The objective of this study was to examine effect of ethylene glycol for semen cryopreservation in Korean Jeju Black Bull. The semen was cryopreserved with extenders containing cryoprotectants (7% glycerol and 3%, 5%, 7% ethylene glycol) and packed to 0.5 ml straws. The semen straws were located above 3 cm of liquid nitrogen for 5 min, 5 cm for 10 min and 8 cm for 10 min. And then frozen straw was plunged into $LN_2$. Post-thawed sperm motility, viability and membrane integrity were significantly higher in 5% ethylene glycol ($72.5{\pm}5.00%$, $54.88{\pm}0.66%$ and $46.00{\pm}2.40%$; p<0.05). Motility and viability were similar between 7% glycerol and 5% ethylene glycol. However, the membrane integrity was significantly higher in 5% ethylene glycol ($34.69{\pm}4.64%$ vs $46.00{\pm}2.40%$; p<0.05). The viability and membrane integrity were significantly higher in 5 cm for 10 min and 8 cm for 10 min than 3 cm for 5 min (viability: $55.81{\pm}2.94$, $55.19{\pm}3.34$ vs $47.94{\pm}3.48%$; p<0.05 and membrane integrity: $44.94{\pm}3.51$, $46.06{\pm}2.25$ vs $40.38{\pm}1.03%$; p<0.05). The percentage of capacitated sperm assessed by CTC staining, percentage of F pattern was higher in 7% glycerol, 5% and 7% ethylene glycerol, and AR pattern was significantly higher in 3% ethylene glycol. F pattern was significantly increased in 5 cm for 10 min and 8 cm for 10 min (p<0.05), but AR pattern was significantly increased in 3 cm for 5 min (p<0.05).
The ultrastructure of spermatozoa in urodeles and primitive anurans was examined and compared. The spermatozoa of urodeles are characterized by seven plesiomorphies in subacrosomal cone, endounclear canal. perforatorium, ring, marginal filament, undulating membrane and tail axis. Most primitive anuran spermatozoa have no marginal filament, subacrosomal cone and ring structure with the exception of having the subacrosomal cone in Ascaphus and the ring in Discohlossus as compared with those of urodeles. Persistence of the subacrosomal cone and the ring structure is typical in most urodeles and is further linked with the primitive anurans. Therefore, these characters are regarded as symplesiomorphies in urodeles and primitive anurans. The organization of sperm tail, endounclear canal and perforatorium indicates a close phylogenetic relationship between urodeles and the primitives anurans.
This study was undertaken to observe the effects of $^{60}Co\;{\gamma}-irradiation$ on the cell of spermatogenic epithelium in the testis of the chicken. 16-week-old chicken were provided as an experimental group and compared with control group. The experimental group was divided into a single irradiation (800, 1000, 1200 rads) and into three partial irradiation group (800/3, 1000/3, 1200/3 rads). The morphological changes of epithelial cell of the testis were observed by means of hematoxyline and eosin stain. Microstructure of spermatocyte and sperm was observed by means of semithin section of electron microscopic specimen. The results obstained are summerized as follows. 1. Spermatogonia and sertoli cells were found to be isolated from the basal membrane of seminiferous tubules as dose of $^{60}Co\;{\gamma}-irradiation$ was increased. 2. Spermatocytes of pachytene stage were seperated from the cytotplasmic process of sertoil cell in case of 1000 rads of $^{60}Co\;{\gamma}-irradiation$. 3. Normal arrangement of the cell of spermatogenic epithelium was found in control group and only the partial irradiation group of 800 rads. Vaculation in the seminiferous was pronounced in case of a single irradiation group of 800 rads, but the irradiation group of 1000 rads and 1200 rads were found to be damaged severely in both a single and a partial dose.
