Rat에서 설파메타진의 대사 및 약물동태학 (Metabolic and pharmacokinetic profiles of sulfamethazine in the rat)

  • We used rats as the experimental animal for the elucidation of metabolic patterns and pharmacokinetic profiles of SMZ in the rat, by use of the urine and plasma from predetermined intervals, respectively. Information herefrom would give some insight into species differences and sex differences in the metabolism and pharamcokinetics of drugs, at least SMZ in particular. Results would be summarized as follows: 1. There were two hydorxy metabolites(5-hydroxysulfamethazine and 6-hydroxyethylsulfamethazine) and an acetyl derivative($N_4$-acetyl sulfamethazine) in the 24h-collected urine, on confirmation with each standard materials. There were also two unknown metabolites therein. 2. In the viewpoint of quantitative aspect, $N_4$-acetylsulfamethazine was the largest, hence it is assumed that the acetyl pathway is the major one in the metabolism of SMZ in the rat. 3. As regards sex difference in the rat, the male had more metabolic capacity than the female in metabolism of SMZ. 4. The concenteration-time curves of sulfamethazine(20mg/kg, po) in the plasma compartment were fitted to a one-compartment open model by use of a computer program(NONLIN). 5. There were significant differences(P<0.05) in the pharmacokinetics of sulfamethazine between two sexes in the rat, with higher disposition rate in the male. 6. The emergence of $N_4AcSMZ$ metabolized from SMZ was fast in the plasma of the rat. Half-life of $N_4AcSMZ$ was also. significantly different(P<0.05) between two sexes, suggesting differences in the eliminatory capacity of $N_4AcSMZ$.

16S rDNA-DGGE를 이용한 2종의 제주도 해양 해면의 공생세균의 군집 구조 (Community Structure of Bacteria Associated with Two Marine Sponges from Jeju Island Based on 16S rDNA-DGGE Profiles)

  • 제주도에 서식하는 2종의 해양 해면, Dictyonella sp.와 Spirastrella abata의 공생세균 군집구조를 16S rDNA-DGGE(denaturing gradient gel electrophoresis) 방법에 의해 분석하였다. 해면으로부터 total genomic DNA를 추출하여 GC clamp가 추가된 세균에 특이적인 341f primer와 518r primer를 이용하여 16S rRNA gene의 V3 부위를 증폭한 후 DGGE 전기 영동하고 재증폭하여 염기서열을 분석하였다. 그 결과 Dictyonella sp.에서 8개, Spirastrella abata에서 7개의 band를 확인할 수 있었다. 공통된 주요 band가 없는 패턴을 나타내었으며, DGGE band로부터 DNA를 추출하여 부분 염기서열을 분석한 결과, NCBI에 등록된 서열들과 93%~98%의 유사도를 나타내었다. Dictyonella sp.의 주요 해면 공생세균은 uncultured Gammaproteobacteria, Spirastrella abata의 경우 uncultured Alphaproteobacteria, Firmicutes에 각각 포함되어 해면 종에 따른 숙주 특이적 분포를 보이는 것으로 나타났다.

Identification of Egr1 Direct Target Genes in the Uterus by In Silico Analyses with Expression Profiles from mRNA Microarray Data

  • Early growth response 1 (Egr1) is a zinc-finger transcription factor to direct second-wave gene expression leading to cell growth, differentiation and/or apoptosis. While it is well-known that Egr1 controls transcription of an array of targets in various cell types, downstream target gene(s) whose transcription is regulated by Egr1 in the uterus has not been identified yet. Thus, we have tried to identify a list of potential target genes of Egr1 in the uterus by performing multi-step in silico promoter analyses. Analyses of mRNA microarray data provided a cohort of genes (102 genes) which were differentially expressed (DEGs) in the uterus between Egr1(+/+) and Egr1(-/-) mice. In mice, the frequency of putative EGR1 binding sites (EBS) in the promoter of DEGs is significantly higher than that of randomly selected non-DEGs, although it is not correlated with expression levels of DEGs. Furthermore, EBS are considerably enriched within -500 bp of DEG's promoters. Comparative analyses for EBS of DEGs with the promoters of other species provided power to distinguish DEGs with higher probability as EGR1 direct target genes. Eleven EBS in the promoters of 9 genes among analyzed DEGs are conserved between various species including human. In conclusion, this study provides evidence that analyses of mRNA expression profiles followed by two-step in silico analyses could provide a list of putative Egr1 direct target genes in the uterus where any known direct target genes are yet reported for further functional studies.

