GC-NPD와 GC-MSD를 이용한 농산물 중 Isotianil의 공정분석법 개발 (Development of analytical method for Isotianil in agricultural commodities using GC-NPD and GC-MSD)

  • Isotianil은 쌀 도열병을 방제하기 위한 새로이 개발된 식물활성 살균제로 다른 식물활성제에 비해 소량으로도 장시간의 효과를 나타내는 특징을 가지고 있다. 신규 농약 Isotianil에 대한 기준은 국내의 경우 2010년 쌀에 대하여 0.1 mg/kg으로 최초 설정되었으며, 이에 따른 식품 중 안전관리를 위해 Isotianil을 분석하기위한 검사법을 개발하였다. 검체 일정량에 acetonitrile을 가하고 균질화하여 대상농약을 추출하였으며, 액-액분배하고 florisil 카트리지를 이용한 고체상 추출 후 GC-NPD 및 GC-MSD로 분석하였다. 분석결과 농산물에 의한 간섭물질은 없었으며, 대표 농산물 현미 등 6 종에 대한 Isotianil의 분석 시험법의 정량한계와 직선성은 각각 0.05 mg/kg과 0.999($r^2$)이었다. 회수율은 대표 농산물에서 70.0~103.9%이었으며, 분석오차는 10% 미만으로 CODEX 잔류분석 기준에 적합하였다.

손바닥 선인장의 항산화활성 및 분말 첨가 쿠키의 품질특성 연구 (The Antioxidant Activity of Korean Cactus (Opuntia humifusa) and the Quality characteristics of Cookies with Cactus Powder added)

  • In order to better understand the quality of Korean cactus, Opuntia humifusa, as the functional foodstuffs, it's seeds, stems and fruits were analyzed. In addition, we performed quality evaluation on cookies manufactured with cactus powder added. The results showed the soluble nitrogen-free component levels in the cactus to be very high. The highest component levels were crude fiber(48.75%) and crude fat(7.38%) in the seeds, crude ash(15.39%) in the stems, and moisture(11.26%) and crude protein(8.79%) in the fruits. The antioxidant activities of the cactus were evaluated based on peroxide values(POV), thiobarbituric acid reactive substances(TBARS value), and electron donating ability(EDA) using the DPPH method. The POVs for soybean oil containing a 0.05% concentrated water extract from each part of the cactus revealed a remarkable antioxidant effects on peroxidation as storage time increased. The TBA values were 63.65% in BHT, 60.67% in the stems, 57.31% in the fruits and 49.16% in the seeds. The EDAs were 30.26% in the stems, 25.89% in the fruits, and 25.65% in the seeds. The total polyphenol contents of the water soluble materials from the cactus were 3.14 g/100 g in the fruits, 2.93 g in the stems and 2.11 g in the seeds. For the cookies, as the amounts of fruit and stem powder increased, the spreadness was significantly lowered, but moisture content was increased compared to the control. The color degree of the cookies changed from dim to intense as more fruit powder was added, resulting from a decrease in the L- and b-values and a significantly increased a-value. With the addition of stem powder, the L- and a-values revealed no significant differences, while the b-value was lowered compared to the control as more stem powder added. For texture changes of the fruit cookies, there were significant differences in hardness, gumminess, cohesiveness, springiness and brittleness. The stem cookies, however, showed only differences in hardness, gumminess, and brittleness, and not in cohesiveness and springiness. Finally, an overall sensory preference was found with the 1% fruit powder and 3% stem powder additions.