Phenolic compounds play a major role in the interaction of plants with their environment. They are thought to have been a feature of higher plants since early colonization of the land. Phenolics are crucial for many important aspects of plant life. They can play structural roles in different supporting or protective tissues, for example in cell walls, they can be involved in defence strategies, and signalling properties particularly in the interactions between plants and their environment. In brown algae, phenolic compounds are contained within membrane bound vesicles known as physodes, and their roles in algae are thought to be similar to those of higher plant phenolics. They can be stained using various histochemical stains, however, none of these stains are phenolic specific so care must be taken during interpretation of such results. Many, but not all phenolics are also autofluorescent under UV or violet light. Physodes are involved in cell wall construction, both in primary and secondary walls in brown algae. They bind together with other wall components to make a tough wall. They have also been found to play a role at fertilization, in blocking polyspermy in some species. Sperm are very quickly rendered immobile after phenolic release from newly fertilized zygotes seconds after fertilization. Phenolic compounds are thought to be important herbivore deterrents in some species due to their astringent nature. Phenolic compounds also offer effective UV protection in the early life stages and also the adults of many algal species. In the future, this factor may also make them an important player in the pharmaceutical and skincare industries.
Ultrastructure of germ cell during gametogenesis of surf clam, Spisula sachalinensis were investigated by electron microscopic observations. Oogonia are oval in shape and 5-6 ${\mu}m$ in diameter. They contain a large nucleus and have several mitochondria in the cytoplasm. Vitellogenic oocytes grew attached by a stalk to the germinal epithelium in the ovarian sac and their cytoplasm contained many yolk granules, lipid granules and mitochondria. Mature oocytes measuring 50-60 ${\mu}m$ in diameter have a nucleus containing an electron dense nucleolus, and a lots of yolk granules, lipid granules, mitochondria, rough endoplasmic reticulum and cortical granules were present in the cytoplasm. The surface of the plasma membrane in mature oocytes was formed of microvilli approximately 1.5 ${\mu}m$ long and enveloped in the vitelline layer. Spematogonia are oval in shape and about 5 ${\mu}m$ in diameter. Sprmatogonia develop into spermatocyte, spermatid and spermatozoon. The spermatozoon consisted of the head, midpiece and tail. The sperm head with diameter of about 1.5 ${\mu}m$ was ovoid, and contained the nucleus which consisted of acrosome. The four mitochondria encircled the centrosome in midpiece. The flagellum measuring about 40 ${\mu}m$ long had the classical 9+2 axoneme structure.
To evaluate the effect of spermatozoa culture on glycosidase activity of frozen-thawed spermatozoa in human, the spermatozoa were treated experimentally and assayed for activities of ${\alpha}$-L-fucosidase, ${\alpha}$-D-mannosidase, ${\beta}$-D-galactosidase and N-acetyl-${\beta}$-D-glucosaminidase (${\beta}$-GlcNAc'ase). The ${\beta}$-GlcNAc'ase activity was at least two-folds higher than other glycosidases regardless of spermatozoa incubation (p<0.05). The spermatozoa motility was decreased with incubation periods, but no effects by different glycosidases on the changes of spermatozoa motility during the various periods of incubation. In all glycosidases, the spermatozoa-zona binding rates in spermatozoa without incubation were higher than in spermatozoa incubated for 2 h (p<0.05). ${\beta}$-GlcNAc'ase is present mainly in the plasma membrane of spermatozoa frozen-thawed in human. It was also shown that the glycosidase activity was increased in all glycosidases in spite of lower sperm-zona binding by spermatozoa incubation.
Fine structures of the testis and vas deferens in the fall-web worm, Hyphantria cunea Drury, are studied with electron microscope. Adult worms have single testis close to the midlines of the abdomen. Testis is composed of 4 follicles which are incompletly separated from each other and bounded together by a peritoneal sheath. The peritoneal sheath consisted of outer cuticular layer and two kinds of inner layers, in which glycogen particles are dispersed commonly. These two layers are divided by the morphology of cytoplasmic granules. Follicular epithelium forming the wall of the follicles have melanin pigment granules, and trachea or tracheoles are extended through this epithelium. In the cysts of adult testis, matured spermatozoa are grouped together in bundles and after releasing the sperm bundles to the vas deferens, lamellar shaped lysosomes appeared in the cytoplasms of the cyst cells. The number of spermatozoa per cyst is exactly 256 ($2^8$), this number is characteristics of the Lepidoperan species. Vas deferens is a tube with a fairly thick bounding epithelium, a basement membrane and a layer of circular muscle outside it. At the apical portion of the epithelial cells, microvilli are well developed. And in the cytoplasms of these cells, numerous excretory granules are observed.

