A long-term study initiated in 1989 at San-born Field, Columbia, Missouri, was designed to evaluate the affect of environmental factors, nitrogen application, and crop rotation on soybean (Glycine max [L.] Merr.) seed composition. Soybeans were grown as part of a four- year rotation which included corn (Zea maize L.), wheat (Triticum aestivum L.), and red clover (Trifolium pratense L.). Results from soil tests made prior to initiation of the study and subsequently every five years, were used to calculate application rates of nitrogen, phosphorus, and potassium necessary for target yield of pursuant crops. In the experimental design, nitrogen was applied to one-half of the plot on which the non-leguminous crop, either corn or wheat was grown. Analysis of soybean seed by near infrared reflectance spectroscopy collected over an 11-year period revealed a linear increase in protein and decrease in oil content. Application of nitrogen fertilizer to non-leguminous crops did not have an apparent effect on total protein or oil content of subsequent soybean crop. Analysis of soybean seed proteins by sodium dodecyl sulfate polyacrylamide gel electrophoresis in conjunction with computerassisted densitometry revealed subtle changes in the accumulation of seed proteins. Immunoblot analysis using antibodies raised against the $\beta-subunit$ of $\beta-conglycinin$ showed a gradual increase in the accumulation of the 7S components during successive years of the experiment. A linear increase in temperature and decrease in rainfall was observed from the onset of data· collection. Higher temperatures during the growing season have been linked to increased protein and diminished oil content of soybean, thus changes observed in this study are possibly related to climatic conditions. However, crop rotation and subsequent changes in soil ecology may contribute to these observed changes in the seed composition.

Pumpkin (Cucurbita moschata Duch.) seed is rich in protein and sulfur-containing amino acids. Tofu is a protein gel made from soybean, which is rich in lysine but lacking in sulfur-containing amino acids. This study was conducted to investigate the use of pumpkin seeds in tofu manufacture and to determine its quality and texture characteristics. Soybean was substituted with pumpkin seed to obtain pumpkin seed tofu at the following ratios: 10%, 30%, 50%, and 70% (P10, P30, P50 and P70). Tofu manufactured only with soybean was used as a control (Con). The higher rate of pumpkin seed substitution significantly decreased the moisture content and yield rate (p<0.05). In contrast, pH value and turbidity were significantly increased with the increase in the amount of pumpkin seed (p<0.05). The L-value (81.74~79.04), a-value (-0.19~-3.89) and b-value (12.40~9.84) of samples significantly decreased with the amount of pumpkin seed (p<0.05). No significant difference in syneresis was found among the samples (p<0.05). The hardness tended to decrease with the increase in the amount of pumpkin seed. The microstructure analysis revealed that the pore size of pumpkin seed tofu was smaller than that of Con. These results suggest that the pumpkin seed protein is a useful ingredient in the manufacture of tofu. Increasing the pumpkin seed substitution levels improves the texture of tofu.
Excessive soil water in paddy field induces growth losses during the vegetative stages of soybean plants. Our objectives were to know growth responses of soybean as affected by the level of ridge heights. A field experiment was conducted at the Research Farm of Korea University near Seoul in 1996. Ten cultivars of soybean (Glycine max (L.) Merrill) were planted at the ridge heights of 10, 30, and 50cm in paddy soil on 27 June. Total dry matter accumulation at the 10cm ridge height was severely decreased until the growth stage of R5 as compared to the ridge heights of 30 and 50cm, and this decrement was mainly due to both reduced leaf and stem dry weights. However, seed dry weight was not significantly decreased at the lower ridge height with the increment of harvest index. Cultivar differences were found on seed dry weight, crop growth rate from R1 to R5, and leaf/stem ratio of R5. From the above results, soybean growth responses to excessive soil water in paddy field were different between seed filling stage and late vegetative to early reproductive growth stages.