Canine Mesenchymal Stem Cells Derived from Bone Marrow: Isolation, Characterization, Multidifferentiation, and Neurotrophic Factor Expression in vitro

  • The purpose of this study is to characterize canine mesenchymal stem cells (MSCs) derived from bone marrow (BM) for use in research on the applications of stem cells in canine models of development, physiology, and disease. BM was harvested antemortem by aspiration from the greater tubercle of the humerus of 30 normal beagle dogs. Canine BM-derived MSCs were isolated according to methods developed for other species and were characterized based on their morphology, growth traits, cell-surface antigen profiles, differentiation repertoire, immunocytochemistry results, and neurotrophic factor expression in vitro. The canine MSCs exhibited a fibroblast-like morphology with a polygonal or spindle-shaped appearance and long processes; further, their cell-surface antigen profiles were similar to those of their counterparts in other species such as rodents and humans. The canine MSCs could differentiate into osteocytes and neurons on incubation with appropriate induction media. RT-PCR analysis revealed that these cells expressed NGF, bFGF, SDF-1, and VEGF. This study demonstrated that isolating canine MSCs from BM, stem-cell technology can be applied to a large variety of organ dysfunctions caused by degenerative diseases and injuries in dogs. Furthermore, our results indicated that canine MSCs constitutively secrete endogenous factors that enhance neurogenesis and angiogenesis. Therefore, these cells are potentially useful for treating dogs affected with various neurodegenerative diseases and spinal-cord injuries.

RAPD 표지자 분석 에 의한 가시아메바속 한국분리주의 유전적 지위 (Genetic status of Acanthamoeba spp. Korean isolates on the basis of RAPD markers)

  • 가시아메바 속(Accnthamoeba spp.)의 DNA 염기 구성 정보와 관계없이 임의의 10개의 엽기로 구성된 프라이머를 사용하여 random amplified polymorphic DNA-polymerase chain reaction(RAPD-PCR)에 의해 게놈 상의 DNA를 무작위로 증폭하여 확인된 표지자로써 한국 분리주 및 외국 분리주와 기존의 알려진 4개 가시아메바 종간의 유전적 근연관계 분석을 통해서 4개 한국 분리주의 분류상의 성상을 규명하였다. 본 연구에서 A. culbertsoni, A. hokchetti, A. triangularis, A. pokphrwc와 한국 분리주인 YM-2 YM-3, YM-4 YM-5 그리고 외국 분리주인 HOV의 게놈 DNA는 18 종류의 프라이머에 의하여 다양한 양상의 증폭산물을 보였으며 그 중 9개 프라이머는 한국 분리쿠간에도 특이성을 보이는 RAPD 표지자를 제공하였다. 총 18개의 프라이머에 대한 증폭 산물을 대상으로 각 시료의 유사도를 조사한 결과, A. culbertsoni는 A. hakhetti, A. triangularis, A. polyphuga와 유사도가 각각 0.300, 0.308, 0.313이었고, A hqkchetti와 A. triangularis간의 유사도는 0.838이었다. 한국 분리주 YM-2, -3, -4 간의 평균 유사도는 0.959이었고, YM-2, -3, -4 와 A. hotchetti, A. triangularis 간의 평균 유사도는 0.832이었다. 한국 분리주 YM-5는 YM-2, -3 -4 간의 비교에서 평균 0.237의 유사도를 보인 반면, A. culbersoni와 유사도 0.857을 보여, 다른 한국 분리주보다 A. culbertsoni와 유전적으로 유사함을 알 수 있었다. UPGMA법에 의한 유전적 근 연관계 분석 결과 phonogram 강에 두개의 분지군이 존재하는데, A. hakchetti, A. triangularis 및 3개 한국 분리주(YM-2, -3 -4)가 하나의 분지군을, A. cuzburtsoni, A. polyphaga HOV주, 및 YM-5가 따른 분지군을 형성하는 것으로 나타났다 게놈 DNA 상의 변이에 근거하여 볼 때, YM-5 주늘 유전적으로 A. culbertsoni와 거의 유사한 분리주이며 한국 분리주는 최소 2종 이상의 가시아메바로 분류할 수 있다고 사료된다.

