Assessment of genetic diversity of Prangos fedtschenkoi (Apiaceae) and its conservation status based on ISSR markers

  • Prangos fedtschenkoi (Regel et Schmalh.) Korovin (Apiaceae) is an endemic species for mountainous Middle Asia, which is both a rare and useful plant. Organic extractions from this species are being used in pharmaceutics and cosmetology. In recent years, P. fedtschenkoi distribution area has considerably decreased, presumably, resulting from human activities such as agriculture, construction works, overgrazing and collection from wild for pharmaceutic purposes. Six populations were found in Uzbekistan and their genetic divergence and differentiation were studied with 10 inter-simple sequence repeat (ISSR) markers, selected out of 101. Totally 166 amplified ISSR fragments (loci) were revealed, of which 164 were polymorphic. Relatively moderate level of polymorphism was found at population level with polymorphic bands ranging from 27.71% to 47.59%. Mean P = 39.05%, $N_a=1.40$, $N_e=1.25$, S.I. = 0.21, and $H_e=0.14$ were revealed for all loci across six populations. AMOVA showed higher variation among populations (62%) than within them (38%). The Bayesian model determined 5 clusters, or genetic groups. The posteriori distribution of the Theta II estimator detected full model identifying high inbreeding, intensified by low gene flow (Nm = 0.3954). Mantel test confined population 6 as distinct cluster corresponding to geographic remoteness (R = 0.5137, $p{\leq}0.005$). Results were used as the bases for developing conserve measures to restore populations.

천연 해조장 생태계 서비스 및 편익지표에 관한 탐색적 연구 (An Exploratory Study on the Ecosystem Service and Benefit Indicators of Natural Seaweed Beds)

  • The purpose of this study is to explore the ecosystem service and benefit indicators of natural seaweed beds. Ecosystems of natural seaweed beds provide a wide range of services and benefits to human society including provisioning services, regulating services, supporting services, and cultural services. Indicators for each of the ecosystem services are chosen by marine plants ecologists and as follows. Ecosystem indicators of natural seaweed beds for provisioning services are well-being food(amount of seaweed harvested/amount of fish landed, fish biomass, area of natural seaweed beds, the number of species, contribution to the second production), raw materials(amount of biomass by breed, amount of aquaculture feed), genetic resources(amount of genetic material extracted, amount of genetic material contained by age and habitat), and medicinal resources(amount of medicinal material extracted). Ecosystem indicators of natural seaweed beds for regulating services are air purification(amount of fine dust/NOx or $SO_2$ captured), climate regulation(amount of $CO_2$ sequestered), waste treatment(amount of N, P stored, biochemical degradation capacity COD), and costal erosion prevention(length and change of natural coast line, amount of sediment prevented). Ecosystem indicators of natural seaweed beds for supporting services are lifecycle and maintenance(primary production, contribution to the second production) and gene pool protection(amount of compositional factors in ecosystem, introduced species). Ecosystem indicators of natural seaweed beds for cultural services are recreation and tourism(the number of visits of an area) and information for cognitive development(amount of time spent in education, research and individual learning about ecosystem of natural seaweed beds).

태안해안국립공원 모항 암반조간대 웅덩이의 어류 종조성과 수직분포 (Species Composition and Vertical Distribution Pattern of Fish in Rock Tidal Pools at Mohang, Taeanhaean National Park)

  • 태안반도 모항항의 암반조간대 조수웅덩이에서 2010년 1월부터 12월까지 매월 어류를 채집하여 고도에 따른 종조성분포를 분석하였다. 조사 기간 동안 채집된 어류는 3목 8과 14속 16종이었으며, 이 가운데 망둑어과의 점망둑(Chaenogobius annularis)이 34.6%로 우점하였고, 아우점종으로는 그물베도라치(Chaenogobius annularis)가 22.0%, 조피볼락 (Chaenogobius annularis)이 6.9%, 두줄망둑 (Chaenogobius annularis)이 6.8%순이었다. 그물베도라치는 고도에 관계없이 주거종으로 출현하였으며, 점망둑은 상부조간대에서 가장 우점하였고, 모든 조사 시기에 채집되었다. 주거종의 개체수는 일시주거종과 기회방문종보다 높았으며, 생체량은 주거종이 61.4%로 가장 높았으며, 일시주거종이 14.2%, 기회방문종이 24.4%였다.