Growth Performance, Carcass and Meat Characteristics of Black Goat Kids Fed Sesame Hulls and Prosopis juliflora Pods

  • Finding alternative feeds, such as sesame hulls and Prosopis juliflora species can attenuate difficulties of feed shortage and reduce the cost of animal feed in arid and semi-arid countries. Thirty-two Black male kids with similar initial weights (BW = $16.7{\pm}0.80\;kg$) and $120{\pm}5\;d$ of age, were used to evaluate the effect of replacing barley grains and soybean meal with Prosopis juliflora pods (PJP) and sesame hulls (SH) on growth performance, digestibility and carcass and meat characteristics. Kids were equally divided into four dietary treatment groups for an 84-d fattening period. Treatment diets had similar crude protein (CP) and metabolizable energy (ME). The treatment groups were: (T1) no PJP nor SH, (T2) 10% PJP and 20% SH, (T3) 15% PJP and 15% SH, and (T4) 20% PJP and 10% SH. A tendency was detected (p<0.08) for dry matter (DM), crude protein (CP), organic matter (OM) and neutral detergent fiber (NDF) intakes that were greater for T2 than T3 and T4 while T1 was not different from all other treatment groups. Ether extract (EE) intake was the greatest (p<0.05) for T2 and the lowest for T1. Acid detergent fiber (ADF) intake was greater (p<0.05) for T2 than T1 while T3 and T4 were intermediate. Final live weight, average daily gain (ADG) and feed conversion ratio (FCR) were comparable among different treatment groups. Digestibility of DM, OM, CP, NDF and ADF were similar among all treatment groups, however, EE digestibility was the lowest (p<0.05) for T1 when compared to other treatments. In addition, nitrogen intake, nitrogen in urine and retained and retention percentages were similar among all treatment groups. However, N loss in feces was higher (p<0.05) for T2 than T3 and T4 while T1 was intermediate. No differences were observed among treatment groups with respect to fasting live weight, hot and cold carcass weights, dressing-out percentages, mesenteric fat, visceral organs, carcass cuts percentages and carcass linear dimensions. No differences were also observed for dissected loin, leg, rack and shoulder tissues except in the total bone % for loin cuts and in the meat to bone ratio for rack cuts. T3 has the greatest total bone % and the lowest meat to bone ratio when compared to all other treatment groups. No differences were observed between treatment groups in all quality characteristics of the longissimus muscle. The present study demonstrates the potential of using PJP and SH for growing kids without adverse effects on growth performance, carcass characteristics and meat quality.

꽃송이버섯(Sparassis crispa)의 배양적 특성 (Cultural Characteristics of Cauliflower Mushroom, Sparassis crispa)

  • 본 시험은 국내외에서 수집한 꽃송이버섯균 8균주에 대하여 균의 배양적 특성 및 균사체 대량배양 조건을 밝히고자 몇 가지 실험을 하였다. 꽃송이버섯균의 고체배양시 HBA(벌꿀바나나추출배지)에서 $53.0{\pm}5.3mm{\cdot}15$$^{-1}$로 균사생장이 빨랐다. 최적배양온도는 $23^{\circ}C$이었으며 pH는 6.0이 었고, 탄소원으로 fructose, 질소원은 glutamine을 잘 이용하였다. 꽃송이버섯균의 균사체 대량배양을 위한 균주로는 통기식 액체배양시 균사체 생산량이 많은 ASI150010을 선발하였다. 액체배양용 추출배지 재료인 대두박추출배지 ($15\;g{\cdot}{\ell}^{-1}$)에서 30일 동안의 균사량은 1.85g으로 감자추출배지 ($200\;g{\cdot}{\ell}^{-1}$)의 1.05g보다 1.7배가 많았다. 곡물을 이용한 꽃송이버섯 균사체 배양시 수분조절 방법으로 현미는 1일간 침수 처리에서 균사체 생장이 빨랐으며, 밀과 보리는 삶는 방법이 좋았다. 그리고 밀은 보리보다 균배양이 빨라서 좋았다.

