Development of a Fungal Spore Aerosol Generator: Test with Cladosporium cladosporioides and Penicillium citrinum

  • As the first step to develop efficient means to control fungal spore bioaerosols, we designed, manufactured, and evaluated a fungal spore aerosol generator. We studied the physical and biological properties of the fungal spore bioaerosols on two common fungal species. The results demonstrated that the fungal spore bioaerosol generator effectively produces fungal spore bioaerosols.

  • Apexes of basidia in Rhodophyllus(synonyum of Entoloma) muraii var. albus are divided into four sections or depressed in the center. A spore is formed by inflation of the apex of the sterigma. The apex of the sterigma is swollen and changed from a papilla through a penisform and a club into a globeform. Six spots of globose spore are regularly or irregularly depressed with hilum axes. Finally the spores are cuboid. Basidia of Entoloma squamiferrum are developed from hymenium layer of crator-shaped parabasidium. Apex of basidium is flat or depressed in the center. Although four sterigmata are developed, only two sterigmata are symmetrically swollen to two spores in certain basidia. It means that two sterigmata among four sterigmata are infertile. The apex of the sterigma is swollen into a paillaform, and then turned into a penisform. It is swollen from a clubform into a globeform. Six spots of the surfaces of globose spore are deperssed with hilum axes. Finally the spore is cuboid, and then it is released from hilum. Four sterigmata of papillaform of E. violaceobrunneum are developed from cartor-shaped basidium. The apex of sterigma is swollen to a small globeform. And then it is swollen to a clubform. The clubform is again swollen to a ellipticalform, and then more than six spots of spore surfaces are ramomly depressed with hium axes. When the depression of surface of a elliptical spore is over, it is the multi-angular spore of the heterometrical-form.

  • The establishment of algal spores plays an essential role in adult kelp distribution and abundance patterns. Sedimentation is a key variable regulating algal spore settlement and success, possibly controlling species-specific dominance $in$ $situ$. Laboratory experiments were used to determine spore attachment and survival rates of two Alaskan canopy-forming kelps, $Nereocystis$ $luetkeana$ (K. Mertens) Postels & Ruprecht and $Eualaria$ $fistulosa$ (Postels & Ruprecht) M. J. Wynne, to various types of sediment loading. Spore attachment for both species was significantly and similarly affected by three sediment treatments: suspended particles; settled sediment covering the substratum; and smothering of attached spores by settling sediment. Spore attachment decreased by approximately 90% at 420 mg sediment $L^{-1}$, the highest sediment load tested here, under all three treatments for both species. These results suggest that increases in sedimentation may constrain the success of the spore stages, but sediment does not seem to be a likely factor explaining species-specific distribution patterns. However, while sedimentation affected spores of both species similarly, timing of spore release in relation to times of maximum sediment load in the water might differ for different species, possibly explaining kelp species distribution patterns.

An Approach for Lactulose Production Using the CotX-Mediated Spore-Displayed β-Galactosidase as a Biocatalyst

  • Currently, enzymatic synthesis of lactulose, a synthetic prebiotic disaccharide, is commonly performed with glycosyl hydrolases. In this work, a new type of lactulose-producing biocatalyst was developed by displaying β-galactosidase from Bacillus stearothermophilus IAM11001 (Bs-β-Gal) on the surface of Bacillus subtilis 168 spores. Localization of β-Gal on the spore surface as a fusion to CotX was verified by western blot analysis, immunofluorescence microscopy, and flow cytometry. The optimum pH and temperature for the resulting spore-displayed β-Gal was 6.0 and 75℃, respectively. Under optimal conditions, it showed maximum activity of 0.42 U/mg spores (dry weight). Moreover, the spore-displayed CotX-β-Gal was employed as a whole cell biocatalyst to produce lactulose, yielding 8.8 g/l from 200 g/l lactose and 100 g/l fructose. Reusability tests showed that the spore-displayed CotX-β-Gal retained around 30.3% of its initial activity after eight successive conversion cycles. These results suggest that the CotX-mediated spore-displayed β-Gal may provide a promising strategy for lactulose production.

When the depression of surface of a elliptical spore is over, the spore is a heterodiametrical spore of multi-angular.

Effects of Light, Desiccation and Salinity for the Spore Discharge of Gracilaria verrucosa (Rhodophyta) in Korea

  • The effects of light, desiccation and salinity on the discharge of spores in Korean agarophyte, Gracilaria verrucosa were studied. Among the examined factors, light after darkness was the most effective for spore discharge. The maximum release of tetraspores was induced at 24 h after the treatment. Desiccation also seems to be conductive to the release of tetraspores. However, its effect, as in treatment of distilled water for salinity, was hardly found in induction of carpospore discharge. This may suggest that spore discharge in this alga is primarily related with photoperiodic rhythm. Also it appears that the amount of light energy received by fertile thalli also significantly affects to the spore release, considering relationship between the amount of the discharged spores and the elapsed time after treatment.

Effect of Ent-norsecurinine, an Alkaloid, on Spore Germination of Some Fungi

  • The inhibitory activity of ent-norsecurinine alkaloid was evaluated against spore germination of some plant pathogenic fungi(Curvularia maculans, Curvularia species, C. palliscens, Colletotrichum gloeosporioides, Colletotrichum species, Afternaria solani, A. brassicae, Fusarium udum, Helminthosporium echinoclova and H. penniseti). It inhibited spore germination of all the test fungi. C. maculans, C. species, and C. palliscens were the most sensitive as complete inhibition of spore germination was observed at 1000 ppm. A. solani was not inhibited by this chemical.

  • Mycelial growth resistant isolates of Botryosphaeria dothidea to benomyl showed 99~79% spore germination on the PSA media supplemented with 200~2,100 $\mu\textrm{g}$/ml of carbendazim and thiophanate-methyl to manifest the high cross-resistance in spore germination. Mycelial growth, 23~9 mm in colony diameter, also manifested the high cross-resistance of mycelial growth together with similarity of spore forming cross-resistance. Benomyl resistant isolates BR1, BR2 and BR3, grew 23~10 mm in colony diameter at 330~3,000 $\mu\textrm{g}$/ml of captafol, captan and oxine-copper showing the high double resistance of mycelial growth and spore formation with minor difference. However, within concentration range of the 3 fungicides tested, germinations of all the tested isolates were completely suppressed to show no double-resistance in the fungal spore germination.

Effect of Tetrahydropalmatine, an Alkaloid on Spore Germination of Some Fungi

  • The tetrahydropahnatine alkaloid was assayed against spore germination of some saprophytic and pathogenic fungi e.g., Alternaria solani, A. brassicicola, A. brassicae, A. alternata, Erysiphe pisi, Curvularia lunata, C. pallescens, C. maculans, Curvularia species, Colletotrichum species, C. musae, Helminthosporium echinoclova, H. pennisetti, H. spiciferum, and Heterosporium sp. It inhibite spore germination of all the fungi tested. Colletotrichum spp. Curvularia lunata, Helminthosporium spiciferum and Heterosporium sp. were most sensitive as complete inhibition of spore germination was observed at very low concentration(200 ppm).

Fed-batch cultivation for cell growth and spore production by probiotic B. polyfermenticus SCD

  • The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.
