Detection and Molecular Characterization of Cryptosporidium spp. from Wild Rodents and Insectivores in South Korea

  • In order to examine the prevalence of Cryptosporidium infection in wild rodents and insectivores of South Korea and to assess their potential role as a source of human cryptosporidiosis, a total of 199 wild rodents and insectivore specimens were collected from 10 regions of South Korea and screened for Cryptosporidium infection over a period of 2 years (2012-2013). A nested-PCR amplification of Cryptosporidium oocyst wall protein (COWP) gene fragment revealed an overall prevalence of 34.2% (68/199). The sequence analysis of 18S rRNA gene locus of Cryptosporidium was performed from the fecal and cecum samples that tested positive by COWP amplification PCR. As a result, we identified 4 species/genotypes; chipmunk genotype I, cervine genotype I, C. muris, and a new genotype which is closely related to the bear genotype. The new genotype isolated from 12 Apodemus agrarius and 2 Apodemus chejuensis was not previously identified as known species or genotype, and therefore, it is supposed to be a novel genotype. In addition, the host spectrum of Cryptosporidium was extended to A. agrarius and Crosidura lasiura, which had not been reported before. In this study, we found that the Korean wild rodents and insectivores were infected with various Cryptosporidium spp. with large intra-genotypic variationa, indicating that they may function as potential reservoirs transmitting zoonotic Cryptosporidium to livestock and humans.

  • Some modern accerelators provide a dual energy for photon beam treatment. The main advantages of dual energy in the treatment of rectosigmoid or rectal cancer are as fellows. 1. Dose in the critical organ such as small intestine, bladder and genital organ are reduced. 2. Presacral and perineal area is fully covered. Dose distribution analysis such as calculation of dose in a target volume, isocenter, $D_{nax}$ and dose spectrum in any region of interest are possible. Examples of plan are given and results are discussed.

Pretense in Wax Gourd (동아의 단백질가수분해효소)

  • Protease activity in matured wax gourd sarcocarp was 0.19unit/0.5ml, immatured wax gourd sarcocarp 0.56unit, and matured wax gourd 24.35 unit, immatured wax gourd core 0.35unit. Protease activity in matured wax gourd sarcocarp to raw meat or raw pork was 13,0 unit, 7.4 unit, respectively, and that in wax gourd core to raw beef was 30.2 unit, and raw pork was 24.5 unit. Thermal stability of pretense in matured wax gourd sarcocarp was stable below 70$\^{C}$ when it was heated for 10 minutes. In case of 80$\^{C}$, the remaining activity was 21%, and at 90$\^{C}$, it was lost entirely. The absorption spectrum showed peak at 280nm. According to the HPLC analysis, casein was hydrolyzed into small size by protease in core or sarcocarp of matured was gourd and immatured wax gourd. Wax gourd diluted by 1/10 showed two peaks, one was from casein being hydrolyzed, and the other was from the increased molecular weight with coagulated casein. On the other hand, the molecular weight didin't increase in immatured wax gourd core diluted by 1/10. The result of dilution of 1/10 showed different pattern from undiluted one, but the peak of sarcocarp in matured wax gourd was 1 and the peak of core in immatured wax gourd was 5, and those of core and sarcocarp of immatured wax gourd were 3 respectively.

A Comparative Analysis of Bibliotherapy Books in the UK, US, and Korea: A Suggestion for the Humanistic Approach to Bibliotherapy (영어권 및 국내 독서치료용 도서 비교 분석 - 인문학적 독서치료의 필요성에 관한 제안 -)

  • This study analyzes the spectrum of recommended books for adults included in the bibliotherapy book lists commonly used in libraries and organizations in the UK, US, and Korea. While the recommended book lists in the UK and US retain a diverse range of books relevant to the practitioners' needs, the Korean bibliotherapy book lists are mostly made up of non-fiction self-help books regardless of where the bibliotherapy service is provided. In the subcategory of the bibliotherapy book lists in the UK and US, aside from fictions, books on "health, fitness, dieting" including medical books rank higher than any other kind, which shows that bibliotherapy claims to have therapeutic effects in those organizations. In Korean bibliotherapy book lists, on the other hand, so-called "psychology books impregnated with self-help ideology" take up more than 60%. With the prospect of bibliotherapy as an interdisciplinary practice in mind, I suggest that bibliotherapy should draw upon the insights from humanities in order to help participants have time for deep introspection and attain inner maturity rather than just try to restore their normal conditions before the crisis.