추파 청예작물의 저온 조건이 질산태 질소의 흡수와 식물조직내의 질소 화합물의 함량에 미치는 영향 (Effects of Low Temperature on Nitrate Uptake and Accumulation of Nitrogenous Compound in Fall-Sowing Species)

  • The experiments were undertaken to investigate the dynamics of $NO_3\;^-$ uptake at low temperature with forage rape(Bmssica napus), barle(Hordeum vulgare) and winter rye (Scale cereale). A net $NO_3\;^-$uptake and the contents of nitrogenous compound in tissues at low temperature ($5^{\circ}C$) were compared with normal temperature ($25^{\circ}C$).The net uptake of $NO_3\;^-$ by Brass ica nqus was the highest among three species at both temperature applied. During 9 hours after temperature treatment, $NO_3\;^-$ uptake of Brassica napus, Hordeum vulgm and Scale c e d e grown at $5^{\circ}C$ decreased to about 31, 50 and 27% compared with those grown at $25^{\circ}C$. $NO_3\;^-$ contents in roots were much sensitively affected by low temperature than those of leaves. Comparing with the plants grown at $25^{\circ}C$, $NO_3\;^-$ contents in roots of increased to 48.3, 38.7 and 53.5% repectively, for Brassica napus, Hordeum vulgare and Scale cereale during 9 days. A relatively less changes in the content of $NH_4\;^+$-N were observed according to the temperature treatment. The ontents of amino acids-N in leaves at $5^{\circ}C$ slightly decreased while that of roods remarkably increased compared with $25^{\circ}C$. Protein-N was the largest pool of nitrogen and its contents in leaves and roots showed a similar response on the temperature treatment. These results suggested that $NO_3\;^-$uptake was inhibited at low temperature. The dynamics of $NO_3\;^-$ uptake could be closely related to the increased concentration of $NO_3\;^-$ and amino acids in roots.

A Study on the Radiation Effect of the Smoke Movement in Room Fires

  • To investigate smoke movement with radiation in a room fires, a numerical and experi-mental analysis were performed. In this paper, results from a field model based on a self-developed SMEP (Smoke Movement Estimating Program) were compared with Stockier's ex-periment and the experiments on various sized pool fires in a room with door The SMEP using PISO algorithm solves conservation equations for mass, momentum, energy and species, together with those for the modified k- $\varepsilon$ turbulence model with buoyancy term. Also it solves the radiation equation using the S-N discrete ordinates method (DOM). The result of the cal-culated smoke temperature considering radiation effect has shown good agreement compared with the experimental data, although there are large discrepancy in the hot smoke layer be-tween the temperature predicted by the SMEP with only convection effect and obtained by the experimental result. This large discrepancy is caused from the radiation effect of $H_2O$ and $CO_2$ gas under smoke productions. Hence the radiation effect under smoke in fire is the point to be specially considered in order to produce more realistic result.

First Record of Amphisiella milnei (Ciliophora, Stichotrichida) from Korea

  • A marine ciliate Amphisiella milnei (Kahl, 1932) $Horv{\acute{a}}th$, 1950 was discovered from the tidal pool of Baekdo Island, South Korea. The existence of extra cirri between leftmost frontal cirrus and buccal cirrus discriminates this species from its congeners. Its morphological features are described as follows: body size in vivo $110-130{\times}35-45{\mu}m$; elongate rectangular to elliptical in shape; two large and several small ring-shaped structures; yellowish cortical granules arranged irregularly on ventral side but longitudinally along dorsal kineties on dorsal side; 34-40 adoral membranelles, 3 frontal cirri, 1 buccal cirrus, 1 parabuccal cirrus, usually 2 extra cirri behind leftmost frontal cirrus, and 3 frontoventral cirri; amphisiellid median cirral row composed of 25-31 cirri with 27-36 left and 27-44 right marginal cirri; usually 5 transverse cirri and 2 pretransverse cirri with 7 dorsal kineties; two macronuclear nodules. In addition to, 18S rDNA sequence of A. milnei was analyzed to understand its phylogenetic relationship.

Extinguishment of Liquid Fuel Fire by Water Mist Containing Additives

  • An experimental study was presented for extinguishing characteristics of liquid fuel fire by water mist($Dv_{0.99}{\leq}200{\mu}m$) containing potassium acetate and sodium acetate trihydrate. To evaluate the extinguishing performance of water mist containing additives, the evaporation characteristics of a water droplet on a heated surface was examined. The evaporation process was recorded by a charge-coupled-device camera. Also, small-scale extinguishing tests were conducted for n-heptane pool fire in ventilated space. During the experiments, flame temperatures were measured, and concentrations of oxygen and carbon monoxide were analyzed by a combustion gas analyzer. The average evaporation rate of water droplet containing additives was lower than that of pure water at a given surface temperature and decreased with the concentration increase due to the precipitation of salt in the liquid-film and change of surface tension. In case of using additives, the fire extinguishing times was shorter than that of pure water at a given discharge pressure and it was because the momentum of a water droplet containing additives was increased. And also dissociated metal atoms, potassium or sodium, were reacted as a scavenger of the major radical species OH, H which were generated for combustion process. Moreover, at a high pressure of 4 MPa, the fire was extinguished through blowing effect as well as primary extinguishing mechanisms.

