  • Sixty five soybean varieties were tested to observe varietal response and to get the basic data for resistant variety breeding to soybean cyst nematode (Heterodera glycines I.). They were classified into five groups, from the most resistant to the most susceptible by decreasing rate of seed weight. A few Korean varieties were included in the most resistant group. In general, early maturing varieties were more susceptible. Total plant weight could be used as an important criterion to select a resistant variety to soybean cyst nematode.

First Report of Pasteuria nishizawae Sayre, Wergin, & Nishizawa Attcaking Heterodera glycines in Korea (국내 미기록 콩씨스트선충 기생세균, Pasteuria nishizawae Sayre, Wergin & Nishizawa의 보고)

  • Obligate bacterial parasite attacking Heterodera glycines was firstly found from Chungju soybean field in Korea. Diameters of sporangium and central body were 5.6 ${\mu}{\textrm}{m}$ and 1.9 ${\mu}{\textrm}{m}$ under light microscopy (LM), and 3.9 ${\mu}{\textrm}{m}$ and 1.8 ${\mu}{\textrm}{m}$ under transmission electron microscopy (TEM). Endospore showed cup-shaped with smooth-type exosporium without peg-like thickening in polar area under SEM and TEM. Bacteria completed its life cycle in female of soybean cyst nematode after adhering on cuticle of second-stage juvenile. From these results, the Pasteuria found from Chungju was identified with P. nishizawae.

Resistance of Soybean Cultivars to Heterodera glycines HG type 2.5 in Korea (국내 콩 장려품종의 콩 씨스트 HG type 2.5에 대한 저항성)

  • A total of 75 soybean cultivars developed in Korea was screened against soybean cyst nematode (SCN), Heterodera glycines HG type 2.5. Cysts developed on soybean cultivars ranged from 104 to 624 cysts per pot. There was no resistant cultivar but 'Jangyeopkong', 'Saealkong', 'Miryangkong', and 'Mansukong' were moderately resistant; 33 cultivars were moderately susceptible and the other cultivars were susceptible. 'Jangyeopkong', 'Saealkong', 'Miryangkong', and 'Mansukong' could be recommended for soybean fields infested with SCN until developing SCN resistant soybean.

  • Soybean cyst nematode (Heterodera glycines Ichinohe; SCN) is an important soybean pest and the use of resistant cultivars is the effective method to reduce or eliminate SCN damage. However, breeding for SCN resistance is difficult and expensive by the oligogenic nature of the resistance and genetic variability in the pathogen. Fortunately, SCN resistance loci, rhg1 and Rhg4 are generally accepted as a necessity for the development of resistant genotypes using any source of resistance. In this study, resistance of 44 Korean soybean cultivars to SCN was tested using two molecular markers. Seonheukkong and Pokwangkong were the homozygous to rhg1 locus. Seven cultivars were susceptible to SCN based on Satt309 marker linked rhg1 locus. All Korean cultivars estimated in this study were recessive homozygous to Rhg4 locus and were susceptible in the PCR reaction using primer 548/563 linked to the Rhg4 locus conferring resistance to SCN race 3. Among 44 cultivars estimated, seven cultivars were susceptible to SCN in both Satt309 and primer 548/563 markers. Based on both Satt309 and primer 548/563 markers, there is no resistant cultivar to SCN in Korea. Therefore, SCN resistant cultivars need to be developed in the future. These two markers can be used for improving SCN resistant cultivars.

Heterodera glycines-Induced Syncytium Structures Related to the Nematode Growth and Reproduction in Susceptible Soybean Cultivars

  • The production of soybean cyst nematode (SCN), Heterodera glycines, including female formation and fecundity was much higher in SCN race 14 (R14) than in race 3 (R3) in susceptible soybean cultivars Bragg (intolerant), Lee74 (moderately tolerant), and PI 97100 (highly tolerant). The nematode body was also significantly larger in R14 than in R3 at 20 days after inoculation, but the further nematode growth appeared to be slower in R14 than in R3, resulting in no significant difference between the two races at 30 days after inoculation. Within each race, no significant difference was observed in the growth and reproduction among the soybean cultivars tested. Syncytial areas near the nematode lip regions (infection sites) were measured for each soybean cultivar-SCN race combination. R14 induced significantly larger syncytia than R3. Bragg had relatively larger syncytia than Lee74 and PI 97100, but the difference among the soybean cultivars was minimal or not significantly different. Syncytium occupation in the stelar region differed only between PI 97100 and the other two cultivars, which may be somewhat, but not exactly, related to tolerance levels. Syncytial cytomplasm was degenerated more with R14 and in Bragg than with R3 and in Lee74 and PI 97100, respectively. In light microscopy, degenerated syncytia were characterized by depleted and loose cytoplasm with less plastids than normal-looking (intact) syncytia which had dense syncytial cytoplasm. Electron microscopy revealed that degenerated syncytia contained highly vacuolated cytoplasm with degenerated plastids. The above results suggest that structural characteristics of syncytia may match the nematode growth and reproduction.

