mRNA의 핵에서 세포질로의 이동에 관여하는 spSac3 유전자의 결실돌연변이 제조와 특성 조사 (Construction of spSac3 Null Mutants Defective in mRNA Export)

  • mRNA의 핵에서 세포질로의 이동에 관여하는 발아효모 Saccharomyces cerevisiae의 SAC3유전자와 유사한 분열효모 Schizosaccharomyces pombe의 유전자 (spSac3로 명명)의 결실돌연변이주 (knockout mutant)를 제조하여 그 특성을 조사하였다. S. pombe의 이배체 (diploid) 균주에서 한 spSac3 유전자만을 결실시킨 후 4분체분석 (tetrad analysis)을 수행한 결과, 이 유전자는 성장에 필수적이었다. 또한 in situ hybridization을 통해 세포 내의 poly(A)+ RNA분포를 살펴본 결과, spSac3 유전자의 결실돌연변이주는 mRNA의 핵에서 세포질로의 이동에 이상이 있었다. 이와 같은 결과들은 spSac3 유전자 역시 mRNA의 핵에서 세포질로의 이동에 매우 중요한 역할을 담당하고 있음을 시사한다.

Sn-1.7Bi-0.7Cu-0.6In 솔더의 특성 연구 (Characteristics of Sn-1.7Bi-0.7Cu-0.6In Lead-free Solder)

  • Characteristics of Sn-1.7%Bi-0.7%Cu-0.6%In (hereafter, SBIC) lead-free solder was investigated in this study. The results from SBIC were compared to other lead-free solders such as Sn-3.5%Ag-0.7%Cu (hereafter, SAC), Sn-0.7%Cu (hereafter, SC), and lead-bearing Sn-37%Pb (hereafter, SP) alloy. Tensile properties of bulk solder, wettability, spreading index, bridge and dross were evaluated. As experimental results, tensile strength and elongation of SBIC was 62.5MPa and 21.5%, respectively. The tensile strength was comparable to that of SP solder. The wetting time of SBIC was 1.2 sec at $250^{\circ}C$, and its wetting properties including wetting force were as good as the SAC alloy. However, wettability of the SC was not so good as the SBIC and SAC. The spreading index of SBIC at $250^{\circ}C$ was 71 %, and it was similar level to those of SAC and SC solders. Bridging was not found for all solders of SBIC, SAC and SC in the range from 240 to $260^{\circ}C$. In dross test at $250^{\circ}C$ for an hour, the amount of dross produced from SBIC was about 57% compared to that from SAC.

Rhizobium sp. SNU003의 nifHD 클로닝 (Molecular Cloning of nifHD from Rhizobium sp. SNU003)

  • 해녀콩 뿌리혹의 질소고정 공생균인 Rhizobium sp. SNU003 균주의 게놈내 7.9 kb 의 EcoRI, 6.5kb 의 SalI, 7.3 kb 의 HindIII 와 4.4 kb 의 PstI 절편에 nifHD 가 존재함을 확인하였다. 람다파아지 EMBL3-BamHI arm 을 사용하여 genomic library 를 제조하였으며 이로부터 nif-유전자를 포함하고 있는 9개의 재조합 파아지 클론을 선별하였다. 이들 중 15.3 kb 의 삽입 DNA 를 가지고 있는 Rnif-6 클론은 7.6 kb 의 BamHI/SacI 절편에 nifHD 가 위치하고 있었다. 따라서 이절편을 pUC19 에 sub cloning 하고 제한효소 지도를 작성한 결과 Rhizobium sp. SNU003 의 nifH 와 nifD 는 4.5 kb 의 BamHI/BglII 절편에 연속배열하고 있다.

세균감염에 의한 초생타조(Struthio camelus camelus)의 쇠약 증후군의 발생 증례 (Ostrich chick fading syndrome(OCFS) caused by bacterial infection of farmed ostrich chicks)

  • The most common cause of death is ostrich chick fading syndrome(OCFS), which is due to bacterial infection during artificial incubation and hatching. Six farmed ostrich chicks aged 3 and 10 days in Chonbuk province, were submitted to Chonbuk Livestock Development and Research Institute for necropsy, Clinically, birds showed hair loss, ocular exudate, lethargy, diarrhea, and subsequently died 3-5 days after onset of clinical signs. Grossly, umbilicus was enlarged. White-yellowish purulent nodules were scattered on the lung and the membrane of air-sac was thickened and had inflamed exudate on the surface in two chicks that died 3 days after hatching. In 10 days-old chick, intestine was shown rodding segmentally. Yolk sac was still retarded and its surface was partially hemorrahgic. The synovial fluid of the leg was yellowish. Microscopically, multifocal purulent exudates were scattered on the lung. Capillary microthrombi in the glomerulus were prominent and tubular epithelia were necrotic. Necrotic hepatocytes were scattered and intestine were congested. Microbiologically, Pseudomonas sp and/or E coli were isolated from air-sac, lung and/or liver. This case suggests that poor hygiene during artificial incubation, hatching or in the first week after hatching may cause high mortality of the ostrich chicks.

한국산 포식선충 (Nematoda : Mononchida) 의 2신종 및 2미기록종기재 (Two New and Two First Recorded Species of Predatory Soil Nematodes (Nematoda : Mononchida) from Korea)

  • 포식선충목(Mononchida) 2 신종과 기록된 2 미기록종을 기재하였다. lotonchus cucumis sp. n.는 체장이 2.0-3.2 mm이며, 음문전후방에 각각 3개의 돌기가 있으며, 각질화 된 조각의 자궁, 긴꼬리에 미선구를 가졌다. Mylonchulus sp. n.은 체장이 1.1-1.2 mm이며 구강의 아복부벽에 2쌍의 치아를 가지고있고, 아중부소치가 2-3열로 정리되어 있고, 매우 짧은 후부자궁낭과 미선구가 있다. Mononchus sinensis와 M. aquaticus는 한국에서 처음으로 기록된다.

