Conformational Dynamics of Heme Pocket in Myoglobin and Hemoglobin

  • The conformational dynamics of heme pocket, a small vacant site near the binding site of heme proteins -myoglobin (Mb) and hemoglobin (Hb), was investigated after photolysis of carbon monoxide from MbCO and HbCO in D$_2$O solution at 283 K by probing time-resolved vibrational spectra of photolyzed CO. Two absorption bands, arising from CO in the heme pocket, evolve nonexponentially in time. The band at higher energy side blue shifts and broadens with time and the one at lower energy side narrows significantly with a negligible shift. These spectral evolutions are induced by protein conformational changes following photolysis that modify structure and electric field of heme pocket, and ligand dynamics in it. The conformational changes affecting the spectrum of photolyzed CO in heme pocket likely modulates ligand-binding activity.

포만트 정보의 동적 변화특성 조사에 관한 연구 (Investigation on Dynamic Behavior of Formant Information)

  • This study reports on the effective way of displaying dynamic formant information on F1-F2 space. Conventional ways of F1-F2 space (different name of vowel triangle or vowel rectangle) have been used for investigating vowel characteristics of a speaker or a language based on statistics of the F1 and F2 values, which were computed by spectral envelope search method. Those methods were dealing mainly with the static information of the formants, not the changes of the formant values (i.e. dynamic information). So a better way of investigating dynamic informations from the formant values of speech signal is suggested so that more convenient and detailed investigation of the dynamic changes can be achieved on F1-F2 space. Suggested method used visualization of static and dynamic information in overlapped way to be able to observe the change of the formant information easily. Finally some examples of the implemented display on some cases of the continuous vowels are shown to prove the usefulness of suggested method.

  • The purpose of the present study is to analyse characteristics of the background noise for the broadband seismic stations in KMA. It is well known that the background noise arises continuously from long period microseism, sea waves, minute changes of atmospheric pressure, seasonal temperature change of the ground surface, culture activities, and etc. The background noise shows spatial and temporal changes and it has various characteristics such as its spectral amplitudes in frequency domain are not constant Such the background noise gives considerable influences on the quality of seismic record. To investigate annual variations, the background noise was separated into high frequency components of above 1Hz More larger average amplitude is found in winter than other seasons. The average amplitude for 12 seismic stations are compared. It is known that the background noise is considerably larger in stations located in island region such as Jeju, Ulleungdo, and Bagryeongdo seismic stations. However the noise is relatively small in inland stations such as Chuncheon, Chungju and Uljin seismic stations.

Simulations of Two-Dimensional Electronic Correlation Spectra

  • Two-dimensional (2D) correlation method, which generates the synchronous and the asynchronous 2D spectrum by complex cross correlation of the Fourier transformed spectra, is an analysis method for the changes of the sample spectrum induced by vari ous perturbations. In the present work, the 2D electronic correlation spectra have been simulated for the cases where the sample spectrum composed of two gaussian bands changes linearly. When only the band amplitudes of the sample spectrum change, the synchronous spectrum shows strong peaks at the band centers of the sample spectrum, but the asynchronous spectrum does not make peaks. When the sample spectrum shifts without changing intensity and width, the synchronous spectrum shows peaks around the initial and final positions of the band maximum and the asynchronous spectrum shows long peaks spanning the shifting range. The band width change produces the complex 2D correlation spectra. When the sample spectrum shifts with band broadening, the width change by 50% of full width at half maximum (FWHM) does not give so large an effect on the correlation spectrum as the spectral shift by one half of FWHM of the sample spectrum.

Mastitis Detection by Near-infrared Spectra of Cows Milk and SIMCA Classification Method

  • Mastitis is a major problem for the global dairy industry and causes substantial economic losses from decreasing milk production and considerable compositional changes in milk, reducing milk quality. The potential of near infrared (NIR) spectroscopy in the region from 1100 to 2500nm and chemometric method for classification to detect milk from mastitic cows was investigated. A total of 189 milk samples from 7 Holstein cows were collected for 27 days, consecutively, and analyzed for somatic cells (SCC). Three of the cows were healthy, and the rest had mastitis periods during the experiment. NIR transflectance milk spectra were obtained by the InfraAlyzer 500 spectrophotometer in the spectral range from 1100 to 2500nm. All samples were divided into calibration set and test set. Class variable was assigned for each sample as follow: healthy (class 1) and mastitic (class 2), based on milk SCC content. The classification of the samples was performed using soft independent modeling of class analogy (SIMCA) and different spectral data pretreatment. Two concentration of SCC - 200 000 cells/ml and 300 000 cells/ml, respectively, were used as thresholds fer separation of healthy and mastitis cows. The best detection accuracy was found for models, obtained using 200 000 cells/ml as threshold and smoothed absorbance data - 98.41% from samples in the calibration set and 87.30% from the samples in the independent test set were correctly classified. SIMCA results for classes, based on 300 000 cells/ml threshold, showed a little lower accuracy of classification. The analysis of changes in the loading of first PC factor for group of healthy milk and group of mastitic milk showed, that separation between classes was indirect and based on influence of mastitis on the milk components. The accuracy of mastitis detection by SIMCA method, based on NIR spectra of milk would allow health screening of cows and differentiation between healthy and mastitic milk samples. Having SIMCA models, mastitis detection would be possible by using only DIR spectra of milk, without any other analyses.