균체 지방산 분석을 이용한 Bacillus anthracis의 동정 (Analysis of Cellular Fatty Acid Methyl Esters (FAMEs) for the Identification of Bacillus anthracis)

  • Bacillus anthracis, the etiological agent of anthrax has been classified into the Bacillus subgroup I with B. cereus, B. mycoides and B. thuringiensis based on morphological and DNA similarity. DNA studies have further indicated that these species have very AT-rich genomes and high homology, indeed it has been proposed that these four sub-species be recognized as members of the one species. Several methods have been developed to obtain good differentiation between these species. However, none of these methods provides the means for an absolutely correct differntiation. The analysis of fatty acid methyl esters (FAMEs) was employed as a quick, simple and reliable method for the identification of 21 B. anthracis strains and closley related strains. The most significant differences were found between B. anthracis and B. anthracis closely related strains in FAMEs profiles. All tested strains of B. anthracis had a branched fatty acid C17:1 Anteiso A, whereas the fraction of unsaturated fatty acid Iso C17:1 w10c was found in B. anthracis closely related strains. By UPGMA clustering analysis of FAMEs profiles, all of the tested strains were classified into two clusters defined at Euclidian distance value of 24.5. The tested strains of B. anthracis were clustered together including Bacillus sp. Kyungjoo 3. However, the isolates of B. anthracis closely related spp. Rho, S10A, 11R1, CAU9910, CAU9911, CAU9912 and CAU9913 were clustered with the other group. On the basis of these results, isolates of B. anthracis Bongchon, Kyungjoo 1, 2 and Bacillus sp. Kyungjoo 3 were reclassified as a B. anthracis. It is concluded that FAMEs analysis provides a sensitive and reliable method for the identification of B. anthracis from closely related taxa.

Comparison of the growth performance, nutrient digestibility, fecal microflora, blood profiles, and meat quality of broilers, Korean native chickens and white semi broilers under an identical breeding environment

  • The purpose of this study was to compare the growth performance, nutrient digestibility, fecal microflora, blood profiles and meat quality of broilers, Korean native chickens and white semi broilers under identical feeding conditions. Sixty 1-d chicks of each type were randomly placed into 12 pens per cage (5 chicks per cage) and fed commercial diets for 28, 49, and 28 days, respectively. The broilers showed a significantly higher (p < 0.05) growth performance from 0 to 4 weeks compared with the other types. Korean native chickens had a significantly higher (p < 0.05) digestibility of dry matter and nitrogen than that of the other species. The blood glucose was significantly higher in the Korean native chickens compared to the other species, and the blood cholesterol was significantly lower. The Lactobacillus content in Korean native chickens was significantly higher (p < 0.05) compared to the other species, and the E. coli content in the broilers was significantly higher (p < 0.05) than in the Korean native chickens and white semi broilers. In terms of meat quality, the breast meat of the broilers had a high water content and pH level. The breast meat of the white semi broilers had a significantly lower (p < 0.05) shear force than that of the other species. In conclusion, there was an interspecific physiological difference due to the age and body weight of the chickens. The broilers had a higher growth performance and meat quality compared to the Korean native chickens and white semi broilers.

부자류 및 Aconitine이 흰쥐 간 내 유전자발현에 미치는 영향 (Effects on Gene Expressions in the Rat Liver of Aconitine and Aconitum Species.)

  • Objectives : Aconitine is one of the toxic components of aconitum species. This study was carried out to evaluate gene expressions of herbal prescriptions containg aconitum species and oriental medicinal plants of aconitum species. Methods : We have measured gene expressions in the liver of aconitine, Aconitum carmichaeli DEBX., Aconitum ciliare DC. Yong Ho Whan using Sprague-Dawley rat. Gene expression in rat liver has been analyzed using codelink 10k microarray. Results : 1. Genes up-regulated over than 4 fold were 118 and down-regulated less than 4 fold were 91 in aconitine 50 ${\mu}g/kg/day$ over control. 2. Genes up-regulated of over than 4 fold were 124 and down-regulated less than 4 fold were 98 in Aconitum ciliare DC. 4g/kg/day and 169 of over than 4 fold, and 110 of less than 4 fold for Aconitum carmichaeli DEBX. 4g/kg/day, respectively. 3. Regulated genes in treatment group of Aconitum carmichaeli DEBX, Aconitum ciliare DC. and aconitine was only 2 different genes, Sulfotransferase family 4A, member 1 and Lin-7 homolog b (C. elegans). Conclusions : Gene expression profiles in liver were different among aconitine, Aconitum carmichaeli DEBX., Aconitum ciliare DC. and herbal prescription YongHo-whan. Furthermore, we can find many new genes related with effects of aconitum species.

