Natural Photodynamic Activity of 5-Aminolevulinic Acid Produced by E. coli Overexpressing ALA Synthase from Bradyrhizobium japonicum

  • The present study was conducted to determine plant growth and physiological responses of corn, barnyardgrass, and soybean to ALA (5-aminolevulinic acid). ALA effect on early seedling growth of test plants was greatly concentration dependant, suggesting that it inhibits at higher concentrations. No significant difference in herbicidal activity of two types of ALA on plant height and weight of test plants was observed. Barnyardgrass was the most sensitive to ALA and followed by corn and soybean, indicating that both crop plants were less affected by ALA concentration as well as different growth stages than barnyardgrass. Greatly reduced chlorophyll contents from leaves of three plant species were observed with increasing of ALA concentration. Compared with untreated controls, higher amounts of three tetrapyrroles were detected from three crop plants, indicating more accumulation in ALA-treated plants. The differential selectivity among plant species would be explained with the differences in tetrapyrrole accumulating capabilities, the susceptibility of various greening groups of plant species to the accumulation of various tetrapyrroles, and their metabolism in various plant tissues. The results indicate that negative biological potential of ALA exhibited differently on plant species, and that the photodynamic herbicidal activity against susceptible plants highly correlated with the extent of tetrapyrrole accumulation by the species.

  • 1. In order to obtain useful mutants such as early maturity, resistance to lodging, high protein and oil content, and capability of high yield, dormant seeds of two soybean varieties, Jang Dan Baik Mok and Clark, were treated with ${\gamma}$-ray, Ethyl Methane Sulfonate(EMS), Ethylene Imine(EI)and combinations of ${\gamma}$-ray and EMS or EI. 2. The germination rate and survival rate in a variety Jang Dan Baik Mok were significantly decreased with ${\gamma}$-ray treatment while it was not the same in the Clark variety. A significant decrease for seedling height measured at 14 and 21 days after sowing was found with the increase of ${\gamma}$-ray dose in both varieties. 3. Germination rates in both varieties were significantly decreased as EI concentration increases, particularly severe damage in germination was observed at 0.008 Mo. concentration. Germination rate damages were found with EMS concentration increases in the variety Jang Dan Baik Mok while no regular responses in seedling height were observed in the variety Clark. 4. Germination rate was significantly lowered with the combined treatment of EMS and ${\gamma}$-ray 24KR than that of EMS alone. In the treatments of ${\gamma}$-ray with three levels of EI concentration, the combined treatments except 24KR+EI 0.002 Mol. resulted in better germination than of EI alone. In both varieties, significant reduction in seedling height was observed in the combined treatments of ${\gamma}$-ray with various concentrations of EMS, whereas stimulation effect on seedling height was found with treatment of ${\gamma}$-ray EI +0.08Mol. 5. Germination rate, survival rate, and plant height as criteria of radio sensitivity, variety Jang Dan Baik Mok is moze sensitive to ${\gamma}$-ray, EMS, and EI than the variety Clark, and the varietal difference in responsibility to mutagen may be due to the genetic-constitution of the varieties.

Effect of Planting Time on Seed Production of Vegetable Soybean at Different Locations (풋콩 재배지에 따른 파종시기가 종자생육에 미치는 영향)

  • This investigation was conducted to determine the effect of the optimum planting date and place for good seed production on growth variation, grain yields, different of variety, infection of seed by fungi, and seed germination and vigor after room storage. Early maturity Korean soybean variety, Keunolkong, and introduced vegetable soybean variety, Fukura-edamame, were planted at field of YAES. and high cool land of Sajapyong in Milyang(altitude, 850m) on May 15 (early), June 15 (mid.), and July 15 (late) in 1991. The emergence and vegetative period gradually increased in the early planting date. The grain yield, seed weight, pod number, healthy grain yield also increased when Keunolkong was planted on early date. The healthy seed rate, Keunolkong increased in the early planting date, whereas that of Fukura also increased in the late planting date. Infection ratio of grain to phomopsis seed decay (Phomopsis spp. ' Diaporthe phaseolorum) in Fukura steadily increased in the early planting date. Infection ratio of grain to purple seed stain (Cercospora kikuchii) generally increased in the mid and late planting date. The seed germination and seedling vigor after room storage from five to six months gradually increased in the late planting date. And seed germination of Fukura rapidly decreased in the early planting date. Therefore, the optimum planting date for good seed production in early maturity vegetable soybean was June 15 in terms of harvesting time avoid a high temprature and humidity.

Effects of Auxins and Cytokinins on Organogenesis of Soybean Glycine max L.

  • To select the section with shoot formation ability, the calli and shoot formation from three sections (first leaf including cotyledonary node, hypocotyl and cotyledon explants) of 5-days-seedlings of soybean were induced on MS medium supplemented with 1.0 mg/L BAP, 3% sucrose, and 0.3% gelrite for one month. The first leaf section exhibited the highest shoot formation rate (51%), followed the hypocotyl section (10%) and the cotyledon section (0%). The shoot formation rates and shoot number of the four excised sections (whole first leaf, a half of the first leaf, a third of the first leaf and only node) of the first leaf were also investigated on the same medium. A half of the first leaf explant and the third of the first leaf explant had higher shoot formation rates (76-80%) and numbers (3-4 / explants) than those in other two explants. Effects of six cytokinins (kinetin, zeatin, BAP, 2iP, PBA, and TDZ) on shoot formation were determined, using the half of the first leaf explants. Zeatin (1.0 mg/L) exhibited the highest in shoot formation rate (94%) and numbers (8 / explant). In addition, the combined effects of three cytokinins (zeatin, BAP, and TDZ; 0.5, 1.0, 2.0 mg/L, respectively) and an auxin (IAA; 0.0, 0.5, 1.0, 2.0 mg/L) were determined. The combination (1:1, v/v) of zeatin (1.0 mg/L) and IAA (1.0 mg/L) exhibited the highest in shoot formation rate (96%) and numbers (16 / explant), twice more than zeatin (1.0 mg/L) alone. The shoot cuttings were transferred and cultivated on the rooting media supplemented with only auxin, IBA at various concentrations. The highest root formation (8 / shoot) was achieved on the medium supplemented with 1.5 mg/L. After 4 weeks of cultivation, the plantlets with an extensive root system were transplanted in pots with a soil mixture of vermiculite and fine sand. Transferred to field, about 75% of the plantlets survived.

