Ecological Characteristics of Collected Small Grain Soybean (Glycine max L.) Varieties (수집(蒐集) 소립(小粒)콩의 주요 생태적(生態的) 특성(特性))

  • Bean sprout is one of the traditional vegetable in Korea and produced by sprouting soybean (Glycine max L.) cultivars. This experiments was conducted to investigate the ecological characteristics in collected small grain soybean lines from Chungnam province region. Thirty varieties were used in this study. Seed coat color of collected small soybean lines showed eight different types. Major seed coat color type was yellow as 8 lines of total 35 lines. The stem height, number of nodes and number of branches averaged 58.5 cm, 16.5 nodes and 11.8 branches, respectively. The average number of main stem pods and number of branching pod were appeared 43.1 and 62.7 respectively. One hundred seed weight averaged 9.9g which was lower than 10~12g of sprouting soybean cultivars as breeding goal of Korea. The average of node width showed 13.2cm and long stem open types and short stem close types were classified 1 line and 14 lines. The flower dates of collecting small soybean lines were between 8 Aug. and 13 Aug. and flowering duration was 74~85 days. The date of maturity were between 12 Sep. and 12 Oct., and maturity duration showed great variation among cultivated soybean lines.

Ecological Characteristics of Color - Soybean Collections (유색콩 수집종의 주요 생태적 특성)

  • This study was carried out to improve utility value as soybean genetic resources and to obtain basic information for color-soybean breeding. The 146 local color soybean lines collected in Kyongbuk province were investigated major agronomic characteristics. Seed coat-color showed seven different types which were black, white mottled on black, white mottled on brown, black saddle on green, green, raddish brown and brown. Major color types were black and green colors. The 90 percent of collected lines were purple in flower color. The 86 lines were green in color of cotyledonary part and 64 lines yellow. Great variations were observed in days to flowering and days from flowering to maturity, 51 to 81 days and 75 to 103 days, respectively. But days to maturity was 150 to 159 days and had smaller variation than days to flowering and days from flowering to maturity in all lines except those with white mottled on black (123 days). The 100 seed weight ranged below 10g to over 36g. Black seed color with green embryo seed was heavier seed weight than black seed color with yellow embryo seed. Seed shape of all collected lines was ellipsodial and average ratio of length: width: thickness of seed was 1:0.87:0.69. Positive correlation coeffieients were obtained among seed length, width, thickness and 100 seed weight.

Diversity in Lipid Contents and Fatty Acid Composition of Soybean Seeds Cultivated in Korea (국내 유통 콩의 지방함량 및 지방산 조성변이)

  • The 117 soybeans seeds were collected from the nine provinces of Korea, and protein and lipid contents, and fatty acid composition levels were evaluated to investigate their relationship. The 100-seed weights of the black soybeans were varied $27.7{\sim}33.1g$, while the 100-seed weight of yellow soybeans were varied $24.6{\sim}36.6g$. Protein and lipid content of the 117 soybean seeds was 38.3% and 17.8%, respectively Protein contents of the 59 black soybean seeds (38.6%) were significantly higher than those of the 58 yellow soybean seeds (37.9%). However, lipid contents of the black soybean seeds (17.6%) were lower than those of the yellow soybean seeds (18.1%). Linoleic and oleic acid composition levels of the 117 soybean seeds were 53.75% and 22.08%. Unsaturated fatty acid levels of soybean seeds showed a statistically significant variability among the nine provinces of Korea, however, the differences were not found in the linoleic (18:2) and oleic acid (18:1) levels. Therefore, it was considered that the significant variability of unsaturated fatty acid were mainly due to the variations of linolenic acid (18:3) level. The composition levels of linoleic, oleic, palmitic (16:0), and linolenic acid in the yellow soybean seeds were 53.43%, 22.73%, 12.23%, and 8.24%, while those of the black soybean seeds were 54.13%, 21.48%, 12.47%, and 8.31%. Obtained results suggested that fatty acid composition levels were varied and possibly influence by the phenotype of seed coat colors. Oleic acid, mono-saturated fatty acid, showed the most remarkable variability between yellow and black soybean seeds, and the composition levels were higher in the yellow soybean seeds. Relationship between unsaturated fatty acid levels and 100-seed weights in the yellow soybean seeds showed a negative correlation (r=-0.513, P<0.01), but no relationship $(r=0.154^{ns})$ was observed in the black soybean seeds.

