Comparative Analysis of Aniline Dioxygenase Genes from Aniline Degrading Bacteria, Burkholderia sp. HY1 and Delftia sp. HY99. (Aniline 분해균주 Burkholderia sp. HY1과 Delftia sp. HY99에서 유래된 Aniline Dioxygenases 유전자의 비교 분석)

  • In this study, aniline dioxygenase genes responsible for initial catabolism of aniline in Burkholderia sp. HY1 and Delftia sp. HY99 were cloned and the amino acid sequences were comparatively analyzed, which already have been reported as bacteria utilizing aniline as a sole source of carbon and nitrogen, B. sp. HY1 was found to have at least a plasmid, and the plasmld-cured strain, B. sp. HY1-PC obtained using mitomycin C was tested with wild type strain to investigate whether the former maintained the degradability for aniline. This proved that the aniline oxygenase gene from B. sp. HY1 was located in chromosomal DNA, not in plasmid DNA. Aniline dioxygenase small subunits from B. sp. HY1 and D. sp. HY99 were found, based on 146 amino acids, to share 79% similarity. Notably, ado2 genes from B. sp. HY1 and D. sp. HY99 which were found to be terminal dioxygenase of aniline dioxygenase small subunit showed 99% similarity in the deduced amino acid sequences with tdnA2 of Frateuria sp. ANA-18 and danA2 of D. sp. AN3, respectively. Besides, enzyme assay and amino acid sequence analysis of catechol dioxygenase supported the previous report that B. sp. HY1 might occupy ortho-cleavage pathway using catechol 1,2-dioxygenase, while D. sp. HY99 might occupy catechol 2,3-dioxygenase for meta-cleavage pathway.

Drug Resistance Profiles of Salmonella sp. and Shigella sp. Isolated from Diarrheal Patients in Pusan, Korea (부산지역의 설사환자로부터 분리한 Salmonella sp. 및 Shigella sp.의 약제내성 유형에 관한 연구)

  • A total of 34 Salmonella sp. an d25 Shigella sp. were isolated from 311 patients with diarrhea. The isolation rates of Salmonella sp. ad Shigella sp. were 10.9% and 8%, respectively. The serogroups of 34 Salmonella sp. were in order of group D(50%), group B(38.25), group E(8.8%) and group C 92, 9%0. the serogroups of 25 Shigella sp. were group D(96%) and group B(4%). Seasonal distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at July, 17.65% and 64%, respectively. Age group distribution of isolated Salmonella sp. and Shigella sp. were shown the most high at twenties and thirties (23.5%), and teens(76%), respectively. The Salmonella isolates were resistant in order of prevalence use of streptomycin(SM) (100%), erythromycin (EM) and movobiocin (NB)(90.6%), penicillin G(PG) (65.6%) and cephalexin (CPX)(46.9%). the isolates of Shigella sp. were resistant in order of prevalence use of EM (95.8%), NB(91.7%), SM(87.5%). Eighteen kinds of resistant patterns of Salmonella ioslates were detected. The multiple resistance patterns of Shigella isolates were mostly SM, EM, NB type (79.2%). The minimum inhibitory concentration of Salmonella sp. and Shigella sp. and Shigella sp. isolated from patients with diarrhea were tabulated.

Variations of Species Composition of Early Feeding Organisms Through Nature Mixed Culture (자연혼합배양에 의한 초기 먹이생물의 종 조성 변화)

  • We were successfully reared young marine ornamental larva fish in a unique process of microalgae blooming culture tank. The marine fish larva was grown and survived in this method. Generally, we called this method as natural mixed culture. Observed planktonic microalgae were 34 species with 19 diatoms (Detonula pumila, Nitzschia sp., Fragilaria oceanica, Chaetoceros curvisetus, Stephanodiscus sp., Chaetoceros decipies, Chaetoceros sp., Thalassiosira rotula, Eucampia zodiacus, Diploneis splendica, Nitzschia longissima, Surirella cuneata, Asterionella glacialis, Nitzschia spp., Chaetoceros debile, Thalassionema nitzschioides, Nitzschia closterium, Skeletonema costatum and Licmophora sp.), 14 flagellates (Euglena, sp., Gonyaulax sp., Pyramimonas sp., Protoperidinium sp., Eutreptia sp., Parapedinella sp., unidentified micrc-flagellate, Gyrodinium sp., Scrippsiell trochoidea, Gymnodinium sanguineum, Chrysochromulina sp., Gymnodinium sp., Prorocentrum triestinum and Micromonas sp.) and 1 ciliate (Mesodinium rubrum) in this culture tank. Dominant microalgae were Chrysochromulina sp. during the larval rearing periods. Blooming condition maintained continuously and stably from 10 to 60 days in this microcosm.