Effects of particle size and adaptation duration on the digestible and metabolizable energy contents and digestibility of various chemical constituents in wheat for finishing pigs determined by the direct or indirect method

  • Objective: This experiment was conducted as a $3{\times}2{\times}2$ factorial design to examine the effects of particle size (mean particle size of 331, 640, or $862{\mu}m$), evaluation method (direct vs indirect method) and adaptation duration (7 or 26 days) on the energy content and the apparent total tract digestibility (ATTD) of various chemical components in wheat when fed to finishing pigs. Methods: Forty-two barrows ($Duroc{\times}Landrace{\times}Yorkshire$) with an initial body weight of $63.0{\pm}0.8kg$ were individually placed in metabolic cages and randomly allotted to 1 of 7 diets with 6 pigs fed each diet. For the indirect method, the pigs were fed either a corn-soybean meal based basal diet or diets in which 38.94% of the basal diet was substituted by wheat of the different particle sizes. In the direct method, the diets contained 97.34% wheat with the different particle sizes. For both the direct and indirect methods, the pigs were adapted to their diets for either 7 or 26 days. Results: A reduction in particle size linearly increased the digestible energy (DE) and metabolizable energy (ME) contents as well as the ATTD of gross energy, crude protein, organic matter, ether extract (EE) and acid detergent fiber (ADF) (p<0.05), and had a trend to increase the ATTD of dry matter of wheat (p = 0.084). The DE, ME contents, and ATTD of gross energy, crude protein, dry matter and organic matter were higher (p<0.05) when determined by the direct method, but the ATTD of ADF, EE, and neutral detergent fiber were higher when determined by the indirect method (p<0.05). Prolongation of the adaption duration decreased the ATTD of neutral detergent fiber (p<0.05) and had a trend to increase the ATTD of EE (p = 0.061). There were no interactions between particle size and the duration of the adaptation duration. The ATTD of EE in wheat was influenced by a trend of interaction between method and adaptation duration (p = 0.074). The ATTD of ADF and EE in wheat was influenced by an interaction between evaluation method and wheat particle size such that there were linear equations (p<0.01) about ATTD of ADF and EE when determined by the direct method but quadratic equations (p = 0.073 and p = 0.088, respectively) about ATTD of ADF and EE when determined by the indirect method. Conclusion: Decreasing particle size can improve the DE and ME contents of wheat; both of the direct and indirect methods of evaluation are suitable for evaluating the DE and ME contents of wheat with different particle sizes; and an adaptation duration of 7 d is sufficient to evaluate DE and ME contents of wheat in finishing pigs.

청국장 발효과정 중 항고혈압성 peptide의 생산 및 분리 (Production and Separation of Anti-hypertensive Peptide during Chunggugjang Fermentation with Bacillus subtilis CH-1023)

  • 한국 전통장류의 기능성 탐색 연구의 일환으로 청국장 발효과정 중 고혈압을 유발하는 angiotensin converting enzyme(ACE)의 저해 peptide를 분리하고 저해효과를 검토하였다. 청국장은 Bacillus subtilis CH-1023을 이용하여 제조하였고, $20^{\circ}C,\;30^{\circ}C,\;40^{\circ}C,\;50^{\circ}C,\;60^{\circ}C$에서 $0{\sim}72$시간동안 배양하면서 protein양, protease activity, ACE 저해율을 측정하고 저해활성을 가지는 peptide를 정제 후 아미노산조성을 분석하였다. 청국장의 발효시간이 경과함에 따라 protein 함량이 증가하여 $40^{\circ}C$, 60시간에서 최대를 나타낸 후 감소하였으며, 발효시간에 따른 protease activity와 ACE 저해율은 $40^{\circ}C$, 60시간에서 최대로 나타난 후 점차 감소하였다. 따라서 Bacillus subtilis CH-1023을 이용한 청국장 제조는 $40^{\circ}C$에서 60시간 배양이 최적의 배양조건이었다. 최적 발효조건에 따라 청국장을 제조한 후 20 mM sodium phosphate buffer(pH 7.0)를 가해 추출한 추출물을 Amicon YM-3 membrane filtration과 Sephadex G-10, G-25를 이용한 gel filtration으로 부분정제 하였다. 또한 정제한 peptide는 첨가함량이 높아질수록 저해활성은 높게 나타났으며, 0.5 mg 정도의 peptide 함량으로 94.3%의 저해율을 나타내었다. 정제한 peptide의 아미노산 조성은 histidine, alanine, phenylalanine 함량이 높은 것으로 나타났다.

