Optimal Conditions for the Growth of Soybean Sprouts by Ozone Water Watering (오존수 살수(撒水)에 의한 콩나물의 성장조건 최적화)

  • This study was conducted to investigate the optimal conditions for the growth of soybean sprouts by ozone water watering. Optimal conditions for ozone water watering of soybean sprouts during cultivation at 2$0^{\circ}C$ were evaluated with ozone concentration(0.1~O.5 ppm) and watering frequency(1~9 times) by response surface methodology. The optimal conditions for growth of soybean sprouts were ozone concentrations of 0.20~0.32 ppm, ozone treatment frequency of 3.0~4.4 times. Germination rates, hypocotyl weights and hypocotyl lengths in the soybean sprouts cultivated under the optimal conditions increased by 13.3, 10.1 and 11.9%, respectively, whereas root weights decreased by 89.0%.

Identification of Factors Controlling Expression of Purple Color in Hypocotyl of Soybean Sprouts (콩나물 재배 시 자색 배축 발현 요인 구명)

  • Soybean sprouts have good digestibility, high isoflavone content compared with soybean seeds, and large amounts of aspartic acid, which is effective in eliminating hangovers. However, the region between the cotyledon and hypocotyl in soybean sprouts appears purple, the product value of soybean sprouts reduces. To date, the scientific correlation among factors such as growth temperature, spray water temperature, and cultivation periods for soybean varieties related to purple color expression between the cotyledon and hypocotyl of soybean sprouts are unknown. The purpose of this study was to identify the factors regulating the expression of purple color between the cotyledon and hypocotyl of soybean sprouts. Of 15 Korean soybean varieties with purple color in the hypocotyls and flowers, 9 varieties, including Sowonkong, Wonhwang, Sinhwa, Eunhakong, Pungsannamulkong, Paldonamulkong, Kwangankong, Shingang, and Jangki showed purple color between the cotyledon and hypocotyl of sprouts. However, the remaining six cultivars, including Dachae, Myeongjunamulkong, Sobaeknamulkong, Sojinnamulkong, Anpyeong, and Jonam, did not show purple color. The proportion of soybean sprouts with purple hypocotyls was the lowest at $26^{\circ}C$ and there was no significant difference at the other three cultivation temperature conditions ($17^{\circ}C$, $20^{\circ}C$, and $23^{\circ}C$). Similar to the results of the cultivation temperature experiment at five different spray water temperatures ($10^{\circ}C$, $17^{\circ}C$, $20^{\circ}C$, $23^{\circ}C$, and $26^{\circ}C$), the proportion of soybean sprouts with purple hypocotyls was the highest at the lowest spray water temperature ($10^{\circ}C$), and lowest at the highest temperature. Additionally, there was no purple color expression in soybean sprout hypocotyls on the $3^{rd}$ day after soybean sprouting. However, the highest expression level was observed on the $4^{th}$ day. Then, on the $5^{th}$ day, the proportion of soybean sprouts with purple hypocotyls reduced significantly, compared with that on the $4^{th}$ day. The results showed that interaction among these factors under cultivation conditions was remarkably effective for the expression of purple color in hypocotyls under the dark condition. These results suggested that cultivation temperature and spray water temperature for purple color expression in soybean sprout hypocotyls are the most important factors in the sprout cultivation environment.

Crop Effects on Soil Microorganism Activity and Community Composition in the Agricultural Environment (농경지에서 재배작물이 토양미생물활성 및 군집구성에 미치는 영향)

  • Soil microorganism activity in an agricultural field is affected by various factors including climate conditions, soil chemical properties, and crop cultivation. In this study, we elucidate the correlation between microorganism activity and agricultural environment factors using the dehydrogenase activity (DHA) value, which is one of the indicators of soil microbial activity. As a result, the various factors noted above were related to the DHA value. Annual rainfall, soil Mg2+, bacterial and fungal diversities, types of crops, developmental stages, seasons, and cultivation status were highly correlated with the DHA value. Furthermore, next-generation sequencing (NGS) analysis was used to identify that the type of crop affected soil microbial compositions of both bacteria and fungi. Soil used for soybean cultivation showed the highest relative abundance for Verrucomicrobia, Planctomycetes, and Acidobacteria but Actinobacteria and Firmicutes had the lowest relative abundance. In the case of soil used for potato cultivation, Actinobacteria had the highest relative abundance but Proteobacteria had the lowest relative abundance. Armatimonadetes showed the highest relative abundance in soil used for cabbage cultivation. Among the fungal communities, Mortierellomycota had the highest relative abundance for soybean cultivation but the lowest relative abundance for cabbage cultivation; further, Rozellomycota, Chytridiomycota, and Cercozoa had the highest relative abundance for cabbage cultivation. Basidiomycota had the highest relative abundance for potato cultivation but the lowest relative abundance for soybean cultivation.