Selection of Soybean Cultivars Resistant to Soybean Cyst Nematode, Heterodera glycines (콩씨스트선충(線蟲) 저항성품종(抵抗性品種) 육성(育成)에 관(關)한 연구(硏究))

  • The races of soybean cyst nematode, Heterodera glycines were investigated at 13 localities in Kyungpook, Chungpook and Cheonnam provinces for the resistant soybean variety selection to soybean cyst nematode. Race C, 1, and 5 were distributed and among them race 5 was dominant in Korea. Native soybean 203 lines and 25 introduced soybean varieties were investigated in field where soybean cyst nematode race 5 infested. As the result, the reproduction of soybean cyst nematode in native soybean lines decreased on entry number 18, 21, 36, 38, 41, 42, 43, 45, 55, 59, 68, 72, 75, 93, 94, 95, 98, 123, 131, 136, 146, 170, 174, 187, 190, 191 than others. And then, in pot experiment, entry number 170 showed moderately resistant with 10 cysts per root, while entry number 55 was decreased reproduction a little and the other 25 line appeared susceptibility. In introduced soybean varieties, Peking, Illsoy and PI90763 showed resistance and not reproduction in field and pot experiment. But Custer, Delmar, Dyer and Jackson represented moderately resistant under 10 cysts, while the other 18 varieties were susceptible. In yield analysis, native soybean line 170 showed the same tendency as Peking and PI90763 known for resistant varieties in decreased rate of branch number, pod number, pod weight per plant and weight of 100 grains. Peking and PI90763 decreased at the rate of under 1.6% in branch number, pod number, pod weight per plant and weight of 100 grains, while Illsoy showed a little higher decreased rate of 3.8% in pod number and 3.4% in pod weight per plant. Custer, Delmar, Dyer and Jackson represented a little higher decreased rate in yield.

Effect of the Density of Heterodera glycines on Soybean Yield (콩씨스트선충(Heterodera glycines)의 밀도가 콩 수량에 미치는 영향)

  • This study was conducted to investigate the yield loss of soybean by initial density of Heterodera glycines in soil. A soybean cultivar named 'Cheongja5ho' was cultivated as test material in Dangjin city, Chungcheongnam-do, Korea. Each seed was planted with distance of 25 cm in 35 plots by initial density of egg of the cyst nematode on June 21st, and allowed to harvest on November 12st in 2019. As a results, the yield of soybean decreased sharply when as the initial density of eggs of cyst nematode increased. The initial density of eggs in soil affected largely on the decreased yields of the soybean by more than 20% in above 2,000 eggs, and 30% in above 6,000 eggs of the cyst. Beyond the 6,000 eggs, the yield loss was more slowly decreased compared with the above cases. Eggs of H. glycines are widely distributed in Korea, resulting in the serious soybean yield loss by H. glycines.

  • In order to find out resistant varieties of the soybean to the cyst nematode. Heterodera glycines Ich. 64 soybean varieties were tested. 1. According to the result of investigations about correlation between numbers of cyst nematodes and soybean yields ; in pot test $Y=36.2-0.63\times(r=-0.74)$. and in field investigation $$Y=10.3-0.32\times(r=-0.56)$. There is a negative correlation between the density of nematodes and the soybean yield. 2. All of the recommended varieties were appeared susceptible; P I-84751. P I-90763. Southern-proripic. Keumgang-sorip. Baektai were appeared comparatively resistant ; and other varieties were appeared to be medium or susceptible. 3. Fewer larvae were developed into adult female cysts in resistant varieties than in susceptible ones.

Reproductivity of Mixtures of Race 3 and Race 4 of Heterodera glycines on Soybean Cultivars (대두품종에서의 콩씨스트 선충 혼합 Race의 생식력)

  • Soybean cultivars were inoculated with mixtures of races 3 and 4 of the soybean cyst nematode(SCN), Heterodera glycines Ichinohe. On a susceptible soybean cultivar Lee, the relative female maturation of the race mixture was not significantly different when compared with the maturation potentials for each of the two races alone. However, female maturation was significantly lower for the mixture than' for race 4 alone on the soybean cultivas Pickett and Mack which are susceptible to only race 4. Selection of SCN populations consisting of race 3 and race 4 resulted in a decrease in race 4 on soybean cultivars Lee and Bragg which are susceptible to all SCN races and an increase in race 4 on soybean cultivars Pickett and Peking which are susceptible to only race 4. A significant reduction of race 4 was also observed on Lee with most mixtures of race' 3 and race' 4 combined in several ratios, suggesting that race 3 was more competitive than race 4 on the susceptible soybean.

Distribution of Races of Soybean Cyst Nematode in Korea (국내 콩씨스트선충의 Race 분포)

  • To study races of Heterodera glycines in Korea, 21 soil samples were collected from eight provinces in 1995. Four races were found; race 3(48%) was a dominant race flowed by race 5(24%), race 1(19%), and race 6(9%). About 30~40% of H. glycines populations reproduced on Pickett and PI88788, while non reproduced on Peking or PI90763. Development of resistant soybean cultivar targeting to race 5 and 6 of H. glycines using PI90763 and PI88788 as a parent is recommended.