Differences of Cesarean Section Rates according to San-Yin-Jiao(SP6) Acupressure for Women in Labor (삼음교 지압에 따른 산부의 제왕절개율 차이)

  • Purpose: The purpose of this study was to explain differences of cesarean section rates according to San- Yin-Jiao(SP6) acupressure for women in labor. Method: A noneqivalent control group pre test - post test design was used to explain differences of cesarean section rates according to SP6 acupressure. The participants were 209 women who were assigned to one of three groups SP6 acupressure(n=86), SP6 touch(n=47), and control group(n=76). For 30 minutes, the SP6 acupressure group received SP6 acupressure, and the SP6 touch group received SP6 touch for the duration of each uterine contraction. The Control group was encouraged to deep breath and relax for the duration of each uterine contraction for 30 minutes. Result: The rates of cesarean section were 12.8%, 29.8%, and 22.4% for the SP6 acupressure group, SP6 touch group, and control group respectively. There was a significant difference among groups (p=0.049). Cesarean section rateswere significantly different between the SP6 acupressure and non-SP6 acupressure group(p=0.035). Conclusion: This finding shows that 30 minutes of SP6 acupressure was effective in decreasing the cesarean section rate. Therefore, SP6 acupressure during labor could be applied as an effective nursing intervention.

Diversity of Fungal Endophytes from Pinus koraiensis Leaves in Korea (한국의 잣나무에서 분리한 내생균의 다양성)

  • The composition of endophytic fungal species from Pinus koraiensis was studied in two areas (Yeongwol and Jincheon), Korea. To identify 113 isolates, rDNA ITS regions were sequenced. On the basis of the ITS sequence, Pestalotiopsis sp. was the most frequently isolated species in both areas. Pestalotiopsis sp. (68.5%) and Lambertella sp. (12.4%) were isolated frequently in Yeongwol, whereas Pestalotiopsis sp. (41.7%), Hypoxylon sp. (20.8%) and Phomopsis sp. (12.4%) were dominant in Jincheon. Hypoxylon sp. and Phomopsis sp. were not found in Yeongwol.

Studies on the morphological and physiological characteristics of isolated strains from rotting ginseng (인삼부패곰팡이의 형태 및 생리학적 특성에 관한 연구)

  • Three kinds of microoganisms were isolated and identified from the ginseng and ginseng products to research the properties of the molds which spoil the ginseng and ginseng products. The results obtained were as follows: (1) The predominant strains on ginseng products were Aspergillus sp., Penicillium sp.-A and Penicillium sp.-B. These predominant fungi deteriorated ginseng products exclusively, (2) Aspergillus sp. showed the greatest mycelial growth at $40^{\circ}C$ and its optimum pH was 5, meanwhile Pencillium sp. showed the greatest mycelial growth at $30^{\circ}C$ and its optimum pH was 3. (3) The growth of the isolated strains was stimulated with the increase in the concentration of saponin at the lower concentration, meanwhile it was inhibited at 1.0% concentration of saponin.

  • The effects of substrate size on the growth of microphytobenthos Achnanthes sp., Amphora sp., Navicula sp. and Nitzschia sp. were examined using glass beads in order for phytoremediation in the benthic layer of coastal waters. The glass beads used in this study were 0.09~0.15 mm (G.B 1), 0.25~0.50 mm (G.B 2), 0.75~1.00 mm (G.B 3) and 1.25~1.65 mm (G.B 4). No addition of glass bead used as control. The specific growth rate and maximum cell density of four microphytobenthos species were increasing with decreasing size of glass beads. Moreover, the control experiment without added attachment substrates showed the lowest specific growth rate and maximum cell density. Therefore, the suitable attachment substrates for mass culture of microphytobenthos seems to be important in order for phytoremediation using microphytobenthos.

Antitumor Activities of Polysaccharudes fractuibuzed from Zoogloea sp. Against Meth A Cells (Zoogloea sp.의 다당체가 Meth A 세포에 의한 종양형성 억제 효과)

  • The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.

  • The vegetative nuclear divisions in hyphae and chromosome numbers were studied with the aid of Giemsa-HCl techniques from 10 strains of Fusarium oxysporum. The entire nuclear division process occurred within an intact nuclear envelope like other fungus. The results confirmed that 2 strains(F. oxysporum S Hongchun D2, F. oxysporum S Jinyang 4) were n=4; 3 strains(F. oxysporum f. sp. lini KFCC 32585, F. oxysporum f. sp. melongenae KFCC 34743 and F. oxysporum f. sp. raphani) n=5; 2 strains(F. oxysporum f. sp. vasinfectum, and F. oxysporum f. sp. mori KFCC 34742) n=6; 3 strains(F. oxysporum f. sp. cucumerium, F. oxysporum f. sp.niveum, and F. oxysporum f. sp. pisi) n=7.