Storability of Soybean Lines for Maintanance of Germplasm (대두유전자원의 안전보존을 위한 계통간 발아수명검정)

  • To develop a long term storage technique of soybean germplasm, fifty lines of soybean seeds were kept in storage room controlled by air-conditioner and dehumidifier at KAERI. The storage room was kept below 25$^{\circ}C$ of temperature and 60% relative humidity. Germinability of stored seeds was evaluated at every three or six months. On the other hand, the newly harvested seeds of the lines were accerelated aging at 40$^{\circ}C$ and 90% of relative humidity, and compared with non-accerelated seeds. The average germinability at the starting time of storage was 97% but the germinability was decreased to 62% at 42 months after storage in the storage room. The germinability seemed to be affected by seed size and seed coat color. The germinability of small seed lot tended to be higher than that of large seed lot, and the germination response in longer term of storage was greater than those of shorter term of storage. The accerelated aging test for 7 days proved to be a useful technique to pretest germinability of soybean seeds for assaying the relative storage potential.

Influence of Seed-filling Temperature on the Seed Quality and Water Soaking Properties of Soybean (등숙온도가 콩의 품질 및 수분흡수 특성에 미치는 영향)

  • Korean soybean varieties, 'Seonyu' and 'Hwangkeum' were planted in 2012, and three temperature gradient, Tc($19.8^{\circ}C$, ambient temperatured), $Tc+1.7^{\circ}C$, and $Tc+2.5^{\circ}C$, were artificially created by controlling the green house system during seed filling period. Mature seeds that developed under these conditions were analyzed for variances in physicochemical properties. The 100-seed weight and seed-coat ratio of soybean were decreased, but small seed rate was increased by high temperature during seed filling period. Protein content was increased, but oil content was decreased significantly with increasing the seed filling temperature. The decrement of carbon to nitrogen ratio (C/N), and the increment of monosaccharide, fructose and sucrose, in seeds explained that carbohydrate assimilation during seed filling was restricted by high temperature. Rapid increments of seed volume and weight were observed in the seeds of high seed filling temperature, but as soaking time increased the highest values were observed in the seeds of ambient seed filling temperature. The 100-seed weight and seed-coat ratio of soybean were closely related not only to the increment of soaking volume and weight, but also the increments of total dissolved solids (TDS) and electro conductivity (EC). Whereas protein content and C/N ratio showed less relationship with the soaking properties, but they had a positive correlation with TDS and EC. From the results, it was considered that high values of TDS and EC in the seeds of high temperature were mainly due to the incomplete conversion of assimilates into storage compounds. However, sugar content showed less influence on the soaking properties and the values of TDS and EC.

Comparison of agronomic characteristics of colored soybean landraces and selection of useful genetic resources