The Study of Cyanobacterial Flora from Geothermal Springs of Bakreswar, West Bengal, India

  • Geothermal springs in India, formed as a result of volcanic or tectonic activities, are characterized by high temperature and relatively abundant reduced compounds. These thermal springs are inhabited by characteristic thermophilic organisms including cyanobacteria. Cyanobacteria are among the few organisms that can occupy high temperature aquatic environments including hot springs. In alkaline and neutral hot springs and streams flowing from them cyanobacteria can form thick colourful mats that exhibit banding patterns. The present investigation involves study of mat forming cyanobacterial flora from hot springs located in Bakreswar, West Bengal, India. The important species found are Synechococcus bigranulatus, S. lividus, Gloeocapsa gelatinosa, G. muralis, Phormidium laminosum, P. frigidum, Oscillatoria princes, O. fragilis, Lyngbya lutea, Pseudanabaena sp., Calothrix thermalis, and Fischerella thermalis. Their distribution pattern in relation to physico-chemical parameters of spring water has also been studied. Three cyanobacterial strains of the above mentioned list were grown in culture and their pigment content and nitrogen fixing capacity were also studied. Nitrogen fixing capacities of Calothrix thermalis, Nostoc sp. (isolated in culture) and Fischerella thermalis are 5.14, 0.29, and 2.60 n mole $C_2H_4/{\mu}g$ of Chl-${\alpha}$/hr respectively. Carotenoid : Chlorophyll-${\alpha}$ ratio of four mat samples collected from Kharkunda, Suryakunda, Dudhkunda and bathing pool are 2.45, 1.60, 1.48, and 1.34, respectively. Higher value of Carotenoid : Chlorophyll-${\alpha}$ ratio coincided with higher temperature.

MULTI-SCALE MODELS AND SIMULATIONS OF NUCLEAR FUELS

  • Theory-based models and high performance simulations are briefly reviewed starting with atomistic methods, such as Electronic Structure calculations, Molecular Dynamics, and Monte Carlo, continuing with meso-scale methods, such as Dislocation Dynamics and Phase Field, and ending with continuum methods that include Finite Element and Finite Volume. Special attention is paid to relating thermo-mechanical and chemical properties of the fuel to reactor parameters. By inserting atomistic models of point defects into continuum thermo-chemical calculations, a model of oxygen diffusivity in $UO_{2+x}$ is developed and used to predict point defect concentrations, oxygen diffusivity, and fuel stoichiometry at various temperatures and oxygen pressures. The simulations of coupled heat transfer and species diffusion demonstrate that including the dependence of thermal conductivity and density on composition can lead to changes in the calculated centerline temperature and thermal expansion displacements that exceed 5%. A review of advanced nuclear fuel performance codes reveals that the many codes are too dedicated to specific fuel forms and make excessive use of empirical correlations in describing properties of materials. The paper ends with a review of international collaborations and a list of lessons learned that includes the importance of education in creating a large pool of experts to cover all necessary theoretical, experimental, and computational tasks.

Mass production and application of activation tagged hairy root lines for functional genomic of secondary metabolism in ginseng

  • Activation tagging that uses T-DNA vectors containing multimerized transcriptional enhancers from the cauliflower mosaic virus (CaMV) 35S gene is a powerful tool to determine gene function in plants. This approach has been successfully applied in screening various types of mutations and cloning the corresponding genes. We generated an activation tagged hairy root pool of ginseng (Panax ginseng C.A. Meyer) in an attempt to isolate genes involved in the biosynthetic pathway of ginsenoside (triterpene saponin), which is known as the major active ingredient of the root. Quantitative and qualitative variation of ginsenoside in activation tagged hairy root lines were profiled using LC/MS. Metabolic profiling data enabled selection of a specific hairy root line which accumulated ginsenoside at a higher level than other lines. The relative expression level of several genes of triterpene biosynthetic pathway in the selected hairy root line was determined by real time RT-PCR. Overall results suggest that the activation tagged ginseng hairy root system described in this study would be useful in isolating genes involved in a complex metabolic pathway from genetically intractable plant species by metabolic profiling.