Synthesis of long afterglow phosphor SrAl2O4 : Eu+2,Dy+3 by skull melting method (스컬용융법에 의한 SrAl2O4 : Eu+2,Dy+3 축광성 형광체 합성)

  • $SrAl_2O_4$ : $Eu^{2+}$,$Dy^{3+}$ phosphorescent phosphors were synthesized by skull melting method. The molar ratio of oxides in the phosphors synthesized by the skull melting technique was $SrCO_3$ : $Al(OH)_3$ : $Eu_2O_3$ : $Dy_2O_3$= 1 : 2 : 0.015 : 0.02. Crystal structure and surface morphology were investigated by scanning electron microscopy (SEM) and X-ray diffraction (XRD) analysis. Optical properties of the synthesized $SrAl_2O_4$ : $Eu^{2+}$,$Dy^{3+}$ were measured by photoluminescence (PL) spectrometer for in-depth study on the excitation, emission and afterglow properties. From the PL measurements, it was found that excitation occurred in the wavelength range from 300 to 420 nm with peak position at 360 nm. The emission spectrum showed a broad curve in the wavelength from 450 to 600 nm with peak position at 530 nm. $SrAl_2O_4$ : $Eu^{2+}$,$Dy^{3+}$ phosphors exhibited afterglow properties with emission that lasted for a long period.

Accuracy Verification of Theoretical Models for Estimating Microwave Reflection from Rough Sea Surfaces (거친 바다표면의 마이크로파 반사 계산을 위한 이론적 모델 정확도 검증)

  • This paper presents the verification of accuracies of theoretical models for calculating the microwave reflections from rough sea surfaces. First of all, the Pierson-Moskowitz ocean spectrum was used to generate the rough sea surfaces. Then the relationship between the significant wave heights, root-mean-square(RMS) heights and wind speed was derived by estimating the significant wave heights and RMS heights of the generated sea surfaces according to various wind speeds, and compared the derived relationship with other measurement data sets. The reflection coefficients of the sea surfaces were calculated by using a numerical method(the moment method). Then, the numerical results were compared with Ament model, PO(Physical Optics) model, GO(Geometrical Optics) model and B-M(Brown-Miller) model for various roughness conditions(wind speed) and incidence angles. It was found that the Ament model is not accurate except for a very low roughness conditions($kh_{rms}$<0.4, k is wavenumber and $h_{rms}$ is RMS height). It was also found that at incidence angles lower than $70^{\circ}$, the PO and the GO models agree well with the numerical results, while the B-M model agrees well with the numerical analysis results at incidence angles higher than $80^{\circ}$ for very rough sea surfaces with $kh_{rms}$>10.

Characterization of Radiation Field in the Steam Generator Water Chambers and Effective Doses to the Workers (증기발생기 수실의 방사선장 특성 및 작업자 유효선량의 평가)

  • Characteristics of radiation field in the steam generator(S/G) water chamber of a PWR were investigated and the anticipated effective dose rates to the worker in the S/G chamber were evaluated by Monte Carlo simulation. The results of crud analysis in the S/G of the Kori nuclear power plant unit 1 were adopted for the source term. The MCNP4A code was used with the MIRD type anthropomorphic sex-specific mathematical phantoms for the calculation of effective doses. The radiation field intensity is dominated by downward rays, from the U-tube region, having approximate cosine distribution with respect to the polar angle. The effective dose rates to adults of nominal body size and of small body size(The phantom for a 15 year-old person was applied for this purpose) appeared to be 36.22 and 37.06 $mSvh^{-1}$) respectively, which implies that the body size effect is negligible. Meanwhile, the equivalent dose rates at three representative positions corresponding to head, chest and lower abdomen of the phantom, calculated using the estimated exposure rates, the energy spectrum and the conversion coefficients given in ICRU47, were 118, 71 and 57 $mSvh^{-1}$, respectively. This implies that the deep dose equivalent or the effective dose obtained from the personal dosimeter reading would be the over-estimate the effective dose by about two times. This justifies, with possible under- or over- response of the dosimeters to radiation of slant incidence, necessity of very careful planning and interpretation for the dosimetry of workers exposed to a non-regular radiation field of high intensity.

