Genetic Variation in Sprout-related Traits and Microsatellite DNA Loci of Soybean

  • Genetic diversity and soybean sprout-related traits were evaluated in a total of 72 soybean accessions (60 Glycine max, 7 Glycine soja, and 5 Glycine gracilis). 100-seed weight (SW) was greatly varied and ranged from 3.2g to 32.3g in 72 soybean accessions. Positive correlation was observed between GR and hypocotyl length (HL), whereas negative correlation was observed between SW and hypocotyl diameter (HD). Re-evaluation by discarding two soybean genotypes characterized with low GR indicated that much higher correlation of sprout yield (SY) with HD and SW. Based on the principal component analysis (PCA) for sprout-related traits, 57 accessions were classified. Soybean genotypes with better traits for sprout, such as small size of seeds and high SY, were characterized with high PCA 1 and PCA 2 values. The seed size in second is small but showed low GR and SY, whereas the third has large seed, high GR and more than 400% SY. In genetic similarity analysis using 60 SSR marker genotyping, 72 accessions were classified into three major and several minor groups. Nine of twelve accessions that were identified as the representatives of soybean for sprout based on PCA were in a group by the SSR marker analysis, indicating the SSR marker selection of parental genotypes for soybean sprout improvement program.

Control of Soybean Sprout Rot Caused by Pythium deliense in Recirculated Production System

  • A soybean-sprout rot epidemic occurred in a mass production soybean sprout factory in 2000 and 2001 in Korea, which caused up to 20% production loss. Among the causal pathogenic bacteria and fungi, Pythium deliense was found to be the dominant pathogen of severe root and hypocotyls rot, particularly in recirculating water system. An average of 90% of the isolated fungi from the rotted sprout on potato dextrose agar were Pythium sp. The fungal density of Pythium in the sampled water was monitored in the recycled water system for 1 year using a selective medium (com meal agar with Pimaricin, 10 mg; Rifampicin, 10 mg; and Ampicillin, 100 mg per 1 liter). The drained water from the soybean-sprout cultivation always had a certain amount of fungus in it. The removal of Pythium from the recycling water system must be thorough, safe, and environment friendly. However, the pathogen in the water was easily found even after ozone and chlorine treatments, which were devised on the recycling system for the removal of microorganisms. 5-$\mu\textrm{m}$ pore size filter was applied and was able to successfully control the disease. As the sprout industry increasingly shifts into mass production, the demand for water will increase continuously. Recycling water for sprout production is eco-friendly. However, a process must be devised to be able to first decompose organic matters before Pythium zoospores are filtered.

  • 1. 콩잎의 세균밀도는 4.60$\times$$10^2$~ 9.10$\times$$10^{5}$CFU/$\textrm{cm}^2$으로, 생육단계가 진전됨에 따라 세균밀도가 증가하는 경향이었다. 2. 콩나물 부패 세균의 밀도는 콩잎에서 0~5.00$\times$$10^3$CFU/$\textrm{cm}^2$으로, 부패세균의 밀도는 생육단계에 관련이 없었으나 재배지역과는 관련이 있었다. 3. 나물 콩 품종과 콩나물 부패세균의 밀도는 품종과 관련이 적었으며 생육단계와 작물의 부위에 따라 변이가 심하였다. 4. 콩잎에서 분리된 콩나물 부패세균은 Erwinia cypripedii, E. carotovora subsp. carotovora, Xanthomonas campestris pv. glycines, Staphylococcus sp., Micrococcus sp. 이며, E. carotovora subsp. carotovora, X. campestris pv. glycines가 밀도가 높았다.다.

환기봉을 이용한 재배용기 내부 콩나물 품온 저하 및 부패 경감 (Utilization of ventilation Pipe to Decrease Commondity Temperature and Rot of Soybean Sprouts)