  • Vie investigated the effect of indole acetic acid (IAA) on diamine oxidase (DAO; EC 1.4.3.6) in the seedling of soybean (Glycine max L. forma Paldalkong). DAO activity was not detected in the resting soybean seeds. During germination it appeared in the elongating zone on day 3, increased up to day 4 and decreased thereafter. Endogenous IAA content has been shown the same pattern as DAO activity. However, cadaverine content was reduced on day 4. To investigate the effect of IAA on DAO activity, the segments of hypocotyl on day 3 were soaked in various concentration of IAA. The activity increased at low concentrations ($10^{-7}-10^{-6}M$) of 1M as compared to that of control but not at high concentrations ($10^{-5}-10^{-4}M$) of IAA. Differing from DAO activity, cadaverine content increased as concentrations of IAA increased. On the other hand, ethylene was induced by IAA at high concentrations. To study the effect of ethylene on DAO activity, we cotreated IAA and 2,S-Norbonadiene (NDE). In this case, DAO activity was not affected.fected.

Comparision of the Activity and Characteristics of Lactate Dehydrogenase Isolated from Different Parts of Soybean Seedling (발아초기의 콩 부위별 Lactate Dehydrogenase 활성변화 및 효소성질 비교)

  • The change of lactate dehydrogenase(LDH) activity and the possibility of the existence of LDH. isozyme were examined with different parts of soybean sprout. The enzyme activity was little changed in cotyledons throughout the early stagy of germination. However, hypocotyls and roots showed the continuous decline of the enzyme activity since the radicle emerged from seeds. It was found that LDH from hypocotyls. and roots was unstable as compared with LDH from cotyledons, even at low temperature. The enzyme from hypocotyls and roots was not purified with a good yield when the purification procedure developed for LDH from cotyledons. was employed. LDH from hypocotyls and roots has the Rm value of 0.29, and 0.25 from cotyledons. The apparent Km value for LDH from cotyledons was 0.45mM with sodium pyruvate, while crude homogenate of hypocotyls or roots showed biphasic phenomenon with two Km values 0.014 and 0.45mM. The results indicate the possibility that crude homogenate of hypocotyls or roots may contain a different LDH isozyme from the LDH of soybean reported previously.

  • In order to determine and to reduce the differences between biochemical test method for the seed viability with T.T.C. (2, 3, 5. Triphenyl tetrazolium Chloride) reagent and germinator method, the topographical diagram of red colored formazan was carried out in 15-16 parts differences. From this resulted information, the classification of the typical staining reaction given for 3 species were derived into 3-4 parts for the germinable in normal seedling, the same as the following results. 1. Corn (Gram inaceae) * Entier embryo stained in bright red color. * Both extremities of scutellum unstained * Both extremities of scuttlium, coleorhiza and non-critial portions of radicle unstained. 2. Soybean (Leguminosae) *Seed completely stained in red color. *Minor unstained areas on cotyledons. *Extreme tip of radicle unstained; minor unstained areas on cotyledons 3. Radish (Cruciferae) *Seed completely stained. *Minor unstained areas on cotyledons. *Outer cotyledon mostly unstained: inner cotyledon completely stained Extreme tip of radicle unstained: large portion of outer cotyledon unstained.

Studies on Bacterial Diseases of Soybean (대두의 세균성병에 관한 연구)

  • Bacterial diseases of soybean has been recognized as a limiting factor of soybean production in Korea as it was estimated to cause around 10 percent of yield losses annually. The purpose of the study is to obtain information on the diseases through proving the kinds of pathogens and epidemiology, The wire brush method and multineedle appeared to be the best way of inoculation under all circumstances. Wire brush method, especially, was effective in shortening the incubation period and manifesting the lesion development by introducing more inoculum per unit of area. In case of spray inoculation it was necessary to apply a small amount(1 : 1,000) of wetting agent, twin-20, otherwise it was unabled to produce the diseases under field conditions. Two kinds of bacterial diseases caused by Pseudomonas glycinea and Xanthomonas phaseoli var. sljense were found from surveyed areas in Kore. Wild fire disease on soybean caused by Pseudomonas tabaci had not detectable during the experiment although there were several reports on the disease from other countries. when the pathogens were introduced into sterile soil, bacterial leaf blight pathogen could exsisted until 30 days while bacterial pustule pathogen survived only 4 days under the natural conditions of later June. Both bacteria, however, could produce the disease after more than 10 months period of storage in refrigerator when they were exsisted in infected plant tissues. In warehouse, non-temperature controlled, the bacteria lose their infectability within 6 months period from October to April even though they exsisted in infected tissues. Surface infested seeds with the pathogens could not produced the diseases on seedling stages of soybean plants when the seeds were planted in sterile soil after inoculation by dipping the seeds into bacterial suspensions, although germination was depressed by the pathogens when the seeds were planted on agar media.