천일염 함유 청국장의 항산화효과 (Antioxidative Effects of Chungkukjang Preparation by Adding Solar Salt)

  • 본 연구에서는 천일염의 아직 밝혀지지 않은 기능성과 청국장의 효과를 감안하여 천일염으로 제조된 청국장의 기능적 우수성을 평가하기 위하여 Bacillus subtilis DJI 균주를 접종하여 발효시킨 무염청국장, 3% 천일염 첨가 청국장, 3% 정제염 첨가 청국장과 3% 정제염이 첨가되어진 시판청국장의 에탄올, 물 및 헥산 추출물의 항산화력을 비교 검토하였다. 청국장 에탄올, 헥산 및 물 추출 수율은 각각 $10.9{\sim}11.8%$, $15.7{\sim}16.9%$, $18.6{\sim}19.4%$ 범위로 청국장 용매별 간에는 서로 유사한 경향이었으며, 물 추출 수율이 가장 높게 나타났다. 추출물의 DPPH에 대한 전자소거능, 아질산염 소거능, linoleic acid에 대한 과산화물가 및 항산화지수 측정에 의한 항산화효과는 에탄올 추출 시료군이 물 추출 시료군에 비하여 높았다. 청국장 추출물의 전자소거능과 아질산염 소거능은 무염청국장과 3% 천일염 첨가 청국장 추출물이 가장 우수하였으나, 합성항산화제에 비해서는 낮은 효능을 보였다. 청국장 추출물 중 항산화지수는 3% 천일염 첨가 청국장 추출물이 가장 우수하였다. Bacillus subtilis DJI 균주를 접종하여 발효시킨 청국장인 무염청국장, 3% 천일염 첨가 청국장 및 3% 정제염 첨가 청국장이 시판청국장에 비하여 항산화효과가 우수하였으며, 균주를 접종하여 발효시킨 청국장 중 무염청국장과 3% 천일염 첨가 청국장이 3% 정제염 첨가 청국장에 비하여 항산화효과가 다소 우수하였다.

기체크로마토그래피법에 의한 식물성 유지내 유리 지방산의 신속한 스크리닝 (Rapid Gas Chromatographic Screening of Vegetable Oils for Free Fatty Acids)

  • 식물성 유지에 존재하는 유리지방산과 유기산들을 고상추출법으로 분리 농축하여 안정하고 휘발성이 큰 TBDMS 유도체로 전환한 후 GC와 GC-MS로 분석하였다. 고상 추출하기 전 유지를 dichloromethane에 용해시키고 $NaHCO_3$, 포화수용액으로 추출하는 방법이 지방만 회수에 매우 효과적이었으며 TBDMS 유도체화하기 전 유기산들을 triethylammonium염으로 만들어 줌으로써 미량의 휘발성 유기산의 손실을 막을 수 있었다. 대두유, 미강유. 참기름, 들기름 등을 분석한 결과 탄소수가 많은 유리지방산들이 주로 동정되었으며 정제하지 않은 참기름에서는 탄소수가 홀수인 지방산들을 포함한 다양한 유기산들이 확인되었다. 아직 보고된 바 없는 아라키드산과 vanillic acid를 각각 들기름과 참기름에서 발견하였으며 참기름에 존재하는 유리상태의 올레산에 대한 리놀레산의 양적비율은 $1.02{\sim}1.18$로 문헌에 보고된 전체지방산 중의 함량비율인 1.19와 상당히 유사했다. 유리상태로서의 올레산과 리놀레산의 함량비는 참기름의 adulteration 여부를 판정하는 지표로 이용될 수 있을 것으로 예상된다. 유지의 유기산 조성을 단순화된 막대그래프 형태의 RI spectra로 표현한 결과 각 유지의 패턴분별이 매우 용이했다.

