Effects of Capsosiphon Fulvescens Extract on Contact Dermatitis in NC/Nga Mice (매생이(莓山苔) 추출물이 NC/Nga 마우스의 접촉성 피부염에 미치는 효과)

  • Objectives : The aim of this study is to report the effect of Capsosiphon fulvescens(Maesangi) extract(MSE) on contact dermatitis(CD). Methods : CD induced by Dermatophagoides farinae body(Dfb) in NC/Nga mice. 4% sodium dodecyl sulfate(SDS) $150{\mu}l$ and Dfb(10mg/ml) ointment 100mg were topically treated twice a week for four weeks. MSE 200 mg/kg was topically treated every 4weeks. Every week, we examined the skin lesions, weight of spleen and lymph node, epidermal thickness, production of Tumor necrosis factor-alpha($TNF-{\alpha}$) and interleukin-6(IL-6). Results : MSE reduced features of Dfb-induced skin lesions, weight of spleen and production of $TNF-{\alpha}$. MSE significantly reduced weight of lymph node, epidermal thickness, number of mast cells and production of IL-6. Conclusions : MSE may have a potential therapeutic effects for CD by suppressing allergic inflammation.

  • Experiment was performed to investigate the immunotoxicity of cadmium administered orally and the effect of ginseng petroleum ether fraction on it. Mice were given 3, 30, or 300 ppm cadmium as cadmium chloride orally in the drinking water and injection of ginseng petroleum ether fraction intraperitoneally for 4 weeks. Mice were sensitized and challenged with sheep red blood cells (S-RBC). Humoral immune response was evaluated by antibody production and Arthus reaction. Body weight gain, spleen weight, thymus weight, and liver weight were also measured. In the present study, cadmium generally suppressed the humoral immunity. There was decrease in the rate of body weight gain and liver weight to body weight by cadmium-administration. Ginseng petroleum ether fraction showed restoring effect, to some extent, on the decrease in PFC and in the rate of liver weight to body weight by cadmium-administration. But it more suppressed Arthus reaction.

The Bisphenol A: A Modulator of Pregnancy in Rats

  • Bisphenol A is used in the manufacture of epoxy, polycarbonate, and corrosion-resistant unsaturated polyester-styrene resins required for food packaging materials in industrial processing. Some reports indicated the possibility of harmful effects on rats. In this study was used a method for the determination of bisphenol A in blood according to the OSHA High Performance Liquid Chromatography (HPLC) guideline. The method involved blood extraction using methylene chloride. And it was evaluated developmental and teratogenic effects in pregnant rats and second generation. The results obtained were as follows. There was a significant increase in the body weights and treated groups F1 female in liver, spleen, kidney, but according to dose-response. F1 female rat's relative body weight and absolute body weight are not different. There was a significant increase liver, spleen, kidney organ weight and reproductive organ weight epididymis, prostate gland in F1 male rats. There was a proestrous in pregnant rat, group 200 $\mu\textrm{g}$/kg, 2000 $\mu\textrm{g}$/kg, 20,000 $\mu\textrm{g}$/kg. The effect on rat treated with bisphenol A decrease organ weight and reproductive organ weight. Identification and quantitation were performed with using HPLC C18 column and using at retention time 5.5 min. The results of the detection of bisphenol A were at 20,000 $\mu\textrm{g}$/kg in average 1 $\mu\textrm{g}$/ml, 200 $\mu\textrm{g}$/kg average in 0.9 $\mu\textrm{g}$/ml blood samples. From those results, it could be concluded that the effects of pregnant rat and second generation(F1) by bisphenol A treatment during lactational period were estrogenic and bisphenol A was remained in serum at low level.