  • 이형흡충류의 하나인 Stictodora속 흡충 2마리가 구충제 및 하제를 투여한 24세 남자환자의 설 사변에서 검출되었다. 총체는 길이 0.90∼0.98 mm, 폭 0.38 mm로 소형이며 특이한 복흡반-생식만(ventrogenital) 장치가 있고 생식공과 별개인 gonotyl(12∼15개의 미소한 갈고리 보유)을 가지고 있었다. 충체는 형태학적으로 Stictodora fuscatum(Onji and Nishio, 1916)과 매우 비슷하였으나 다수의 충체를 획득할 때까지 확정적인 종 동정은 보류하였다. 환자는 숭어, 농어 등 반염수산 어류를 즐겨 생식한다고 하였다. 이들이 Stirtodora sp.는 물론 따로 보고한 3종류 이형홉충의 감염원이었을 것으로 추측된다. 이 중례는 문헌상 Stictodora속 홉충의 최초 인체기생례에 해당된다.

  • A new haplosplanchnid cercaria, Cercaria kuwaitae VI sp. n., was found in the prosobranch snail Cerithidea cincingulata in the Kuwait Bay Details are presented on the morphology and behavior of the cercaria and the encystment process. The new cercaria is a biocellate, distome, with a prominent single sac-like intestinal cecum extending well posterior to the ventral sucker and develops in simple sporocysts. It differs from known haplosplanchnid cercariae in the absence of finger-like processes on the tail, and the presence of V-shaped excretory vesicle extending beyond ventral sucker and the presence of cervical glands. The surface topography of the cercaria and its sporocyst is examined by scanning electron microscopy. This is the first haplosplanchnid cercaria to be described from a Cerithidea species.

타조(Struthio camelus)에서 발생한 아스퍼질러스증 (Aspergillosis in an Ostrich (Struthio camelus))

  • A nine-month-old male ostrich (Struthio camelus) exhibited loss of appetite and dyspnea for 10 days, followed by emaciation and death. Grossly, multiple white nodules measuring 1-3 mm in diameter were observed in the surface of and inside of the lung and in the mucosa of the air sac. Microscopically, the granuloma formations were observed in the lung and air sac. The core of granuloma consisted mainly of macrophages and fibroblasts. The thin layer comprising the giant cells and macrophages surrounded the granuloma. By Periodic acid Schiff reaction, mycelia were detected especially in the core of granuloma. From the present results, the causative agent inducing the death of an ostrich was thought to be Aspergillus sp. This is the first report of the occurrence of acute Aspergillus pneumonia (brooder pneumonia) in an adult male ostrich in Korea.

A new species of Brianola Monard, 1926 (Copepoda: Harpacticoida: Canuellidae) from Rawai Beach, Phuket Island, Thailand

  • A new species of the canuellid genus Brianola Monard, 1926 (Copepoda: Harpacticoida) was collected during a meiofauna survey in July 2021 at the intertidal muddy sand of Phuket Island, Thailand. Currently, there are only eight known species of Brianola: B. stebleri, B. exigua, B. curvirostris, B. elegans, B. sydneyensis, B. vangoethemi, B. hamondi, and B. haliensis. Thus, the Brianola rawaiensis sp. nov. from Thailand is the ninth species of the genus. The new species, Brianola rawaiensis is most closely related to B. haliensis by sharing a four-segmented antennule in females and five-segmented antennule in males, eight-segmented antennary exopod and three-segmented antennary endopod, and P1 exp-3 with 5 spines/setae. However, B. rawaiensis sp. nov. is different from B. haliensis by having: 1) P1 exp-1 with 1 outer pinnate spine, 2) P1 exp-2 with inner plumose seta, 3) P2 exp-3 with 3 setae/spines, 4) P2 enp-1 with one inner plumose seta, 5) P4 enp-1 with inner seta. Sexual dimorphism is expressed in the antennule and segmentation of the urosome. Ovigerous female bearing one egg sac with 20 eggs.

Bradyrhizobium sp. SNU001 nod 유전자 클로닝 (Molecular Cloning of nod Genes from Bradyrhizobium sp. SNU001)

  • 대두 (Glycine max) 뿌리혹의 질소고정 공생균주 Bradyrhizobium japonicum SNU001 의 nod 유전자를 클로닝하였다. Rhizobium meliloti 의 4.5 kb EcoRI/ HindIII 절편을 탐침으로 한 게놈 혼성화 반응을 분리균주의 게놈상에 nod 유전자가 존재함을 확인하고, lambda EMBL3-BamHI vector 를 이용하여 genomic library 를 작성하였다. 작성된 library 로부터 1, 2 차 선별과정을 통해 nod 유전자가 있는 클론 1-5 를 선별하고, 클론 2로부터 nod DABC 탐침과 lambda DNA 탐침을 사용한 혼성화반응을 수행하여 삽입된 genomic DNA 에 대한 부분적인 제한효소 지도를 작성하였다. nod DABC 탐침과 가장 강한 혼성화반응을 보인 phage 클론 lambda CNS-1 의 3.9 kb BamHI 절편을 pBS KS(+) vector 에 subcloning 하고 동일한 탐침을 이용한 혼성화반응을 통해 subclone pBjCNS-1 을 선별하였다. 이 subclone 에 대한 부분적인 제한효소 지도를 작성하여 nod DABC 가 1.8 KpnI/SacI 절편에 존재함을 확인하였다.