Cluster Analysis of Korean Domestic Soybeans based on Yields Characters in Paddly Field Cultivation (논 재배 시 한국 콩 품종의 수량성 관련요인분석에 의한 품종군 분류)

  • In order to select the suitable soybean cultivars for paddly field cultivation, growth response and yields characters were investigated with 37 Korean domestic soybean cultivars, in Yeoncheon, Gyeonggi province. The results can be summarized as follows ; When the soybeans were cultivated in paddy field, significant differences were observed in growth responses at each growth stage and yields among the cultivars. Yields of six cultivars, such as Danbaekkong, Paldokong, Pungsannanulkong, Myeongjunamulkong, Saeolkong, and Namhaekong, were less than 200kg/10a, however, seven cultivars, such as Shinpaldalkong 2, Sohokong, Dormikong, keumkangkong, Bukangkong, Dajangkong, and Geomjeongkong 2, showed highest yields ranged from 301~385kg/10a. At V5 stage, highly positive correlation(r=091$^{{\ast}{\ast}}$) was observed between the top dry weight and leaf area while negative correlation was existed between T/R ratio and root dry weight. However, at R2 age, leaf area was correlated with both top and root dry weight while T/R ratio was correlated with leaf area and top dry weight. Thirty-seven soybean cultivars used in this experiments were classified into 3 different clusters based on 19 important agronomic characters, and genotypic properties of the cultivars were similar in each cluster. In results, high yields could be achieved by selected cultivation of suitable soybean cultivars such as Shinpaldalkong 2 which showed early maturation date and highly ripening ratio.

Changes in Isoflavone Content and Mass Balance During Soybean Processing

  • We analyzed the isoflavone content of domestic soybeans during steaming, boiling, fermentation, germinating cultivation, fermentation, and soybean curd production. The isoflavone content of the beans was reduced by steaming and boiling, and overall reductions ranging from 16.0 to 65.0% of initial isoflavone values were detected. After 4 days of germinating cultivation, the total isoflavones of Eunhakong increased from 1,341 to $2,017\;{\mu}g/g$ and the total isoflavones of Guinunikong increased from 1,284 to $1,535\;{\mu}g/g$. The isoflavone content of the vinegar beans produced from Hwangkeumkong and Black No.1 increased from 1,877 to $1,956\;{\mu}g/g$, and from 885 to $1,956\;{\mu}g/g$ after 8 days of immersion in 4% acetic acid, respectively. During soybean curd production, significant amounts of isoflavones were lost in the whey (30-31 %) and soybean curd residue (15-20%). Only 37.4% of the isoflavones present in the original soybeans remained in the soybean curd with the hot extraction method, and 50.7% of them with the cold extraction method. Soybean curd prepared with whole soybean method, however, retained 80.7% of the initial isoflavones.

  • Monacolin K (MK) was produced on soybean using Monascus ruber GM011 by a two-stage-fermentation process. The optimal temperature was identified as $28^{\circ}C$. Higher yield was obtained by multiple-level-temperature cultivation than by single-level-temperature cultivation. The highest yield of total MK, 4.810 mg/g dry soybean product, was attained after 30 days of solid-state fermentation. No citrinin could be detected in the fermented soybean.