  • 재배중인 콩나물의 용기 내 품온의 저하 및 부패 경감을 통하여 콩나물 생산성을 증대코다 콩나물 재배 용기(30$\times$28$\times$25cm) 내부에 다공성 PVC 환기봉(지름 32mm$\times$높이25cm ; 128개의 지름 2mm 환기구)을 1, 3, 5개씩 수직방향으로 설치하고 6일간 콩나물을 재배하며 품온의 변화 및 수량, 상품성 개체비율, 부패율 등을 조사한 결과 환기봉 설치에 의하여 용기 내부의 온도가 저하됨을 알 수 있었는데, 특히 바닥에서 5cm 높이의 하층 및 10cm 높이의 증측 부위가 15cm 상층 부위(3.$0^{\circ}C$)에서 보다 효율적으로 온도가 저하되어 각각 4.7$^{\circ}C$ 및 3.9$^{\circ}C$의 온도 저하효과가 나타났으며, 용기 내부 상/중/하층의 온도 편차가 감소되어 균일한 콩나물 생산이 가능할 것으로 사료되며 수확된 콩나물의 성상 및 수량 조사결과 수율과 상품성 개체 비율이 무처리에 비해 각각 3.9%, 4.0% 증대되고 부패율은 4% 감소되는 등 콩나물 재배시 환기봉을 사용함으로써 농약을 사용치 않으면서 콩나물의 재배 용기 내 품온 저하를 통한 부패 경감과 콩나물생산성 향상이 가능할 것으로 나타났다.

조리방법에 따른 상용채소의 무기질 함량 변화 -무청, 콩나물, 취나물을 중심으로- (Changes on Mineral Contents of Vegetables by Various Cooking Methods)

  • This study was conducted to investigate the changes of mineral contents in radish leaves, soybean sprout and chwi namul by cooking method(boiling, steaming, sauting) and cooking time(1, 3, 5 min.). The residual rate of minerals(such as Calcium, Sodium, Potassium, Iron and Magnesium) in radish leaves, soybean sprout and chwi namul by 1 minute sauting was the range of 83.2 to 99.1%. It was shown that sauting was desirable method for all three vegetables. The residual rate by the cooking method was sauting, steaming, boiling, in that order, but boiling in radish leaves and chwi namul were more desirable method than steaming for Calcium use. At the cooking method and cooking time, Potassium content in soybean sprout was reduced remarkably in 5 minutes boiling. Boiling of radish leaves and soybean sprout showed that Sodium was reduced remarkably after 5 minutes of boiling, 54.4% for radish leaves and 19.9% for soybean sprout, respectively.

콩나물의 Asparagine이 숙취에 미치는 영향 (Effects of Soy-sprout Asparagine on Hangover)

  • This experiment was conducted to determine the amount of asparagine content in soybean sprout soup as well as its effect on the regulation of blood alcohol concentration in human body. Asparagine content and alcohol concentration in blood were analyzed using the amino-acid analyzer and alcohol tester, respectively. Asparagine content in soybean sprout soup was highly significantly different among sprout soups sampled from different restaurants, although the boiling time and amount of sprout used were different among the samples. Significantly higher asparagine content was observed in soups with higher amounts of sprouts (2.81% in 500 grams of sprout) and 10 minutes after boiling with other solid ingredients in the soup. Lower concentration of alcohol in blood was recorded in persons with higher body weight. The asparagine+soybean extract (eaten immediately after drink) lowered the blood alcohol concentration significantly followed by only asparagine and control. The blood alcohol concentration after drinking become zero earlier (by 30 minutes), when asparagine+soybean extract or only asparagine was consumed as compared to control.

환경요인이 콩나물 무름병 발생에 미치는 영향 (Effects of Environmental Conditions on Incidence of Bacterial Soft Rot in Soybean Sprout)

  • Erwinia rarotovora subsp carotovora에 의한 콩나물 무름병 발생을 인위적인 조건하에서 조사하였다. 콩나물의 재배 온도가 콩나물 무름병 발생과 생장에 미치는 영향은 3$0^{\circ}C$이상의 고온에서 무름병 발생율이 높아졌으며 35$^{\circ}C$에서는 약 22%의 발병율을 나타내었다. 종피와 자엽 부위에 인위적인 상처를 낸 경우 70%이상의 발병율을 보였으며 콩나물의 신장을 억제하였다. 습도는 90%이상 유지시 33%의 발병율을 보였으며 콩나물의 생장을 억제시켰다. 콩나물 재배 배출액을 관수액으로 재이용하였을 때 무름병의 발병율이 증가하였다.