목향(Saussurea lappa) 유래 Dehydrocostus lactone의 경엽 접촉 살초 활성 (Foliage Contact Herbicidal Activity of Dehydrocostus lactone Derived from Saussurea lappa)

  • 목향(Saussurea lappa) 뿌리 n-hexane 추출물의 ethyl ether 분획물로부터 경엽 살초 활성을 나타내는 물질로 dehydrocostus lactone (3aS,6aR,9aR,9bS)-3,6,9-trimethylidene-3a,4,5,6a,7,8,9a,9b-octahydroazuleno[5,4-d]furan-2-one)(DHCL)을 분리 동정하였다. DHCL 4,000ppm의 경엽처리 결과, 바랭이, 옥수수 및 콩에 대하여 85% 이상의 엽신 고사율을 보인 반면에 까마중에서는 약 40%의 저해율을 나타내어, 화본과 및 광엽식물 간 선택성 차이는 나타나지 않았으나, 검정식물 초종 간 반응성 차이를 나타내었다. 까마중에서의 반응성 차이는 다른 검정식물과 비교하여 까마중 잎의 비교적 높은 cuticle 함량에 의한 흡수량 감소 때문이었다. 광은 DHCL의 살초활성 발현과 관련이 없었으며, 종자 발아 및 세포 생장 억제 효과는 나타내지 않았다. DHCL 처리에 의한 엽절편의 전도도 증가 및 acetyl-CoA carboxylase의 활성 저해효과로부터 DHCL의 살초 활성 발현은 일정 부분 지방산 생합성 저해에 따른 세포막 약화에 의해 나타날 가능성이 있는 것으로 나타났다.

By-product of Tropical Vermicelli Waste as a Novel Alternative Feedstuff in Broiler Diets

  • Two experiments were conducted to determine physical and chemical properties of vermicelli waste (VW) and effect of VW inclusion levels on growth performance of broilers. In experiment 1, VW samples were randomly collected from vermicelli industry in Thailand to analyze nutritional composition. Vermicelli waste contained 9.96% moisture, 12.06% CP, 32.30% crude fiber (CF), and 0.57% ether extract (EE), as DM basis. The ratio of insoluble:soluble non-starch polysaccharide (NSP) was 43.4:8.9. A total of 120 chicks (6 pens per treatment and 10 chicks per pen) were fed a corn-soybean meal-based diet or 20% VW substituted diet to determine the apparent metabolizable energy corrected for nitrogen retention ($AME_n$) of VW. The $AME_n$ of VW was $1,844.7{\pm}130.71$ kcal/kg. In experiment 2, a total of 1,200 chicks were randomly allotted to 1 of 4 dietary treatments for 42-d growth assay. There were 300 chicks with 6 pens per treatment and 50 chicks per pen. The dietary treatments contained 0%, 5%, 10%, or 15% VW, respectively. All diets were formulated to be isocaloric and isonitrogenous. From 0 to 18 d of age chicks fed VW diets had higher (p<0.001) feed conversion ratio (FCR) compared with those fed the control diet. No difference was observed during grower and finisher phase (19 to 42 d). Chicks fed VW diets had lower relative weight of abdominal fat (p<0.001) but higher relative weight of gizzard (p<0.05) than those of chicks fed the control diet. Increasing VW inclusion levels increased ileal digesta viscosity (p<0.05) and intestinal villus height of chicks (p<0.001). For apparent total tract digestibility assay, there were 4 metabolic cages of 6 chicks that were fed experimental treatment diets (the same as in the growth assay) in a 10-d total excreta collection. Increasing VW inclusion levels linearly decreased (p<0.05) apparent total tract digestibility of DM and CF.