Effects of Sophorae Radix on skin condition in mice with contact dermatitis induced by Dinitrofluorobenzene (고삼(苦蔘) 추출물이 접촉피부염이 유발된 생쥐의 피부에 미치는 영향)

  • Objectives : The dried root of Sophora flavescens Aiton have been used to treat patient with skin diseases such as eczema in Eastern countries. S. flavescens can clear away heat and dry dampness and purge sthenic-fire from the liver and gallbladder. Recently, anti-allergic effect of S. flavescens has been reported. However, the effect on skin condition of contact dermatitis still remains unknown. Thus, the purpose of this study is to evaluate the efficacy of S. flavescens on contact dermatitis. Methods : In order to investigate the effects of S. flavescens (methanol extract of S. flavescens, MESR) on skin condition, the contact dermatitis was induced in mice skin by using 1-fluoro-2,4-dinitrofluorobenzene (DNFB). As the effective endpoints, skin thickness, skin weight, histopathological change were checked. In addition, effects on skin lesion, body weight and spleen/body weight ratio were also investigated. Results : Topical application of MESR ($500{\mu}g/day$) lowered skin thickness (P < 0.05) and skin weight (P < 0.05), respectively. MESR-treated group showed diminished spongiosis and immune cell infiltration in skin tissues compared to those of non-treated control group. The inhibition of skin lesions was also observed in MESR-treated group. In addition, MESR did not affect body weight gain and spleen/body weight ratio in contrast with those in dexamethasone-treated group. Conclusion : These data suggest that Sophorae Radix could improve skin lesion of contact dermatitis. This indicates the possibility for Sophorae Radix to be used to patients with skin diseases such as contact dermatitis.

Effects of Partial Cervus elaphus Linne' Extract on Antitumoral Immune Response in Melanoma-induced Mice (부위별(部位別) 녹용(鹿茸)이 흑색종(黑色腫) 유발(誘發) 생쥐의 종양억제(腫瘍抑制)와 면역기능(免疫機能)에 미치는 영향(影響))

  • Introduction: In order to investigate the effects of partial Cervus elaphus Linne' extract on antitumoral immunological, a group of mice are melanoma-induced and observed reponses in terms of the number of lymphocyte, CD4+ count, CD8+ count, CD4+/CD8+ ratio in blood and spleen, change in body weight, melanoma weight, spleen index, NK cell activity and productivity of IL-2 in each mouse Methodology: Male C57BL/6 mice were chosen as an experimental object and were divided into 5 groups randomly selection. The normal group did not receive any induction. The control group was treated with normal saline in melanoma-induced' mice. Sample I group induced the upper part of Cervus elaphus Linne' extract in melanoma-induced mice. Sample II group was induced the middle part of Cervus elaphus Linne' extract in melanoma-induced mice. Sample III group was induced the lower part of Cervus elaphus Linne' extract in melanoma-induced mice. The dosage of medication was 0.2cc daily for 14days. Results: 1. There was a significant difference in the number of lymphocyte in spleen in the sample I (upper part of Cervus elaphus Linne' extract induced) and the sample II (middle part of Cervus elaphus Linne' extract induced) compared to the control group and the sample III (lower part of Cervus elaphus Linne' extract induced). On the other hand, no significant difference was observed in the number of lymphocyte in blood in the control group and all sample groups. 2. In the CD4+ T cell ratio in blood, all three sample groups showed differences compared to the control group, though there was no significant difference between sample groups. In the CD4+ T cell ratio in spleen, there was a difference between the sample I and the control group, while the sample II and the sample III had significant difference to the control group. And also, it has been observed there were differences between the sample I and the other samples. 3. In the CD8+ T cell ratio in spleen, all three sample groups showed significant differences compared to the control group, while there was no difference between groups in the ratio in blood. 4. In the CD4+/CD8+ T cell ratio in blood, the sample I showed a significant difference compared to the control group, while the sample 1I and sample III showed differences compared to the control group. In the CD4+/CD8+ T cell ratio in spleen, all three samples showed a significant difference compared to the control group, when the sample I had a difference to the other sample groups. 5. The spleen index of the sample I and the sample II showed a significant difference compared to the sample III and the control group. In comparison between the sample groups, the sample I and the sample II showed a significant difference to the sample III. 6. In terms of the change in body weight and melanoma weight, all three sample groups showed a significant difference compared to the control group, while the comparison between the sample groups showed the sample and the sample II had a significant difference to the sample Ⅲ. 7. In comparison of NK cell activity, the sample I had a difference compared to the other groups when the effector to target cell ratio was 2.5:1. With the ratio of 5:1, the sample I and sample II showed significant differences compared to the control group, while the sample Ⅲ showed a difference. When the effector to target cell ratio was 10:1, there was no difference between groups. 8. In the productivity of IL-2, all three sample groups showed significant differences compared to the control group. In comparison between sample groups, there were significant differences between each sample groups in order of the sample I , the sample II and the sample Ⅲ. Conclusion: As one can witness from the above results, administration of partial Cervus elaphus Linne extract played important role in antitumoral immune response in melanoma-induced mice, and it could be suggested that sample I and sample II groups have prominent antitumoral immune effect.