Natto is a soybean fermented food from Japan, which is made from steamed soybean, Bacillus natto and water. Demand of Natto has increased recently because it does not have smell compared to Cheonggukfang, which is Korean traditional fermented food. Currently, Pungsannamulkong is the most commonly used in Korea for Natto. Four candidate varieties of soybeans were investigated and compared in terms of Agronomic Traits, quality characteristics, hard seed rate, and water absorption rate in order to determine more suitable Korean soybean variety than pungsannamulkong. 'Haewon' had higher yield than other three varieties in Goesan-gun and Jinan-gun. The infected seed rate which affects soybean processing is higher in pungsannamulkong. 'Haewon' showed low 100 seed weight with 8.6 g in Goesan-gun and 9.5 g in Jinan-gun, which was the smallest of four soybean varieties. The water absorption rate was higher in 'Haewon' than in pungsannamulkong which is desirable characteristics for Natto process. The hard seed rate of pungsannamulkong was 6~7%, which is considered to be unsuitable to Natto process. The yield of Natto was significantly different for each variety. 'Haewon' showed the highest amino nitrogen content with 575.0 mg%. These results suggested that 'Haewon' can be considered as suitable candidates for yield and quality of Natto compared to pungsannamulkong.
Soybean seeds contain many biologically active secondary metabolites, such as proteins, saponins, isoflavones, phytic acids, trypsin inhibitors and phytosterols. Among them, saponins in soybeans have attracted considerable interest because of their health benefits. Soyasaponin A and B are the most abundant types of saponins found in soybeans along with soyasapogenol (aglycone), which is a precursor of soyasaponin. The main purpose of this experiment was to determine the concentration of soyasapogenol in soybean seeds and sprouts as a function of seed size, usage, seed coat color and seed cotyledon color. The 79 Korean soybean varieties were cultivated at Yesan of Chungnam in 2006 for the analysis of soyasapogenol using HPLC with Evaporative Light Scattering Detection (ELSD). The total average concentration of soyasapogenol was $1313.52{\mu}g\;g^{-1}$ in soybean seeds and $1377.22{\mu}g\;g^{-1}$ in soybean sprouts. Soybean sprouts were about 5% higher than soybean seeds in average total soyasapogenol concentration. In the process of sprouting, the average soyasapogenol A content decreased by approximately 1.6%, but soyasapogenol B and total soyasapogenol increased by 8.31% and 4.88%, based on the content of soybean seeds. When classified according to the size of seeds, the total soyasapogenol concentration of soybean seeds were not significantly different (p<0.05) On average, small soybean seeds were increased by as much as $103.14{\mu}g\;g^{-1}$ in sprouting process. As a function of the use of the seeds, The total soyasapogenol in soybean seeds were significantly different (p<0.05). While, the soybean sprouts were not significant different (p<0.05). Altogether, sprout soybean seeds show the greatest change in content during the germination process. When seeds with different coat colors were compared, the total soyasapogenol concentration of soybean with yellow seed coats ($1357.30\mu g\;g^{1}$) was slightly higher than that of soybean with black ($1260.30{\mu}g\;g^{-1}$) or brown ($1263.62{\mu}g\;g^{-1}$) seed coats. For the color of the cotyledon, the total soyasapogenol concentration was significantly increased in green cotyledon during the germination and seedling process. The results of this study suggest the functional characteristics of soybeans through quantitative analysis of soyasapogenol. In addition, the concentration of soyasapogenol exhibited a change during the germination process, which was evaluated by the nutritional value of the soybean sprouts.

The antimicrobial effect of mixed solutions of grapefruit seed extract(250ppm) and ascorbic acid(1%) on the spoilage microorganisms such as Staphylococcus epidermis, Escherichia coli, Pseudomonas syringae, Candida albicans and Corynebacterium xerosis isolated from the spoiled soybean sprouts were investigated. Cell wall and membrane were partially destroyed and the contents of the destroyed cell were exuded after treatment. Packages with 30 m cast polypropylene(CPP), 16 m polyolefin(RD 106) and 10 m high density polyethylene(HDPE) were applied for soybean sprouts dipped in mixed solutions respectively. Oxygen and carbon dioxide concentration inside packages were dependent on the kind of films during storage at 5oC. The antimicrobial activity of mixed solutions was maintained for 5 days at CPP package. Package with HDPE showed a severe browning than the others after 5 days. Ascorbic acid content of mixed solution treatment was higher than that of control for each package.