Somatic cell counts determination in cow milk by near infrared spectroscopy: A new diagnostic tool

  • Milk somatic cell count (SCC) is a recognized indicator of cow health and milk quality. The potential of near infrared (NIR) spectroscopy in the region from 1100 to 2500nm to measure SCC content of cow milk was investigated. A total of 196 milk samples from 7 Holstein cows were collected for 28 days, consecutively, and analyzed for fat, protein, lactose and SCC. Three of the cows were healthy, and the rest had mastitis periods during the experiment. NIR transflectance milk spectra were obtained by the InfraAlyzer 500 spectrophotometer in a wavelength range from 1100 to 2500 nm. The calibration for logSCC was performed using partial least square (PLS) regression and different spectral data pretreatment. The best accuracy of determination was found for equation, obtained using smoothed absorbance data and 10 PLS factors. The standard error of calibration was 0.361, calibration coefficient of multiple correlation 0.868, standard error of prediction for independent validation set of samples 0.382, correlation coefficient 0.854 and the variation coefficient 7.63%. The accuracy of logSCC determination by NIR spectroscopy would allow health screening of cows, and differentiation between healthy and mastitic milk samples. When the spectral information was studied it has been found that SCC determination by NIR milk spectra was indirect and based on the related changes in milk composition. In the case of mastitis, when the disease occurred, the most significant factors that simultaneously influenced milk spectra were alteration of milk proteins and changes in ionic concentration of milk.

원격탐사 자료를 이용한 근소만 갯벌 퇴적환경 및 저서미세조류 환경 분석 (Analysis on the Sedimentary Environment and Microphytobenthos Distribution in the Geunso Bay Tidal Flat Using Remotely Sensed Data)

  • 근소만 갯벌에 대해 위성영상 자료를 적용하여 갯벌 표층의 퇴적상과 저서미세조류군집의 변화를 관측하였다. 현장조사 자료 분석을 통해 갯벌 표층 퇴적상 분포를 확인하고, 저서미세조류의 분포 지역이 위성영상에서 어떠한 반사도 특성으로 나타나는지 관측하였다. 근소만의 표층은 $3.5{\Phi}-6.5{\Phi}$ 까지 크기의 입자가 주로 분포하여 공간적인 퇴적상의 분포가 뚜렷하지 않으므로 중저해상도의 영상에서는 퇴적환경 변화의 양상 분석이 적합하지 않았다. 그러나 10월부터 2월까지 갯벌 표층에 나타난 저서미세조류의 번성 현상을 뚜렷하게 관측할 수 있었으며, 중저해상도의 영상을 활용하여 계절별 저서생태계의 분포 변화 관측이 가능함을 알 수 있었다. 고해상도 영상에서는 모래성분이 우세한 지역이 잘 구분되어 나타났으며, 잘피의 분포지역을 뚜렷이 관측할 수 있었다. 따라서 국내 고해상도 위성영상인 KOMPSAT-2 자료가 갯벌 표층 퇴적상 및 정밀한 생태환경 주제도 작성에 효과적으로 활용될 수 있을 것으로 판단된다.