Effect of Magnetic Field Exposure on Semen Characteristic and Organ Weight in Mice (자기장이 웅성 생쥐의 정액성상과 장기무게에 미치는 영향)

  • This study were performed to investigate the effect of magnetic field exposure on semen characteristic and the weights of body, reproductive organs and liver, kidney and spleen in mice. In magnetic field exposure for 15 days, sperm concentrations and viability were significantly lower in magnetic field(15.7$\times$10$^{6}$ $m\ell$, 29.3%) than that in control group(25.1$\times$10$^{6}$ $m\ell$, 34.4%)(P<0.05). The proportion of sperm abnormality were significantly increased in magnetic field exposure groups for 15 days than that in control group(P<0.05). The exposure of magnetic field in mice didn't affect the body and reproductive organ weight such as testis, epididymis, vasicular gland and coagulatin gland. The weight in liver and kidney were not affect in magnetic field exposure groups. However, the spleen weight were significantly higher(P<0.05) in group exposed with than without magnetic field. This studies indicate the short or long term magnetic field exposure in mice were noxious effects on the sperm characteristics and spleen weight, but didn't affect body, reproductive organs, and liver and kidney weight.

Effects of vitamin E and selenium on the ovariectomized rat (Vitamin E와 selenium이 난소가 제거된 흰쥐에 미치는 영향)

  • The present study was devised to determine the effects of body weight, organ weight, hematological values and biochemical parameters by vitamin E and selenium (Selevit) in the ovariectomized (OVX) rats. The animals were divided into 4 groups. Intact group (n=10) received no treatment and operation. Sham group (n=10) received only sham operation and no treatment. OVX group received operation and no treatment. OVX+Selevit received operation and Selevit. The body weights of the all group increased, and that of OVX+Selevit group was the lowest rates. There were significant differences (P<0.01, P<0.001) of body weights between OVX+Selevit group and all other groups. Also, organ weights such as heart, liver, spleen and kidney were measured. The heart and liver weight were significantly lower (P<0.01, P<0.001) in the OVX+Selevit group than in the Intact and Sham group. Also, the kidney weight was significantly lower (P<0.05, P<0.01) in the OVX+Selevit group than in the all other group. On the other hand, there was no significantly difference in the organ weight of spleen between the OVX+Selevit group and the other groups. The number of WBC was significantly higher (P<0.05) in the OVX+Selevit group than in the all other groups. The hematological values of RBC, MCV, MCH and MCHC were no significant differences in any groups. The biochemical parameters of serum total protein and alkaline phosphatase increased significantly in the OVX+Selevit group as compared to that in the OVX group. But, there were no significant differences in AST and ALT in any groups. We conclude that vitamin E and selenium were significantly decreased the body weights in the ovariectomized rats. Our findings suggest that vitamin E and selenium may influence the process of lipid packaging and absorption in the ovariectomized rats.

Anti-Stress Effects of Ginseng in Immobilization-Stressed Rats

  • Stress is a global menace exacerbated by the advancement of industrialization. Failure of stress management is to a breakdown of the psychological and physiological protection mechanisms against stress. The aim of present study was to investigate the anti-stress potential of ginseng against immobilization stress. Male Sprague-Dawley rats (n=24) were divided into three groups; (i) control, (ii) immobilization stress (2hr daily, for 2 weeks), and (iii) immobilization stress (2 hr daily, for 2 weeks) plus oral administration of ginseng (200 mg/kg BW Id). Immobilization stress resulted in a significant inhibition of body weight gain by 45 % and a significant decrease in the tissue weights of thymus and spleen (p < 0.05). The concentrations of blood GOT and GPT were significantly increased in the immobilization-stressed group compared to the control group (p < 0.05). There were no differences in the blood cholesterol levels among groups. Ginseng administration in the immobilization-stressed group tended to reverse the lack of body weight gain and food intake, though not significantly. The ginseng-administered group showed a significant reversal in the stress-induced effect on spleen and thymus weight, increasing the tissue weights by 16% and 20%, respectively, compared to immobilization-stressed group (p<0.05). The plasma corticosterone level was significantly increased in the stressed group by 39 % compared to the control group (p<0.05), but ginseng administration significantly reversed the stress-induced increase in plasma corticosterone by 15 % compared to the immobilization-stressed group. The present study suggests that the anti-stress effect of ginseng is mediated by normalization of stress-induced changes in the circulating hormones and a reversal of tissue weight loss, thereby returning the body to normal homeostasis.