The experiment was carried out to study the variations and geographical distribution of $\beta$-amylase isozyme by isoelectric focusing (IEF) within Korean, Chinese and Japanese soybean land races. In pH 3-10 gel of IEF, the amylase of soybean accessions was separated into low pI group isozymes (TEX>${$Sp_1$}^b$) and high pI group isozymes(${$Sp_1$}^a$). In pH4-6.5 gel, isoelectric points were at 5.07, 5.15, 5.25, 5.40, and 5.94, and h, j, and k bands also were found. The distribution of Sp$_1$$^{a}$ allele (high pI type) was 29.3% in soybean accessions from Korea, 10.1 % in those from China, and 6.9% in Japanese accessions. The percentage of ${$Sp_1$}^a$) allele was the highest in soybean accessions from Kyungsang province (35 %) in Korea, then central China (32 %) in China, and Honshu (10%) in Japan.
Soybean [Glycine max (L.) Merr.] seed is an important dietary source of protein, oil, carbohydrates, isoflavones, and other nutrients for humans and animals. But, antinutritional factors in the raw mature soybean are exist. Kunitz trypsin inhibitor (KTI) protein and stachyose are main antinutritional factors in soybean seed. The genetic removal of the antinutritional factors will improve the nutritional value of soybean seed. The objective of this research was to breed a new yellow soybean strains (rs2rs2titi genotype) with the traits of lacking of KTI protein and low content of stachyose. Breeding population was developed from the cross of "Jinyangkong" and 15G1 parents. Presence or absence of KTI protein was detected based on Western Blot technique. Content of stachyose in mature seed was detected by HPLC. Total four new strains (603-1, 603-2, 625, and 694) with KTI protein free and low content of stachyose were selected. Four strains (603-1, 603-2, 625, and 694) have yellow seed coat and hilum. Plant height of 603-1 strain was 65 cm and 100-seed weight was 29.2 g. Plant height of 603-2 strain was 66 cm and 100-seed weight was 26.2 g. Plant height of 625 strain was 64 cm and 100-seed weight was 27.1 g. Content of stachyose for four new strains was 3.0~3.50 g/kg. Four strains selected in this research will be used to improve new yellow soybean cultivar with KTI protein free, and low content of stachyose.
This experiment was carried out to examine ecological response and soybean quality as affected by environmental cultivation for producing high seed quality in domestic soybean variety. The results are as follows: Under equal cumulative temperature condition, soybean plants grown in Muju showed longer days to flowering, which was an effect of the long day-length on high latitudes, and longer duration of reproductive stage as a result of low temperature within that period. Considering apparent seed quality, 100 seed weight of soybeans grown in Muju was heavier than Miryang. Ratio of seed crack and disease-damaged seeds was lower in Muju, and these parameters decreases as planting was delayed. The protein contents did not show significant difference in terms of altitude and planting date, however, crude oil contents were higher in Miryang. An opposite trend was observed in C18:1 and C18:3. In the fatty acid composition, the proportion of C18:1 decreased as seeding date was delayed, and was higher in Miryang. Opposite observations were obtained from C18:3. The anthocyanin contents were highest on June 10 planting and higher in Muju than in Miryang. Isoflavone content was higher as seeding date was delayed and is similar accross seeding dates in Muju. As a summary, for high seed quality production the optimum planting date was June 10, and Muju was more suitable region than Miryang.
Soybean seeds deteriorate even before harvest, especially under high temperature and high relative humidity conditions. This study was conducted to determine the effect of harvest date on seed quality in early maturing soybean cultivars. Soybean cultivars used in the experiment were Damyang-native, having small seed with yellow color, and Yuhsuzumi, having large seed with green color. The length of physiological maturity to grain harvest was 24 days in Yuhsuzumi and 8 days in Damyang-native. The moisture contents of seed were 25.3 % in Yuhsuzumi and 14.5 % in Damyang-native cultivar when pod reached its mature pod color. The seed color of Yuhsuzumi started to turn yellow, and its germination rate was greatly decreased when harvest was delayed after maturity, However, Damyang-native cultivars produced seed of high quality regardless of delayed harvest.