Objectives: Epichlorohydrin is a material that has been predicted to have high volatility and strong toxicity and is used normally in working area. Therefore, the hazardous and dangerous level and the foreign management system about epichlorohydrin should be invested at home and abroad and through hazard assessment for occupational environment measurement and exposure status of industrial workers in domestic working area. Methods: To assess risk and to suggest Development and Adoption to prevent health damage of workers owing to the epichlorohydrin exposure, the hazardousness and dangerousness of epichlorohydrin and its practical examples and regulation level for domestic and abroad health impairment are researched on the base of various references. Results: The epichlorohydrin caused skin and mucus membrane irritation, respiratory paralysis, kidney and live damage under the influence of acute toxicity and in animal study, it was confirmed as a doubtful carcinogenic substance to trigger reducement of sperm number and reproduction ability, abnormal spermatogenesis, mutagen, increase of forestomach epithelium and occurrence of papilloma and so on, as well as it induced stimulus asthma and allergic contact dermatitis for exposure workers. Conclusions: Epichlorohydrin was found to occur allergic contact dermatitis, carcinogenesis doubt and reproduction toxicity and was verified as a material which would be established reinforcement of management level to care health of handlers, such as denotement dangerousness of skin absorption.
Ultrastructure and function of testis somatic cells in freshwater prawns Macrobrachium nipponense were studied. The paired testes of the prawn were elongated, united at their anterior end, which lay between the dorsal surface of the hepatopancreas and the heart. Each testis consisted of a large number of seminiferous cords compactly held together by connective tissue. A seminiferous cord was composed of an outer layer of simple squamous epithelium, a basement membrane, the closely packed germ cells and sustentacular cells of the germinal ridge, and an inner layer of simple cuboidal epithelial cells. Leydig cell-like cells in an angular areas filling the space of the seminiferous cords were observed. The nuclei of leydig cell-like cells were characterized by a distinct nucleolus. The simple squamous epithelial layer was composed of flattened cells tying on a basement membrane. The nuclei of the flattened cells were often overlapped in a layer, and the cytoplasm of the cells was observed just near the nuclei. The sustentacular cells were complex in morphology. These cells had relatively small cell bodies from which long cytoplasmic extensions ramified reached the space of germ cells in the germinal ridge. The nuclei of sustentacular cells usually exhibited angular profiles and located most commonly at the periphery of the cords. Cells of simple cuboidal epithelium located between germinal ridge and lumen of seminiferous cord, and part of the cells were adjacent to basal lamina, The cuboidal epithelial cells contained numerous mitochondria, the well-developed rER, the well-developed Golgi complex, and irregularly shaped nuclei. Transition vesicles appeared on the cis side of the Golgi complex. The large vesicles on the trans side of the complex appeared to fuse to form a membrane-bound structure. A number of pits on the cell apex suggested exocytotic activity for secretion of the sperm supporting matrix.
Cryopreservation of boar semen is continually researched in reproductive technologies and genetic resource banking in breed conservation. For evaluating the boar semen quality, sperm motility (MOT) is an important parameter because the movement of spermatozoa indicates active metabolism, membrane integrity and fertilizing capacity. Various researches have been trying to improve the quality of semen post-thawed in boar. Recently, polymorphism (g.358A>T) of cluster-of-differentiation antigen 9 (CD9) gene reported to be significant association with MOT. Also, CD9 gene was expressed in the male germ line stem cells is crucial for sperm-egg fusion, and was therefore selected as candidate gene for boar semen. This study was conducted to evaluate the pig SNP (g.358A>T) of CD9 gene as a positional controlling for semen parameters of post-thawed boar semen. To results, the g.358A>T SNP of the CD9 gene was significantly associated with the traits such as MOT, curve linear velocity, straight line velocity, average path velocity and amplitude of lateral head displacement. Particularly, the g.358A>T SNP significantly has the highest association with MOT and animals with AA genotype (p<0.001). Therefore, we suggest that the g.358A>T in the intron 6 region of the porcine CD9 may be used as a molecular marker for Duroc boar Post-thawed semen quality, although its functional effect was not defined yet.