Effects of Samjawhadam-jeon on Immune Cells in Ovalbumin-induced Asthmatic Mice (삼자화담전(三子化痰煎)이 천식모델 생쥐의 면역세포에 미치는 영향)

  • Objective: The purpose of this study was to evaluate the effect of Samjawhadam-jeon (SJHDJ; 三子化痰煎) on immune cells in OVA-induced asthmatic mice. Material and Methods : C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (four times a week) for asthma induction. Two experimental groups were treated with different concentrations of SJHDJ group (400 mg/kg) extracts and cyclosporin A (10 mg/kg) for the latter 8 weeks. At the end of the experiment, the mouse lung. peripheral lymph node (PLN) and spleen were removed and immune cells were analyzed by flow cytometer. Results : Lung weight, total cells in lung, PLN, and spleen of the SJHDJ group decreased significantly compared with that of the control group. Number of $CD3e^+/CD69^+$, $CD3e^+/DX5^+$ cells in lung, PLN and spleen, number of $CD3^+$ cells in PLN and spleen, number of $CD3e^-/CCR3^+$ cells in lung and PLN, and number of $B220^+/IgE^+$ cells in PLN of the SJHDJ group decreased compared with that of the control group. Number of $CD4^+/CD25^+$ cells in PLN and spleen of the SJHDJ group increased compared with that of the control group. Conclusion : These results demonstrate that SJHDJ will be a desirable alternative therapy for allergic asthma by inhibiting the expression of immune cells.

Effect of Green Tea Probiotics on the Growth Performance, Meat Quality and Immune Response in Finishing Pigs

  • The objective of this study was to determine the effects of green tea probiotics on growth performance, meat quality and immune response in finishing pigs, and to assess the possibility of substituting green tea probiotics for antibiotics in diets of finishing pigs. This green tea probiotics is made by mixing green tea powder and excipients (defatted rice bran and wheat bran) and fermenting the mixture with beneficial bacteria. A total of 90 crossbreed "Landrace$\times$Yorkshire" finishing pigs with an average body weight of $72.5{\pm}2.5kg$ were assigned to 5 dietary treatments in a completely randomized design. Each treatment had 3 replications with 6 pigs per replication. The five dietary treatments were control, antibiotic (0.003% chlortetracycline added) and 0.1, 0.5 and 1.0% of green tea probiotics. There were no significant differences in final body weight, daily weight gain, daily feed intake and feed conversion ratio in the green tea probiotics and antibiotic treatments (p>0.05). Crude protein content was significantly increased in the 0.1 and 1.0% green tea probiotics treatment groups (p<0.05) and there was no significant difference in crude fat content of the meat among the treatments. The TBA value of meat was significantly lowered with 0.5 and 1.0% green tea probiotics treatments compared to that of controls and statistically similar to the antibiotic treatment after 3 weeks of storage (p<0.05). The growth of spleen cells stimulated with Con A (0.1 and $1.0{\mu}g/ml$) was significantly increased with 1.0% green tea probiotics treatment compared to that of the control treatment (p<0.05). The growth of spleen cells stimulated with LPS (1.0, 3.0 and $10{\mu}g/ml$) was significantly increased in the 0.5% green tea probiotics group compared to the antibiotic group (p<0.05). In Con A ($1.0{\mu}g/ml$) medium, IL-6 production of spleen cells was significantly increased with 1.0% green tea probiotics treatment compared to that of the control (p<0.05). In LPS ($10.0{\mu}g/ml$) medium, TNF-${\alpha}$ production of spleen cells increased significantly in all green tea probiotics treatment groups compared to that of the control (p<0.05). Finally it can be summarized that addition of green tea probiotic has a positive effect similar to antibiotic and 0.5% is the suitable dietary supplementation dose for finishing pig production.