Phylogenetics, Safety and In Vitro Functional Properties of Bacillus Species Isolated from Iru, a Nigerian Fermented Condiment

  • Bacillus species were isolated from iru, a traditional fermented condiment in Nigeria. Polyphasic approach was used to evaluate the phylogenetic relationship and strain sub-type of the isolated species. Additionally, the phylogenetic profiles of the species isolated from iru were compared with those of bacilli isolated from different continents. The phylogenetic diversity analysis was performed using the combination of 16S rRNA gene sequencing, ITS-PCR, ITS-PCR-RFLP, and M13 RAPD-PCR. The analysis revealed that Bacillus subtilis U170B and B. subtilis U146A isolated from iru were the closest relatives of strains belonging to the phylogeny of B. subtilis sensu stricto and were related to other bacilli isolated from different continents that had functional benefits. The two isolated species exhibited resistance to acidic pH (pH 2.0). The survival rates of B. subtilis U170B, B. subtilis U146A, and B. clausii UBBC-07 (commercial probiotic strain) cultured at pH 2.0 for 3 h were 33.45, 12.44, and 9.53%, respectively. The strains were highly tolerant to bile salts [0.3% (w/v)]. B. subtilis U170B exhibited the highest cell viability (43.45%) when cultured for 3 h in the presence of bile salts, followed by B. subtilis U146A (25%) and B. clausii UBBC-07 (18.94%). B. subtilis U170B and B. subtilis U146A did not exhibit haemolytic activity and were susceptible to different antibiotics. Additionally, these two strains exhibited weak antagonistic activity against B. cereus. The diverse wild strains of B. subtilis can be used as a safe multifunctional starter culture for the industrial production of condiments with health benefits.

환경오염 방지를 위한 식물의 생태학적 연구(III) -도로변 식생과 중금속 함량 및 오염에 관한 연구 (Ecological Studies of Plants for Control of Environmental Pollution, III -The Studies on the Content and Contamination of Heavy Metals and Vegetation of Roadside-)

  • Some ecological attributes of perennial plants and Pb contamination were analyzed for study plots near an entrance of Nevade Test Site at Mercury Valley, Nye County, Nevada. The surface of the desert pavement soil was composed of stones (1 to 4cm diameter). The underside of each stone was coated with coarse and fine sand (about 90%). The profiles of soil were constituted with the A-horizon and C-horizon only. The soil pH at the plots ranges from 7.6 to 8.5, C/N was 13 and cation exchange capacity showed 15me/100g. Nine species and 42 number of individuals were found in all plots. Franseria dumosa and Larrea divaricata were dominant species. The discrete clumps of vegetation were consisted of 9 species of common perennials and these were covered about 25% on desert pavement, on the other words, bare area without vegetation was about 75%. The size and spacing of the plants was irregular. Community coefficient as comparison between shrub species in these study area and those in near the low elevation desert indicated a low degree of similarity. Density, cover and productivity in the study plots as compared with those in the nearest study areas in Mercury Valley showed a higher value. The soils in the studied area involved high heavy metal contents in the plant tissue was higher than those of its soil. The leavds of Lycium andersonii tended to accumulate more Zn and Mo than those of the other species. Larrea divaricata leaves accumulated very high leaves of Fe and Ephedra nevadensis were generally high in Mn. Lead contamination was apparent in foliage of desert vegetation collected alongside the roadway, reflecting the variation in traffic volume. Lead contents greater than fifteen-fold of normal (low traffic) were found in plant foliage alongside the heavily traveled roadway. Lead content of old foliage by the heavily traveled roadway was as much as 129 ppm but that of new foliage 17 ppm only.