  • Estimated dose rates in spent fuel pool storage with the extended fuel cycle core management were reviewed and compared with design limit after calculation with the aid of DLC-23/CASK(22 n, 18 g) nuclear data and ANISN code. Radioactivity and gamma spectrum within spent fuel assemblies were calculated with ORIGEN code by extended fuel cycle model. In the calculation of dose rate, the fuel pool geometry was assumed to be infinite slab. Also, composition materials and radiation source within assemblies which are being stored in pool storage were assumed to be uniformly distributed throughout all the assemblies. As a result of culculation of dose rate from stored assemblies and waterborne radionuclides in pool water, the calculated dose rates appear to be lower than design basis limit under normal condition as well as abnormal condition.

Reinterpretation of the protein identification process for proteomics data

  • Introduction: In the mass spectrometry-based proteomics, biological samples are analyzed to identify proteins by mass spectrometer and database search. Database search is the process to select the best matches to the experimental mass spectra among the amino acid sequence database and we identify the protein as the matched sequence. The match score is defined to find the matches from the database and declare the highest scored hit as the most probable protein. According to the score definition, search result varies. In this study, the difference among search results of different search engines or different databases was investigated, in order to suggest a better way to identify more proteins with higher reliability. Materials and Methods: The protein extract of human mesenchymal stem cell was separated by several bands by one-dimensional electrophorysis. One-dimensional gel was excised one by one, digested by trypsin and analyzed by a mass spectrometer, FT LTQ. The tandem mass (MS/MS) spectra of peptide ions were applied to the database search of X!Tandem, Mascot and Sequest search engines with IPI human database and SwissProt database. The search result was filtered by several threshold probability values of the Trans-Proteomic Pipeline (TPP) of the Institute for Systems Biology. The analysis of the output which was generated from TPP was performed. Results and Discussion: For each MS/MS spectrum, the peptide sequences which were identified from different conditions such as search engines, threshold probability, and sequence database were compared. The main difference of peptide identification at high threshold probability was caused by not the difference of sequence database but the difference of the score. As the threshold probability decreases, the missed peptides appeared. Conversely, in the extremely high threshold level, we missed many true assignments. Conclusion and Prospects: The different identification result of the search engines was mainly caused by the different scoring algorithms. Usually in proteomics high-scored peptides are selected and low-scored peptides are discarded. Many of them are true negatives. By integrating the search results from different parameter and different search engines, the protein identification process can be improved.

Spectral Characteristics of Heavy Metal Contaminated Soils in the Vicinity of Boksu Mine (복수광산 주변 중금속 오염 토양의 분광학적 특성)

  • This study investigated spectral characteristics of heavy metal contaminated soil samples in the vicinity of abandoned Boksu mine. Heavy metal concentrations including arsenic, lead, zinc, copper and cadmium were analyzed by XRF analysis. As a result, all of the soil samples excluding control sample were over-contaminated based on the counter measure standard. The XRD results revealed that quartz, kaolinite and smectite were detected for all of the soil samples and heavy metals in soil were adsorbed on clay minerals such as kaolinite and smectite. The spectral analyses confirmed that spectral reflectance of near-infrared and shorter portion of shortwave-infrared spectrum decreases as heavy metal concentration increases. Moreover, absorption depths at 2312 nm and 2380 nm, the absorption features of clay minerals, decreases with higher heavy metal concentration indicating adsorption of heavy metal ions with clay minerals. It indicates that spectral features and heavy metal contamination of soil samples have high correlations.