  • 야생종의 콩나물 관련형질의 특성 및 주요성분을 재배종과 비교하여 야생종을 소립 나물콩 육성을 위한 교배친으로서의 이용 가능성을 검토하기 위해 실시한 결과를 요약하면 다음과 같다. 1. 야생종과 재배종의 콩나물 특성을 비교한 결과 하배축의 길이와 수량에는 차이가 없었으나, 콩나물의 전체길이, 하배축 두께, 개체당 무게, 상품률에서는 재배종이 우수하였고, 잔뿌리의 수와 뿌리의 길이에서는 야생종이 작고 짧아 야정종이 우수한 것으로 평가되었다. 2. 조지방 및 sucrose함량은 재배종이, 조단백질 함량은 야생종이 높았으며, 지방산중 oleic acid는 재배종이 linoleic acid는 야생종이 높았고, isoflavone함량에서는 두 종간 차이가 없었다. 17개의 아미노산 중 유의적인 차이가 인정된 aspartic acid, lysine, arginine등 10개 아미노산은 모두 야생종에서 높았다. 3. 콩나물 하배축의 색차는 야생종에서 백색도가 높고 적색도가 낮아 재배종보다는 야생종이 우수한 것으로 평가되었다.

고콜레스테롤 식이로 사육한 흰쥐 혈청 지질의 성분에 미치는 대두 배아의 영향 (Effects of Soybean Germ on the Lipid Composition of Serum in Cholesterol Fed Rats)

  • 대두(Glycin max Merrill) 배아의 기능적 활성을 조사하기 위하여 대두를 발아시켜 만든 배아(soybean sprout)을 시료로 하여 0.75% 콜레스테롤 첨가식이로 유발된 고지혈증 Sprague Dawley계 수컷의 흰쥐에 대두 배아의 유기용매 추출물을 급여시킨 후 혈청의 지질 개선효과를 알아보기 위하여 4주간 동안 대두 배아를 섭취시켰다. 식이 급여가 대조군의 체중 및 장기 무게에 미치는 영향은 거의 찾아볼 수 없었으며 혈청 총 콜레스테롤 및 LDL-콜레스테롤의 농도는 대조군에 비하여 낮았고. 콜레스테롤 식이군에 비하여 콜레스테롤 식이와 대두 배아 메탄올 추출물의 혼합투여시 낮게 나타났다. 총 콜레스테롤에 대한 HDL-콜레스테롤의 비는 대두 배아 추출물 급여군이 높게 나타났으며, 동맥경화 지수는 고콜레스테롤 식이군에 비하여 대두 배아 추출물이 낮았다. 또한 혈청중의 유리 콜레스테롤 및 고콜레스테롤 에스테르의 농도는 고콜레스테롤 식이군에 비해 대두 배아의 혼합 급여군이 감소하였고, 인지질의 농도도 콜레스테롤 식이군 보다 콜레스테롤 식이와 대두 배아 급여군이 낮은 경향을 나타내었다.

Elucidation of Cause of Cotyledon Black-Decay of Soybean Sprout by Bean Bug, Riptortus clavatus

  • Cotyledon Black Decay (CBD) on soybean sprout mimics the black spot due to microbial infection. CBD, not visible or predictable at seedlot state, for some reason, shows up exclusively on cotyledon of soybean sprout during sprouting process. Such an incidence rate fluctuated from 0.8 to 19.5% over three years from 2004. We suspected some pod-infecting anthracnose fungi and/or pod-blight pathogen, or pod-sucking bean bug, one of the major pests of soybean, might have involved, of which we ruled out fungal pathogen because it was preventable through heat treatment, a proven method for seedlot disinfestation. The healthy seeds artificially fed by bean bug for one to seven days were sprouted, and 6 to 41% of the soybean sprout revealed the CBD mimic to those occurred in soybean sprout from previous commercial seedlot screening experiments. This finding is the first report to confirm that bean bug damage to pod at $R_8$ stage is directly responsible for the CBD, which did not concur with any other deleterious effects on sprouting such as reduction in hypocotyls elongation and rooting except unsightly sprout quality. However, earlier feeding either at green pod or greenish yellow pod stage ($R_6$ -early $R_7$ stage) resulted in rather severe damages, which strikingly reduced hypocotyls growth to about one forth to about two third, as well as the reduction in rates of seed germination.