  • This study was conducted to compare the agronomic traits of well-known colored native soybean germplasms. Recently, we are increasingly interested in colored as various functional ingredient of soybeans have revealed. We used a total of 396 soybean genetic resources, consisting of ten "Seonbijabikong", 110 "Jyinunikong", 276 "Seoritaekong". We sowed on 10th June 2018 at the field of Nongsaensmyeongro in Jeonju city. The average number of days to flowering, days to maturing and days to growth of colored soybean were 53, 84 and 136 days, respectively. Days to flowering of "Seonbijabikong" were ranged from 41 to 50 days with an average 48 days, those of "Jyinunikong" were ranged from 39 to 72 days with an average 52 days. Days to flowering of "Seoritaekong" were ranged from 35 to 63 days with an average 54 days, which were earlier in "Seonbijabikong", and similar with "Seoritaekong" and "Jyinunikong". Days to growth of "Seonbijabikong" were ranged from 125 to 137 days with an average 132 days, those of "Jyinunikong" were ranged from 91 to 144 days with an average 130 days and those of "Seoritaekong" were ranged from 99 to 150 days with an average 139 days. they were shortest in "Jyinunikong" and longest in "Seoritaekong". The distribution of maturity period was from 6th September to 5th November. The maturity period was as early as September and yields were more than 100g per plant, which were all three accessions(IT161905, IT162602, IT269617), "Seoritaekong". They would be useful as breeding materials of colored soybean with early maturity. The 100-seed weight is important characteristics that distinguish the usage of soybeans. "Seoritaekong" and "Seonbijabikong" have large seed characteristics for cooking with rice, "Jyinunikong" has small seed it for medicine. The average 100 seed weight was 35.0g of "Seonbijabikong", 30.8g of "Seoritaekong" and 13.4g of "Jyinunikong", respectively. As for seed coat lust, the ratio of dull was as high as 100% of "Seonbijabikong" and 91% of "Seoritaekong", that of shiny was as high as 77% of "Jyinunikong". Cotyledon color of "Seonbijabikong" were all yellow, that of "Seoritaekong" were 94% of green. The other name of "Seoritaekong" is "Sokcheong", which means that cotyledon color is green. Therefore if cotyledon color of Seoritaekong is not green, it might be misidentified. In the future, we will increase the utilization through evaluation of functional component such as isoflavones and anthocyanins of colored soybean landraces.

Change of asparagine content in soybean sprouts by variety, root growth, and cultivation period (콩나물의 품종, 뿌리발생 및 재배일수에 따른 asparagine 함량 변이)

  • The varietal difference, effect of root amounts and cultivation duration on the asparagine content of soybean sprouts, which is known to have the excellent detoxifying effect of ethanol, were investigated for developed varieties and indigenous sprout lines to establish cultivation methods of increasing the asparagine content and to develop soybean varieties having high asparagine content. Some of the research results obtained are summarized as follows; 1. Range and mean of asparagine content of 174 germplasm were 0.38~1.67% and 0.99%, respectively, on fresh weight basis. 2. Developed sprout-soybean varieties showed somewhat higher asparagine content of 1.29% than that of indigenous sprout lines of 0.96% on fresh weight basis. 3. No significant difference in asparagine content among the seed size groups was recognized though the highest asparagine content, 1.02% on fresh weight basis was observed in the seed size of 8.1~10.0g/100 seeds. 4. Among the seed coat color groups, soybean of brown seed coat color showed the highest asparagine content (1.15%) on fresh weight basis. No difference was observed among other groups of seed coat color. 5. Range of asparagine content of 174 varieties was 4.08~6.24% on dry weight basis. Soybean varieties that showed high asparagine content were Dawonkong, Sobacknamulkong, Sowonkong, and Somyungkong, with the contents of 6.24%, 6.21%, 5.95%, and 5.85%, respectively. 6. Amount of roots which have the highest asparagine content out of sprout parts was greatly increased up to 10 days of sprout cultivation. 7. Highly significant difference in asparagne content of sprout was recognized between those grown for five and ten days, with the asparagine content of 0.68% and 1.21%, respectively, on fresh weight basis. 8. Asparagine content of hypocotyle part was also highly proportional to days to cultivation; it increased from 2.91% at 2 days of cultivation to 15.68% on fresh weight basis at 14 days of cultivation.

