knop‘s medium with the lowest mineral content, a rapid prothallium senescence was observed; in 1/4MS medium, prothallium development was delayed. Germination rate increased with the increase in temperature and reached its maximum, 86.7%, at 30℃; however, at this temperature, the prothallia were thinner and abnormal development of rhizoids was observed compared to normally developed prothallia and rhizoids at 25℃. Therefore, the results suggested that the optimal temperature for L. japonicum spore germination was 25℃. The rate of germination was also measured under different light conditions, and the highest rate of 90.6% was observed under LED red light compared to fluorescent (77.2%) or LED blue (5.4%) lights. The germinated spores developed into heart-shaped prothallia under LED red light; however, 15 days after seeding, prothallium development decreased and the became elongated. In contrast, a normal and continuous development of heart-shaped prothallia was observed under fluorescent light.
From soil samples, 380 antagonistic microorgnisms were isolated. Among the isolates, 42 strains had mycelia growing inhibition ability against Fusariun solani, ginseng root rot causing pathogen. Isolates CHA 1 and S-PFHR 6 were proposed as antagonists for this study and they were identified as Promicromonospora sp. and Pseudomonas pseudoalcaligenes respectively. As an antagonism against hyphae of F. solani in dual culture test, CHA 1 and S-PFHR 6 inhibited linear growing, caused abnormal branching, and the membrane projection which formed by cell wall destruction. The secondary metabolites contained in the culture filtrates which prepared from PD broth and Nutrient broth inhibited the spore germination to 14.3%. The culture filtrate of S-PFHR 6 which prepared by a little amount of soil extract addition to nutrient rich medium had more strongly. inhibited the spore germination and spore germination decreased to less than 4.0% in it. The soil used in this study had fungistasis and the germination rate of macroconidia and chlamydospore of F.solani was 19.4% and 17.7% respectively. The steam sterilized soil lost fungistasis and germination rate of conidia increased to more than 97.9%. The soils amended with the propagule of CHA 1 and S-PFHR 6 increased fungistasis and the germination rate of macroconidia decreased to 14.7% and 11.7% respectively in each treatments. But the soil ammended with glucose and asparagine annulled fungistatic ability and the germination rate of macroconidia increased to more than 48.0%. As an antagonistic activity of the secondary metabolites of two antagonistic isolates in soil, the germination rate of macroconidia of F. solani was 9.3% in the soil amended with the culture filtrate of CHA 1 but the culture filtrate of S-PFHR 6 had no such activity. In the soil which treated with antagonist propagule or culture filtrate, the chlamydospore germination rate was lower than that in natural soil. The addition of glucose and asparagine to antagonist propagule treated soil did not enhanced the chlamydospore germination.
The commercial product "pyto-patch" prepared as nano sized silver particle less than 5 nm, has effective antifungal activity against Collectotrichum gloeosporioides, Botrytis cinerea, Sclerotinia sclerotiorum in vitro. As a fungal growth inhibiton mechanism, it can reduce spore germination rate and mycelial growth. As s promising fungicide, Phyto-patch can control anthracnose effectively. The spore of C. gloeosporioides dipped in 5 ppm phyto-patch dilute suppressed germination rate to 13.2%, and mycelial growth stopped for 15 days at 10 ppm. The spore postinoculated on 10 ppm phyto-path smeared PDA surface could not germinate for 3 days and prohibit pathogen infection effectively. In field test, the anthracnose development of 4 ppm phyto-patch treated plot was less than 7% after 21 days compaired to 40% of it in untreated plot. In heavy rainfall season, pepper anthracnose effectivly contrrolled by regular 10 ppm phytopatch spraying every 7 days. The diseased pepper fruit decreased to 5.8% compaired to 94.6% in untreated plot. During drying period, the diseased pepper fruit havested in phyto-patch treated plot was 24.2%, but pepper fruit havested in untreated plot destroyed to 100% within 3 days. The nano silver particle coated on multching textile prevented late blight of pepper effectively and disease occurance delayed about month.

This study was conducted to investigate the optimal conditions for spore germination, prothallus propagation, sporophyte formation, and seedling growth in Polystichum braunii (Spenn.) $F{\acute{e}}e$. The rate of spore germination and early prothalium development was high in Knop (41.2%), which had low mineral content. The optimal medium for prothallus propagation and sexual organ formation was 2MS medium (2% sucrose). Among the various mixtures of cultivation soil (bedding soil, peat moss, perlite, and decomposed granite), a mixture of bedding soil and decomposed granite at a ratio of 2:1 (v:v) had a positive effect on sporophyte formation (276.3 ea/$7.5m^2$). The most efficient conditions for promoting the growth of sporophytes were pots filled with only bedding soil.
Fusarium verticillioides is an important pathogen of maize, being responsible for ear rots, stalk rots, and seedling blight worldwide. During the past decade, F. verticillioides has caused several severe epidemics of maize seedling blight in many areas of China, which lead to significant losses. In order to understand the molecular mechanisms regulating fungal development and pathogenicity in this pathogen, we isolated and characterized the gene fpk1 (GenBank Accession No. EF405959) encoding a homolog of the cAMP-dependent protein kinase catalytic subunit, which included a 1,854-bp DNA sequence from ATG to TAA, with a 1,680-bp coding region, and three introns (lengths: 66 bp, 54 bp, and 54 bp), and the predicated protein precursor had 559 aa. The mutant ${\Delta}fpk1$, which was disrupted of the fpkl gene, showed reduced vegetative growth, fewer and shorter aerial mycelia, strongly impaired conidiation, and reduced spore germination rate. After germinating, the fresh hypha was stubby and lacking of branch. When inoculated in susceptible maize varieties, the infection of the mutant ${\Delta}fpk1$ was delayed and the infection efficiency was reduced compared with that of the wild-type strain. AU this indicated that gene fpk1 participated in hyphal growth, conidiophore production, spore germination, and virulence in F. verticillioides.
Hyphal growth by Fusarium moniliforme was best at -14 bars osmotic water potential. Hyphal growth was prevented at -94 bars. The production of microconidia was best at -14 bars osmotic potential and prevented at -84 bars regardless of Strain. In contrast, this fungus sporulated macroconidia best at -1.4 bars and progressively less with each increment drop in water potential below that of basal media. The rate of spore germination followed a similar pattern with all of the spores; uniformly maximal at about -1.4 bars and progressively slower as the water potential was lowered from -1.4 bars to -42 bars. Under the natural conditions, plants infected by F. moniliforme produce microconidia on the dead tissues instead of producing macroconidia. This phenomenon agrees well with the water potential experiment since the dead plant tissues have a lower water potential than the living plant.