Identification of New Isolates of Phytophthora sojae and the Reactions of Korean Soybean Cultivars Following Hypocotyl Inoculation

  • Phytophthora root and stem rot (PRSR) caused by Phytophthora sojae is one of the most destructive diseases of soybean. PRSR recently became an issue as soybean cultivation in paddy fields increased in South Korea. The management of PRSR mainly involves R-gene-mediated resistance, however, little is known about the resistance in Korean cultivars. Major Korean soybean cultivars were investigated for the presence or absence of R-gene-mediated resistance to four P. sojae isolates, two of which were new isolates. Isolate-specific reactions were observed following P. sojae inoculation. Of 21 cultivars, 15-20 cultivars (71.4-95.2%) showed susceptible reaction for each isolate. Ten cultivars were susceptible to all the isolates, and six cultivars were identified to have R-gene-mediated resistance to one or two isolates. The results of this study would provide a framework for the discovery of resistant cultivars, development of new cultivars resistant to P. sojae, and investigation of pathogenic diversity of P. sojae population in South Korea.

Effect of Chain Type Mole-Drainer Attached to Tractor for Drainage Improvement (배수개선을 위한 트랙터 부착식 체인형 암거 천공기의 적용 효과)

  • The area of paddy fields for upland crop cultivation is gradually increasing. In this context, we developed a chain type mole drainer that can be attached to a tractor to prevent the wet injury of crops. To conduct a field test, we formed underdrains at 2 m intervals in a paddy field wherein soybeans are cultivated, and we investigated the status of the soil moisture, groundwater level, and soybean growth during the cultivation period. The soil moisture content decreased by 22.3~26.8% in the test plot and by 5.9~6.9% in the control plot during a period of 57~88 hour after a rainfall of 41.5~157.0 mm. The effect of underdrainage was clear as the hourly groundwater level decreased approximately 2.8 times faster in the test plot compared with the control plot. Moreover, the soybean yield was greater by 78.6 kg/10a in the test plot than in the control plot. Therefore, for soybean cultivation in paddy fields, the use of the developed mole drainer is expected to help in improving the drainage and increasing the soybean yield.

Amino Acid Composition Changes in Soybean Sprouts during Cultivation (재배기간에 따른 콩나물의 아미노산 조성 변화)

  • We examined changes in the amino acid composition of soybean sprouts of four different Korean soybean cultivars (Yutae, Jinunee, Subaktae, and Baktae). Total amino acids levels in all samples 1 day after cultivation were lower than those in soybeans but increased at cultivation times greater then 3 days. Glutamic and aspartic acids, the most abundant amino acids in all soybean cultivars studied at day 0 of sprouting, constituted more than 30% of all amino acids. Aspartic acid content of hypocotyls markedly increased, but glutamic acid levels decreased 5-day-old sprouts of Jinunee and 3-day-old sprouts of the other cultivars. Moreover, compositions of aspartic acid decreased in the order hypocotyl > root > cotyledon and Yutae > Baktae > Jinunee > Subaktae.

Effect of Ozone Treatment on the Quality of Soybean Sprouts 2. Optimum Conditions of Ozone Treatment during Soaking of Soybean for Quality Enhancement of Soybean Sprouts (콩나물의 품질에 미치는 오존처리 효과 2. 콩의 수침중 오존처리의 최적화)

  • For the purpose of enhanceing soybean-sprouts quality, the optimum conditions for ozone treatment of soybean during soaking before cultivation at 18~2$0^{\circ}C$ were evaluated with ozone concentration, treatment time and treatment frequency by response surface methodology. Germination rates of cleaned soybean by ozone water in the conditions of solubilized-ozone concentrations of 0.15 to 0.35ppm, ozone-treatment frequency of 1.5 to 2.3 times and ozone-treatment time of 30 to 36min. increased 18.8 to 24.0% for the control products. And, length of hypocotyl in conditions of 0.12 to 0.33ppm, 1.7 to 2.7 times and 45 to 90min. were also increased by 69.36 to 79.40%. On the other hand, weight of roots with ozone treatment were decreased in the conditions of solubilized ozone concentrations of 0.1 to 0.2ppm and ozone-treatment time of 30 to 57min. But, ozone-treatment frequency did not affected root growth. Putrefaction rates of the control were 5 to 15%, but those of ozone-treated samples during cultivation did not show. The overall optimum conditions for above 16% germination rates, above 9% hypocotyl yields compared to the control samples and below 98% of the control root weight were solubilized-ozone concentrations of 0.25 to 0.30ppm, ozone-treatment time of 43 to 49min. and ozone-treatment frequency of one time.