Landsat TM 자료와 표충퇴적물 분석을 통한 천수만 간석지 퇴적물 분류 (Classification of Tidal Flat Deposits in the Cheonsu-bay using Landsat TM Data and Surface Sediment Analysis)

  • This study aimed at verifying the grain-sized distribution of surface deposits in a tidal flat using multi-spectral Landsat TM. In this study, we employed the grain-sized analysis, PCA and unsupervised classification techniques for analyzing the distribution of deposits. As a result in this study, the unsupervised classification method using PCA image was found to be most useful in classifying tidal flat deposits using satellite data. This method is considerably effective in analyzing not only the aspects of distribution in terms of accumulated deposits and erosion, but also the changes in seaside topography and shoreline. The grain-sized distribution analysis indicates that the mud flat inside the Cheonsu-bay tidal flat is distributed, the mixed flat located in the middle, and the sand flat distributed near the sea. The sand flat is dominant around the southern part of Seomot isle and its beach. On the other hand, the mud and mixed flat is dominant on the western part. Likewise, the western coast of Seomot isle and its beach is significantly affected by waves facing the offshore. However, the eastern side of the bay could be a site for the evolution of tidal flat made of fine materials where it is less affected by ocean waves. These results show that multi-spectral satellite data are effective for the classification of distribution materials and environmental impact assessment and continuous monitoring. In particular, the research on environmental deposits can provide important decision-supporting information for decision-making on seaside development, by analyzing the progress of deposits and environmental changes.

Structural Changes in Isothermal Crystallization Processes of Synthetic Polymers Studied by Time-Resolved Measurements of Synchrotron-Sourced X-Ray Scatterings and Vibrational Spectra

  • The structural changes occurring in the isothermal crystallization processes of polyethylene (PE), poly-oxymethylene (POM), and vinylidene fluoridetrifluoroethylene (VDFTrFE) copolymer have been reviewed on the basis of our recent experimental data collected by the time-resolved measurements of synchrotron-sourced wide-angle (WAXS) and small-angle X-ray scatterings (SAXS) and infrared spectra. The temperature jump from the melt to a crystallization temperature could be measured at a cooling rate of 600-1,000 $^{\circ}C$/min, during which we collected the WAXS, SAXS, and infrared spectral data successfully at time intervals of ca. 10 sec. In the case of PE, the infrared spectral data clarified the generation of chain segments of partially disordered trans conformations immediately after the jump. These segments then became transformed into more-regular all-trans-zigzag forms, followed by the formation of an orthorhombic crystal lattice. At this stage, the generation of a stacked lamella structure having an 800-${\AA}$-long period was detected in the SAXS data. This structure was found to transfer successively to a more densely packed lamella structure having a 400-${\AA}$-long period as a result of the secondary crystallization of the amorphous region in-between the original lamellae. As for POM, the formation process of a stacked lamella structure was essentially the same as that mentioned above for PE, as evidenced from the analysis of SAXS and WAXS data. The observation of morphology-sensitive infrared bands revealed the evolution of fully extended helical chains after the generation of lamella having folded chain structures. We speculate that these extended chains exist as taut tie chains passing continuously through the neighboring lamellae. In the isothermal crystallization of VDFTrFE copolymer from the melt, a paraelectric high-temperature phase was detected at first and then it transferred into the ferroelectric low-temperature phase at a later stage. By analyzing the reflection profile of the WAXS data, the structural ordering in the high-temperature phase and the ferroelectric phase transition to the low-temperature phase of the multi-domain structure were traced successfully.

Spectral and Thermal Studies of Transition Metal PSSA Ionomers

  • Transition metal PSSA ionomers containing Co(II), Ni(II), Cr(III), Ru(III), and Rh(III) are investigated by IR, Far-IR, UV-Vis and DSC. Reliable IR Spectroscopic criteria are established for assessing the degree of ion-exchange of PSSA ionomers and the local structures around metal cations in them. In the hydrated transition metal PSSA ionomers, the ionic groups are solvated by water molecules and there is no significant interactions between sulfonate group and metal cations. The visible spectra indicated that metal cations are present as [M$(H_2O)_6$]$^{n+}$ with Oh symmetry. Their $T_g$ values increase as the extent of ionic site concentration increases, but there is no direct dependence of $T_g$ on the nature of metal cations or their oxidation states. Thus, the water content in PSSA ionomer is found to have dominant influence on $T_g$ of hydrated transition metal PSSA ionomers. Dehydration of the hydrated transition metal PSSA ionomers results in direct interaction between ionic groups and significant color changes of the ionomers due to the changes of the local structures around metal cations. On the base of spectral data, their local structures are discussed. In case of dehydrated 12.8 and 15.8 mol % transition metal PSSA ionomers, no glass transition is observed in 25-$250^{\circ}C$ region and this is believed to arise from the formation of highly crosslinked structures caused by direct coordination of sulfonate groups of metal cations. In the 6.9 mol % transition metal PSSA ionomers, the glass transition is always observed whether they are hydrated or dehydrated and this is though to be caused by the sufficient segmental mobility of the polymer backbone.