A New Soybean Cultivar, "Galchae" for Sprout with Brown Seed Coat, Small Seed Size and High Sprout Yielding (갈색종피.소립 고수율 나물용 콩 신품종 "갈채")

  • A new sprout-soybean cultivar, "Galchae" was developed at the National Institute of Crop Science (NICS) in 2008. Galchae was selected from a cross between YS1287 and Jinju#1. The preliminary, advanced, and regional yield trials to evaluate the performance of Iksan 64 were carried out from 2005 to 2008. This cultivar has a determinate growth habit with purple flower, brown pubescence, brown seed coat, brown hilum, rhomboid leaflet shape and small seed size (8.4 grams per 100 seeds). The maturity date of "Galchae" is 6 days later than the check variety, "Dawon". It has good seed quality for soybean-sprout and resistance to lodging. This cultivar has resistance to soybean mosaic virus (SMV) and necrotic symptom (SMV-N). The average yield of "Galchae" was 2.51 ton per hectare in the regional yield trials (RYT) for double cropping carried out for two years from 2007 to 2008.

Black soybean anthocyanins attenuate inflammatory responses by suppressing reactive oxygen species production and mitogen activated protein kinases signaling in lipopolysaccharide-stimulated macrophages

  • BACKGROUND/OBJECTIVES: Oxidative stress is closely related with inflammation and development of many diseases. Black soybean seed coat contains high amount of anthocyanins, which are well-known for free radical scavenging activities. This study investigated inflammatory response and action mechanism of black soybean anthocyanins with regard to antioxidant activity in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. MATERIALS/METHODS: RAW 264.7 cells were treated with anthocyanins extracted from black soybean seed coats in a concentration range of 12.5 to $100{\mu}g/mL$. The production of reactive oxygen species (ROS), secretion of pro-inflammatory mediators and cytokines, and the signaling in the mitogen activated protein kinases (MAPKs) pathway were examined. RESULTS: Black soybean anthocyanins significantly decreased LPS-stimulated production of ROS, inflammatory mediators such as nitric oxide (NO) and prostaglandin $E_2$, and pro-inflammatory cytokines, including tumor necrosis factor ${\alpha}$ and interleukin-6, in a dose-dependent manner without cytotoxicity (P < 0.001). Black soybean anthocyanins downregulated the expression of inducible NO synthase and cyclooxygenase-2 in LPS-stimulated RAW 264.7 cells (P < 0.001). Moreover, black soybean anthocyanins inhibited LPS-induced phosphorylation of MAPKs, including extracellular signal-regulated kinase, c-Jun N-terminal kinase, and p38 (P < 0.001). CONCLUSION: These results suggest that black soybean anthocyanins exert anti-inflammatory activity by inhibiting ROS generation and subsequent MAPKs signaling, thereby inhibiting inflammatory responses.

Agronomic Traits of Soybean Breeding Lines with Low Stachyose and Raffinose Contents (Stachyose 및 Raffinose 저함량 콩 선발계통의 농업적 형질)

  • Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. Raffinose and stachyose are the main antinutritional factors in soybean seed. They are carbohydrates belonging to the raffinose family of oligosaccharides, which are not readily digested in humans and cause flatulence or diarrhea. The genetic reduction of the raffinose and stachyose contents in mature soybean seeds will improve the nutritional value of soybean. The objective of this research was to evaluate agronomic traits with 10 $F_6$ strains selected from breeding populations derived from a cross among seven parents. The contents of raffinose and stachyose in mature seeds were detected by high-performance liquid chromatography. Agronomic traits such as flower color, flowering date, harvesting date, lodging, plant height, seed coat color, hilum color, 100 seed weight, and yield were evaluated. Ten intermediate parents showed low raffinose and stachyose contents. The intermediate parent 883-1 had a small seed size, six intermediate parents (15A1, 15D1, RS-5, RS-33, RS-64, and RS-70) had a medium seed size, and two intermediate parents (14G20 and RS-21) had a large seed size. The intermediate parent RS-21 had a black seed coat and a green cotyledon. Four intermediate parents (883-1, 14G20, RS-5, and RS-21) had elite agronomic traits. The new intermediate parents developed through this study will be used to develop improved soybean cultivars with low contents of raffinose and stachyose.
